Screening of photocatalysts by scanning electrochemical microscopy.

A method for rapid screening of photocatalysts employing a form of scanning electrochemical microscopy (SECM) is described. A piezoelectric dispenser was used to deposit arrays composed of approximately 300-microm-size photocatalyst spots with different compositions onto conducting glass, fluorine-doped tin oxide substrate. The scanning tip of the SECM was replaced by a fiber optic connected to a xenon lamp and was rapidly scanned over the array. In this arrangement, the photocatalytic performance of the spots was evaluated by measuring the photocurrent at the substrate of the array. A fiber o

Scanning electrochemical microscopy of individual catalytic nanoparticles.

Electrochemistry at individual metal nanoparticles (NPs) can provide new insights into their electrocatalytic behavior. Herein, the electrochemical activity of single AuNPs attached to the catalytically inert carbon surface is mapped by using extremely small (≥3 nm radius) polished nanoelectrodes as tips in the scanning electrochemical microscope (SECM). The use of such small probes resulted in the spatial resolution significantly higher than in previously reported electrochemical images. The currents produced by either rapid electron transfer or the electrocatalytic hydrogen evolution reactio

Scanning electrochemical microscopy with slightly recessed nanotips.

Slightly recessed nanoelectrodes were prepared by controlled etching of nanometer-sized, flat Pt electrodes. By using high-frequency (e.g., 2 MHz) ac voltage, the layer of Pt as thin as greater, approximately >3 nm was removed to produce a cylindrical cavity inside the insulating glass sheath. The etched electrodes were characterized by combination of voltammetry and scanning electrochemical microscopy (SECM) to determine the radius and the effective depth of the recess. The theory was developed for current versus distance curves obtained with a recessed tip approaching either a conductive or

Tunneling Mode of Scanning Electrochemical Microscopy: Probing Electrochemical Processes at Single Nanoparticles.

Electrochemical experiments at individual nanoparticles (NPs) can provide new insights into their structure-activity relationships. By using small nanoelectrodes as tips in a scanning electrochemical microscope (SECM), we recently imaged individual surface-bound 10-50 nm metal NPs. Herein, we introduce a new mode of SECM operation based on tunneling between the tip and a nanoparticle immobilized on the insulating surface. The obtained current vs. distance curves show the transition from the conventional feedback response to electron tunneling between the tip and the NP at separation distances

Scanning electrochemical microscopy based irreversible destruction of living cells.

In this research, scanning electrochemical microscopy combined with electrochemical impedance spectroscopy has been applied to irreversible electroporation of active yeast cells by causing cell death. This finding is important for the development of irreversible electroporation technique, which could be suitable for the curing of cancerous tissues, because during this research cell death has been achieved using relatively low ultramicro-electrode (UME) voltage, precisely of 2.0 V vs Ag/AgCl,Cl-sat. It was determined that the irreversibly electroporated area of immobilized yeast cells was locat

Focused-Ion-Beam-Milled Carbon Nanoelectrodes for Scanning Electrochemical Microscopy.

Nanoscale scanning electrochemical microscopy (SECM) has emerged as a powerful electrochemical method that enables the study of interfacial reactions with unprecedentedly high spatial and kinetic resolution. In this work, we develop carbon nanoprobes with high electrochemical reactivity and well-controlled size and geometry based on chemical vapor deposition of carbon in quartz nanopipets. Carbon-filled nanopipets are milled by focused ion beam (FIB) technology to yield a flat disk tip with a thin quartz sheath as confirmed by transmission electron microscopy. The extremely high electroactivit

Menadione metabolism to thiodione in hepatoblastoma by scanning electrochemical microscopy.

The cytotoxicity of menadione on hepatocytes was studied by using the substrate generation/tip collection mode of scanning electrochemical microscopy by exposing the cells to menadione and detecting the menadione-S-glutathione conjugate (thiodione) that is formed during the cellular detoxication process and is exported from the cell by an ATP-dependent pump. This efflux was electrochemically detected and allowed scanning electrochemical microscopy monitoring and imaging of single cells and groups of highly confluent live cells. Based on a constant flux model, approximately 6 x 10(6) molecules

Scanning Electrochemical Microscopy Imaging during Respiratory Burst in Human Cell.

Phagocytic cells, such as neutrophils and monocytes, consume oxygen and generate reactive oxygen species (ROS) in response to external stimuli. Among the various ROS, the superoxide anion radical is known to be primarily produced by nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) oxidase. In the current study, we attempt to evaluate the respiratory burst by monitoring the rapid consumption of oxygen by using scanning electrochemical microscopy (SECM) imaging. The respiratory burst was measured in a human monocytic cell line (THP-1 cells) derived from an acute monocytic leukemia pa

Photo-Scanning Electrochemical Microscopy on the Nanoscale with Through-Tip Illumination.

Scanning electrochemical microscopy (SECM) has previously been employed in probing photoelectrochemical processes at semiconductor surfaces. However, the spatial resolution of these studies has not yet matched the nanoscale SECM resolution attained without substrate illumination. Herein, we introduce nanoscale photo-SECM with a glass-sealed, polished tip simultaneously serving as a nanoelectrode and a light guide to produce a microscopic light spot on the substrate surface. The advantages of this approach are demonstrated by comparing current transients obtained using through-tip and global il

TEM-Assisted Fabrication of Sub-10 nm Scanning Electrochemical Microscopy Tips.

