A 36-year-old woman with a history of redo mitral valve replacement (bileaflet mechanical mitral valve prosthesis) in 2002 for prosthetic mitral valve endocarditis presented to the emergency department with a history of recurrent palpitations. A transthoracic echocardiogram was performed and falsely indicated a diagnosis of prosthetic mitral valve thrombosis. However, review of the echocardiogram by an experienced echocardiologist revised the diagnosis to microbubbles formation phenomenon with mechanical mitral prosthesis. Frame-by-frame analysis or reducing the frame rate of the recorded echo

Lipid-coated microbubbles (MBs) are widely used as ultrasound contrast agents and drug delivery vehicles, yet the microscale organization of their shell constituents remains poorly understood. In this study, we investigated how lipid shell composition and environmental factors influence the formation of domains in the shell of microbubbles composed of 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC) and polyoxyethylene-40-stearate (PEG40St) from 9:1 to 1:9 molar ratios. Fluorescence microscopy images revealed that these shells formed polymorphic, heterogeneous domain structures rather than un

Animal-derived lung surfactants have saved millions of lives of preterm neonates with neonatal Respiratory Distress Syndrome (nRDS). However, a replacement for animal-derived lung surfactants has been sought for decades due to its high manufacturing cost, inaccessibility in low-income countries, and failure to show efficacy when nebulized. This study investigated the use of lipid-coated microbubbles as potential replacements for exogenous lung surfactants. Three different formulations of microbubbles (DPPC with/out PEG40-stearate and poractant alfa) were prepared, and their equilibrium and dyn

BACKGROUND AND OBJECTIVE: Sonothrombolysis employs ultrasound waves to induce inertial cavitation of microbubbles, generating high-speed microjets that mechanically erode blood clots. While clot lysis efficacy is closely linked to the proximity of microbubbles to the clot surface, experimental quantification remains challenging due to blood opacity and small timescales.
METHODS: This study utilises a computational modelling strategy to evaluate the impact of microbubble standoff distance (10-70μm) at ultrasound pressure amplitudes (600-1500 kPa) on sonothrombolysis efficacy, as quantified by

Ultrasound and microbubble-mediated gene delivery is emerging as a powerful nonviral gene delivery approach due to its ability to target various tissues. Since microbubble cavitation plays a crucial role in gene delivery, factors affecting cavitation, such as microbubble composition, size, ultrasound pressure, frequency, and pulse interval, can directly affect the efficiency of gene delivery. The effect of ultrasound parameters on gene delivery efficiency has been systematically investigated in numerous studies. However, relatively few studies have investigated the influence of different micro

OBJECTIVE: This study aimed to develop targeted cationic microbubbles conjugated with a CD105 antibody (CMB105) for use in targeted vascular endothelial cell gene therapy and ultrasound imaging. We compared the results with untargeted cationic microbubbles (CMB) and neutral microbubbles (NMB).
METHODS: CMB105 were prepared and compared with untargeted CMB and NMB. First, the microbubbles were characterized in terms of size, zeta-potential, antibody binding ability and plasmid DNA loading capacity. A tumor model of subcutaneous breast cancer in nude mice was used for our experiments. The abili

OBJECTIVE: Endobronchial ultrasound (EBUS) is commonly used to guide transbronchial needle biopsies for the staging of lymph nodes in non-small cell lung cancer patients. Although contrast-enhanced ultrasound (CEUS) and microbubbles (MBs) can improve the diagnostic accuracy in tumors, the ability of contrast-enhanced EBUS (CE-EBUS) to image MBs has not yet been comprehensively evaluated. In this study, we assessed the ability of a CE-EBUS system (Olympus EU-ME2 PREMIER and BF-UC180F bronchoscope) to detect laboratory-synthesized MBs in comparison to clinical (Toshiba Aplio SSA-790A) and pre-cl

In recent work, oxygen microbubbles (OMB) have been shown to oxygenate hypoxic tumors, increase radio-sensitivity and improve tumor control by radiation therapy. Compared to intra-tumoral injection, intravenous delivery of adjuvant agents such as OMBs for radiotherapy offers an attractive means of achieving true theranostic function in a minimally invasive manner via contrast-enhanced ultrasound (CEUS), while reducing the risk of injury, infection or displacing tumor cells. However, short intravascular circulation times with conventional DSPC-lipid OMBs may lead to premature off-target dissolu

Gaseous molecules including nitric oxide, hydrogen sulfide, carbon monoxide and oxygen mediate numerous cell signaling pathways and have important physiological roles. Several noble gasses have been shown to elicit biological responses. These bioactive gasses hold great therapeutic potential, however, their controlled delivery remains a significant challenge. Recently, researchers have begun using microbubbles and liposomes to encapsulate such gasses for parenteral delivery. The resultant particles are acoustically active, and ultrasound can be used to stimulate and/or image gas release in a t

Microbubbles (MBs) are gas-filled hollow microparticles. Under the ultrasound field, the difference of acoustic characteristics between gas and plasma in the blood vessel will cause strong impedance mismatch and echogenicity to enhance ultrasonic grey scale imaging. The linear or nonlinear resonance of MB shell in the ultrasound field would lead to repeated expansion-contraction till destruction. In this case, the pre-loaded drug in the MB shell will be released in an accelerated manner triggered by the ultrasound at a probe-given site. Sonoporation effect of the cell membrane would also help

