Thermodynamics of alligator metmyoglobin unfolding.

The conformational free energy of alligator metmyoglobin was examined over a pH range of 4.4-8.0, a temperature range of 18-48 degrees C, and a guanidinium chloride concentration of 0-2.3 M. For isothermal unfolding at 25 degrees C essentially the same value was obtained for the conformational free energy from all the data: 7.0 +/- 0.5 kcal/mol. The cooperativity of the unfolding with respect to denaturant is considerably less than for mammalian myoglobins. On unfolding three to four side chains with a pKa of 6.3 in the unfolded protein are protonated instead of the six expected. The temperatu

Inhibition of methemoglobin and metmyoglobin reduction by cobalt.

Because cobalt compounds tend to form stable complexes, there has been continued interest in the use of the salts and chelates of cobalt in cyanide poisoning, and continued uncertainty about the precise nature of their protective effects. We have found that cobalt ions inhibit the enzymatic reduction of both methemoglobin and metmyoglobin. Virtually total inhibition of methemoglobin and metmyoglobin reductase activity occurred with the addition of 2.5 mM cobalt acetate to the assay system. Both enzymes were inhibited by lower levels of cobalt in a dose-dependent manner. The similarity in susce

Metmyoglobin reductase. Identification and purification of a reduced nicotinamide adenine dinucleotide-dependent enzyme from bovine heart which reduces metmyoglobin.

Beef heart muscle has been found to contain an enzyme which will rapidly and directly reduce metmyoglobin in vitro. Reduction rates are far greater than any previously reported for nonspecific or nonenzymatic systems. The enzyme is NADH-dependent and requires the presence of ferrocyanide ion for in vitro assay. The artificial electron carriers, dichlorophenolindophenol and methylene blue, are not required. Nonenzymatic reduction of metmyoglobin, which has previously been reported, was not encountered under the assay conditions described herein. Demonstration of enzymatic activity is dependent

Volume changes in binding of ligands to methemoglobin and metmyoglobin.

The volume changes for the binding of various ligands to metmyoglobin and methemoglobin have been determined from the effect of pressure on the binding constants (for metmyoglobin) and by direct dilatometry (for methemoglobin). The volume changes associated with the binding of cyanide and azide ions to methemoglobin are pH-dependent. The volume change for the binding reaction is evidently affected by the same subtle structural variations that have been judged to be present from the variation with pH of enthalpy and entropy for the binding reactions in these proteins. Hydration changes and spin

Autocatalytic Mechanism in the Anaerobic Reduction of Metmyoglobin by Sulfide Species.

The mechanism of the metal centered reduction of metmyoglobin (MbFeIII) by sulfide species (H2S/HS-) under an argon atmosphere has been studied by a combination of spectroscopic, kinetic, and computational methods. Asymmetric S-shaped time-traces for the formation of MbFeII at varying ratios of excess sulfide were observed at pH 5.3 < pH < 8.0 and 25 °C, suggesting an autocatalytic reaction mechanism. An increased rate at more alkaline pHs points to HS- as relevant reactive species for the reduction. The formation of the sulfanyl radical (HS•) in the slow initial phase was assessed using the s

Ligand binding to catalase and metmyoglobin. Direct measurements of proton involvement.

1. Spectroscopic measurements of the binding of the weak-acid ligands formate, azide and cyanide to catalase indicate interaction of a primary haem-binding site with the undissociated forms of the ligands between pH 5 and 8. A similar conclusion can be drawn from observations on the pH dependence of catalase activity in the presence of cyanide and formate. 2. Direct measurements of pH changes during catalase interaction with these ligands also indicate stoichiometric uptake of one proton per haem group upon addition of formate or azide at pH 6, but at greater pH values the proton uptake declin

Binding mechanism of disulfide species to ferric hemeproteins: The case of metmyoglobin.

The interactions of the heme iron of hemeproteins with sulfide and disulfide compounds are of potential interest as physiological signaling processes. While the interaction with hydrogen sulfide has been described computationally and experimentally, the reaction with disulfide, and specifically the molecular mechanism for ligand binding has not been studied in detail. In this work, we study the association process for disulfane and its conjugate base disulfanide at different pH conditions. Additionally, by means of advanced sampling techniques based on multiple steered molecular dynamics, we p

Structural factors governing azide and cyanide binding to mammalian metmyoglobins.

The structural factors governing azide and cyanide binding have been examined by measuring the effects of 46 mutations at key topological positions in the distal pocket in sperm whale, pig, and human myoglobin. Replacement of His64 (E7) with smaller amino acids results in dramatic increases in the association rate constant for azide binding primarily due to relief of steric hindrance imposed by the imidazole side chain. Gln64 and His64 (native) metmyoglobins have abnormally low rate constants for azide dissociation (0.1-0.3 s-1) due to direct hydrogen bonding between the N epsilon atoms of the

Co-oxidation of salicylate and cholesterol during the oxidation of metmyoglobin by H2O2.

