Effects of microcystins on fish.

Microcystins (MCs) are hepatotoxic heptapeptides released into water during or on senescence of cyanobacterial blooms. This review details the different effects of the MCs on fish and discusses their potential consequences in aquatic food webs. In early life stages, exposure to MCs causes, in a dose-dependent manner, perturbations to embryonic hatching, decrease in survival and growth rate, as well as histopathological effects (enlarged and opaque yolk sac, small head, curved body and tail, hepatobiliary abnormalities, ultrastructural alterations in hepatocytes, heart rate perturbations). In a

A Review of Nephrotoxicity of Microcystins.

Cyanobacterial blooms triggered by eutrophication and climate change have become a global public health issue. The toxic metabolites microcystins (MCs) generated by cyanobacteria can accumulate in food chain and contaminate water, thus posing a potential threat to human and animals health. Studies have suggested that aside liver, the kidney may be another target organ of MCs intoxication. Therefore, this review provides various evidences on the nephrotoxicity of MCs. The review concludes that nephrotoxicity of MCs may be related to inhibition of protein phosphatases and excessive production of

Microcystins and Reproductive Dysfunction: Mechanisms and Consequences.

Accelerating eutrophication of aquatic ecosystems worldwide has increased concern regarding cyanotoxin exposure as an emerging environmental and public health issue, with Microcystin-LR (MC-LR) among the most extensively studied congeners due to its widespread occurrence and high toxicity. Evidence from experimental animal and cellular studies indicates that MC-LR elicits pronounced toxic impacts on both the male and female reproductive systems. In males, MC-LR induces overt testicular injury, compromises the structural and functional integrity of the blood-testis barrier, and triggers severe

Adverse effects of microcystins on sperm: A systematic review.

Eutrophication of water bodies can lead to cyanobacterial blooms, with the resultant release of microcystins (MCs), posing a threat to the ecosystem and human health. MCs are environmental toxins with male reproductive toxicity. However, there is a dearth of reviews focusing on sperm or spermatogenesis. In this paper, studies on sperm toxicity caused by MCs in recent 20 years were collected and summarized, aiming at revealing the toxic effects and potential mechanisms of MCs on sperm. Based on the previous findings, MCs can decline sperm quality and count, and cause malformation in vertebrates

Protein phosphatase inhibition and in vivo hepatotoxicity of microcystins.

Administration of microcystin (MCYST)-YM or -LR (peptide hepatotoxins produced by the cyanobacterium Microcystis aeruginosa) to mice resulted in the inhibition of liver protein phosphatase 1 and 2A activity. In all cases significant inhibition preceded or accompanied clinical changes due to MCYST intoxication. Fifteen minutes after intraperitoneal injection of lethal doses of MCYST-YM protein phosphatase activity was already decreased to 44% of controls, and by 60 min was further decreased to 22% of controls. The inhibition was dose dependent: intraperitoneal injection with 84 nmol/kg of MCYST

Vertical transmission of microcystins to Nile crocodile (Crocodylus niloticus) eggs.

Cyanobacteria or blue green algae are known for their extensive and highly visible blooms in eutrophic, stagnant freshwater bodies. Climate change and global warming have also contributed to a rise in toxic cyanobacterial blooms. One of the most important cyanobacteria is Microcystis aeruginosa, which can synthesize various microcystins that can affect the health of terrestrial and aquatic animals. Commercial Nile crocodile (Crocodylus niloticus) farming in South Africa is based on keeping breeders (adult males and females) in big dams on farms (captive-bred approach). Unfortunately, cyanobact

Novel insights into cyanobacterial (microcystins) neurotoxicity in rats: hepatic encephalopathy.

Microcystis, a commonly occurring genus of bloom-forming cyanobacteria, can produce numerous secondary metabolites, including microcystins (MCs), which are hepatotoxic and neurotoxic to humans and animals. However, the mechanisms of cyanobacterial neurotoxicity associated with MCs have not yet been clarified. This study reports the first observations of hepatic encephalopathy (HE) after exposure to Microcystis bloom extracts (MEs), which contained MCs. Mechanisms of toxicity were studied in rats exposed to MEs by use of a single intraperitoneal injection of 80 μg MC-LR equivalents/kg, body mas

Nephrotoxic effects of chronic administration of microcystins -LR and -YR.

