Light sheet microscopy with acoustic sample confinement.

Contactless sample confinement would enable a whole host of new studies in developmental biology and neuroscience, in particular, when combined with long-term, wide-field optical imaging. To achieve this goal, we demonstrate a contactless acoustic gradient force trap for sample confinement in light sheet microscopy. Our approach allows the integration of real-time environmentally controlled experiments with wide-field low photo-toxic imaging, which we demonstrate on a variety of marine animal embryos and larvae. To illustrate the key advantages of our approach, we provide quantitative data for

Study of cellular adhesion with scanning acoustic microscopy.

A mechanical scanning acoustic reflection microscope was applied to living cells (e.g., osteoblasts) to observe their undisguised shapes and to evaluate their adhesive conditions at a substrate interface. A conditioned medium was collected from a bone-metastatic breast cancer cell line, MDA-MB-231, and cultured with an immature osteoblast cell line, MC3T3-E1. To characterize the cellular adhesion, MC3T3-E1 osteoblasts were cultured with or without MDA-MB-231 conditioned medium for 2 days, then assayed with the scanning acoustic reflection microscope. At 600 MHz the scanning acoustic reflection

Scanning acoustic microscopy of neoplastic and inflammatory cutaneous tissue specimens.

Acoustic microscopy utilizes high frequency ultrasound to generate microscopic images. The current study was designed to examine representative disorders of the skin by use of a reflective scanning acoustic microscope (R-SAM), and to determine whether the obtainable resolution was sufficient to render a microscopic diagnosis. An Olympus UH3 Scanning Acoustic Microscope was utilized with lenses producing burst wave frequencies at 600 and 800 MHz (600 and 800 million cylces/sec). Cutaneous tissue specimens representing 12 different neoplastic and inflammatory disorders were examined. Acoustic im

Potential protective effect of resveratrol on acoustic trauma: electron microscopy study.

According to expert consensus, dystonia can be classified as focal, segmental, multifocal, and generalized, based on the affected body distribution. To provide an empirical and data-driven approach to categorizing these distributions, we used a data-driven clustering approach to compare frequency and co-occurrence rates of non-focal dystonia in pre-defined body regions using the Dystonia Coalition (DC) dataset. We analyzed 1,618 participants with isolated non-focal dystonia from the DC database. The analytic approach included construction of frequency tables, variable-wise analysis using hiera

Observations of amyloid breakdown by proteases over time using scanning acoustic microscopy.

Amyloid consists of insoluble beta-fibrillar proteins with stable structures. The Congo red staining method for histologically detecting amyloid is unsuitable for quantitatively assessing amyloid fibers. Scanning acoustic microscopy (SAM) detects the attenuation of sound (AOS) through sections. This study aimed to clarify whether AOS values reflected the amount of amyloid fibril degradation in tissues. Formalin-fixed paraffin-embedded unstained sections of various types of amyloidosis were digested with different endopeptidases. The AOS images after digestion were observed over time via SAM. T

Quantification of bush-cricket acoustic trachea mechanics using Atomic Force Microscopy nanoindentation.

Derived from the respiratory tracheae, bush-crickets' acoustic tracheae (or ear canals) are hollow tubes evolved to transmit sounds from the external environment to the interior ear. Due to the location of the ears in the forelegs, the acoustic trachea serves as a structural element that can withstand large stresses during locomotion. In this study, we report a new Atomic Force Microscopy Force Spectroscopy (AFM-FS) approach to quantify the mechanics of taenidia in the bush-cricket Mecopoda elongata. Mechanical properties were examined over the longitudinal axis of hydrated taenidia, by indent

Evaluation of acetic acid-induced gastric ulcers in rats by scanning acoustic microscopy.

To clarify the mechanism of gastric ulcer recurrence, we studied acetic acid-induced gastric ulcers. The healing of these gastric ulcers was then studied for 4-21 weeks after the injection. Forty-eight ulcers were examined by macroscopic observation and scanning acoustic microscopy (SAM) and by histologic study of the ulcers. SAM has a higher radio frequency and provides more detailed information about the gastric wall than endoscopic ultrasonography (EUS). Thirteen healed and 35 non-healed gastric ulcers were observed by macroscopic observation over 4-21 weeks. SAM enabled us to observe the n

Scanning Acoustic Microscopy and Time-Resolved Fluorescence Spectroscopy for Characterization of Atherosclerotic Plaques.

Atherosclerotic plaques constitute the primary cause of heart attack and stroke. However, we still lack a clear identification of the plaques. Here, we evaluate the feasibility of scanning acoustic microscopy (SAM) and time-resolved fluorescence spectroscopy (TRFS) in atherosclerotic plaque characterization. We perform dual-modality microscopic imaging of the human carotid atherosclerotic plaques. We first show that the acoustic impedance values are statistically higher in calcified regions compared with the collagen-rich areas. We then use CdTe/CdS quantum dots for imaging the atherosclerotic

A Theoretical Approach in Applying High-Frequency Acoustic and Elasticity Microscopy to Assess Cells and Tissues.

