Comparison of laboratory methods for identifying members of the family Mycobacteriaceae.

BACKGROUND: The introduction of a method based on matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-ToF mass spectrometry) into the practice of laboratories significantly increased the identification of acid-resistant bacteria (ARB). METHODS: Seventy-four nontuberculous mycobacteria (NTM) cultures identified by deoxyribonucleic acid (DNA) hybridization, polymerase chain reaction, Sanger sequencing, and MALDI-ToF mass spectrometry. RESULTS: Analysis of the identification results obtained by the methods of DNA hybridization and Sanger sequencing showed a compl

Development of an in situ hybridization assay for the diagnosis of Mycobacteriaceae infections of veterinary importance.

Mycobacteria (Mycobacteriaceae family) comprise five genera (Mycobacterium, Mycolicibacterium, Mycolicibacter, Mycolicibacillus, and Mycobacteroides), which include relevant animal and human pathogens. Histology is a rapid method for preemptively diagnosing mycobacteriosis, contributing to surveillance, control, and eradication. A constraint on histology is the limited sensitivity and specificity of acid-fast stains, as the number of detectable bacilli in formalin-fixed paraffin-embedded (FFPE) tissue varies and other microorganisms are acid-fast positive. Immunohistochemistry has low specific

Mycobacterium gordonae (Actinomycetota: Mycobacteriales: Mycobacteriaceae) Infecting Captive-Reared Laurel Dace, Chrosomus saylori (Cypriniformes: Leuciscidae) in North America.

A captive-reared cohort of Laurel Dace, Chrosomus saylori (Skelton 2001) (Cypriniformes: Leuciscidae), a federally-endangered species endemic to several streams in eastern Tennessee's Cumberland Plateau, exhibited signs of morbidity and trickling mortalities in a recirculating aquaculture system. Ziehl-Neelsen (ZN) staining revealed acid-fast rod-shaped bacteria within necrotic and intracellular lesions affecting the eye, swim bladder, spleen, and ovary. Histopathology showed granulomatous inflammation characterised by macrophages, multinucleated giant cells, and eosinophilic exudates. The eye

Comparative in silico analysis of mce operons across Mycobacteriaceae and Nocardiaceae: Insights into genetic arrangements and regulatory mechanisms.

The mammalian cell entry (Mce) proteins are critical for the entry and persistence of Mycobacterium tuberculosis (Mtb) within alveolar macrophages. These proteins assemble into an ATP-binding cassette transporter comprising six substrate-binding proteins (MceA-F), two transmembrane domains (YrbEAB), and two nucleotide-binding domains (MceG2). Although Mtb harbours four distinct mce operons, the regulatory mechanisms governing differential operon expression remain insufficiently understood. In this study, we analyzed the genetic organization, regulatory features, and functional roles of operons

Comparative performance of reference-based metagenomic tools to identify species-level taxa among families of bacteria: benchmarking Mycobacteriaceae and Neisseriaceae.

UNLABELLED: Hypotheses concerning the ecology and evolution of bacteria commonly relate to the presence and abundance of species in various settings and conditions. Shotgun metagenomics may address these hypotheses, which previously relied on PCR or culture. However, the problem of determining the presence/absence of a given species of interest is not trivial, particularly when closely related species are present in the reference database or metagenomic sample. Reference-based methods for detecting species-level taxa mostly rely on thresholding of aligned reads or mapped k-mers, or derivative

A novel phage satellite class induces prophage excision in Mycolicibacterium aichiense.

Phage satellites are mobile genetic elements that parasitize a helper phage to complete their life cycle. While phage satellites are widespread in diverse bacterial hosts, none have yet been isolated in Mycobacteriaceae. Here, we report the first phage satellites isolated and characterized in Mycobacteriaceae, Extracellular Prophage-Inducing Particles (EPIPs). EPIPs induce a helper phage-HerbertWM, a Mycolicibacterium aichiense prophage-upon infection. Genomic sequencing of thirteen isolates revealed a genome size of ∼11 kb and ∼83.3% nucleotide similarity calculated using Virus Intergenomic D

Systemic visceral talc granulomatosis associated with miliary tuberculosis in a drug addict.

