The respiratory metabolism of Mycobacterium lepraemurium.

The respiratory metabolism of Mycobacterium lepraemurium isolated from Sprague-Dawley rats lepromata using several substrates was investigated. None of the intermediates of the glycolysis cycle as well as of the tricarboxylic acid cycle except succinate was oxidized by purified whole suspensions of M. lepraemurium. Likewise, many sulfur compounds such as cystine, thiourea, thioacetate, thiodiglycol, mercaptoact and some sulfhydryl compounds, e.g., cysteine, dithioerythritol, dithiorthritol, and penicillamine were readily oxidized by murine bacillary suspensions, whereas thioglycolate, thiogluc

[Oxidative phosphorylation in Mycobacterium lepraemurium].

The generation of ATP by cell-free extracts of Mycobacterium lepraemurium isolated from Sprague-Dawley rats was investigated. Cell-free preparations catalyzed phosphorylation coupled to the oxidation of NADH and succinate yielding P/O ratios of 0.6 and 0.4, respectively. Ascorbate oxidation did not result in ATP formation. The oxidative phosphorylation was uncoupled by 2,4- dinitrophenol and pentachlorophenol. Phosphate esterification coupled to NADH oxidation was inhibited by rotenone which had no effect on ATP synthesis associated with succinate oxidation. Antimycin A and cyanide completely

Mycobacterium leprae and Mycobacterium lepraemurium infections in domestic and wild animals.

Mycobacterium leprae, the aetiological agent of leprosy in humans, gives rise to a chronic granulomatous disease that affects primarily the skin and peripheral nerves, and secondarily some internal organs such as the testis and the eye; viscera are seldom involved. Depending on host resistance, leprosy may present as a benign disease (tuberculoid leprosy) or as a malignant disease (lepromatous leprosy), with a spectrum of intermediate stages appearing between the two. Immunity against leprosy depends on the cell-mediated immunity of the host, and this is severely compromised in the malignant (

[Energy production in Mycobacterium lepraemurium cultured in vitro].

Cell-free extracts prepared from in vitro cultured Mycobacterium lepraemurium catalysed phosphorylation coupled to the oxidation of NADH and succinate, yielding P/O ratios of 0.52 and 0.34, respectively. No ATP synthesis occurred during oxidation of ascorbate. Oxidative phosphorylation was uncoupled by dinitrophenol and dibromophenol. Oxidation of NADH and coupled phosphorylation was markedly inhibited by rotenone, whereas this inhibitor had no effect on succinate oxidation and associated ATP synthesis. Oxidative phosphorylations and coupled oxidations of NADH and succinate were strongly inhib

In vitro sensitivity of Mycobacterium lepraemurium for antimycobacterial drugs.

The sensitivity of Mycobacterium lepraemurium for isoniazide, dapsone, ethionamide, pyrazinamide, rifampicin, p-aminosalicylic acid, ethambutol and various sulfonamides was determined using Ogawa medium. The sensitivities of M. lepraemurium for these drugs are different from those obtained with M. leprae. Our results illustrate that M. lepraemurium is not a model for the drug sensitivities of M. leprae.

Antibacterial resistance in mice infected with Mycobacterium lepraemurium.

The differences in susceptibility among C57Bl/6, DBA/2 mice and their F1 hybrids to infections with M. lepraemurium were shown to depend upon the route of infection and size of the inoculum. A method was developed to measure the ability of lymphocytes obtained from M. lepraemurium-infected donors to effect adoptive immunization of syngeneic naive mice against infection with M. tuberculosis. This required sublethal irradiation of recipient mice prior to cell transfer and bacterial challenge. Using this method, it was found that mice infected subcutaneously generated antituberculous immune mecha

Palmitate oxidation by in vivo and in vitro grown Mycobacterium lepraemurium.

Oxidation of palmitate by Mycobacterium lepraemurium isolated from C3H mice lepromata (in vivo) and also grown on Ogawa egg-yolk medium (in vitro) was investigated. Palmitate was found to be oxidized, after a lag period of about 8 h, by both the in vivo and in vitro grown bacilli. Cell-free extracts prepared from in vivo and in vitro grown cells catalysed an active oxidation of palmitate after a lag period of 3-4 h. The amount of ATP increased, with the increase in time during oxidation of palmitate by the cell-free extracts. The generation of ATP was strongly inhibited by the inhibitors roten

Experimental leprosy with Mycobacterium lepraemurium in hairless mice (Mus musculus).

Forty-three homozygous albino hairless mice (Mus musculus) were inoculated intradermally on the head, back, sides and base of the tail with suspensions of lepromatous tissue containing numerous Mycobacterium lepraemurium (Hawaiian strain). Visible nodules were noted in all mice. These lepromatous nodules enlarged slowly and often merged with each other, covering up to two thirds of the dorsum. Several mice became blind during the course of the infection. All of the mice died or were killed 68 to 287 days following inoculation. Post-mortem examinations on representative mice showed, in addition

Effect of reactive oxygen intermediaries on the viability and infectivity of Mycobacterium lepraemurium.

Murine leprosy is a natural disease of the mouse, the most popular model animal used in biomedical research; the disease is caused by Mycobacterium lepraemurium (MLM), a successful parasite of macrophages. The aim of the study was to test the hypothesis that MLM survives within macrophages because it highly resists the toxic effects of the reactive oxygen intermediaries produced by these cells in response to infection by the microorganism. MLM cells were incubated in the presence of horseradish peroxidase (HRPO)-H(2)O(2)-halide for several periods of time. The peroxidative effect of this syste

Oxidation of reduced nicotinamide adenine dinucleotide by particles from Mycobacterium lepraemurium.

