Experimental biology and medicine (Maywood, N.J.)2017eng
Gastrointestinal diseases are a significant health care and economic burden. Prevention and treatment of these diseases have been limited by the available human biologic models. Microphysiological systems comprise organ-specific human cultures that recapitulate many structural, biological, and functional properties of the organ in smaller scale including aspects of flow, shear stress and chemical gradients. The development of intestinal microphysiological system platforms represents a critical component in improving our understanding, prevention, and treatment of gastrointestinal diseases. Thi

Microphysiological systems (MPS) that incorporate engineered blood vasculatures have enabled new opportunities to study human physiology and disease, offering platforms for drug development, tissue modeling, and regenerative medicine. However, most human tissues also contain an equally complex yet underrepresented secondary vascular network, the lymphatic system. Lymphatics play indispensable roles in interstitial fluid drainage, immune cell trafficking, and antigen presentation, and are central to the pathophysiology of diseases such as lymphedema, chronic inflammation, and cancer metastasis.

The pharmaceutical industry continues to face a critical translational bottleneck, with the majority of drug candidates failing in clinical trials despite promising preclinical results, largely due to the limited predictive power of conventional in vitro systems and animal models. Microphysiological systems (MPS), including organ-on-chip and organoid-on-chip platforms, have emerged as transformative human-relevant technologies capable of recapitulating organ-level structure, function, and disease dynamics under precisely controlled microenvironments. By integrating advances in microfabrication

Patient-derived microphysiological systems (P-MPS) have emerged as powerful tools in precision medicine that provide valuable insight into individual patient characteristics. This review discusses the development of P-MPS as an integration of patient-derived samples, including patient-derived cells, organoids, and induced pluripotent stem cells, into well-defined MPSs. Emphasizing the necessity of P-MPS development, its significance as a nonclinical assessment approach that bridges the gap between traditional in vitro models and clinical outcomes is highlighted. Additionally, guidance is provi

A number of major disease states involve skeletal muscle, including type 2 diabetes, muscular dystrophy, sarcopenia and cachexia arising from cancer or heart disease. Animals do not accurately represent many of these disease states. Human skeletal muscle microphysiological systems derived from primary or induced pluripotent stem cells (hPSCs) can provide an in vitro model of genetic and chronic diseases and assess individual variations. Three-dimensional culture systems more accurately represent skeletal muscle function than do two-dimensional cultures. While muscle biopsies enable culture of

Microphysiological Systems (MPS) have emerged as a transformative platform in biomedical research, enabling the investigation of disease mechanisms, drug screening, and toxicity prediction by closely simulating human physiological functions. However, the rapid advancement of MPS technology has raised a series of complex ethical challenges. These include the sourcing and application of human-derived stem cells, the protection of donors' personal and genetic data, the potential for brain organoids to develop consciousness-like characteristics, and the challenges to species boundaries posed by hu

Neutrophils are the most abundant immune cells in human blood and serve as the first line of defense against infection. However, the role of neutrophils in cancer progression is complex and context-dependent, and they are underexplored as therapeutic targets for cancer compared with other immune cell types. Microphysiological systems, also known as complex in vitro models, have emerged as powerful tools for deciphering immune cell-cancer interactions and preclinical drug testing due to the advantages of real-time visualization of cell behaviors, precise control of microenvironments, and incorp

Microphysiological Systems (MPS) feature cells, often cultured in 3D, within a microfluidic circuit aiming to mimic the circulation and cross-talk between organs found in the human body. These platforms have proven useful in different contexts, ranging from basic biology research to disease modelling and drug development - all with the underlying aim of reducing our reliance on animal experiments. In addition to dedicating a “cross-journal collection [https://www.nature.com/collections/fgjeddegjc]” to this topic, we wanted to delve deeper into the key steps to translating MPS from concept to a

The global increase in chronic diseases and comorbidities due to aging populations is placing significant strain on healthcare systems. Chronic kidney disease (CKD) and osteoarthritis (OA) are among the most prevalent conditions in the elderly, with complex interconnections driven by shared risk factors such as systemic inflammation and metabolic dysregulation. Despite the critical need to understand these interactions, traditional animal models often fail to adequately capture the multidirectional crosstalk between human comorbid conditions, limiting insights into their mechanisms and complic

Perfusable hydrogels have garnered substantial attention in recent years for the fabrication of microphysiological systems. However, current methodologies to fabricate microchannels in hydrogel platforms involve sophisticated equipment and techniques, which hinder progress of the field. In this protocol, we present a cost-effective, simple, versatile and ultrafast method to create perfusable microchannels of complex shapes in photopolymerizable hydrogels. Our method uses one-step UV photocross-linking and a photomask printed on inexpensive transparent films, to photopattern both synthetic (PEG

