Metschnikowia pulcherrima clade yeasts produce pulcherriminic acid, which binds with iron to form pulcherrimin, a red pigment. However, the functions of pulcherriminic acid in yeasts and bacteria that synthesize it remain incompletely understood. To gain deeper understanding of these functions the responses of pulcherriminic acid-secreting Metschnikowia yeast in the presence of various solid iron sources (metallic iron, magnetite, and lepidocrocite) containing iron in different valence states were examined. By applying the classical plate method, the formation of pulcherrimin, was observed bot

Two novel yeast species belonging to the genus Metschnikowia were isolated from flowers and young fruits in Okinawa, Japan. Five strains (JCM 35539, JCM 36740T, JCM 36743, JCM 36744, and JCM 36747) and four strains (JCM 35542, JCM 35544T, JCM 36745, and JCM 38279) were repeatedly isolated from samples collected over 2-3 y. Because BLAST analysis of D1/D2 large subunit ribosomal DNA sequences did not clearly resolve their phylogenetic placement and intragenomic polymorphisms were detected in the internal transcribed spacer region, their relationships were further assessed by whole-genome phylog

Metschnikowia bicuspidata, a pathogenic yeast species, is responsible for significant disease outbreaks, including the devastating milky disease in aquaculture systems. This review consolidates current knowledge of its pathogenesis, genomic characteristics, histopathology, and treatments. The yeast's psychrotolerant nature and ability to exploit aquatic hosts even under adverse conditions have heightened its impact, particularly in high-density farming environments. Potential treatments, such as antibiotics, killer toxins, and Massoia lactone, show promising antifungal activity, yet each prese

Metschnikowia pulcherrima, an ascomycetous yeast, has been frequently isolated from various substrates associated with agriculture such as nutrient-rich plant materials (fruit, fruit juices and fermenting grape must), soil and with various insects including fruit flies. Due to the growing consumer demand for wines with reduced levels of additives such as sulphites, there has been great interest in this yeast species for its ability to compete with other fungi. Despite having similar phenotypes, many species related to M. pulcherrima have been described in the last 20 years based on the sequenc

The spoilage of wine caused by Brettanomyces bruxellensis and Hanseniaspora uvarum poses a significant challenge for winemakers, necessitating the development of effective and reliable strategies to control the growth of these yeasts, such as grape must bioprotection. Despite evidence that certain microorganisms can inhibit the growth of Brettanomyces bruxellensis and Hanseniaspora uvarum, the specific mechanisms driving this inhibition remain unclear. The primary objective of this study is to elucidate the underlying mechanisms responsible for this inhibitory effect. We analyzed one Metschnik

Metschnikowia pulcherrima has been studied and applied in winemaking in recent years, but its application in icewine fermentation has rarely been reported. In this study, the tolerance and β-glucosidase activities of indigenous M. pulcherrima strain were first detected. The results show that, except for a relatively low tolerance to ethanol [limited to 4% (v/v)], the other tolerance is good to the fermentation of icewine; the cell wall-bound and extracellular β-glucosidase activities of M. pulcherrima strain were 35.93 nmol/mL (1.28 U) and 14.31 nmol/mL (0.42 U), respectively. M. pulcherrima a

Carotenoids are valuable natural pigments with antioxidant and health-promoting properties. Growing concerns regarding the toxicity and environmental persistence of synthetic colorants have increased interest in safe and sustainable natural alternatives. Compared with plant-derived carotenoids, microbial production offers several practical advantages, including rapid growth, year-round cultivation, utilization of agro-industrial substrates, and simplified downstream processing. This study aimed to isolate carotenoid-producing microorganisms from environmental samples, identify the most product

This study addresses the inherent limitations of Metschnikowia pulcherrima (Mp), specifically its limited fermentation capacity and low ethanol tolerance, by developing biocapsules immobilized with Aspergillus oryzae (Ao), referred to as Mpb. Quantitative analysis demonstrated that Ao achieved an immobilization efficiency of 89.57 ∼ 93.53% for the Mp strain Kb01-1, resulting in the successful preparation of uniform spherical Mpb with a consistent diameter of 1.83 mm and a total of 535 capsules. Scanning electron microscopy revealed that Mpb exhibited a porous and dense composite structure. The

INTRODUCTION: The interest toward the use of non-Saccharomyces yeasts in the winemaking process has been increasing because it has been demonstrated that they can contribute positively to the quality of wines; however, there is a gap in the literature on holistic approaches showing the effective contribution of non-Saccharomyces yeasts in sequential fermentations.
METHODS AND RESULTS: Two commercial strains of Metschnikowia pulcherrima (Mp) and Torulaspora delbrueckii (Td) were used in sequential fermentations with Saccharomyces cerevisiae (Sc). The fermentations were monitored by evaluating