Particles from Mycobacterium lepraemurium catalysed the oxidation of NADH with oxygen as the terminal electron acceptor. The preparations contained cytochromes of the a + a3'b and c types, as well as CO-binding pigments. The NADH oxidase activity was sensitive to inhibitors of the flavoprotein system as well as to HQNO and antimycin A. In addition, a cytochrome oxidase sensitive to cyanide was also present. The system was inhibited by the thiol-binding agent, PCMB, and thus indicated the involvement of sulphydryl group in the enzymatic oxidation of NADH. The sensitivity of the NADH oxidase sys

CBA mice inoculated i.v. with 2 x 10(8) viable MLM were observed at intervals for 5 months. In the bone marrow, haematopoietic cells were progressively displaced by MLM-laden phagocytes; depletion of erythroid cells began earlier and was more pronounced than that of myeloid cells. Transiently, mild anaemia and profound leucopenia were noted. The spleen was enlarged and the site of increasing histiocytosis and extramedullary haematopoiesis which was accompanied by displacement of splenic parenchyma. The liver was enlarged and its parenchyma contained scattered islands of haematopoietic elements

[Characterization of experimental infection with Mycobacterium lepraemurium in 2 strains of mice].

A model of experimental leprosy in two strains of mice, namely CBA/J and CBi, has been developed based on: 1) the histological examination of a granuloma in the hind foot pad 200 days after inoculation of 0.30 microliter of Mycobacterium lepraemurium (6 x 10(8) MLm/ml); 2) the assessment of T lymphocytes in the granuloma identified by the alpha-naphthyl acetate method for esterase, and c) dissemination of the infection. The histological findings in the low resistance CBA/J strain included positive acid fast bacilli vacuolated cells, without lymphocytic infiltration, scarce number of T lymphocy

Propagation of Mycobacterium lepraemurium on supplemented minimal medium and its experimental pathogenesis.

The splenic tissue of a mouse experimentally infected with M. lepraemurium (Hawaiian strain, M-65) and developing 'rat leprosy', yielded a pure culture of an acid - fast bacterium having all the characteristics of M. lepraemurium on mineral salt minimal medium supplemented with simple sources of C and N, e.g., NH4 -salts, liquid paraffin, urea, gelatin etc. This could be maintained, by serial passages in vitro with good growth. Its indefinite propagation with tissue - free washed, small inoculum on complex media including Ogawa medium was difficult, and its serial sub-culture was practically i

Development of Anxiolytic and Depression-like Behavior in Mice Infected with Mycobacterium lepraemurium.

Murine leprosy is a systemic infectious disease of mice caused by Mycobacterium lepraemurium (MLM) in which the central nervous system (CNS) is not infected; nevertheless, diseased animals show measurable cognitive alterations. For this reason, in this study, we explored the neurobehavioral changes in mice chronically infected with MLM. BALB/c mice were infected with MLM, and 120 days later, the alterations in mice were evaluated based on immunologic, histologic, endocrine, neurochemical, and behavioral traits. We found increases in the levels of IL-4 and IL-10 associated with high bacillary l

Immune response to Mycobacterium lepraemurium: assessment of the humoral immune response in mice.

Intraperitoneal infection of mice with Mycobacterium lepraemurium produced a primary immune response as represented by direct (IgM) plaque-forming cells at the splenic level within a short period after infection. This response was enhanced when animals were administered a second infection fifteen days after the first one. An identical situation was observed with the developed (IgG) plaques, although the magnitude of these plaques in animals given the second challenge was of a higher order than those given only one infection. In both instances, the number of plaques declined within a very short

At varying periods of time following the successful establishment of systemic infections with Mycobacterium leprae or M. lepraemurium in the mouse and the nine-banded armadillo eyes were examined by light microscopy. Inoculation of bacilli was by the intravenous or intraperitoneal route or directly into the hind footpads; eyes were not directly inoculated in this study. During periods of up to 3 years under laboratory conditions no animal showed evidence of impaired vision or blindness, and the external appearance of both eyes was normal. Immune response in BALB/c mice following immunization with heat-killed Mycobacterium lepraemurium.

BALB/c mice were immunized with 1 X 10(7) heat-killed Mycobacterium lepraemurium (Mlm) via the hind foot pad. Four weeks later, the animals were infected with 1 X 10(9) Mlm intraperitoneally. Skin test studies, using footpad swelling as a parameter, indicated the development of skin reactivity to Mlm and M. leprae cell extracts. Immunized animals that were infected showed positive reactions to both antigens by the second week. This persisted up to fourteen weeks, at which time, bacillary restriction was also observed in the spleens and livers. Non-immunized infected animals, on the other hand

Effect of BCG vaccination on Mycobacterium lepraemurium infection in a highly susceptible inbred mouse strain.

Upon infection with Mycobacterium lepraemurium (MLM) C3H mice develop a disease that has features in common with lepromatous leprosy in man. Intraperitoneal vaccination with a single dose of BCG four weeks before inoculation with MLM in the footpad significantly reduced the total bacillary load of the animals. In vaccinated animals there was a delay in the dissemination of bacilli to the popliteal lymph node, liver, and spleen. The growth rate of MLM in the footpad and the popliteal lymph node was not altered by BCG vaccination. Reduced dissemination of the bacilli seems to be a sensitive para
