Faulkes et al. recently showed that naked mole-rats (NMRs) have a very distinctive cardiac gene expression profile among other African mole-rats, as well as metabolic variations that result from their chronic exposure to a hypoxic environment. These adaptations might underlie their resistance to cardiac ischemic injuries.

BACKGROUND: Naked mole-rats (Heterocephalus glaber, NMRs) and blind mole-rats (Spalax galili, BMRs) are representative subterranean rodents that have evolved many extraordinary traits, including hypoxia tolerance, longevity, and cancer resistance. Although multiple candidate loci responsible for these traits have been uncovered by genomic studies, many of them are limited to functional changes to amino acid sequence and little is known about the contributions of other genetic events. To address this issue, we focused on gene losses (unitary pseudogenes) and systematically analyzed gene losses

Naked mole rats are a long-lived animal model that age much like humans, but that can also withstand oxidative damage, cancer, neurodegenerative diseases, and severe hypoxic conditions, which is of particular interest to this study. The conditions of their underground burrows result in competition for oxygen consumption, yet despite this oxygen deprivation they emerge unscathed. To understand the mechanisms in place to facilitate neuronal preservation during hypoxia, we investigated the protein levels of well-known cell-stress factors. We found that under hypoxic conditions, nearly half of the

Life underground often leads to animals having specialized auditory systems to accommodate the constraints of acoustic transmission in tunnels. Despite living underground, naked mole-rats use a highly vocal communication system, implying that they rely on central auditory processing. However, little is known about these animals' central auditory system, and whether it follows a similar developmental time course as other rodents. Naked mole-rats show slowed development in the hippocampus suggesting they have altered brain development compared to other rodents. Here, we measured morphological ch

Hypoxia is one of the strongest environmental drivers of cellular and physiological adaptation. Although most mammals are largely intolerant of hypoxia, some specialized species have evolved mitigative strategies to tolerate hypoxic niches. Among the most hypoxia-tolerant mammals are naked mole-rats (Heterocephalus glaber), a eusocial species of subterranean rodent native to eastern Africa. In hypoxia, naked mole-rats maintain consciousness and remain active despite a robust and rapid suppression of metabolic rate, which is mediated by numerous behavioural, physiological and cellular strategie

Mole-rats are known to use their protruding, chisel-like incisors to dig underground networks of tunnels, but it remains unknown how these incisors are used to break and displace the soil. Theoretically, different excavation strategies can be used. Mole-rats could either use their head depressor muscles to power scooping motions of the upper incisors (by nose-down head rotations) or the lower incisors (by nose-up head rotations), or their jaw adductors to grab and break the soil after penetrating both sets of incisors into the ground, or a combination of these mechanisms. To identify how chise

Renowned for extreme longevity and extraordinary cancer resistance, the naked mole-rat ( Heterocephalus glaber) serves as a powerful biological model to uncover novel healthy aging mechanisms. Fourteen hallmarks have been proposed to describe the core features of biological aging; however, whether their direct enhancement contributes to longevity remains unclear. Here, we summarize current knowledge on the differential regulation of these aging hallmarks in the naked mole-rat and their potential roles in its exceptional lifespan. We also discuss current research limitations and suggest future

Naked mole-rats share some sensory characteristics with other subterraneans, including lack of object vision, retention of the ability to entrain their circadian rhythm to light, and poor hearing. On the other hand, a characteristic that may be specialized in the naked mole-rat is their exquisite orienting responses to the touch of even a single body vibrissa. They have about 100 whisker-like body vibrissae on their otherwise furless bodies. They are also insensitive to chemical and inflammatory pain, likely an adaptation to living in an atmosphere that is high in carbon dioxide, a result of m

Studies on mammalian species with naturally reduced eyes can provide valuable insights into the evolutionary developmental mechanisms underlying the reduction of the eye structures. Because few naturally microphthalmic animals have been studied and eye reduction must have evolved independently in many of the modern groups, novel evolutionary developmental models for eye research have to be sought. Here, we present a first report on embryonic eye development in the Cape dune mole rat, Bathyergus suillus. The eyes of these animals contain all the internal structures characteristic of the normal

Osteoarthritis (OA) is the most prevalent disabling disease, affecting quality of life and contributing to morbidity, particularly during aging. Current treatments for OA are limited to palliation: pain management and surgery for end-stage disease. Innovative approaches and animal models are needed to develop curative treatments for OA. Here, we investigated the naked mole-rat (NMR) as a potential model of OA resistance. NMR is a small rodent with the maximum lifespan of over 30 years, resistant to a wide range of age-related diseases. NMR tissues accumulate large quantities of unique, very hi

