Vincristine impairs musculoskeletal development in pediatric mice.

BACKGROUND: Over 85% of children diagnosed with cancer now survive their disease, yet cancer and chemotherapy frequently associate with long-term health complications, including impaired musculoskeletal development. Despite high survival rates, there is limited research on how pediatric chemotherapy affects muscle and bone physiology. Vincristine, a vinca alkaloid chemotherapeutic, is widely used in pediatric oncology, but its systemic effects on the developing musculoskeletal system remain poorly understood. This study aimed to investigate the musculoskeletal consequences of vincristine in a

Anatomical Considerations of Embryology and Development of the Musculoskeletal System: Basic Notions for Musculoskeletal Radiologists.

The musculoskeletal (MSK) system begins to form in the third week of intrauterine development. Multiple genes are involved in the complex different processes to form the skeleton, muscles and joints. The embryonic period, from the third to the eighth week of development, is critical for normal development and therefore the time when most structural defects are induced. Many of these defects have a genetic origin, but environmental factors may also play a very important role. This review summarizes the embryology of the different components of the MSK system and their configuration as an organ-

Cellular Plasticity in Musculoskeletal Development, Regeneration, and Disease.

In this review, we highlight themes from a recent workshop focused on "Plasticity of Cell Fate in Musculoskeletal Tissues" held at the Orthopaedic Research Society's 2019 annual meeting. Experts in the field provided examples of mesenchymal cell plasticity during normal musculoskeletal development, regeneration, and disease. A thorough understanding of the biology underpinning mesenchymal cell plasticity may offer a roadmap for promoting regeneration while attenuating pathologic differentiation. © 2019 Orthopaedic Research Society. Development and assessment of a telesonography system for musculoskeletal imaging.

BACKGROUND: Telesonography systems have been developed to overcome barriers to accessing diagnostic ultrasound for patients in rural and remote communities. However, most previous telesonography systems have been designed for performing only abdominal and obstetrical exams. In this paper, we describe the development and assessment of a musculoskeletal (MSK) telesonography system. METHODS: We developed a 4-degrees-of-freedom (DOF) robot to manipulate an ultrasound probe. The robot was remotely controlled by a radiologist operating a joystick at the master site. The telesonography system was us

Musculoskeletal adaptations to weightlessness and development of effective countermeasures.

A Research Roundtable, organized by the American College of Sports Medicine with sponsorship from the National Aeronautics and Space Administration, met in November 1995 to define research strategies for effective exercise countermeasures to weightlessness. Exercise was considered both independently of, and in conjunction with, other therapeutic modalities (e.g., pharmacological nutritional, hormonal, and growth-related factors) that could prevent or minimize the structural and functional deficits involving skeletal muscle and bone in response to chronic exposure to weightlessness, as well as

Regulated cell death in musculoskeletal development, homeostasis, and diseases.

Regulated cell death (RCD) is a fundamental biological process that plays critical roles in regulating organogenesis and maintaining homeostasis under physiological conditions, and when cells are exposed to detrimental microenvironment, it contributes to the initiation and progression of pathological changes. Musculoskeletal disorders and diseases, such as osteoporosis, osteoarthritis, and intervertebral disk degeneration, have become public health problems worldwide, owing to their high prevalence and heavy economic burden. RCD is closely associated with the pathogenesis of skeletal disorders

Development and validation of a novel ankle joint musculoskeletal model.

An improved understanding of contact mechanics in the ankle joint is paramount for implant design and ankle disorder treatment. However, existing models generally simplify the ankle joint as a revolute joint that cannot predict contact characteristics. The current study aimed to develop a novel musculoskeletal ankle joint model that can predict contact in the ankle joint, together with muscle and joint reaction forces. We modelled the ankle joint as a multi-axial joint and simulated contact mechanics between the tibia, fibula and talus bones in OpenSim. The developed model was validated with r

Age-related changes in the musculoskeletal system and the development of osteoarthritis.

Osteoarthritis (OA) is the most common cause of chronic disability in older adults. Although classically considered a "wear and tear" degenerative condition of articular joints, recent studies have demonstrated an inflammatory component to OA that includes increased activity of several cytokines and chemokines in joint tissues that drive production of matrix-degrading enzymes. Rather than directly causing OA, aging changes in the musculoskeletal system contribute to the development of OA by making the joint more susceptible to the effects of other OA risk factors that include abnormal biomecha

[Homeostasis and Disorder of Musculoskeletal System.Mechanisms of cartilage development and homeostasis.].

