Deep-ultraviolet microscopy reveals biomolecular spatiotemporal intracellular dynamics.

Intracellular dynamics span a broad range of time scales and biomolecular processes, offering insights into cell health, functional state, phenotype, and response to external perturbations. Several label-free optical imaging approaches have been used to capture intracellular dynamics but are limited by spatiotemporal resolution and biomolecular specificity required to distinguish unique subcellular and metabolic processes. In this work, we demonstrate deep-ultraviolet (UV) microscopy as a powerful, label-free, high-resolution approach for quantifying multiscale intracellular dynamics with biom

Enhancing the pathological diagnosis of oral cancers using broadband ultraviolet photoacoustic microscopy.

Conventional histopathological diagnostic methods, such as hematoxylin-eosin (H&E) staining and immunohistochemistry (IHC), face significant challenges in intraoperatively diagnosing early-stage oral cancer and certain cancer types. To address this, we propose a novel pathological diagnostic method based on a broadband ultraviolet photoacoustic microscope (bUV-PAM). This method capitalizes on the unique optical absorption properties of nuclei and high-frequency photoacoustic spectral variations, enabling the simultaneous performance of photoacoustic morphological imaging (PAMI) and photoacoust

Microscopy with ultraviolet surface excitation for wide-area pathology of breast surgical margins.

Intraoperative assessment of breast surgical margins will be of value for reducing the rate of re-excision surgeries for lumpectomy patients. While frozen-section histology is used for intraoperative guidance of certain cancers, it provides limited sampling of the margin surface (typically <1  %   of the margin) and is inferior to gold-standard histology, especially for fatty tissues that do not freeze well, such as breast specimens. Microscopy with ultraviolet surface excitation (MUSE) is a nondestructive superficial optical-sectioning technique that has the potential to enable rapid, high-re

Reflectance confocal microscopy: an effective tool for monitoring ultraviolet B phototherapy in psoriasis.

BACKGROUND: In vivo reflectance confocal microscopy (RCM) is a novel, noninvasive imaging technique which enables imaging of skin at a cellular resolution comparable to conventional microscopy. OBJECTIVES: We performed a pilot study to evaluate RCM as a noninvasive tool for monitoring ultraviolet (UV) B phototherapy in psoriasis. METHODS: In six patients with psoriasis, lesional and nonlesional skin was selected for RCM imaging using a standardized protocol. Well-known histological features of psoriasis were visualized: parakeratosis, acanthosis, agranulosis, papillomatosis, presence of epid

A Label-Free Ultraviolet Photoacoustic Microscopy Enables Nuclear Imaging and Toxicity Assessment in an Intact Brain Organoid.

Fetal alcohol spectrum disorders (FASDs) are caused by prenatal exposure to ethanol (EtOH), leading to developmental brain abnormalities. Cortical organoids derived from human-induced pluripotent stem cells provide physiologically relevant models to study such neurotoxic effects. However, accurate imaging of nuclear morphology, a key indicator of cytotoxicity and developmental impairment, remains difficult. Traditional fluorescence-based techniques rely on staining and sectioning, limiting throughput and potentially altering native structures. Herein, we present an ultraviolet photoacoustic mi

Shedding new light on Cyclospora: how the use of ultraviolet fluorescence microscopy can improve diagnosis of cyclosporiasis.

UNLABELLED: Cyclosporiasis has historically been underdiagnosed due to a lack of clinical recognition and challenges in laboratory detection. Microscopic detection of Cyclospora requires either the modified Kinyoun's acid-fast or safranin stains, which are not part of the standard ova-and-parasite examination. Alternatively, the use of fluorescence microscopy for the detection of Cyclospora due to the organism's autofluorescent properties has been an available tool in the field of clinical parasitology. However, it is unknown how ultraviolet (UV) fluorescence performs compared to permanent sta

Label-free ultraviolet photoacoustic microscopy for multiscale structural phenotyping of epithelial morphogenesis in organoid-derived intestinal epithelial wrinkle tissue.