Medical ultrasound is a diagnostic imaging modality used for visualizing internal organs; the frequencies typically used are 2-10 MHz. Scanning acoustic microscopy (SAM) is a form of ultrasound where frequencies typically exceed 50 MHz. Increasing the acoustic frequency increases the specimen's spatial resolution but reduces the imaging depth. The advantages of using SAM over conventional light and electron microscopy include the ability to image cells and tissues without any preparation that could kill or alter them, providing a more accurate representation of the specimen. After scanning the

Preservation of microelastic properties of dentin and tooth enamel in vitro--a scanning acoustic microscopy study.

OBJECTIVES: The aim of this study was to examine the influence of storage media on the elastic properties of dentin and tooth enamel with respect to the storage period. Several reports suggest that saline and other aqueous solutions may induce chemical reactions and the dissolution of minerals, which in turn may cause alterations of elastic tissue properties. METHODS: Three non-erupted human wisdom teeth were extracted and divided in three slices. Sections were stored in three different media for a maximum period of 21 days. During this time all sections were inspected by time-resolved 50-MHz

Selective deletion or preservation of tissue components via enzymatic digestion monitored by scanning acoustic microscopy.

Detecting specific tissue components is valuable in histology. Scanning acoustic microscopy (SAM) measures the attenuation-of-sound (AOS) through tissue sections, enabling the generation of histological images without staining. AOS values decrease as tissues degrade. In this study, we enzymatically digested target components and monitored the process using AOS imaging over time. Additionally, we applied specific dyes and antibodies to inhibit enzyme activity and preserve target component. Collagenase digested the bone to clearly visualise the internal structure. The target component showed a d

Ultrasound Microscopy-Based Identification of Enamel and Restorative Materials: An Ex Vivo Acoustic Impedance Study.

INTRODUCTION AND AIMS: Differentiating restorative materials from enamel during dental examinations is challenging because of their similar appearances. Even with ultrasound microscopy, the acoustic properties of restorative materials remain unassessed. This study investigated the potential of ultrasound microscopy to differentiate between enamel, composite resin, and glass ionomer. METHODS: Extracted third molars served as the tooth model: a 1.2 mm-diameter cylindrical cavity drilled into the enamel and restored with either composite resin (flowable bulk-fill or paste) or glass-ionomer cemen

Diagnostic System for Early In Situ Melanoma Detection Using Acoustic Microscopy and Infrared Spectroscopic Mapping Imaging.

This study proposes a novel diagnostic system for the early detection of cutaneous melanoma based on morphological and biochemical changes during tumor formation. The methods used in this system are acoustic microscopy and infrared (IR) spectroscopy. The former identifies the anatomical parameters of the developing tumor, whilst the latter identifies its biochemical features, both at the micron scale. To implement this diagnostic method, an animal model that mimics human melanoma was developed. The results of this investigation show that using high-frequency (>20 MHz) acoustic microscopy in co

Cortical bone properties in the Brtl/+ mouse model of Osteogenesis imperfecta as evidenced by acoustic transmission microscopy.

Higher skeletal fragility has been established for the Brtl/+ mouse model of osteogenesis imperfecta at the whole bone level, but previous investigations of mechanical properties at the bone material level were inconclusive. Bone material was analyzed separately at endosteal (ER) and periosteal regions (PR) on transverse femoral midshaft sections for 2-month old mice (wild-type n = 6; Brtl/+ n = 6). Quantitative backscattered electron imaging revealed that the mass density computed from mineral density maps was higher in PR than in ER for both wild-type (+2.1%, p < 0.05) and Brtl/+ mice (+1.8%

Time sequence of degeneration pattern of the organ of Corti after acoustic overstimulation. A transmission electron microscopy study.

Female pigmented guinea pigs were exposed to a 3.85 kHz pure tone of 120 dB SPL for 22.5 min. The exposed animals were sacrificed after varying post-exposure intervals (5 min, 4 and 24 h, 5 and 28 days). Transmission electron microscopical examination was performed after serial-sectioning in areas of maximum damage and both basal to and apical to this region. By serial-sectioning, the degradation pattern of the individual cell structures in the organ of Corti could be studied in relation to post-exposure time and proximity to the well-demarcated area of maximum damage in the organ of Corti. Degenerated coracoacromial ligament in shoulders with rotator cuff tears shows higher elastic modulus: measurement with scanning acoustic microscopy.

PURPOSE: The purpose of this study was to determine the elasticity of the coracoacromial ligament in shoulders with and without rotator cuff tears. METHODS: The coracoacromial ligaments from 20 cadaveric shoulders (average patient age 79.5 years; 8 men, 12 women) were divided into six portions--three portions (acromial, central, and coracoid) in two layers (superficial and deep). A total of 120 samples were studied. First, the samples were classified by the collagen fiber orientation into three degeneration patterns: wavy, straight, irregular. For each pattern, the tissue sound speed, which s

Novel Water Probe for High-Frequency Focused Transducer Applied to Scanning Acoustic Microscopy System: Simulation and Experimental Investigation.

A scanning acoustic microscopy (SAM) system is a common non-destructive instrument which is used to evaluate the material quality in scientific and industrial applications. Technically, the tested sample is immersed in water during the scanning process. Therefore, a robot arm is incorporated into the SAM system to transfer the sample for in-line inspection, which makes the system complex and increases time consumption. The main aim of this study is to develop a novel water probe for the SAM system, that is, a waterstream. During the scanning process, water was supplied using a waterstream inst
