Comparison of the pathogensis of two isolates of Besnoitia caprae in inbred BALB/c mice.

This study was performed to evaluate the infectivity of bradyzoites of two Besnoitia caprae isolates, BC-1 and BC-2, to inbred BALB/c mice. Each group of inbred BALB/c mice was inoculated intraperitoneally with 1 x 10(3), 1 x 10(4), 1 x 10(5), 5 x 10(5) and 1 x 10(6) of one of the two isolates of B. caprae bradyzoites. The mice were monitored daily for a period of 40 days for survival. After death of each mice, several passages from its peritoneal washing and tissues were analyzed using ribosomal DNA-specific PCR assay. Marked differences in pathogenicity between the isolates were seen. All th

Comparative pathogenicity of three genetically distinct Trypanosoma congolense-types in inbred Balb/c mice.

Inbred Balb/c mice were infected with three clones of Trypanosoma congolense (Sam.28.1, Dind.3.1 and K60.1A) corresponding, respectively, to the three genetically distinct types (savannah, forest and kilifi) defined within this species, for the purpose of comparing their pathogenicity for a better understanding of the epidemiology of African trypanosomosis. Another clone of savannah type, IL 3000, was also tested simultaneously to study a probable strain variation. Both the clones of savannah type were found of extreme virulence with loss of appetite, rough hair, rapid respiration, lethargy, a

Characterization of Mouse-Adapted Marburg and Ravn Viruses in Inbred BALB/c and Outbred CD-1 Mice.

Marburgviruses cause severe hemorrhagic disease in humans, yet characterization of rodent models for pathogenesis and medical countermeasure development remains limited. Here, we investigated clinical course, blood chemistry, viral dissemination, and mucosal viral detection in inbred BALB/c and outbred CD-1 mice infected with mouse-adapted (MA)-Marburg virus (MARV)/Angola or MA-Ravn virus (RAVV) to further mouse model development. Regardless of virus or mouse strain, infection resulted in widespread viral dissemination in animals sampled up to 5 days post-infection. However, in animals followe

Lactoferrin enhances efficacy of the BCG vaccine: comparison between two inbred mice strains (C57BL/6 and BALB/c).

The current vaccine for tuberculosis (TB), an attenuated strain of Mycobacterium bovis Bacillus Calmette Guerin (BCG), is effective to prevent childhood onset of the disease, but its efficacy is reduced in adults. One strategy to improve the existing vaccine is to develop more effective adjuvants. Lactoferrin, an iron-binding glycoprotein possessing immune modulatory activities, is a promising adjuvant candidate. The studies presented here examine the effect of lactoferrin to enhance efficacy of the BCG vaccine using a vaccination/challenge protocol (8 weeks boost and challenge at 12 weeks pos

Comparative Analysis of the Effect of Sex and Age on the Hematological and Biochemical Profile of BALB/c and C57BL/6 Inbred Mice.

Mice are the most commonly used models of infectious disease, and disease in mice is similar to that of humans. As a consequence, standard hematology and biochemistry reference values in mice are essential to evaluate functional changes caused by experimental treatments, although very few data in the literature provide a comparative reference range. The aim of this investigation was to establish the reference intervals for major hematology and biochemistry analytes in 2 inbred mouse strains, BALB/c and C57BL/6, at 3 different age ranges. Parameters were assessed in 600 mice (300 male and 300 f

Relationship between parasite load and immune responses in early stages of Leishmania donovani infection in inbred BALB/c mice.

The aim of this study was to determine if a correlation exists between Leishmania donovani parasite load and the corresponding humoral and cellular mediated immune responses in the early stages of Leishmania donovani infection in inbred BALB/c mice. Five groups of ten BALB/c mice each were inoculated intraperitoneally with stationary phase metacyclic promastigotes of Leishmania donovani at doses 1 x 10(2), 1 x 10(4), 10(6) and 1 x 10(8) respectively per mouse. Group five mice were not manipulated in any way and were left to serve as control. At weekly intervals, for five weeks, the mice were a

Infection of inbred BALB/c and C57BL/6 and outbred Institute of Cancer Research mice with the emerging H7N9 avian influenza virus.

