Methods in molecular biology (Clifton, N.J.)2017eng
Advanced tools for cell imaging are of particular interest as they can detect, localize and quantify molecular targets like abnormal glycosylation sites that are biomarkers of cancer and infection. Targeting these biomarkers is often challenging due to a lack of receptor materials. Molecularly imprinted polymers (MIPs) are promising artificial receptors; they can be tailored to bind targets specifically, be labeled easily, and are physically and chemically stable. Herein, we demonstrate the application of MIPs as artificial antibodies for selective labeling and imaging of cellular targets, on

The abuse and residues of antibiotics have a great impact on the environment and organisms, and their determination has become very important. Due to their low contents, varieties and complex matrices, effective recognition, separation and enrichment are usually required prior to determination. Molecularly imprinted polymers (MIPs), a kind of highly selective polymer prepared via molecular imprinting technology (MIT), are used widely in the analytical detection of antibiotics, as adsorbents of solid-phase extraction (SPE) and as recognition elements of sensors. Herein, recent advances in MIPs

Normally, antibodies against influenza A have been prepared from viable virus or an engineered strain in certain hosts or cultured media. Two factors concerning antibody production are obvious. The obtaining antibody that is a kind of biomolecule has to be handled carefully, e.g., to be kept in a refrigerator. Furthermore, when the virus strain is highly pathogenic, such as H5N1, antibody production has to be done carefully in a high-level biosafety lab. Here, we show how to produce an antibody against H5N1 from a polymeric material using inactivated virus which can be conducted in a low-level

The therapeutic efficacy of cancer therapeutics is frequently limited by poor tumour selectivity, systemic toxicity, and the emergence of drug resistance, underscoring the need for advanced nanoscale drug delivery systems (DDSs) capable of precise molecular targeting and controlled release. Molecularly imprinted polymers (MIPs) have emerged as a promising class of synthetic nanocarriers that combine highly selective and programmable molecular recognition with the robustness, tunability, and scalability of polymeric materials. This review examines recent advances in the nanoscale engineering of

Molecularly imprinted polymers (MIPs) are biomimetic counterparts of biomacromolecules that are synthesized by templating the target within a polymeric network. While the affinity of MIPs can compete with that of antibodies, the specific activity of natural enzymes remains superior to that of catalytically active MIPs. Although the mechanisms underlying the generation of antibodies and MIPs are fundamentally different, important principles have been transferred from biology to artificial binders. Templating a fragment of the target instead of the entire molecule or particle simplifies MIP synt

Imprinting polymerization is a flexible method to make resins specific for different compounds. Imprinting polymerization involves the polymerization of the resin in the presence of a template, here cadmium ions or arsenate. The template is then removed by washing, leaving specific binding sites in the resin. In water treatment, the removal of toxic metal ions is difficult due to the limited affinity of these ions to ion exchange resins. Imprinting polymerization of ion-exchange resins is used to develop resins with high capacity and some selectivity for cadmium ions or arsenate for water trea

Xylazine adulterated Fentanyl is considered one of the deadliest drug threats ever faced in the United States. However, due to the complexity of the illegal drug supply, xylazine detection in the frame of drug-checking (harm reduction) remains an unsolved challenge, one which has cost numerous lives across North America. In this work, we present two sensor platforms for detecting xylazine in drug-checking applications. Both platforms are based on molecularly imprinted polymers (MIPs) using vinyl phosphonic acid as the main monomer. By combining the MIPs with thermometric and colorimetric reado

Polyaniline (PANI)-based molecularly imprinted polymers were investigated for their efficacy in sensing phenylalanine (Phe) when fabricated on both glassy carbon electrode (GCE) and indium tin oxide (ITO) sheets. This study highlights the superior performance of PANI-MIP/ITO over PANI-MIP/GCE for sensing Phe, with clear and distinct redox responses. Molecular computation helps to understand the interaction mechanism between PANI and Phe, where molecular crowding, aggregated clusters, hydrogen bonding, and π-π stacking facilitate stable interactions. We tested the specificity of Phe sensing by

The extensive use of antibiotics and their persistence in the environment make them emerging pollutants of global concern, posing serious risks to ecosystems and public health. Among them, the broad-spectrum fluoroquinolone antibiotic levofloxacin (LEV) is widely prescribed for bacterial infections and has frequently been detected in freshwater systems and environmental matrices. Its presence is linked to the development of antibiotic resistance and ecological toxicity, underscoring the urgent need for efficient removal strategies. In this study, levofloxacin-imprinted polymers (LEV-MIPs) were

