The structure of endometrial microvessels.

Recent studies have provided substantial evidence to highlight abnormalities in the structure of endometrial microvessels in users of progestogen-only contraception. Structural changes alone are unlikely to lead to breakthrough bleeding, but appear to be associated with a reduction in vessel integrity, and may reflect alterations in the control and growth of endometrial microvessels in those exposed to exogenous progestogens. In users of low-dose progestogens, immunohistochemical studies have demonstrated changes in superficial endometrial vascular morphology, density and in endometrial migrat

Structural changes in Alzheimer's disease brain microvessels.

Brain microvascular alterations are thought to contribute to the development of stroke and dementia. Structural changes in capillaries of elderly patients correlate positively with advanced age and dementia. The objective of this study is to use laser-induced fluorescence spectroscopy to compare structural (collagen content) and functional (apoptosis) parameters in brain tissues and isolated vessels of AD patients to age-matched controls. Our results show significantly higher fluorescent labeling for apoptosis in AD vessels compared to controls. Also, there is significantly higher autofluoresc

Electron microscopy of amyloid fibrils and microvessels.

Senile plaques and microvessels in the cortices of brains with Alzheimer's disease were examined using serial sections under light and electron microscopes. In addition, observations were carried out by using an immunostaining method. The results obtained were as follows: (1) Every senile, plaque contains some amyloid fibrils, and many senile plaques have degenerated capillaries with amyloid fibrils. (2) Amyloid fibrils without senile plaques continue directly to the capillaries. (3) A considerable number of preamyloids are observed surrounding the capillaries when a methenamine silver stain i

Microvessel Rupture and Intraplaque Hemorrhage.

BACKGROUND: Intraplaque hemorrhage (IPH) is an important mechanism of plaque progression and destabilization, primarily attributed to disruption of fragile intraplaque microvessels; however, in vivo demonstration of this process remains limited. CASE SUMMARY: A 56-year-old woman presented with unstable angina. Coronary computed tomography angiography demonstrated a noncalcified, low-attenuation plaque with positive remodeling in the proximal right coronary artery. Optical coherence tomography (OCT) revealed intraplaque microvessels, a crescent-shaped hypointense plaque region consistent with

Reinforcing endothelial junctions prevents microvessel permeability increase and tumor cell adhesion in microvessels in vivo.

Tumor cell adhesion to the microvessel wall is a critical step during tumor metastasis. Vascular endothelial growth factor (VEGF), a secretion of tumor cells, can increase microvessel permeability and tumor cell adhesion in the microvessel. To test the hypothesis that inhibiting permeability increase can reduce tumor cell adhesion, we used in vivo fluorescence microscopy to measure both microvessel permeability and adhesion rates of human mammary carcinoma MDA-MB-231 cells in post-capillary venules of rat mesentery under the treatment of VEGF and a cAMP analog, 8-bromo-cAMP, which can decrease

Shock associated with endothelial dysfunction in omental microvessels.

BACKGROUND: Impaired microvascular function leads to a poor outcome in a variety of medical conditions. Our aim was to determine whether vasodilator responses to acetylcholine (Ach) are impaired in human omental arterioles from patients with severe trauma. MATERIALS AND METHODS: Patients with massive blood loss and severe shock requiring damage control procedures were included. Tissues were collected at the first (FEL) and the second explorative laparotomy (SEL). Control tissues were collected from nontrauma patients. Freshly isolated 50-200-μm-diameter omental arterioles were analysed using

Isolation of villous microvessels from the human placenta.

A method for isolating the microvessels of the human placental villi has been developed in order to culture perivascular cells. It consists of an initial selection of the villi by serial sieving. The villi retained by the 75 microns sieve were digested by collagenase-dispase. A Percoll gradient permitted the isolation of microvessels still surrounded by stromal fibres and cells. Another digestion by collagenase-dispase eliminated the contaminant elements and allowed, after a new Percoll gradient, microvessels with endothelium, basement membrane and a few perivascular cells to be obtained. Each

Permeability of intestinal microvessels in chronic arterial hypertension.