A new avian-origin influenza virus A (H7N9) recently crossed the species barrier and infected humans; therefore, there is an urgent need to establish mammalian animal models for studying the pathogenic mechanism of this strain and the immunological response. In this study, we attempted to develop mouse models of H7N9 infection because mice are traditionally the most convenient models for studying influenza viruses. We showed that the novel A (H7N9) virus isolated from a patient could infect inbred BALB/c and C57BL/6 mice as well as outbred Institute of Cancer Research (ICR) mice. The amount of

Genetically inbred Balb/c mice differ from outbred Swiss Webster mice on discrete measures of sociability: relevance to a genetic mouse model of autism spectrum disorders.

The Balb/c mouse is proposed as a model of human disorders with prominent deficits of sociability, such as autism spectrum disorders (ASDs) that may involve pathophysiological disruption of NMDA receptor-mediated neurotransmission. A standard procedure was used to measure sociability in 8-week-old male genetically inbred Balb/c and outbred Swiss Webster mice. Moreover, because impaired sociability may influence the social behavior of stimulus mice, we also measured the proportion of total episodes of social approach made by the stimulus mouse while test and stimulus mice were allowed to intera

Effects of a hybrid maternal environment on brain growth and corpus callosum defects of inbred BALB/c mice: a study using ovarian grafting.

The corpus callosum of many but not all BALB/c mice is either abnormally small or absent. The basis for the defect is hereditary, but the proportion of mice expressing the anomaly can be modified by changing the early environment. The present study investigated the effects of an F1 hybrid maternal environment, which is known to promote better fetal development than an inbred environment, on body and brain growth in BALB/c and on the incidence of the corpus callosal defect. Ovarian follicle cells from BALB/cWah (albino) mice were grafted into pigmented F1 hybrid, BALB/cWah, and pigmented BALB f

An inbred epilepsy-prone substrain of BALB/c mice shows absence of the corpus callosum, an abnormal projection to the basal forebrain, and bilateral projections to the thalamus.

BALB/c mice lack a corpus callosum in about 11% of the population. Two inbred substrains of BALB/c mice, epilepsy-prone (EP) and epilepsy-resistant (ER), have been examined to determine whether these substrains differ in regard to corpus callosum morphology. Further, this study addressed the issue of whether misrouted cortical axons form an aberrant pathway instead of the corpus callosum. Initial studies that examined fresh brain tissue of adult animals revealed normal corpora callosa in all ER mice but deficient or absent corpora callosa in all EP mice. Subsequently, Dil crystals were placed

[Differences in susceptibility to peripheral infection with Japanese encephalitis virus among inbred strains of mouse].

We compared susceptibility of inbred mouse strains against intracerebral as well as peripheral challenge of a flavivirus, Japanese encephalitis (JE) virus, and the results were summarized as follows: (1) Seven inbred mouse strains (C3H/He, C57BL/6, BALB/c, AKR/N, NC, NZB, DBA/2) could be classified into following 3 groups by their mortality and infection rate when they were subcutaneously challenged with a wild JE virus strain isolated from field-caught mosquitoes at a single passage in mouse (B18AM, B-1): (1) high mortality and high infection rate (C3H/He, C57BL/6, AKR/N), (2) low mortality b

Role of genetic resistance in invasive pneumococcal infection: identification and study of susceptibility and resistance in inbred mouse strains.

From a panel of nine inbred mice strains intranasally infected with Streptococcus pneumoniae type 2 strain, BALB/c mice were resistant and CBA/Ca and SJL mice were susceptible to infection. Further investigation revealed that BALB/c mice were able to prevent proliferation of pneumococci in the lungs and blood, whereas CBA/Ca mice showed no bacterial clearance. Rapidly increasing numbers of bacteria in the blood was a feature of CBA/Ca but not BALB/c mice. In the lungs, BALB/c mice recruited significantly more neutrophils than CBA/Ca mice at 12 and 24 h postinfection. Inflammatory lesions in BA

Trypanosoma rhodesiense: analysis of the genetic control of resistance among mice.

