Kinetic analysis of gene expression during mycelium to yeast transition and yeast to mycelium germination in Paracoccidioides brasiliensis.

INTRODUCTION: Paracoccidioidomycosis is an endemic systemic mycosis caused by Paracoccidioides brasiliensis, a thermally dimorphic fungus that in tissues and cultures at 37°C grows as a yeast while at lower temperatures (less than 24°C) it becomes a mold; however the genes that rule these processes and their expression are poorly understood. OBJECTIVE: This research focused on the kinetic expression of certain genes in P. brasiliensis throughout the dimorphic process, one that involves the transition from the mycelium to yeast forms and the germination from the yeast to mycelium form. Effects of Humidity on Mycelium-Based Leather.

Leather is a product that has been used for millennia. While it is a natural material, its production raises serious environmental and ethical concerns. To mitigate those, the engineering of sustainable biobased leather substitutes has become a trend over the past few years. Among the biobased materials, mycelium, the fungal "root" of a mushroom, is one of the promising alternatives to animal leather, as a material with tunable physicomechanical properties. Understanding the effect of humidity on mycelium-based leather material properties is essential to the production of durable, competitive,

Mycelium-Doped Straw Biochars for Antibiotic Control.

Straw, a predominant agricultural residue, represents a significant waste product. Harnessing its potential is of paramount importance both in terms of research and economic value. In this study, chemically pretreated corn straw was infused with distinct microbial fungal mycelium variants and subsequently transformed into a series of biochars through a process involving carbonization and activation. The findings revealed enhancements in the specific surface area and total pore volume of mycelium-doped straw biochars compared to the original corn straw biochar (BCS). Additionally, discernible d

Ca2+ fluxes in developing Trichoderma viride mycelium.

The properties of both Ca2+ influx and efflux in the mycelium during the life cycle of Trichoderma viride were studied by means of 45Ca2+ and by X-ray fluorescence spectroscopy measurements. The properties of the 45Ca2+ influx and effluxes indicate that they are mediated by different transport systems. The Ca2+ influx could be mediated by an electrogenic Ca2+/nH+ antiport, or by an Ca2+ uniport system. Both Ca2+ influx and efflux were stimulated by the uncouplers (and the treatment leading to the suppression of energy metabolism) and by azalomycin F, an antifungal agent. Salicylate stimulated

Morphologically tunable mycelium chips for physical reservoir computing.

We introduce a neuromorphic computing substrate based on PEDOT:PSS-infused mycelium, a biofabricated, morphologically tunable material that can be engineered into electrically active components including resistors, capacitors, and non-linear elements. Leveraging the principles of physical reservoir computing, we demonstrate that hyphal networks grown under controlled environmental conditions can transform time-varying inputs into nonlinear, high-dimensional state trajectories, enabling machine learning tasks such as NARMA-10 sequence prediction. The chips are produced using a "design-grow-comp

Self-grown mycelium in confined geometries as nanofluidic devices.

Precise control of ion and molecular transport at the nanoscale underpins next-generation nanofluidic technologies. However, current approaches such as top-down fabrication and bottom-up assembly remain constrained by cost, scalability, or limited programmability. Fungal mycelium-the largest natural ion transport network in soil-offers a living bio-derived route to nanofluidics. Here, we harness mycelium's self-growth and hyphal anastomosis to construct nanofluidic structures that autonomously conform to confined geometries. With interconnected fibrous networks, nanoscale porosity, and negativ

Fabrication of Living Entangled Network Composites Enabled by Mycelium.

Organic polymer-based composite materials with favorable mechanical performance and functionalities are keystones to various modern industries; however, the environmental pollution stemming from their processing poses a great challenge. In this study, by finding an autonomous phase separating ability of fungal mycelium, a new material fabrication approach is introduced that leverages such biological metabolism-driven, mycelial growth-induced phase separation to bypass high-energy cost and labor-intensive synthetic methods. The resulting self-regenerative composites, featuring an entangled netw

Hericium erinaceus Mycelium Ameliorates In Vivo Progression of Osteoarthritis.

Osteoarthritis (OA) is an age-related disorder that affects the joints and causes functional disability. Hericium erinaceus is a large edible mushroom with several known medicinal functions. However, the therapeutic effects of H. erinaceus in OA are unknown. In this study, data from Sprague-Dawley rats with knee OA induced by anterior cruciate ligament transection (ACLT) indicated that H. erinaceus mycelium improves ACLT-induced weight-bearing asymmetry and minimizes pain. ACLT-induced increases in articular cartilage degradation and bone erosion were significantly reduced by treatment with H.