We studied changes in intestinal microvascular permeability resulting from chronic arterial hypertension. Normotensive dogs and dogs made chronically hypertensive utilizing the one-kidney, one clip Goldblatt technique were used to obtain values for: arterial pressure, portal pressure, intestinal lymph flow, and the lymph-to-plasma protein concentration ratio (CL/Cp). Values for the normotensive dogs were 111 mm Hg, 7.1 mm Hg, 6.2 ml/hr, and 0.64, respectively, while values for the chronically hypertensive dogs were 165 mm Hg, 7.3 mm Hg, 12.5 ml/hr, and 0.66, respectively. Control lymph flow in

Bioengineered 3D Microvessels for Investigating Plasmodium falciparum Pathogenesis.

Plasmodium falciparum pathogenesis is complex and intimately connected to vascular physiology. This is exemplified by cerebral malaria (CM), a neurovascular complication that accounts for most of the malaria deaths worldwide. P. falciparum sequestration in the brain microvasculature is a hallmark of CM and is not replicated in animal models. Numerous aspects of the disease are challenging to fully understand from clinical studies, such as parasite binding tropism or causal pathways in blood-brain barrier breakdown. Recent bioengineering approaches allow for the generation of 3D microvessels an

Plasma expanders stabilize human microvessels in microfluidic scaffolds.

Plasma expanders such as dextran and hydroxyethyl starch (HES) are important components of solutions designed to maintain vascular volume in the clinical setting and to preserve organs ex vivo before transplantation. Here, we show that these polymers also exert stabilizing effects on engineered microvessels in microfluidic type I collagen and fibrin scaffolds. Standard growth media, which did not contain dextran or HES, led to severe leakage, vascular collapse, and catastrophic failure of perfusion. Remarkably, vessels that were provided with 3% dextran or 5% HES had few focal leaks, maintaine

Noninvasive visualization of microvessels using diffraction enhanced imaging.

The architecture of the vascular network is crucial because almost every disease alters the morphology of the blood vessel. Although existing imaging technique can provide vessel images, microvessels will not be discerned without the use of contrast agents. Furthermore, conventional imaging technique can only provide the outline of the vessels due to the restriction of spatial resolution. In this study, a novel imaging technique, diffraction enhanced imaging (DEI), is applied to the observation of microvessels in mouse hepatic fibrosis samples. With high spatial resolution and contrast of biol

Interaction of angiogenic microvessels with the extracellular matrix.

The extracellular matrix (ECM) plays a critical role in angiogenesis by providing biochemical and positional cues, as well as by mechanically influencing microvessel cell behavior. Considerable information is known concerning the biochemical cues relevant to angiogenesis, but less is known about the mechanical dynamics during active angiogenesis. The objective of this study was to characterize changes in the material properties of a simple angiogenic tissue before and during angiogenesis. During sprouting, there was an overall decrease in tissue stiffness followed by an increase during neovess

Radiation-Induced Macrovessel/Microvessel Disease.

Radiation therapy (RT) is a cornerstone in cancer treatment (used in 50% of cases), yet challenges persist because damage to normal tissue through direct impact of radiation or bystander effects is inevitable. Injury of macrovessels by RT manifests as obstructive disease, which is akin to atherosclerotic disease. Historically observed in coronary arteries of patients treated for breast cancer and lymphoma, it also affects patients receiving contemporary therapy for lung and chest cancers. Moreover, radiation at various sites can lead to peripheral vascular disease. An aspect of radiation-induc

The microvessel density of the hypospadiac prepuce in children.

OBJECTIVE: The blood supply of the hypospadiac prepuce is crucial for success in surgery. Although the vascular anatomy of the hypospadiac prepuce has been previously documented, data on microvessel density are missing. It was aimed to document the microvessel density of the normal and hypospadiac prepuce. METHODS: After ethical approval of the study, prepuces from 24 children with hypospadias undergoing surgical repair and from 9 healthy age-matched controls were stained by immunohistochemical methods using panendothelial cell antigen CD31 in order to assay their microvessel density.
