We present a photo-gated corona microfluidics (PGCM) platform for programmable manipulation of diverse microscale objects on open, electrode-pattern-free surfaces. In PGCM, near-infrared laser irradiation locally triggers a reversible solid-liquid phase transition in the paraffin layer, switching the charge-transport regime from diffusion-limited to convection-dominated and reconstructing highly confined electric-field gradients. Quantitative charge measurements confirm that droplets acquire net positive charges via corona discharge, establishing Coulombic force as the dominant actuation mecha

Microfluidic technologies have significantly advanced whole blood analysis by enabling precise, rapid, and cost-effective diagnostic solutions. These platforms have made significant progress in overcoming longstanding challenges such as clogging caused by red blood cells (RBCs), white blood cells (WBCs), and platelets, as well as managing the non-Newtonian viscosity of blood, which affects flow stability and device performance. The direct processing of undiluted whole blood has enabled the isolation of rare cells such as circulating tumor cells (CTCs) with recovery rates of approaching 90%, an

Since the discovery of inertial focusing in 1961, numerous theories have been put forward to explain the migration of particles in inertial flows, but a complete understanding is still lacking. Recently, computational approaches have been utilized to obtain better insights into the underlying physics. In particular, fundamental aspects of particle focusing inside straight and curved microchannels have been explored in detail to determine the dependence of focusing behavior on particle size, channel shape, and flow Reynolds number. In this review, we differentiate between the models developed f

In native tissues, various cell types organize and spatiotemporally function and communicate with neighboring or remote cells in a highly regulated way. How can we replicate these amazing functional structures in vitro? From the view of a chemist, the heterogeneous cells and extracellular matrix (ECM) could be regarded as various chemical substrate materials for "synthetic" reactions during tissue engineering. But how can we accelerate these reactions? Microfluidics provides ideal solutions. Microfluidics could be metaphorically regarded as a miniature "biofactory", whereas the on-chip critica

Infertility is a growing global health issue with far-reaching socioeconomic implications. A downward trend in male fertility highlights the acute need for affordable and accessible diagnosis and treatment. Assisted reproductive technologies are effective in treating male infertility, but their success rate has plateaued at ∼33% per cycle. Many emerging opportunities exist for microfluidics - a mature technology in other biomedical areas - in male infertility diagnosis and treatment, and promising microfluidic approaches are under investigation for addressing male infertility. Microfluidics opens new avenues for materials engineering, as it enables scalable synthesis and provides highly controllable environments for reactions. Herein, we leverage microfluidics to engineer the properties of (bioactive) metal-phenolic network nanoparticles (MPN NPs), an emerging and highly modular nanoparticle platform for the incorporation and delivery of bioactive cargo. By varying the microfluidics operating parameters (flow rate ratio, total flow rate, temperature) and NP composition, we assemble MPN NPs, which consist of poly(ethylene glycol), biomacromolecules, metal ions, and p

Stereolithography (SLA) 3D printing has become a standard tool to produce microfluidic structures, thanks to advancements in both 3D printer technology and resin materials. Used across many applications, SLA-printed microfluidics are often exposed to a variety of solvents, giving rise to potential leaching from the printed structure (in the form of unreacted resin components) that can potentially interfere with device functionality. In this study, we investigated the leaching behavior of SLA-printed structures from both external surfaces (under static conditions) and microfluidic surfaces (und

Microfluidic paper-based analytical devices (μPADs) have experienced an unprecedented story of success. In particular, as of today, most people have likely come into contact with one of their two most famous examples─the pregnancy or the SARS-CoV-2 antigen test. However, their sensing performance is constrained by the optical readout of nanoparticle agglomeration, which typically allows only qualitative measurements. In contrast, single-impact electrochemistry offers the possibility to quantify species concentrations beyond the pM range by resolving collisions of individual species on a microe

Fifteen percent of couples of reproductive age suffer from infertility globally and the burden of infertility disproportionately impacts residents of developing countries. Assisted reproductive technologies (ARTs), including in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI), have been successful in overcoming various reasons for infertility including borderline and severe male factor infertility which consists of 20%-30% of all infertile cases. Approximately half of male infertility cases stem from suboptimal sperm parameters. Therefore, healthy/normal sperm enrichment a

Point-of-care (POC) diagnostics has attracted increasing attention in biomedical research. However, it remains challenging in low resources setting. Origami-paper microfluidics has emerged as a promising approach for point-of-need (PON) analysis by leveraging the advantages of paper, including low cost, zero power, easy to access, biodegradability, and capillary-driven fluid transport. These features enable the development of PON platforms suitable for resource-limited environments without centralized instrumentation and trained personnel. This perspective highlights recent advances in origami

Osteoarthritis (OA) is a complex degenerative joint disease perpetuated by a vicious cycle of biomechanical, biochemical, and chondrocyte homeostasis imbalances, ultimately leading to progressive cartilage degradation. A simultaneous approach to biomechanical disturbances, chondrocyte metabolic imbalances, and maintaining cartilage homeostasis simultaneously holds promise in tackling this challenge. Herein, we report a microfluidics-enabled nanoparticle, termed ZCMC, formed by grafting a magnesium-modified chondroitin sulfate polymer layer onto a zein core preloaded with cannabidiol (CBD), for

Current single-cell RNA-sequencing approaches have limitations that stem from the microfluidic devices or fluid handling steps required for sample processing. We develop a method that does not require specialized microfluidic devices, expertise or hardware. Our approach is based on particle-templated emulsification, which allows single-cell encapsulation and barcoding of cDNA in uniform droplet emulsions with only a vortexer. Particle-templated instant partition sequencing (PIP-seq) accommodates a wide range of emulsification formats, including microwell plates and large-volume conical tubes,

We study rheotaxis of bull sperm inside microchannels to characterize the effects of flow and wall shape on sperm swimming behavior. We found that a large percentage of sperm cells, 80 to 84%, exhibited positive rheotaxis (sperm cells swimming against the flow) within flow velocities of 33 to 134 μm s(-1). Sperm cells were also found to reverse their swimming direction when the liquid flow direction was reversed. Time taken by sperm cells to reverse their swimming direction was inversely proportional to the flow velocity. Sperm behavior was significantly affected by the sperm position with res

Sub-nanoliter droplets produced in microfluidic devices have gained enormous importance for performing all kinds of biochemical assays. One of the main reasons is that the amounts of reagents employed can be reduced by approximately five orders of magnitude compared with conventional microplate assays. This chapter describes how to design, fabricate, and operate a microfluidic device to perform enzyme kinetics and enzyme inhibition assays in droplets. This procedure can be used effectively to screen a small library of compounds. Then, we describe how to use this droplet microfluidic setup to s