Improved treatment of acute ischemic stroke with ultrasound and microbubbles in combination with thrombolytic drugs shows great promise, but the optimal techniques, indications, and contraindications have not yet been well defined. Moreover, details such as microbubble dosage, delivery, required thrombolytic drug dosage, and optimal ultrasound characteristics all remain uncertain. Recent results suggest that ultrasound and microbubbles may be effective in clot lysis of ischemic stroke without additional thrombolytic drugs. Moreover, targeting thrombus with specific immunobubbles may improve th

The development of ultrasound contrast agents, containing encapsulated microbubbles, has increased the possibilities for diagnostic imaging. Ultrasound contrast agents are currently used to enhance left ventricular opacification, increase Doppler signal intensity, and in myocardial perfusion imaging. Diagnostic imaging with contrast agents is performed with low acoustic pressure using non-linear reflection of ultrasound waves by microbubbles. Ultrasound causes bubble destruction, which lowers the threshold for cavitation, resulting in microstreaming and increased permeability of cell membranes

Localized delivery could decrease the systemic side effects of toxic chemotherapy drugs. The unique delivery agents we examine consist of microbubbles with an outer lipid coating, an oil layer, and a perfluorobutane gas core. These structures are 0.5-12 microm in radius at rest. Oil layers of these acoustically active lipospheres (AALs) range from 0.3-1.5 microm in thickness and thus the agents can carry a large payload compared to nano-scale drug delivery systems. We show that triacetin-based drug-delivery vehicles can be fragmented using ultrasound. Compared with a lipid-shelled contrast age

AIMS: This study investigated vancomycin-microbubbles (Vm-MBs) and meropenem (Mp)-MBs with ultrasound-targeted microbubble destruction (UTMD) to disrupt biofilms and improve bactericidal efficiency, providing a new and promising strategy for the treatment of device-related infections (DRIs).
METHODS: A film hydration method was used to prepare Vm-MBs and Mp-MBs and examine their characterization. Biofilms of methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli were treated with different groups. Biofilm biomass differences were determined by staining. Thickness and bacteria

OBJECTIVE: The response of ultrasound contrast agents is sensitive to ambient pressure, especially via their scattered subharmonic signal, which makes them a promising candidate for non-invasive pressure measurements in vivo. This work aimed to understand the sensitivity to ambient pressure of subharmonic oscillations from single microbubbles.
METHODS: The subharmonic oscillation amplitude of single microbubbles in response to varying ambient pressure was studied both experimentally and numerically. In experiment, approximately 2200 single microbubbles from a monodisperse population were meas

During laser atherectomy, the use of contrast can lead to the formation of microbubbles, a common but potentially problematic occurrence. This can result in distal embolization and significant hemodynamic compromise. We would like to share a case of a 64-year-old female who had severe calcified in-stent restenosis with stent underexpansion, which was treated with excimer laser coronary atherectomy. The procedure was complicated by the formation of macrobubbles, which caused slow reflow with temporary hypotension and bradycardia. Laser atherectomy can cause macrobubble formation with distal emb

Molecular markers associated with diseases can be visualized and quantified noninvasively with targeted ultrasound contrast agent (t-UCA) consisting of microbubbles (MBs) that can bind to specific molecular targets. Techniques used for quantifying t-UCA assume that all unbound MBs are taken out of the blood pool few minutes after injection and only MBs bound to the molecular markers remain. However, differences in physiology, diseases, and experimental conditions can increase the longevity of unbound MBs. In such conditions, unbound MBs will falsely be quantified as bound MBs. We have develope

Microbubbles offer unique properties as combined carriers of therapeutic payloads and diagnostic agents. Here we report on the development of novel microbubble architectures that in addition to the usual lipid shell have an actin cytoskeletal cortex assembled on their exterior. We show, using atomic force microscopy that this biomimetic coating creates a thin mesh that allows tuning of the mechanical properties of microbubbles and that the nature of actin assembly is determined by the fluidity of the lipid layer. Further, we show that it is possible to attach payloads and targeting-ligands to

A four-year-old, female neutered English springer spaniel presented for evaluation of exercise intolerance, panting and lingual cyanosis. Echocardiography revealed a structurally and functionally normal heart with the presence of spontaneous intracardiac microbubbles entering the right side of the heart from the caudal vena cava. A cause for the clinical signs could not be identified; routine blood work, urinalysis and electrocardiography were unremarkable. Testing for tick-borne diseases, clotting times, cardiac biomarkers, thyroid function and basal cortisol were all within normal limits. Fa

A non-viral gene delivery approach with nano/microbubbles and ultrasound offers opportunities for targeting soft tissues for gene therapy. The periodontium is a complex structure comprised of hard (cementum, alveolar bone) and soft tissues (periodontal ligament, gingivae). We hypothesized that our established gene delivery method would allow the periodontal tissue to be targeted for transfection for gene therapy. Expression kinetics and sites of transfection sites with this approach were investigated in rat periodontal tissue. Bioluminescence imaging revealed that transient gene expression was