Alpha-amylase inhibitors from mycelium of an oyster mushroom.

The α-Amylase and α-glucosidase are two main enzymes involved in carbohydrate metabolism. This study was aimed at detecting alpha-amylase inhibitory activity from edible mushroom mycelia. Oyster mushroom was collected from a natural source, from Indian Institute of Technology (Banaras Hindu University) campus and was maintained in vitro in mycelial form. Chloroform, acetone, methanol, and water were used separately for extraction of an active constituent from mycelial cells grown, for 7 days, in potato dextrose broth. The extracts were tested for alpha-amylase inhibitory activity. Chloroform,

Effect of temperature on the mycelium to yeast transformation of Paracoccidioides brasiliensis.

The influence of temperature on Paracoccidioides brasiliensis mycelium to yeast transformation was studied by sequential microscopic observations of slide cultures. When incubated at temperatures above 28 degrees C, the mycelial elements gradually produced round to oval chlamydospores and later on, exhibited multiple budding. A sizeable proportion of mycelial elements transformed at 34 degrees C; however, multiple budding was important only at 37 degrees C.

A mycelium-based carbon dioxide source for trapping vector mosquitoes.

Carbon dioxide (CO2) is a key activator and attractant for mosquitoes during host-seeking and is routinely used to bait traps. However, deploying CO2 via dry ice, compressed gas, sugar-yeast reactions, or carbonate-acid systems can be expensive, short-lived, and labor-intensive. We evaluated the effectiveness of a commercially available, mycelium-based CO2 source (ExHale bag) for attracting mosquitoes to passive and active traps. Originally developed for greenhouse use, the ExHale bag provides sustained CO2 release for up to 6 mo. Under semi-field conditions, we compared Biogents gravid Aedes

Cordyceps sinensis mycelium protects mice from group A streptococcal infection.

Group A streptococcus (GAS) infection can cause severe invasive diseases, including necrotizing fasciitis and streptococcal toxic shock syndrome. Cordyceps sinensis, a Chinese herbal medicine, is an immunomodulator. In this study the air-pouch bacterial inoculation model was used to investigate the protective efficacy of C. sinensis mycelium extract against GAS infection. Force-feeding mice with C. sinensis mycelium extract for 3 consecutive days before GAS infection increased the survival rate and reduced local skin-tissue injury compared with mice fed PBS. Bacterial numbers in the air pouch

Bacillus subtilis is crucial for the normal growth of mycelium for Stropharia rugosoannulata.

Stropharia rugosoannulata is an important commercially cultivated edible mushroom worldwide. It grows slowly, and even does not grow at all on completely sterilized substrates, which has limited the spawn manufacture. In this article, it was found that the mycelium growth rate of S. rugosoannulata on non-sterilized or incompletely sterilized sawdust medium is 90%-125% faster than that on completely sterilized sawdust medium. The strain T2-19 with promoting effect on the mycelium growth was isolated from the substrate colonized by S. rugosoannulata. The phylogenetic analysis based on 16S rRNA g

Effects of Blue Light on Hypocrellin A Production in Shiraia Mycelium Cultures.

Blue light is a crucial environmental cue for fungi. Hypocrellin A (HA) is a photoactive perylenequinone from Shiraia with strong antimicrobial and anticancer properties. In this study, effects of the illumination of blue-light-emitting diode (LED) at 470 nm on Shiraia sp. S8 were investigated. Blue light at 50-200 lx and 4-6 h day-1 could enhance HA content in the mycelia, but suppress it at 300-400 lx or with longer exposure (8-24 h day-1 ). The intermittent blue light (6 h day-1 ) at 200 lx not only enhanced the fungal conidiation but also stimulated HA production without any growth retarda

The volatile organic compounds from the mycelium of Tuber borchii Vitt.

The mycelium of T. borchii (characterized by DNA analysis) grown in sterile liquid medium produced some VOCs. The VOCs were retained on carbographs by passing a flow of helium, isolated and characterized in a GC-MS equipment after a thermal desorption. The compounds present in the VOCs from the mycelium cultures, but not in the VOCs from the control cultures, contained 29 compounds. The main compounds were 1,3-ditertbutylbenzene (16.1 ng/l), 3-methylheptane (9.2 ng/l), butan-2-one (8.8 ng/l), ethynylbenzene (5.6 ng/l), and octan-3-one (4.9 ng/l).
