Moorella species are thermophilic acetogens that primarily produce acetate from one-carbon (C1) compounds including CO, CO2 (+H2), methanol and formate. Notably, Moorella caeni DSM 21394T displays a hydrogenogenic metabolism on CO and an acetogenic metabolism on methanol. Furthermore, M. caeni is unable to use CO2 (+H2) and grows only on formate in the presence of a methanogen or when thiosulfate is added as an electron acceptor. Presently, all theoretical frameworks for C1 metabolism in Moorella species are derived from experimental and genomic analyses of Moorella thermoacetica, which exhibi

Moorella thermoacetica can grow with H₂ and CO₂, forming acetic acid from 2 CO₂ via the Wood-Ljungdahl pathway. All enzymes involved in this pathway have been characterized to date, except for methylenetetrahydrofolate reductase (MetF). We report here that the M. thermoacetica gene that putatively encodes this enzyme, metF, is part of a transcription unit also containing the genes hdrCBA, mvhD, and metV. MetF copurified with the other five proteins encoded in the unit in a hexaheteromeric complex with an apparent molecular mass in the 320-kDa range. The 40-fold-enriched preparation contained p

Microbial electrosynthesis systems (MES) are promising devices in which microbes obtain electrons from electrodes to produce extracellular multicarbon compounds. This study investigates whether improvement in cell permeability can enhance electrosynthesis performance of Gram-positive Moorella thermoautotrophica in MES. Results showed that when ≤30mg/L penicillin was added, the cell permeability was doubled, and the electron uptake per biomass (including both cathode-associated biomass and suspended biomass) was 1.84 times that of the control, while formate and acetate production rates per biom

Methanol derived from pectin degradation is common in oxic and anoxic sediments and is used by various aerobic and anaerobic microorganisms such as acetogenic bacteria or methanogenic archaea as a carbon and energy source. Acetogens use the Wood-Ljungdahl pathway to disproportionate methanol to CO2 and acetate. Methanol metabolism has been studied primarily in mesophilic acetogens such as Acetobacterium woodii and Eubacterium callanderi, that do not have cytochromes or quinones. Moorella thermoacetica, the model organism for elucidation of acetogenesis, is a thermophilic, cytochrome- and quino

Artificial photosynthesis by microbe-semiconductor biohybrid systems has been demonstrated as a valuable strategy in providing sustainable energy and in carbon fixation. However, most of the developed biohybrid systems for light harvesting employ heavy metal materials, especially cadmium sulfide (CdS), which normally cause environmental pollution and restrict the widespread of the systems. Herein, we constructed an environmentally friendly biohybirid system based on a typical acetogenic bacteria, Moorella thermoacetica, coupling with a carbon-based semiconductor, graphitic carbon nitride (g-C3

In hydrothermal ecosystems, the dissolution of sulfur dioxide in water results in the formation of sulfite, which can be used in microbial metabolism. A limited number of thermophiles have been isolated using sulfite as an electron acceptor. From a terrestrial thermal spring, Sakhalin Island, Russia, we isolated a thermophilic anaerobic bacterium (strain SLA38T). Cells of strain SLA38T were spore-forming straight rods. Growth was observed at temperatures 45-65 °C (optimum at 60 °C) and pH 5.5-9.0 (optimum at pH 6.5-7.0). The novel isolate was capable of anaerobic respiration with sulfite, thio

Thermophilic acetogenic bacteria have attracted attention as promising candidates for biotechnological applications such as syngas fermentation, microbial electrosynthesis, and methanol conversion. Here, we aimed to isolate and characterize novel thermophilic acetogens from diverse environments. Enrichment of heterotrophic and autotrophic acetogens was monitored by 16S rRNA gene-based bacterial community analysis. Seven novel Moorella strains were isolated and characterized by genomic and physiological analyses. Two Moorella humiferrea isolates showed considerable differences during autotrophi