Inbred mouse strains differ in their resistance to infection with the human pathogen Trypanosoma rhodesiense. Of the strains tested, C57BL/6 (B6) mice were the most resistant, and BALB/c (C) mice were among the most susceptible. The genetic basis underlying the different susceptibility of these two strains was analyzed. (CXB6)F1 progeny of either sex were more resistant than the BALB/c parent. Also, the backcross of F1 mice to the susceptible male or female BALB/c parent resulted in 52.0% susceptible (i.e., death on or before day 24) progeny, as compared with only 0.64% susceptible F1 progeny.

Susceptibility Analysis in Several Mouse Strains Reveals Robust T-Cell Responses After Mycoplasma pneumoniae Infection in DBA/2 Mice.

Mycoplasma pneumoniae (Mp) is a highly contagious respiratory pathogen responsible for human community-acquired pneumonia. The number of antibiotic-resistant Mp strains is increasing; therefore, to develop novel therapeutics, it is crucial to precisely understand the pathogenesis of mycoplasma pneumonia. Herein, we examined the susceptibility and response to Mp among eight inbred mouse strains. Following infection, the bacterial load in the bronchoalveolar lavage fluid (BALF) from DBA/2 mice was higher than that in the other tested strains such as BALB/c mice, which are frequently used in Mp r

BALB/c mice: low sociability and other phenotypes that may be relevant to autism.

Low sociability is one of the most prominent and disabling symptoms of autism. The biology of sociability is not well understood, and there is no available treatment that adequately improves social functioning in most autistic patients. The development of animal models of reduced sociability can aid in the elucidation of the biology of social behaviors, and may ultimately shed light on the biology of autism. This paper will review evidence that mice of the BALB/c inbred strain show relatively low levels of social interaction in various settings and across various stages of development, includi

Strain differences of cerebral ventricles in mice: can the MRL/MpJ mouse be a model for hydrocephalus?

Hydrocephalus is an intractable disease characterized by the excessive accumulation of cerebrospinal fluid (CSF) in the cerebral ventricles. There are many cases in both human and animals; however, the cause and mechanism of it's development is not clearly understood. In this study, differences of cerebral ventricles in 5 inbred mice strains (MRL/MpJ, C57BL/6, C3H/He, DBA/2 and BALB/c) were investigated by histological techniques to determine the possibility of a new animal model for hydrocephalus. Our analysis showed that significant differences in the volume and the surface area of lateral v

Nematospiroides dubius: stimulation of acquired immunity in inbred strains of mice.

The development of immunity to Nematospiroides dubius was studied in three strains of inbred mice (BALB/c, C3H and NIH). Although a primary infection in NIH mice persisted for two months without evidence of a reduction in worm numbers, female mice of this strain readily developed resistance to reinfection. The degree of resistance was enhanced when an immunizing infection of 600 larvae was administered as 6 separate doses of 100 larvae given between days 0 and 11, and the worms removed by anthelmintic treatment given on days 15, 21, 28 and 35. Induction and expression of cell-mediated immune responses in inbred mice infected with Coccidioides immitis.

Comparisons of the course of coccidioidomycosis in two strains of inbred mice established that BALB/c mice are significantly more susceptible to pulmonary infection with Coccidioides immitis than are DBA/2 mice. The susceptibility of BALB/c mice does not reside in their inability to mount a delayed-type hypersensitivity response to C. immitis antigen. That is, BALB/c mice manifested footpad hypersensitivity to coccidioidin early during the course of disease, to a level comparable to that of DBA/2 mice. In contrast to the more resistant DBA/2 mouse strain, however, BALB/c mice developed anergy

NMDA receptor antagonism differentially reduces acquisition and expression of sucrose- and fructose-conditioned flavor preferences in BALB/c and SWR mice.

Conditioned flavor preferences (CFP) are elicited by sucrose and fructose relative to saccharin in rats and inbred mice. Whereas dopamine, but not opioid receptor antagonists interfere with the acquisition (learning) and expression (maintenance) of sugar-CFP in rats, these antagonists differentially affect acquisition and expression of sucrose- and fructose-CFP in BALB/c and SWR inbred mice. Given that NMDA receptor antagonism with MK-801 blocks acquisition, but not expression of fructose-CFP in rats, the present study examined whether MK-801 altered the expression and acquisition of sucrose-
