Thermoresponsive copolymer microgels.

Colloidal aqueous microgels have attracted notable attention due to their possible applications in both polymer chemistry and the (bio)material sector. Due to their soft internal network structure and their response to different external stimuli, they are suitable as carrier systems for different kinds of molecules (e.g. nanoparticles, drugs, diagnostic agents, and enzymes), as optically active materials or even as model systems for fundamental research on soft condensed matter. Within the class of microgels, a variety of molecular structures is possible. The aim of this manuscript is to revie

Microgels Sopping Up Toxins-GM1a-Functionalized Microgels as Scavengers for Cholera Toxin.

Vibrio cholerae is a Gram-negative bacterium that causes secretory diarrhea and constitutes a major health threat in the industrialized world and even more in developing countries. Its main virulence factor is the cholera toxin, which is internalized by intestinal epithelial cells after binding to the glycosphingolipid receptor GM1a on their apical surface. A potential future solution to dampen complications of cholera infection is by scavenging the cholera toxin by presenting competitive binding motifs to diminish the in vivo toxicity of V. cholerae. Here, we generate GM1a-functionalized and

Microgel PAINT - nanoscopic polarity imaging of adaptive microgels without covalent labelling.

Polymer nanostructures have enormous potential for various applications in materials and life sciences. In order to exploit and understand their full capabilities, a detailed analysis of their structures and the environmental conditions in them is essential on the nanoscopic scale. With a super-resolution fluorescence microscopy technique known as PAINT (Points Accumulation for Imaging in Nanoscale Topography), we imaged colloidal hydrogel networks, so-called microgels, having a hydrodynamic radius smaller than the diffraction limit, gaining unprecedented insight into their full 3D structure w

Gels and microgels for nanotechnological applications.

In recent years, "smart" materials have been the focus of considerable interest, from both fundamental and applied perspectives. Polymer gels are within this category; they respond to specific environmental stimuli by changing their size. Thus, the internal structure, the refractive index, and the mechanical properties of the polymer network change. They are considered super absorbent materials, as they can absorb solvent up to several hundred times their own weight. They respond rapidly to local environmental variations, an important fact in device miniaturization and microsensor developments

Self-Induced Buckling in Hollow Microgels.

Hollow microgels are elastic polymer shells easily realizable in experiments. Recent works have shown the emergence of buckling events in dilute hollow microgels under the effect of an added osmotic pressure. Here, we perform large-scale simulations to show that these microgels at high enough packing fractions undergo spontaneous symmetry-breaking deformations ranging from single large dents to multiple indentations, even in the absence of any externally applied stress. This self-induced buckling phenomenon is thus solely driven by interparticle crowding. We construct a phase diagram inspired

Colloidal microgels in drug delivery applications.

Colloidal microgels have recently received attention as environmentally responsive systems and now are increasingly used in applications as carriers for therapeutic drugs and diagnostic agents. Synthetic microgels consist of a crosslinked polymer network that provides a depot for loaded drugs, protection against environmental hazards and template for post-synthetic modification or vectorization of the drug carriers. The aim of this manuscript is to review recent attempts to develop new microgel formulations for oral drug delivery, to design metal-containing microgels for diagnostic and therape

Facile Physicochemical Reprogramming of PEG-Dithiolane Microgels.

Granular biomaterials have found widespread applications in tissue engineering, in part because of their inherent porosity, tunable properties, injectability, and 3D printability. However, the assembly of granular hydrogels typically relies on spherical microparticles and more complex particle geometries have been limited in scope, often requiring templating of individual microgels by microfluidics or in-mold polymerization. Here, we use dithiolane-functionalized synthetic macromolecules to fabricate photopolymerized microgels via batch emulsion, and then harness the dynamic disulfide crosslin

Supramolecular microgels for Pickering emulsion stabilization in the limited coalescence regime: The key role of microgel structure.

Oil-in-water Pickering emulsions can be stabilized by poly-N-Isopropylacrylamide microgels that adsorb, spread and entangle at the droplets surface. Several studies have tuned the microgels size and deformability to understand the relation between microgel structural parameters, their packing and conformation at the droplet surface and the resulting properties of emulsions. However, both the expected key role of the microgel structure and the mechanism that stops droplets coalescence in the particle-poor regime have never been deciphered. Here, we investigate an unprecedented range of structur

Hollow microgels: Interplay of structure, mechanics, and transport.

Responsive hollow nano- and microgels, cross-linked polymer networks containing a solvent-filled cavity, have attracted significant interest in both fundamental science and emerging technologies. The presence of these internal cavities imparts enhanced softness and deformability, enabling pronounced and tunable size and shape transformations in response to external stimuli. As a result, these systems are particularly well suited for biomedical applications such as drug delivery, controlled release, and encapsulation. By tuning synthesis conditions and incorporating appropriate functional moiet

Biofabrication and Characterization of Vascularizing PEG-Norbornene Microgels.

Establishing a robust, functional microvascular network remains a critical challenge for both the revascularization of damaged or diseased tissues and the development of engineered biological materials. Vascularizing microgels may aid in efforts to develop complex, multiphasic tissues by providing discrete, vascularized tissue modules that can be distributed throughout engineered constructs to vascularize large volumes. Here, we fabricated poly(ethylene glycol)-norbornene (PEGNB) microgels containing endothelial and stromal cells via flow-focusing microfluidic droplet generation. When embedded

Selenium-silk microgels as antifungal and antibacterial agents.