Light and scanning electron microscopic study of tissues obtained at biopsy and at autopsy from a 64-year-old male drug addict revealed the presence of foreign body granulomas associated with birefringent crystals in the lungs, liver, bone marrow, spleen, and lymph nodes. Caseating granulomas associated with Mycobacteriaceae tuberculosis were also present. Energy dispersive X-ray analysis identified the crystals as talc. Minute talc crystals were also visualized in urine sediment, by scanning electron microscopy and identified by energy dispersive X-ray analysis.

The Anti-tuberculosis Battle in Greece in the 1800s and 1900s.

Tuberculosis is an infectious disease that mainly affects the lungs (known as pulmonary tuberculosis). Mycobacterium tuberculosis is a species of pathogenic bacteria in the family of Mycobacteriaceae and the causative agent of tuberculosis; it was discovered by Robert Koch in 1882. From about 1918 to 1939, tuberculosis in Greece was characterized as a social disease because it seemed to spread among the lower social classes, including displaced people living in refugee camps. The battle against tuberculosis involved private initiatives aimed at educating people on hygiene and establishing anti

Exploring the evolutionary divergence of cyclic di-nucleotide signaling in diverse mycobacterial species.

Cyclic dinucleotides (CDNs), such as cyclic di-AMP (c-di-AMP) and cyclic di-GMP (c-di-GMP), are key second messengers that regulate fundamental bacterial processes, including cell wall synthesis, biofilm formation, antibiotic resistance, stress response, and virulence. These pathways are particularly relevant in major pathogens like Mycobacterium tuberculosis. Increasing evidence highlights the pathogenic potential of non-tuberculous mycobacteria (NTM), originally environmental species that are emerging as significant human pathogens. Understanding the evolution of CDN signaling may therefore

Tuberculous Spondylitis Caused by Intravesical Bacillus Calmette-Guerin Therapy.

A 74-year-old man underwent intravesical bacillus Calmette-Guerin (BCG) therapy for bladder cancer and later presented with lower left back pain. Magnetic resonance imaging of the spine showed high signal intensity, diagnosed as a cystic lesion in the epidural and bilateral intestinal psoas muscle. A computed tomography-guided needle biopsy and histological examination revealed bacteria from the family Mycobacteriaceae, and Mycobacterium bovis was identified using multiplex polymerase chain reaction. If lower back pain appears in a patient who has undergone BCG therapy, it is necessary to test

Crystal structure of FAD-independent methylene-tetrahydrofolate reductase from Mycobacterium hassiacum.

FAD-independent methylene-tetrahydrofolate (methylene-H4 F) reductase (Mfr), recently identified in mycobacteria, catalyzes the reduction of methylene-H4 F to methyl-H4 F with NADH as hydride donor by a ternary complex mechanism. This biochemical reaction corresponds to that of the ubiquitous FAD-dependent methylene-H4 F reductase (MTHFR), although the latter uses a ping-pong mechanism with the prosthetic group as intermediate hydride carrier. Comparative genomics and genetic analyses indicated that Mfr is indispensable for the growth of Mycobacterium tuberculosis, which lacks the MTHFR encodi

Direct Conversion of Phytosterol to Testosterone via an Integrated Microbial-Enzymatic Strategy.

Testosterone is an essential androgenic steroid traditionally produced from 4-androstenedione through multistep chemical or biocatalytic processes. Here, we report a streamlined and sustainable "two-stage" bioconversion strategy for direct testosterone production from phytosterols. In the first stage, Mycobacterium neoaurum NRRL B-3805 was metabolically reprogrammed to maximize 4-androstenedione accumulation by deleting kstd1 and kshB1 and overexpressing hsd4A, generating strain MnBΔkstd1/ΔkshB1/p261-hsd4A. This engineered strain produced 12.3 g L- 1 of 4-androstenedione within 6 days, corresp

Evaluation of the resistome and gut microbiome composition of hospitalized patients in a health unit of southern Brazil coming from a high animal husbandry production region.