Microphysiological systems (MPS) are 2D or 3D multicellular constructs able to mimic tissue microenvironments. The latest models encompass a range of techniques, including co-culturing of various cell types, utilization of scaffolds and extracellular matrix materials, perfusion systems, 3D culture methods, 3D bioprinting, organ-on-a-chip technology, and examination of tissue structures. Several human brain 3D cultures or brain MPS (BMPS) have emerged in the last decade. These organoids or spheroids are 3D culture systems derived from induced pluripotent cells or embryonic stem cells that conta

pioids are commonly used for treating chronic pain. However, with continued use, they may induce tolerance and/or hyperalgesia, which limits therapeutic efficacy. The human mechanisms of opioid-induced tolerance and hyperalgesia are significantly understudied, in part, because current models cannot fully recapitulate human pathology. Here, we engineered novel human spinal microphysiological systems (MPSs) integrated with plug-and-play neural activity sensing for modeling human nociception and opioid-induced tolerance. Each spinal MPS consists of a flattened human spinal cord organoid derived f

Pancreatic Ductal Adenocarcinoma (PDAC), one of the deadliest cancer types, is dominated by a tumor microenvironment (TME) that is desmoplastic, immunosuppressive, and generally unresponsive to conventional cancer treatments. Cancer-Associated Fibroblasts (CAFs) are thought to drive many of the features of PDAC that contribute to treatment resistance, yet CAFs exhibit wide phenotypic heterogeneity with unclear biological and clinical implications. Recent work has identified CAF subtypes - tumor-restraining CAFs (RestCAFs) or tumor-promoting CAFs (ProCAFs) - that correlate with overall survival

The FDA Modernization Act has accelerated the transition from animal-based testing toward human-relevant new approach methodologies in drug development, shifting attention from technological feasibility to regulatory implementation. Among these approaches, microphysiological systems (MPS) can generate evidence that more directly reflects human physiology, but their regulatory readiness varies substantially across applications. This Review summarizes recent regulatory developments and evaluates the readiness of major organ-specific and multi-organ MPS. Liver MPS have advanced furthest through q

Biological barriers are multicellular structures that precisely regulate the transport of ions, biomolecules, drugs, cells, and other organisms. Transendothelial/epithelial electrical resistance (TEER) is a label-free method for predicting the properties of biological barriers. Understanding the mechanisms that control TEER significantly enhances our knowledge of the physiopathology of different diseases and aids in the development of new drugs. Measuring TEER values within microphysiological systems called organ-on-a-chip devices that simulate the microenvironment, architecture, and physiolog

The small intestine and liver play important role in determining oral drug's fate. Both organs are also interconnected through enterohepatic circulation, which imply there are crosstalk through circulating factors such as signaling molecules or metabolites that may affect drug metabolism. Coculture of hepatocytes and intestinal cells have shown to increase hepatic drug metabolism, yet its crosstalk mechanism is still unclear. In this study, we aim to elucidate such crosstalk by coculturing primary human hepatocytes harvested from chimeric mouse (PXB-cells) and iPSc-derived intestinal cells in

New drug modalities offer life-saving benefits for patients through access to previously undruggable targets. Yet these modalities pose a challenge for the pharmaceutical industry, as side effects are complex, unpredictable, and often uniquely human. With animal studies having limited predictive value due to translatability challenges, the pharmaceutical industry seeks out new approach methodologies. Microphysiological systems (MPS) offer important features that enable complex toxicological processes to be modeled in vitro such as (a) an adjustable complexity of cellular components, including

Perfluorooctane sulfonate (PFOS), a class of synthetic chemicals detected in various environmental compartments, has been associated with dysfunctions of the human central nervous system (CNS). However, the underlying neurotoxicology of PFOS exposure is largely understudied due to the lack of relevant human models. Here, we report bioengineered human midbrain organoid microphysiological systems (hMO-MPSs) to recapitulate the response of a fetal human brain to multiple concurrent PFOS exposure conditions. Each hMO-MPS consists of an hMO on a fully 3D printed holder device with a perfusable orga

Microphysiologic systems (MPS), including new organ-on-a-chip technologies, recapitulate tissue microenvironments by employing specially designed tissue or cell culturing techniques and microfluidic flow. Such systems are designed to incorporate physiologic factors that conventional 2D or even 3D systems cannot, such as the multicellular dynamics of a tissue-tissue interface or physical forces like fluid sheer stress. The female reproductive system is a series of interconnected organs that are necessary to produce eggs, support embryo development and female health, and impact the functioning o

Recent innovations in skin microphysiological systems (MPSs) have gained momentum following regulatory advances such as the FDA Modernization Act 2.0 and the global shift toward alternatives to animal testing. This review highlights the development of three major technologies-3D bioprinting, skin organoids, and skin-on-a-chip-and their roles in replicating human skin physiology for research and preclinical applications. We examine how these platforms model complex skin functions, including epidermal barrier formation, vascular and immune interactions, and disease phenotypes such as psoriasis,