Antimicrobial resistance is a leading threat to global health. Alternative therapeutics to combat the rise in drug-resistant strains of bacteria and fungi are thus needed, but the development of new classes of small molecule therapeutics has remained challenging. Here, we explore an orthogonal approach and address this issue by synthesising micro-scale, protein colloidal particles that possess potent antimicrobial properties. We describe an approach for forming silk-based microgels that contain selenium nanoparticles embedded within the protein scaffold. We demonstrate that these materials hav

Effective Interactions and Dynamical Properties of Thermoresponsive Hollow Microgels.

Hollow microgelspolymer network shellsare soft colloids whose collective behavior is governed by their intrinsic network structure, their unique topology, and solvent quality. Here, we investigate the effective interactions and the dynamics of hollow microgel suspensions at different temperatures just below the volume phase transition. Despite the particles becoming mechanically stiffer as solvent quality worsens, the extracted effective interactions appear to be softer with increasing temperature. This counterintuitive behavior results from a decrease in monomer repulsion, consistent with a

Ionically annealed zwitterionic microgels for bioprinting of cartilaginous constructs.

Foreign body response (FBR) is a pervasive problem for biomaterials used in tissue engineering. Zwitterionic hydrogels have emerged as an effective solution to this problem, due to their ultra-low fouling properties, which enable them to effectively inhibit FBRin vivo. However, no versatile zwitterionic bioink that allows for high resolution extrusion bioprinting of tissue implants has thus far been reported. In this work, we introduce a simple, novel method for producing zwitterionic microgel bioink, using alginate methacrylate (AlgMA) as crosslinker and mechanical fragmentation as a microgel

Fungal Keratitis Treatment Using Drug-Loaded Hyaluronic Acid Microgels.

Antifungal drug-loaded hyaluronic acid (HA) microgels using conjugation and encapsulation drug-loading techniques were utilized in the treatment of fungal keratitis. Natamycin (NAT) and amphotericin B (AMB) drugs were chemically linked to HA microgels by employing a chemical coupling agent to obtain conjugated (C-) HA:NAT and HA:AMB microgels. Also, these drugs were loaded into the HA microgel network during HA microgel preparation to attain encapsulated (E-) HA:NAT and HA:AMB microgels. The conjugation of drug molecules was confirmed by FT-IR spectra of bare and drug-loaded HA microgels. It w

Switchable photoluminescence of CdTe nanocrystals by temperature-responsive microgels.

In the present study, we report a method for preparing a fluorescent thermosensitive hybrid material based on monodisperse, thermosensitive poly( N-isopropyl acrylamide) (PNIPAM) microgels covered with CdTe nanocrystals of 3.2 nm diameter. The CdTe nanocrystals were covalently immobilized on the surface of PNIPAM microgels. The chemical environment around the CdTe nanocrystals was modified by changing the temperature and inducing the microgel volume-phase transition. This change provoked a steep variation in the nanocrystal photoluminescence (PL) intensity in such a way that when the temperatu

Anisotropic microgels for spatiotemporal co-delivery of anticancer drugs.

Codelivery of chemotherapeutic agents is often challenged by physicochemical incompatibility between co-administered drugs and the inability of conventional systems to achieve spatiotemporal control over drug release. Herein, anisotropic microgels were fabricated by an electrohydrodynamic co-jetting (EHDC) technique using PLGA as a sustained-release matrix and polyethyleneimine (PEI), along with a crosslinker, 1,2,3,4-butanetetracarboxylic dianhydride (BTDA), as a pH-responsive compartment, potentially enabling the compartmentalization of physicochemically incompatible drugs within a single pl

Spatially Controlled Release from Printable Microgels for Embedded Bioprinting.

Granular hydrogels composed of jammed microgels have emerged as promising biomaterials for 3D bioprinting due to their tunable viscoelastic properties and ability to incorporate bioactive molecules and encapsulate cells within the interstitial spaces between the particles. Here, we present a new strategy to achieve spatially controlled release of bioactive molecules using κ-carrageenan microgels having a triple role as a printable bioink, a supportive matrix, and a carrier for bioactive molecules. To enable molecule incorporation, we employed a freeze-drying and absorption loading technique an

Transforming sustainable plant proteins into high performance lubricating microgels.

With the resource-intensive meat industry accounting for over 50% of food-linked emissions, plant protein consumption is an inevitable need of the hour. Despite its significance, the key barrier to adoption of plant proteins is their astringent off-sensation, typically associated with high friction and consequently poor lubrication performance. Herein, we demonstrate that by transforming plant proteins into physically cross-linked microgels, it is possible to improve their lubricity remarkably, dependent on their volume fractions, as evidenced by combining tribology using biomimetic tongue-lik

Controlled release of therapeutic antibody using hydrolytically degradable microgels.

Monoclonal antibodies have gained significant interest as potential therapeutics for treating various diseases. However, these therapies are not always effective due to poor treatment compliance associated with multiple administrations and drug resistance. Thus, there is a growing interest in developing advanced monoclonal antibody delivery systems that can customize pharmacokinetics to enhance therapeutic outcomes. This work aimed to engineer hydrolytic 4-arm PEG maleimide (PEG-4MAL) microgels for the controlled delivery of therapeutic antibodies, specifically anti-angiogenic bevacizumab, to

Chemical Orthogonality in Surface-Patterned Poly(ethylene glycol) Microgels.

Because of its widely known antifouling properties, a variety of lithographic approaches has been used to pattern surfaces with poly(ethylene glycol) (PEG) to control surface interactions with biomolecules and cells over micro- and nanolength scales. Often, however, particular applications need additional functions within PEG-patterned surfaces. Monofunctional films can be generated using PEG modified to include a chemically functional group. We show that patterning with focused electron beams, in addition to cross-linking a monofunctional PEG homopolymer thin-film precursor and grafting the r