The reaction between metmyoglobin and H2O2 proceeds with oxidation of the hemo-protein iron to a higher valence state and consumption of the peroxide. This reaction is further associated with (a) O2 evolution; (b) hydroxylation of the aromatic compound salicylate to yield a set of dihydroxybenzoic acid derivatives (analyzed by HPLC with electrochemical detection); (c) autoxidation of cholesterol with formation of 3 beta-hydroxy-5-alpha-cholest-6-ene-5-hydroperoxide; and (d) formation of electronically excited states detected by low-level chemiluminescence. Determination of metmyoglobin in cooked tan mutton using Vis/NIR hyperspectral imaging system.

In this study, the ENVI 4.6 software was used to obtain the spectral reflection value of samples. The outlier samples were eliminated by the Monte Carlo method, and then SPXY (sample set partitioning based on be x-y distances) was used to divide the calibration set and prediction set. The spectral images were pretreated and characteristic wavelengths were extracted. The spectral models of full and pretreated spectra and characteristic bands were established by partial least squares regression (PLSR) and principle component regression (PCR), and the optimal modeling combination was selected. Intracellular imaging of metmyoglobin and oxygen using new dual purpose probe EYFP-Myoglobin-mCherry.

The biological relevance of nitric oxide (NO) and reactive oxygen species (ROS) in signaling, metabolic regulation, and disease treatment has become abundantly clear. The dramatic change in NO/ROS processing that accompanies a changing oxygen landscape calls for new imaging tools that can provide cellular details about both [O2 ] and the production of reactive species. Myoglobin oxidation to the met state by NO/ROS is a known sensor with absorbance changes in the visible range. We previously employed Förster resonance energy transfer to read out the deoxygenation/oxygenation of myoglobin, crea

Conformation of biological macromolecules. Circular dichroism and magnetic circular dichroism studies of metmyoglobin and its derivatives.

The circular dichroism (CD) and magnetic circular dichroism (MCD) spectra of horse heart metmyoglobin and the following derivatives were measured in the Soret and near ultraviolet regions: metmyoglobin and its peroxide compound, and hydroxide, cyanide, azide, and fluoride derivatives. The heme-related CD bands in the Soret and near ultraviolet wavelength regions were altered by ligand substitution, though their relationships to the magnetic moment were quite different. In the Soret region, the CD peak had no definite relation to the magnetic moment, while in the near ultraviolet region the mag

Nitroxides catalytically inhibit nitrite oxidation and heme inactivation induced by H2O2, nitrite and metmyoglobin or methemoglobin.

Stable nitroxide radicals have multiple biological effects, although the mechanisms underlying them are not fully understood. Their protective effect against oxidative damage has been mainly attributed to scavenging deleterious radicals, oxidizing reduced metal ions and reducing oxyferryl centers of heme proteins. Yet, the potential of nitroxides to protect heme proteins against inactivation while suppressing or enhancing their catalytic activities has been largely overlooked. We have studied the effect of nitroxides, including TPO (2,2,6,6-tetramethylpiperidin-N-oxyl), 4-OH-TPO, 4-oxo-TPO and

Nitroxide radicals as research tools: Elucidating the kinetics and mechanisms of catalase-like and "suicide inactivation" of metmyoglobin.

BACKGROUND: Metmyoglobin (MbFe(III)) reaction with H(2)O(2) has been a subject of study over many years. H(2)O(2) alone promotes heme destruction frequently denoted "suicide inactivation," yet the mechanism underlying H(2)O(2) dismutation associated with MbFe(III) inactivation remains obscure. METHODS: MbFe(III) reaction with excess H(2)O(2) in the absence and presence of the nitroxide was studied at pH 5.3-8.1 and 25°C by direct determination of reaction rate constants using rapid-mixing stopped-flow technique, by following H(2)O(2) depletion, O(2) evolution, spectral changes of the heme pro

X-ray absorption spectroscopic studies of a transient intermediate in the reaction of cyanide metmyoglobin with dithionite by using rapid freezing.

The reaction of cyanide metmyoglobin (Mb+CN-) with dithionite produces a transient intermediate, supposed to be cyanide-ligated ferrous myoglobin. The Fe K-edge X-ray absorption spectrum of the intermediate has been measured by using rapid freezing and compared with those of Mb+CN- and deoxymyoglobin (deoxyMb). The shapes of the XANES (X-ray Absorption Near Edge Structure) spectra of Mb+CN- and the intermediate are very similar, including the intensity ratios of the peak C1 to D. This indicates that CN- remains bound with a linear Fe-C-N configuration in the intermediate. The absorption edge o