Acute intoxication with MC-LR induces cytoskeletal alterations, apoptosis and necrosis of hepatocytes resulting in intrahepatic hemorrhage. Preliminary results have shown that chronic treatment of rats with intraperitoneal injections of sublethal doses of microcystins MC-LR and MC-YR could induce not only liver, but also kidney injuries. We aimed to investigate whether the induction of the cytoskeletal changes, apoptosis and necrosis could be the mechanisms involved in the injury of kidney cells in the chronic model of microcystin intoxication. Experimental rats were receiving intraperitoneal

Detection and quantification of microcystins in the mucilage of colonial cyanobacteria.

This study investigated the detection and quantification of microcystins (MCs) within the mucilage of a Microcystis-dominated bloom, employing methods to separate and quantify attached extracellular polymeric substances (EPS) from cyanobacterial cells. The lowest MCs concentration was measured on the filter by classical filtration (112.44 ± 4.94 μg L-1) and methanol extraction, representing free MCs (intracellular and associated with EPS of the mucilage) and MCs adsorbed onto large environmental particles. Centrifugation followed by lyophilization of the pellets significantly elevated particul

Potential Endocrine Disruption of Cyanobacterial Toxins, Microcystins and Cylindrospermopsin: A Review.

Microcystins (MCs) and cylindrospermopsin (CYN), although classified as hepatotoxins and cytotoxins, respectively, have been shown to also induce toxic effects in many other systems and organs. Among them, their potential endocrine disruption (ED) activity has been scarcely investigated. Considering the increasing relevance of ED on humans, mammals, and aquatic organisms, this work aimed to review the state-of-the-art regarding the toxic effects of MCs and CYN at this level. It has been evidenced that MCs have been more extensively investigated than CYN. Reported results are contradictory, wit

[Effect of phosphorus on the production of microcystin].

Effect of phosphorus on the production of microcystin was researched. The effects of soluble reactive phosphorus (SRP) on the growth of cells and on the production of Microcystin were studied. In addition, the efficiency of four different phosphorus compounds was researched. The results showed that microcystin increased with the increase of SRP, and c(TP) = 0.55 mg x L(-1) was the best growth concentration. When c(TP) < or = 0.55 mg x L(-1), the microcystin production increased with the increase of phosphorus concentration and was the lowest without phosphorus. Moreover, when c(TP) > 0.55 mg x

Effect of microwave, frying and boiling on lettuce contaminated with microcystins.

Microcystins inevitably exist in crops by irrigating, potentially threatening food safety. To assess thermal treatment on the fate of microcystins, we evaluated effect of routine cooking practice (microwave, frying and boiling) on health risk of lettuce that was previously exposed to 20, 50 and 100 μg·L-1 microcystins before harvesting. All 3 cooking methods decreased the concentration of microcystins in lettuce, and the decreased degree caused by boiling (43.73%) was largest, followed by frying (40.28%) and microwave (37.71%). Moreover, the results of in vitro digestion coupled with Caco-2 ce

Acute exposure to microcystins affects hypothalamic-pituitary axes of male rats.

Microcystins (MCs) produced by some cyanobacteria can cause toxicity in animals and humans. In recent years, growing evidence suggests that MCs can act as endocrine disruptors. This research systematically investigated effects of microcystin-LR (MC-LR) on endocrine organs, biosynthesis of hormones and positive/negative feedback of the endocrine system in rats. Male, Sprague-Dawley rats were acutely administrated MC-LR by a single intraperitoneal injection at doses of 45, 67.5 or 90 μg MC-LR/kg body mass (bm), and then euthanized 24 h after exposure. In exposed rats, histological damage of hypo

Microcystins-Loaded Aged Nanoplastics Provoke a Metabolic Shift in Human Liver Cells.