INTRODUCTION: Antimicrobial resistance (AMR) poses a significant threat to global public health. The One Health approach, which integrates human, animal, and environmental health, highlights the roles of agricultural and hospital settings in the propagation of AMR. This study aimed to analyze the resistome and gut microbiome composition of individuals from a high-intensity animal husbandry area in the western region of Santa Catarina, Southern Brazil, who were subsequently admitted to the University Hospital in the city of Florianopolis, located in the eastern part of the same state. METHODS:

Mycobacterium vaccae NCTC 11659, a Soil-Derived Bacterium with Stress Resilience Properties, Modulates the Proinflammatory Effects of LPS in Macrophages.

Inflammatory conditions, including allergic asthma and conditions in which chronic low-grade inflammation is a risk factor, such as stress-related psychiatric disorders, are prevalent and are a significant cause of disability worldwide. Novel approaches for the prevention and treatment of these disorders are needed. One approach is the use of immunoregulatory microorganisms, such as Mycobacterium vaccae NCTC 11659, which have anti-inflammatory, immunoregulatory, and stress-resilience properties. However, little is known about how M. vaccae NCTC 11659 affects specific immune cell targets, inclu

Fatal outcome of catheter-related bloodstream infection caused by Multidrug-Resistant Mycobacterium mucogenicum.

Mycobacteria mucogenicum (M. mucogenicum) is a rarely isolated pathogen. It has emerged as a significant pathogen in immunocompromised patients including those with cancer, organ transplant, or patients on immunosuppressive medication. Chemotherapy may reduce the ability of the bone marrow of these to respond to infection, and patients will be at risk for neutropenic sepsis, which leads to fatal complications. Here, we report a case of an 18-year-old boy was seen at Hospital Raja Perempuan Zainab II, Kelantan with acute lymphoblastic leukaemia (ALL) who presented with catheter-related bloodstr

Fitness Cost and Compensatory Evolution in Levofloxacin-Resistant Mycobacterium aurum.

We isolated spontaneous levofloxacin-resistant strains of Mycobacterium aurum to study the fitness cost and compensatory evolution of fluoroquinolone resistance in mycobacteria. Five of six mutant strains with substantial growth defects showed restored fitness after being serially passaged for 18 growth cycles, along with increased cellular ATP level. Whole-genome sequencing identified putative compensatory mutations in the glgC gene that restored the fitness of the resistant strains, presumably by altering the bacterial energy metabolism.

Case report: Mycobacterium neoaurum infection during ICI therapy in a hepatocellular carcinoma patient with psoriasis.

We report here a patient with advanced hepatocellular carcinoma (HCC) and psoriasis treated with immune checkpoint inhibitor (ICI) therapy who experienced tumor partial response and psoriatic exacerbation. Meanwhile, the patient contracted mycobacterium neoaurum during the treatment period, while it was an opportunistic infection and mainly happened in immunosuppressed patients. We discussed the possibility that this infection was an ICI-associated infection independent of immunosuppression due to dysregulated immunity, which was the result of the effects of immunotherapy and autoimmune diseas

Bioballs carrying a syntrophic Rhodococcus and Mycolicibacterium consortium for simultaneous sorption and biodegradation of fuel oil in contaminated freshwater.

Nonpathogenic effective bacterial hydrocarbon degraders, Rhodococcus ruber S103, Mycolicibacterium parafortuitum J101 and Mycolicibacterium austroafricanum Y502, were isolated from mixed polycyclic aromatic hydrocarbon (PAH)-enriched river sediments. They possessed broad substrate specificities toward various PAHs and aliphatic compounds as sole carbon sources. These strains exhibited promising characteristics, including biosurfactant production, high cell hydrophobicity, biofilm formation and no antagonistic interactions, and contained genes encoding hydrocarbon-degrading enzymes. Mycobacterium setense isolated from a cat with atypical mycobacterial panniculitis.

Atypical mycobacterial panniculitis was diagnosed in a cat. Mycobacterium setense was identified as causative agent by 16 S rRNA gene sequence analysis. This a gram-positive rod-shaped acid-fast bacterium belonging to Mycobacterium fortuitum group was never reported before in diseased animals. Resistance to doxycycline and clarithromycin was detected. During treatment with pradofloxacin, additional resistance to fluoroquinolones developed which was due to a mutation in the gyrase gene gyrA (S90W exchange). Despite of antimicrobial treatment for 33 months the patient did not fully recover. Spec