Synthetic cannabinoids (SCs), a prominent class of new psychoactive substances, pose growing challenges to public health due to their severe toxic effects and widespread global presence. In this study, we employed computational methods to develop molecularly imprinted polymers (MIPs) for the selective recognition of seven SCs, chosen based on seizure reports from the Narcotics Examination Unit of the Scientific Police of the State of São Paulo. Density functional theory and extended tight binding for geometry, frequency, and noncovalent model 2 (GFN2-xTB) calculations were used to optimize the

The use of molecularly imprinted polymers (MIPs) in chemical and bioanalytical applications has been gaining in interest in recent years. Compared to their biological receptor counterparts, MIPs are easy to prepare, have long shelf stability and can be used under a variety of harsh conditions. The majority of MIPs currently used are produced by traditional free radical polymerization. One drawback with the use of standard free radical initiators is that little control can be exerted over the chemical processes that form the final imprinted cavities. In this study we set out to investigate the

Molecularly imprinted polymers (MIPs) have emerged as powerful synthetic receptors capable of recognizing specific (bio)molecules through tailor-made binding sites. Traditionally based on poly(methyl acrylates) and hydrogen bonding, their extension to π-conjugated systems offers new opportunities for functional integration. In this work, we explore this avenue by combining a plasmonic platform composed of gold nanoparticle dimers fabricated by electron-beam lithography with a PEDOT-based imprinted polymer. The polymer growth is preferentially confined within the nanogaps of the dimers, forming

Depression and anxiety are two common mental health issues that require serious attention, as they have significant impacts on human well-being, with both being emotionally and physically reflected in the increasing number of suicide cases globally. The World Health Organization (WHO) estimated that about 322 million people around the world experienced mental illnesses in 2017, and this number continues to increase. Cortisol is a major stress-controlled hormone that is regulated by the hypothalamic-pituitary-adrenal (HPA) axis. The HPA axis has three main components, including the hypothalamus

Molecularly imprinted polymers (MIPs) have numerous applications in the sensing field, the detection/recognition of virus, the structure determination of proteins, drug delivery, artificial/biomimetic antibodies, drug discovery, and cell culturing. There are lots of conventional methods routinely deployed for the analysis/detection of viral infections and pathogenic viruses, namely enzyme immunoassays, immunofluorescence microscopy, polymerase chain reaction (PCR) and virus isolation. However, they typically suffer from higher costs, low selectivity/specificity, false negative/positive results

Herein, we report on molecularly imprinted polymers (MIPs) for detecting formaldehyde vapors in air streams. A copolymer thin film consisting of styrene, methacrylic acid, and ethylene glycol dimethacrylate on quartz crystal microbalance (QCM) yielded a detection limit of 500 ppb formaldehyde in dry air. Surprisingly, these MIPs showed specific behavior when tested against a range of volatile organic compounds (VOCs), such as acetaldehyde, methanol, formic acid, and dichloromethane. Despite thus being a suitable receptor in principle, the MIPs were not useful for measurements at 50% humidity d

Molecular imprinting is a technique for creating artificial receptor sites in a polymer. Molecularly imprinted polymers (MIPs) are produced by forming a polymer around a molecule that is used as the template. Upon removal of the template, molecular holes remain which are specific in shape and size to the target molecule. In this research, a MIP was formed for theophylline using a copolymer of methacrylic acid and ethylene glycol dimethacrylate. The theophylline MIP was formed on two platforms: indium tin oxide (ITO) and silicon, which were used as the working electrode for cyclic voltammetry m

Clinical manifestations owing to ascorbic acid deficiency demand an easy-to-use, rapid, robust and inexpensive technique, which can measure serum ascorbic acid at ultratrace level to attend the problem of hypovitaminosis C and acute cases of scurvy. In the present work, a novel molecularly imprinted polymer (MIP)-coated solid-phase microextraction (SPME) fiber that could be coupled to a complementary MIP-sensor was prepared with ascorbic acid as a template molecule. The characteristics and applications of this fiber were investigated. The same MIP receptor for both SPME and the corresponding s