Although the Wood-Ljungdahl pathway, a wide-spread pathway for CO2 fixation in anaerobic microorganisms, was elucidated in the thermophilic acetogenic bacterium Moorella thermoacetica, still relatively little is known about the enzymes involved in hydrogen oxidation, CO2 fixation, energy conservation and the role of quinones and cytochromes. Here, we have used transcriptomics, enzyme assays and genome analyses to identify missing links. NADPH, generated by a [FeFe] hydrogenase, is the reductant for CO2 reduction to formate, a key reaction in CO2 fixation. This reaction is slightly endergonic u

Gaseous fermentation for converting carbon-dioxide (CO2) from biogas with H2 into value-added chemicals such as acetic acid offers a sustainable solution compared to chemical upgradation. In this study we compare the performance of Moorella thermoacetica, a thermophilic homoacetogen employing the Wood-Ljungdahl pathway, with a mixed rumen culture enriched for homoacetogenic activity. Both cultures were evaluated for CO2/H2 conversion efficiency, acetic acid production, and upgrading of biogas and added H2 to renewable natural gas (RNG) using a trickle bed reactor (TBR). The mixed culture achie

Biohybrids composed of microorganisms and nanoparticles have emerged as potential systems for bioenergy and high-value compound production from CO2 and light energy, yet the cellular and metabolic processes within the biological component of this system are still elusive. Here we dissect the biohybrid composed of the anaerobic acetogenic bacterium Moorella thermoacetica and cadmium sulphide nanoparticles (CdS) in terms of physiology, metabolism, enzymatics and transcriptomic profiling. Our analyses show that while the organism does not grow on L-cysteine, it is metabolized to acetate in the bi

Enabling the nonphotosynthetic bacterium Moorella thermoacetica (M. thermoacetica) to form a photosynthetic biohybrid system could transform carbon dioxide (CO2) into value-added products. However, to ensure an efficient, mechanistic electron-transfer pathway to microbial catalysts, a pivotal nanoplatform that involves multicomponent composite systems for photoexcited electron transfer from the photosensitizer to the bacteria, as well as for enhancing light-harvesting capabilities and efficient electron/hole separation, is crucial. Here, we design panchromatic ternary polymer dots (Pdots) as b

Improving natural photosynthesis can enable the sustainable production of chemicals. However, neither purely artificial nor purely biological approaches seem poised to realize the potential of solar-to-chemical synthesis. We developed a hybrid approach, whereby we combined the highly efficient light harvesting of inorganic semiconductors with the high specificity, low cost, and self-replication and -repair of biocatalysts. We induced the self-photosensitization of a nonphotosynthetic bacterium, Moorella thermoacetica, with cadmium sulfide nanoparticles, enabling the photosynthesis of acetic ac

Solar-driven conversion of carbon dioxide to value-added carbon products is an ambitious objective of ongoing research efforts. However, high overpotential, low selectivity and poor CO2 mass transfer plague purely inorganic electrocatalysts. In this instance, we can consider a class of biological organisms that have evolved to achieve CO2 fixation. We can harness and combine the streamlined CO2 fixation pathways of these whole organisms with the exceptional ability of semiconducting nanomaterials to harvest solar energy. A novel nanomaterial-biological interface has been pioneered in which lig

Tandem "Z-scheme" approaches to solar-to-chemical production afford the ability to independently develop and optimize reductive photocatalysts for CO2 reduction to multicarbon compounds and oxidative photocatalysts for O2 evolution. To connect the two redox processes, molecular redox shuttles, reminiscent of biological electron transfer, offer an additional level of facile chemical tunability that eliminates the need for solid-state semiconductor junction engineering. In this work, we report a tandem inorganic-biological hybrid system capable of oxygenic photosynthesis of acetic acid from CO2.

Nickel-containing carbon monoxide dehydrogenases (CODHs) reversibly catalyze the oxidation of carbon monoxide to carbon dioxide and are of vital importance in the global carbon cycle. The unusual catalytic CODH C-cluster has been crystallographically characterized as either a NiFe(4)S(4) or a NiFe(4)S(5) metal center, the latter containing a fifth, additional sulfide that bridges Ni and a unique Fe site. To determine whether this bridging sulfide is catalytically relevant and to further explore the mechanism of the C-cluster, we obtained crystal structures of the 310 kDa bifunctional CODH/acet