Naked mole-rats are among the few mammals with the ability to endure severe hypoxia. These unique rodents use metabolic rate depression along with various molecular mechanisms to successfully overcome the challenges of oxygen-limitation, which they experience in their underground borrows. While studies have reported that naked mole-rats exhibit inherently higher levels of oxidative damage across their lifespan as compared to mice, it has yet to be determined whether naked mole-rats are vulnerable to oxidative damage during periods of low oxygen exposure. To investigate this phenomenon, we exam

Environmental hypoxia imposes severe constraints on aerobic metabolism, yet the naked mole-rat (Heterocephalus glaber) achieves remarkable resilience against low oxygen stress through profound metabolic suppression and substrate flexibility. To investigate the molecular basis of glucose regulation under low oxygen, we examined the insulin and insulin-like growth factor (IGF) signaling pathways across developmental stages and social castes. Juvenile, subordinate adult, and queen NMRs were exposed to 1 h of normoxia (21% O₂) or acute hypoxia (3% O₂), and tissues were analyzed for the expression

Mole-rats are subterranean rodents with sensory and physiological adaptations to life in underground tunnels, yet their neurobiology remains largely unexplored. We present methods for in vivo electrophysiological recordings in awake, head-fixed, and freely moving African mole-rats (Fukomys anselli and Fukomys micklemi), addressing their unique anatomy and physiology. We report anesthesia, analgesia, and surgical procedures and find that elevated carbon dioxide concentrations are beneficial for recovery, likely due to a mutation in the potassium-chloride symporter KCC2. We also present a stereo

Extreme environments are powerful drivers of physiological adaptation. Naked mole-rats offer an informative example of this relationship as they putatively encounter intermittent hypoxia and hypercapnia in their subterranean habitat. This has presumably driven the evolution of a suite of cellular and physiological adaptations that enable life in these conditions. Recently, my laboratory and others have begun to examine physiological responses to environmental hypoxia and hypercapnia in naked mole-rats, and the underlying cellular and molecular mechanisms that differentiate the responses of thi

Naked mole-rats are extremely tolerant to low concentrations of oxygen (hypoxia) and high concentrations of carbon dioxide (hypercapnia), which is consistent with the environment that they inhabit. Naked mole-rats combine subterranean living with living in very densely populated colonies where oxygen becomes depleted and carbon dioxide accumulates. In the laboratory, naked mole-rats fully recover from 5 h exposure to 5% O2 and 5 h exposure to 80% CO2, whereas both conditions are rapidly lethal to similarly sized laboratory mice. During anoxia (0% O2) naked mole-rats enter a suspended animation

Social interactions rely on the integration of information across multiple sensory modalities. In many species, individuals recognize conspecifics through combinations of auditory, olfactory, tactile, and visual cues, reflecting the capacity of neural systems to integrate diverse streams of sensory input. The naked mole-rat (Heterocephalus glaber) provides a powerful model system for understanding how social living shapes the evolution of such communication systems, particularly under conditions where visual information is largely absent. Naked mole-rats represent a rare example of mammalian e

A variety of rodents have been used as experimental animals in metabolic studies of plasma lipids and lipoproteins. These studies have included understanding the functional role of apolipoprotein A-I, the major protein on the surface of HDL. Reviewing the genomic database for entries for rodent apoA-I genes, it was discovered that the naked mole-rat (Heterocephalus glaber) gene encoded a protein with a cysteine at residue 28. Previously, two cases have been reported in which human heterozygotes had apoA-I with cysteine at residues 173 (apoA-I Milano) or at 151 (apoA-I Paris). Interestingly, bo

Pedomorphy, maintenance of juvenile traits throughout life, is most pronounced in extraordinarily long-lived naked mole-rats. Many of these traits (e.g., slow growth rates, low hormone levels, and delayed sexual maturity) are shared with spontaneously mutated, long-lived dwarf mice. Although some youthful traits likely evolved as adaptations to subterranean habitats (e.g., thermolability), the nature of these intrinsic pedomorphic features may also contribute to their prolonged youthfulness, longevity, and healthspan.

Bite force is a simple trait indicating an animal's performance related to foraging, social dominance, and defence, all of which influence individual reproductive success. We examine the effect of breeding status on bite force in four social species of Fukomys, a genus of subterranean African rodents (Bathyergidae). These species are cooperative breeders, where reproduction is limited typically to a breeding pair. We collected in vivo bite force data, head width, and upper incisors width from 404 individuals from 75 families and tested whether breeders exhibit stronger bite force. We reveal th