High-resolution scanning electrochemical microscopy (SECM) is a powerful technique for mapping surface topography and reactivity on the nanoscale and investigating heterogeneous processes at the level of single nanoparticles. The ability to fabricate ultrasmall nanoelectrode tips is critical for the progress in nano-SECM. Despite long-term efforts to improve previously developed procedures, the preparation and characterization of disk-type polished tips with the radius <∼25 nm remains challenging and unpredictable. One of the problems is that the geometry of such tips is hard to characterize b

Scanning electrochemical microscopy imaging of rhodochrosite dissolution using gold amalgam microelectrodes.

Gold/mercury amalgam (Au/Hg) microelectrodes with a diameter of 25 microm were developed for the detection of environmentally relevant analytes such as manganese and iron by scanning electrochemical microscopy (SECM), and applied to investigate the controlled dissolution of manganese carbonate (MnCO(3); rhodochrosite) in acidic conditions. Characterization of the amalgam electrode geometry via approach curves recorded during SECM experiments revealed Au/Hg microelectrodes with sphere cap geometry. Quantitative determination of Mn(2+) has been achieved by calibration of the Au/Hg microelectrode

Electrochemical Size Measurement and Characterization of Electrodeposited Platinum Nanoparticles at Nanometer Resolution with Scanning Electrochemical Microscopy.

The properties of nanoparticles (NPs) are determined by their size and geometric structures. A reliable determination of NP dimension is critical for understanding their physical and chemical properties, but sizing ultrasmall particles on the order of nanometer (nm) scale in the solution is still challenging. Here, we report the size measurement of PtNP at nanometer resolution by in situ scanning electrochemical microscopy (SECM), performed with the electrochemical generation and removal of H2 bubble at a reasonably small distance between tip and substrate electrodes in 200 or 500 mM HClO4 sol

Scanning electrochemical microscopy in the 21st century. Update 1: Five years after.

This Perspective is an update to our more extensive survey of scanning electrochemical microscopy (SECM) published in 2007. During this time, the SECM field retained its momentum by expanding into new areas and meeting the emerging scientific and technological challenges. Here we focus on most prominent developments such as high-resolution imaging, investigation of structures and processes on the nanoscale, alternative energy applications, and new approaches to solving "real world" problems. The fabrication of novel SECM probes and related theoretical advances are also discussed.

High-resolution studies of photo(electro)catalysts by electrochemical scanning probe microscopy.

Photocatalytic processes are central to many energy and environmental applications; however, mechanistic understanding of these processes is incomplete. Traditional techniques provide bulk-averaged data, missing crucial activity variations between and within individual photocatalyst particles, which are challenging to probe due to their small size. Scanning electrochemical microscopy (SECM) and related techniques provide a unique spatially resolved perspective, enabling deeper insights into photocatalyst performance at the microscale and the nanoscale. In this article, we review applications o

Scanning Electrochemical Microscopy of Single Spherical Nanoparticles: Theory and Particle Size Evaluation.

Experiments at individual metal nanoparticles (NPs) can provide important information about their electrochemical and catalytic properties. The scanning electrochemical microscope (SECM) equipped with a nanometer-sized tip was recently used to image single 10 or 20 nm gold particles and quantitatively investigate electrochemical reactions occurring at their surfaces. In this Article, the theory is developed for SECM current vs distance curves obtained with a disk-shaped tip approaching a comparably sized, surface-bound conductive or insulating spherical NP. The possibility of evaluating the si

Beyond Imaging: Localized Synthesis, Fabrication, and Surface Modification via Scanning Electrochemical Cell Microscopy.

Meniscus-confined electrochemical deposition (MCED) enables highly localized material synthesis, transformation, and surface modification. The Perspective highlights scanning electrochemical cell microscopy (SECCM) as a highly versatile MCED platform. While primarily developed for electrochemical imaging, SECCM also functions as a direct-write tool capable of producing complex architectures and probing associated localized electrochemical phenomena. We focus on a concise technical overview to position SECCM within a broader MCED landscape. We then examine its application across key material pr

Impact of Droplet Wettability on Scanning Electrochemical Cell Microscopy Performance in Stainless Steels.

Variations in droplet wettability affect localized corrosion during scanning electrochemical cell microscopy (SECCM) on stainless steel. The droplet dynamics are influenced by stainless-steel microstructural features and surface conditions-such as surface roughness, inclusions, and the addition of an oil layer. As opposed to previous work on aluminum alloys, droplet spreading is promoted by oil immersion, which leads to an increase in the cathodic currents. Rougher surfaces hinder droplet spreading, largely due to the droplet pinning effect, and exhibit higher pitting corrosion incidences comp

Surface interrogation of advanced electrocatalysts by scanning electrochemical microscopy: fundamentals, progress and perspectives.

In situ quantitative analysis of electrode surfaces, particularly the identification of active sites and reaction intermediates, is essential for elucidating structure-activity relationships and understanding the mechanisms underlying electrocatalytic reactions. Although conventional spectroscopic techniques are widely used to probe catalytic intermediates, they often face limitations in sensitivity, spatial and temporal resolution, and the ability to capture transient phenomena. These constraints hinder the precise quantification of catalytically active sites and the accurate assessment of el

Selective Initiation of Corrosion Pits in Stainless Steel Using Scanning Electrochemical Cell Microscopy.

Scanning electrochemical cell microscopy is a useful technique for determining variations in corrosion behavior across a surface. However, the numerous options for experimental parameters and little understanding of their effect on the corroding system render comparisons of results between studies difficult. Herein, we explore changes in corrosion behavior of two martensitic stainless steels, a cast CA6NM and a wrought S41500, as a result of the chosen experimental parameters, including scan rate, approach potential, surface oil immersion, and tip aperture diameter. The study demonstrates that