Studies concerning the toxicity of pollutant-loaded nanoplastics (NPs) toward humans are still in their infancy. Here, we evaluated the adsorption of microcystins (MCs) by pristine and aged polystyrene nanoplastics (PSNPs), prepared MCs-loaded aged PSNPS (1, 5, 10, 15, and 19 μg/mg), and systematically mapped the key molecular changes induced by aged and MCs-loaded PSNPs to human hepatoblastoma (HepG2) cells. According to the results, MC-LR adsorption is increased 2.64-fold by aging, and PSNP accumulation is detected in HepG2 cells. The cytotoxicity of the MC-LR-loaded aged PSNPs showed a posi

Microcystin: From Blooms to Brain Toxicity.

An increase in the temperature of lakes and ponds facilitates the over-growth of photosynthetic cyanobacteria that produce a class of toxins called cyanotoxins. The abundance of cyanobacteria poses a significant threat to drinking and irrigation water supplies, and therefore, cyanotoxins have become a major class of environmental pollutants. Microcystins, the most common cyanotoxins, are cyclic peptides produced by cyanobacteria through non-ribosomal peptide synthases, and currently, approximately 279 microcystins have been identified to date. Exposure to microcystins can cause liver and brain

Serologic evaluation of human microcystin exposure.

Microcystins are among the most commonly detected toxins associated with cyanobacteria blooms worldwide. Two episodes of intravenous microcystin exposures occurred among kidney dialysis patients during 1996 and 2001. Analysis of serum samples collected during these episodes suggests that microcystins are detectable as free and bound forms in human serum. Our goal was to characterize the biochemical evidence for human exposure to microcystins, to identify uncertainties associated with interpretation of these observed results, and to identify research needs. We analyzed serum samples using enzym

Analysis of free and metabolized microcystins in samples following a bird mortality event.

In the summer of 2012, over 750 dead and dying birds were observed at the Paul S. Sarbanes Ecosystem Restoration Project at Poplar Island, Maryland, USA (Chesapeake Bay). Clinical signs suggested avian botulism, but an ongoing dense Microcystis bloom was present in an impoundment on the island. Enzyme-linked immunosorbent assay (ELISA) analysis of a water sample indicated 6000 ng mL-1 of microcystins (MCs). LC-UV/MS analysis confirmed the presence of MC-LR and a high concentration of an unknown MC congener (m/z 1037.5). The unknown MC was purified and confirmed to be [D-Leu1]MC-LR using NMR sp

UV-Induced Degradation of Microcystin-LR Mediated by a Ratiometric Fluorescence Probe for the Visual Detection of Microcystin-LR.

Microcystin-LR (MC-LR), a natural toxin produced by cyanobacteria, poses a significant threat to human health and ecological systems. Rapid and accurate quantification of MC-LR plays a critical role in the early warning systems and environmental risk assessment of eutrophic water bodies. Herein, we identified a unique UV-induced degradation mechanism of MC-LR mediated by a newly developed ratiometric fluorescence probe ASS12. Under UV irradiation, the probe undergoes a specific reaction with MC-LR, during which its fluorescence color changes sequentially from yellow to red and ultimately to bl

Advances in the Intestinal Toxicity of Microcystin-LR.

Microcystin-LR (MC-LR), the most prevalent and toxic microcystin congener, is a persistent cyanotoxin that reaches humans mainly through contaminated water and food. Because the intestine is the first tissue exposed after ingestion, it is both a portal of systemic uptake and a direct toxicological target. This review integrates evidence on intestinal distribution and absorption, exposure-dependent toxic phenotypes, and molecular mechanisms. Acute or high-dose exposure predominantly causes villus erosion, epithelial apoptosis, cytoskeletal collapse, and rapid barrier failure, whereas repeated e

Effects of microcystin-LR on mammalian ovaries.

The ovaries play critical roles in regulating oocyte maturation and sex steroid hormone production and thus are critical for female reproduction. Ovarian function relies on hormone receptors and signaling pathways, making the ovaries potential targets for environmental factors, such as microcystins (MCs). MCs are a diverse group of cyanobacterial toxins generally found in eutrophic water or algal blooms. Here, we review relevant research on the associations between MC exposure and ovarian dysfunction, including their effects on ovarian morphology, folliculogenesis, steroid production, oxidativ