Mechanical wrinkling of epithelial tissues encodes key features of tissue morphogenesis and mechanotransductive responses, yet its quantitative, multiscale characterization remains challenging in organoid-based in vitro models due to reliance on fluorescence labelling and sectioning. Here, we develop label-free ultraviolet photoacoustic microscopy (UV-PAM) for multiscale imaging and quantitative analysis of human organoid-derived epithelial wrinkle tissues. By exploiting intrinsic nucleic-acid absorption at 266 nm, UV-PAM visualizes wrinkle structures and nuclear morphology without labeling or

Efficacy of a Daily Protective Moisturizer with High UVB and UVA Photoprotection in Decreasing Ultraviolet Damage: Evaluation by Reflectance Confocal Microscopy.

Patients with photodermatoses or actinic keratosis benefit from very high ultraviolet B-ultraviolet A (UVB-UVA) photoprotection. However, poor compliance is an issue that jeopardizes adequate protection, leading to disease recurrence. This study evaluated the efficacy of a daily protective moisturizer with high UVB and UVA photoprotection applied 8 h before irradiation. A monocentric, open-label, prospective, control pilot study was performed including 10 patients. Patients were irradiated with UVB and UVA before and 8 h after topical application of the product. NerveTracker: a Python-based software toolkit for visualizing and tracking groups of nerve fibers in serial block-face microscopy with ultraviolet surface excitation images.

SIGNIFICANCE: Information about the spatial organization of fibers within a nerve is crucial to our understanding of nerve anatomy and its response to neuromodulation therapies. A serial block-face microscopy method [three-dimensional microscopy with ultraviolet surface excitation (3D-MUSE)] has been developed to image nerves over extended depths ex vivo. To routinely visualize and track nerve fibers in these datasets, a dedicated and customizable software tool is required. AIM: Our objective was to develop custom software that includes image processing and visualization methods to perform mi

Ultrastructure study of hair damage after ultraviolet irradiation.

BACKGROUND: Natural ultraviolet exposure induces hair damage, which is difficult to avoid. Most of the research work is focused on the effect of ultraviolet on the epidermis, dermis as well as the immune system, whereas the long-term effect of ultraviolet on hair has not been investigated. AIM: we performed our experiment to find out the changes induced in hair follicle and shaft in those patients exposed to high doses of ultraviolet (A and B) during treatment of other skin conditions. Ultraviolet irradiation induces apoptosis in human immature, but not in skin mast cells.

As diverse pruritic cutaneous diseases respond to ultraviolet treatment, we have examined whether ultraviolet light is capable of inducing apoptosis in mast cells. Human mast cell line 1 (HMC1) derived from a patient with malignant mastocytosis and purified skin mast cells were irradiated with single doses of ultraviolet B or ultraviolet A1, or pretreated with 8-methoxypsoralen prior to ultraviolet A1 exposure. After 0 to 48 h of incubation, the percentage of apoptotic and dead cells was assessed. In HMC1 cells, morphologic features of apoptosis were further evaluated by electron microscopy. A

Intravital and electron microscopic observation of Ito cells in rat hepatic microcirculation.

Intralobular distribution of Ito cells (fat-storing cells) in hepatic microcirculatory units was investigated through intravital fluorescence microscopy. By using a low-level ultraviolet epi-illumination and a sensitive silicon intensified target camera, the intrahepatic autofluorescence was visualized and digitally processed. In rats fed an ordinary diet, the ultraviolet-excited autofluorescence was composed of at least two different origins that could not be spectrophotometrically distinguished, namely, multiple patchy autofluorescence activities along the sinusoids and terminal hepatic venu

Topical treatment with liposomes containing T4 endonuclease V protects human skin in vivo from ultraviolet-induced upregulation of interleukin-10 and tumor necrosis factor-alpha.

Exposing human skin to ultraviolet radiation causes DNA damage, sunburn, immune alterations, and eventually, skin cancer. We wished to determine whether liposomes containing a DNA repair enzyme could prevent any of the acute effects of irradiation when applied after ultraviolet exposure. Fifteen human patients with a prior history of skin cancer were exposed to two minimal erythema doses of ultraviolet radiation on their buttock skin. Liposomes containing T4 endonuclease V or heat-inactivated enzyme were applied immediately and at 2, 4, and 5 h after ultraviolet irradiation. Transmission elect