The fungal pathogen Metschnikowia bicuspidata causes "milky disease" in the Chinese mitten crab (Eriocheir sinensis), which poses substantial challenges to sustainable aquaculture development considering the current lack of effective treatment interventions. To address this issue, in laboratory validation, we developed two rapid recombinase polymerase amplification (RPA) detection methods for M. bicuspidata in E. sinensis targeting the histone acetyltransferase B-type subunit 2 gene (HAT-B2): an electrophoretic assay (RPA-AGE) and a colloidal gold lateral flow dipstick assay (RPA-LFD). We opti

Metschnikowia pulcherrima is a promising biological control agent (BCA), particularly intended for postharvest fruit protection. The research presents pulcherriminic acid (PA) producing isolates obtained from organic fruit surfaces that are antagonistic towards Leptosphaeria maculans, the causal agent of phoma stem canker. A highly antagonistic strain identified as M. pulcherrima was selected for studying the interactions with L. maculans in various in vitro culture techniques. In solid co-cultures, increased iron bioavailability restricted PA diffusion in the yeast colony microenvironment, si

Gray mold in grapes during the postharvest period is primarily caused by Botrytis cinerea, with synthetic fungicides being the main method of control. Antagonistic yeasts offer a viable alternative to conventional fungicides for the management of pathogens post-harvest. The study analyzed the effectiveness of 14 Metschnikowia pulcherrima yeast strains against gray mold in grapes and examined their potential mechanisms of action. In vitro analyses demonstrated that all evaluated yeast strains (100 μL, 1 × 104 CFU/mL) significantly inhibited the mycelial growth of B. cinerea, with inhibition rat

Candidiasis is one of the most frequent infections worldwide. In this study, the antimicrobial properties of six strains belonging to the Metschnikowia pulcherrima clade were evaluated against twenty Candida and Candida-related Filobasidiella neoformans var. bacillispora (syn. Cryptococcus neoformans) of different origins, employing the agar cross method. The toxic effect of pulcherrimin, a red metabolite that is responsible for the antimicrobial activities of Metschnikowia spp., was evaluated in various experimental models. The results of agar tests showed that the selected M. pulcherrima str

Brettanomyces bruxellensis is a major spoilage yeast in winemaking due to its persistence in winery environments and its ability to produce volatile phenols responsible for sensory defects. This study evaluated the biocontrol potential of selected Metschnikowia pulcherrima strains, focusing on the role of pulcherriminic acid production and its application in winery-relevant conditions. Twelve M. pulcherrima strains were screened for pulcherriminic acid production under different iron conditions, revealing strong strain-dependent variability. Three strains (1521, 1558, and DSM 70336) were ident

Non-Saccharomyces wine yeasts have a promising role in biotechnological approaches to enhance wine complexity, particularly by influencing the aromatic profile. Metschnikowia pulcherrima is a non-Saccharomyces yeast that is notable for its antimicrobial activity and diverse enzymatic activities. These properties make this yeast a valid candidate for application as a starter culture in winemaking. This study evaluated the application of M. pulcherrima 62 in sequential inoculation with S. cerevisiae for the production of Kallmet wine, delivered from the indigenous Kallmet grape variety tradition

Two male patients with ankylosing spondylitis and upper lobe fibrosis and cavitation are described. A pneumonic disease in one was associated with mycological and serological evidence of infection with Aspergillus terreus but no other aspergillus species. A large pulmonary mycetoma developed in the second patient and among a number of other fungal isolates was found the yeast Metschnikowia pulcherrima. The association of ankylosing spondylitis with bronchopulmonary aspergillosis is considered; A. terreus is described for the first time as a human pulmonary pathogen, and the possible pathogenic

Using non-Saccharomyces yeasts is a promising way to modulate wine composition, particularly in warm regions where high sugar levels result in wines with high alcohol content and low acidity. This study examined the impact of various inoculation methods involving Lachancea thermotolerans and Metschnikowia pulcherrima on the chemical and aromatic profiles of Tempranillo wines. Five treatments were tested: a control with Saccharomyces cerevisiae; two sequential inoculations, with each non-Saccharomyces species followed by S. cerevisiae; and two co-inoculations of both species at different ratios

The capability of Metschnikowia pulcherrima to control postharvest citrus diseases was analyzed, with its action mechanisms and preservation performance during storage further explored. In vitro and in vivo tests demonstrated M. pulcherrima significantly inhibited the growth of Penicillium digitatum (P.d), Penicillium italicum (P.i), and Geotrichum citri-aurantii (G.c), and markedly controlled the incidence and progression of these three postharvest diseases. The non-cellular components of M. pulcherrima did not significantly inhibit three pathogens' growth, while its volatile compounds (VOCs)

This study systematically investigated how Metschnikowia pulcherrima XX04 competes with Penicillium digitatum, Penicillium italicum, and Geotrichum citri-aurantii for iron ions to control these major postharvest citrus pathogens. Exogenous Fe3+ addition significantly reduced the inhibitory activity of the pigment-producing yeast against these pathogens on both agar medium and fruit. Pearson correlation analysis revealed a strong positive relationship between pigment production and antagonistic effectiveness of M. pulcherrima XX04. Comparative analysis of iron-capturing proficiency between M. p
