(-)-α-bisabolol, a valuable sesquiterpene with anti-inflammatory and antibacterial properties, was sustainably produced via de novo high-titer biosynthesis in Candida tropicalis using engineered cell factories. To minimize resource-intensive extraction methods, we employed green metabolic engineering strategies, including an optimized Scaffold-Mediated Assembly Regulation Tool for coexpressing MrBBS and ERG20 genes, enhancing (-)-α-bisabolol titer 3.24-fold over the control strain. A novel Linker- and Scaffold-Mediated Assembly Regulation Tool further improved pathway efficiency by coordinatin

Doronicum is a relatively small genus belonging to the tribe Senecioneae (Fam. Asteraceae), distributed in Asia, Europe, and North Africa. Some of its species are considered toxic due to the presence of pyrrolizidine alkaloids whereas some other ones are largely utilised in the ethnopharmacology of several countries. In the present study, the essential oil composition of a Sicilian accession of Doronicum caucasicum M. Bieb (syn. D. orientale Hoffm.), not previously investigated, is particularly rich in sesquiterpene hydrocarbons (80.3%) with germacrene D (58.9%), α-humulene (8.8%), and β-caryo

The wild tomato sesquiterpene (4R,7R)-zingiberene (6a) acts as a potent repellent of the economically important agricultural pest, the silverleaf whitefly (Bemisia tabaci). While its biosynthesis is catalyzed by 7-epi-zingiberene synthase from the noncanonical terpene precursor (2Z,6Z)-farnesyl pyrophosphate (5a), the precise enzymatic mechanism has remained undetermined. Using deuterated substrates generated chemoenzymatically, the 1,6-cyclization mechanism was established to proceed via a 1,3-hydride shift from the C1 position of the initial bisabolyl carbocationic intermediate. α-Bisabolol is a naturally derived monocyclic sesquiterpene, abundantly present in German chamomile (Matricaria recutita) and various other aromatic plants and is also increasingly accessible via metabolic engineering platforms. The physicochemical profile and ADMET characteristics of α-bisabolol, suggest a high gastrointestinal absorption, and minimal P-glycoprotein and CYP450 interactions, which validate its drug-like potential. The multi-protein target engagement and predicted activity spectrum of α-bisabolol have been discussed in the manuscript by using in silico tools (SwissTargetPredict

Chemical analysis of the chloroform extract of the aerial parts of the Algerian plant Pulicaria undulata exhibiting anti-inflammatory activity resulted in the isolation of 10 new humulene sesquiterpenoids, 1-10, belonging to the asteriscunolide family of compounds. The structure and relative configuration have been defined by NMR data, whereas the absolute configuration has been established by ECD analysis. Compounds 1-10 include enantiomers of the known asteriscunolides A-D and tetrahydroasteriscunolide previously reported from the genera Asteriscus and Nauplius. Compounds 1 and 10 showed in

Meroterpenoid natural products derived from the 11-membered macrocycle humulene are of great interest due to their potent biological activities and intricate architectures. Whereas molecules derived from the most available (E,E,E)-humulene isomer have been thoroughly investigated, little is known of the chemistry and biological properties of natural products derived from the (E,E,Z)-isomer and derivatives thereof. Herein, we detail the first synthesis of (E,E,Z)-(10S)-hydroxyhumulene through a selective oxidative double-bond isomerization, and using this key building block along with (E,E,Z)-h

BACKGROUND: Hops (Humulus lupulus L.) are a dioecious climbing perennial, with the dried mature "cones" (strobili) of the pistillate/female inflorescences being widely used as both a bittering agent and to enhance the flavour of beer. The glandular trichomes of the bract and bracteole flowering structures of the cones produce an abundance of secondary metabolites, such as terpenoids, bitter acids and prenylated phenolics depending on plant genetics, developmental stage and environment. More knowledge is required on the functional and allelic diversity of terpene synthase (TPS) genes responsibl

(-)-α-Bisabolol is one of the important derivatives in the mevalonate (MVA) pathway. Yeast not only possesses a complete MVA metabolic pathway but also has mature modification and optimization strategies, among which transcription factor regulation is a very effective method. In this study, by screening transcription factors associated with the MVA pathway, a new regulatory factor, ECM22, was discovered that can be used for terpenoid synthesis, and ECM22 proved to be the most effective compared to other screened transcription factors. Previous studies have indicated that ECM22 is related to st

Chlamydia are obligate intracellular bacterial pathogens affecting humans and animals, causing miscarriage, stillbirth, or weak fetuses in the late stages of pregnancy of goats and sheep. Because there is no commercial vaccine for chlamydia in animals, drug treatment has become the most effective curative method. Natural products, also known as secondary metabolites, are becoming one of the main sources used in new drug development because of their structural diversity and biodiversity. In natural products, plant sources play a major role in the development process of new drugs. In this study,