Chondrocytes, which are originated from undifferentiated mesenchymal stem cells, play roles in skeletal development and growth in mammal, and smooth movement of joints. Endochondral ossification is necessary for skeletal development, and the multiple and complex biological events are precisely regulated by several hormones, cytokines, and their downstream signaling and transcriptional regulation. On the other hands, articular chondrocytes physiologically retains their features during a lifetime. Numerous molecules involved in endochondral ossification have been identified and investigation of

Effect of musculoskeletal development on the prediction of body density in females.

The purpose of this study was to determine if musculoskeletal development had a significant effect on the prediction of body density in females. Subjects consisted of 156 females aged 17 to 44 years. Subjects were divided into three groups on the basis of musculoskeletal development as determined by the Heath-Carter Anthropometric Somatotype mesomorphy rating. Anthropometric measurements included eight skinfold measures (tricep, subscapula, chest, midaxillary, abdomen, suprailiac, thigh, and calf), body diameters of the humerus and femur, and circumferences at five locations (waist, forearm, u

Changes in Glenohumeral Musculoskeletal Development Following Brachial Plexus Birth Injury.

Brachial plexus birth injury (BPBI), one of the most common nerve injuries in children, often leads to impaired shoulder development, resulting in sustained postural and bone deformity and muscle weakness. Despite the substantial long-term consequences, clinical consensus is lacking for what BPBI treatments are optimal in terms of timing and approach, primarily because BPBI sequelae are complex, involving stunted muscle growth, muscle denervation, and limb disuse that can disrupt glenohumeral joint development. Injury can occur as nerve rupture (postganglionic injury) or nerve avulsion (pregan

Musculoskeletal tuberculosis - imaging using low-end and advanced modalities for developing and developed countries.

Musculoskeletal involvement by TB is rare in comparison to other forms of the disease. It most commonly involves the spine but can also involve large weight-bearing joints, long bones, the skull and the soft tissues. Characteristic appearances of musculoskeletal TB are described for numerous imaging modalities in this paper but it is also highlighted that when based on imaging appearances alone, there is always a differential diagnosis, including other infections and malignancies. Awareness of of TB as a possible cause of vertebral body lesions, arthritis and synovitis, long bone lesions and s

Evaluating the Accuracy of the Frysian Questionnaire for Differentiation of Musculoskeletal Complaints for Triage of Musculoskeletal Diseases: Algorithm Development and Validation Study.

BACKGROUND: Inflammatory rheumatic diseases (IRDs) affect 5% of the general population, whereas 35% of the population experiences musculoskeletal concerns. IRDs cause early disability, reduced life expectancy, and considerable health care costs. Early diagnosis is essential to prevent long-term damage. Similarly important is the early identification of patients with musculoskeletal concerns without IRDs to prevent unnecessary health care expenses. Of the population referred to the rheumatologist, 60% have noninflammatory musculoskeletal concerns, whereas only 20% of patients with an IRD see a

The Impact of AI on the Development of Multimodal Wearable Devices in Musculoskeletal Medicine.

As wearables are becoming an increasingly important part of wellness and everyday life for many people, their potential in healthcare is also expanding, particularly in personalized and remote healthcare. However, many wearables lack sophistication, relying on simple sensors such as accelerometers and pulse meters to measure heart rate, body composition, and daily activity. Such basic metrics are insufficient for musculoskeletal disease diagnosis, which requires more detailed, multimodal neuromusculoskeletal monitoring. A major challenge in wearables development is the need for precise electro

Pannexins in the musculoskeletal system: new targets for development and disease progression.

During cell differentiation, growth, and development, cells can respond to extracellular stimuli through communication channels. Pannexin (Panx) family and connexin (Cx) family are two important types of channel-forming proteins. Panx family contains three members (Panx1-3) and is expressed widely in bone, cartilage and muscle. Although there is no sequence homology between Panx family and Cx family, they exhibit similar configurations and functions. Similar to Cxs, the key roles of Panxs in the maintenance of physiological functions of the musculoskeletal system and disease progression were g
