Atomic force microscopy of electrochemical nanoelectrodes.

Nanometer-sized electrodes have recently been used to investigate important chemical and biological systems on the nanoscale. Although nanoelectrodes offer a number of advantages over macroscopic electrochemical probes, visualization of their surfaces remains challenging. Thus, the interpretation of the electrochemical response relies on assumptions about the electrode shape and size prior to the experiment and the changes induced by surface reactions (e.g., electrodeposition). In this paper, we present first AFM images of nanoelectrodes, which provide detailed and unambiguous information abou

Unravelling friction anisotropy by atomic force microscopy.

Friction plays a crucial role in both natural phenomena, from the flow of blood cells to earthquakes, and technological applications, from car engines to wind turbines. One of the most fundamental aspects of tribology is friction anisotropy, that is, the dependence of the friction force vector on the direction of sliding. Even when two identical surfaces slide against each other, differences in sliding direction can lead to significantly different friction forces. While atomistic and semi-empirical models provide a good understanding of friction and friction anisotropy at the molecular and mac

Retina microrheology via oscillatory atomic force microscopy.

Viscoelastic properties of tissues, including elasticity and viscosity, are crucial for understanding development and disease progression. However, traditional atomic force microscopy (AFM) indentation methods provide limited insight into these complex tissue properties. This study establishes microrheology via oscillatory AFM to assess both the elastic and viscous components of tissue mechanics. We first compared indentation AFM to oscillatory AFM on mouse retinal tissue and found that the Young's modulus of indentation AFM (956.8 Pa) was statistically similar to the elastic component (storag

Towards standardization of conductive atomic force microscopy.

This study evaluates the consistency of calibrations across different Conductive Atomic Force Microscopy (C-AFM) systems for resistance measurements and investigates the comparability of nanoscale current measurements. The results demonstrate that, irrespective of the C-AFM instrument, current amplifier, or probe type, calibration of the complete measurement system enables accurate resistance measurements over the range from 1 MΩ to 1 TΩ, with standard uncertainties below 1%. For all participants an agreement at the level of 4.8% was found over the same resistance range. Furthermore, calibrate

Artifacts and Practical Issues in Atomic Force Microscopy.

As with any other microscopic technique, in atomic force microscopy (AFM), artifacts and problems can arise. Some of these happen due to improper use of the microscope by the operator, and some are due to particular characteristics of the sample. Some occur depending on the type of instrument or from probe damage. Some of them are artifacts inherent in the technique. Knowledge of these issues is important for correct data acquisition and interpretation, and in many cases, training in AFM is inadequate. In this chapter, we show examples of common artifacts in AFM and describe, where possible, h

On averaging force curves over heterogeneous surfaces in atomic force microscopy.

Atomic force microscopy (AFM) can be used to study mechanics at the nanoscale. Biological surfaces and nanocomposites have typically heterogeneous surfaces, both mechanically and chemically. When studying such surfaces with AFM, one needs to collect a large amount of data to make statistically sound conclusions. It is time- and resource-consuming to process each force curve separately. The analysis of an averaged raw force data is a simple and time saving option, which also averages out the noise and measurement artifacts of the force curves being analyzed. Moreover, some biomedical applicatio

Atomic force microscopy investigation of vocal fold collagen.

OBJECTIVES: Collagen is an important constituent of the vocal fold extracellular matrix and is necessary for providing tensile strength and maintaining tissue geometry. Traditional investigations of vocal fold collagen using light and electron microscopy do not provide information on the organization and mechanical properties of collagen in native topographic state. The primary objective of this study was to use Atomic Force Microscopy (AFM) to examine the surface characteristics and organization of collagen in the deep layer of the lamina propria at nanoscale resolution. Numerical chromosomal abnormalities detected by atomic force microscopy.

The numerical abnormalities of human metaphase chromosomes, fixed according to standard procedures for optical microscopy but not treated for banding, were detected by atomic force microscopy (AFM). High-resolution AFM imaging of chromosomes in trisomy 13, 21, and Klinefelter syndrome can be compared directly with the traditional optical image. The unbanded metaphase chromosomes, including the extra ones in trisomic patients showed a structural pattern very similar to G-banding. Comparison of AFM images with light microscopic data allows the identification of specific chromosomes, and images o

Robust Reconstruction of Atomic Force Microscopy Tip Shape From Force-Indentation Curves.

A robust method, termed Piecewise Inverse Force Curve Analysis (PIFCA), is introduced for reconstructing the shape of atomic force microscopy (AFM) tips. The approach relies on piecewise linear axisymmetric approximation of the tip shape based on the analysis of force versus indentation depth curves acquired on a soft reference sample. PIFCA was validated using force curves collected on polydimethylsiloxane samples and was able to reconstruct nearly axisymmetric tip shapes with high goodness-of-fit (R2 ≥ 0.98). PIFCA does not require any additional measurements such as electron microscopy or b

Atomic force microscopy imaging of RBL-2H3 cell degranulation.

Understanding the biophysical mechanisms of degranulation is crucial for managing allergic diseases, yet the spatiotemporal coordination of membrane and cytoskeletal dynamics during these reactions remains incompletely understood. In this study, we utilized Atomic Force Microscopy (AFM) to conduct a time-resolved investigation of RBL-2H3 cells during anti-DNP IgE-induced activation. By systematically mapping the cell surface, we quantified phase-specific changes in morphology, surface adhesion, and cortical stiffness (Young's modulus). Our results reveal a striking temporal asynchronicity, wit

Cleaning and hydrophilization of atomic force microscopy silicon probes.

The silicon surface of commercial atomic force microscopy (AFM) probes loses its hydrophilicity by adsorption of airborne and package-released hydrophobic organic contaminants. Cleaning of the probes by acid piranha solution or discharge plasma removes the contaminants and renders very hydrophilic probe surfaces. Time-of-flight secondary-ion mass spectroscopy and X-ray photoelectron spectroscopy investigations showed that the native silicon oxide films on the AFM probe surfaces are completely covered by organic contaminants for the as-received AFM probes, while the cleaning methods effectively

Determination of a translocation chromosome by atomic force microscopy.

Atomic force microscopy (AFM) has been used to study the translocation involving chromosomes 11 and 13. An amniocentesis procedure was performed at 18 weeks of pregnancy on a familial balanced translocation carrier mother whose karyotype was 46,XX,t(11;13) (q23;q34). After harvesting the tissue cultures, light microscopy studies (LM) have indicated that the fetus had the same translocation. A 0.3 microm gap region on the derivative chromosome 13 was determined by AFM; it was equivalent to a mid-sized G-band. The enhanced resolution of AFM with respect to its line measure analysis and three-dim

Imaging Artificial Membranes Using High-Speed Atomic Force Microscopy.

Supported lipid bilayers represent a very attractive way to mimic biological membranes, especially to investigate molecular mechanisms associated with the lateral segregation of membrane components. Observation of these model membranes with high-speed atomic force microscopy (HS-AFM) allows the capture of both topography and dynamics of membrane components, with a spatial resolution in the nanometer range and image capture time of less than 1 s. In this context, we have developed new protocols adapted for HS-AFM to form supported lipid bilayers on small mica disks using the vesicle fusion or L
