Molecular methods for the diagnosis and characterization of Cryptococcus: a review.

Cryptococcosis is a fungal infection caused by yeasts of the genus Cryptococcus, with Cryptococcus neoformans and Cryptococcus gattii as the primary pathogenic species. This disease is a threat to immunocompromised patients, especially those who have AIDS. However, the disease has also been described in healthy individuals. The tests used to identify these microorganisms have limitations that make final diagnosis difficult. However, currently there are specific gene sequences that can be used to detect C. neoformans and C. gattii from clinical specimens and cultures. These sequences can be use

[Comparison of Phoenix™ Yeast ID Panel and API® ID 32C commercial systems for the identification of Candida species isolated from clinical samples].

Opportunistic fungal pathogens are one of the important causes of nosocomial infections, and several different types of yeasts, especially Candida species are increasingly recovered from immunocompromised patients. Since many of the yeasts are resistant to the commonly used antifungal agents, the introduction of appropriate therapy depends on rapid and accurate identification. The aims of this study were to compare the commercial identification systems namely API® ID 32C (bioMerieux, France) and Phoenix™ Yeast ID Panel (Becton Dickinson Diagnostics, USA) for the identification of Candida speci

Identification of Candida species in the clinical laboratory: a review of conventional, commercial, and molecular techniques.

In healthy individuals, Candida species are considered commensal yeasts of the oral cavity. However, these microorganisms can also act as opportunist pathogens, particularly the so-called non-albicans Candida species that are increasingly recognized as important agents of human infection. Several surveys have documented increased rates of C. glabrata, C. tropicalis, C. guilliermondii, C. dubliniensis, C. parapsilosis, and C. krusei in local and systemic fungal infections. Some of these species are resistant to antifungal agents. Consequently, rapid and correct identification of species can pla

Feline meningitis due to Cryptococcus neoformans var. neoformans and review of feline cryptococcosis.

Cryptococcus neoformans var. neoformans was ascertained as the prime cause of fatal meningitis in a young-adult female domestic cat who was suspected of rabies. The pathogen was isolated in heavy growth from the infected brain specimen on simplified sunflower seed (Helianthus annuus) medium at 30 degrees C. Periodic acid-Schiff stained impression smear revealed numerous cryptococcal cells. Histologically, pia-arachnoid showed thickening along with many circular yeast cells with and without budding morphologically consistent with Cr. neoformans. The detailed typing of the strain indicated that

Association between fertility and molecular sub-type of global isolates of Cryptococcus gattii molecular type VGII.

The basidiomycetous yeast Cryptococcus gattii, is a primary pathogen which causes disease in apparently healthy humans and a wide range of animals. Recently, an outbreak of cryptococcosis caused by a previously uncommon genotype of C. gattii, VGII, emerged on Vancouver Island, British Columbia, Canada. Two pathogenic sub-types of VGII (designated VGIIa and VGIIb) were identified among these isolates. All of the isolates proved to be mating type alpha and had exceptionally high sporulation capacity. The common subtype, VGIIa, was more virulent than VGIIb in mice, suggesting a linkage between su

Identification of mucorales from clinical specimens: a 4-year experience in a single institution.

Mucormycosis, a fatal opportunistic infection in immunocompromised hosts, is caused by fungi belonging to the order Mucorales. Early diagnosis based on exact identification and multidisciplinary treatments is critical. However, identification of Mucorales fungi is difficult and often delayed, resulting in poor prognosis. This study aimed to compare the results of phenotypic and molecular identification of 12 Mucorales isolates collected from 4-yr-accumulated data. All isolates were identified on the basis of phenotypic characteristics such as growth rate, colony morphology, and reproductive st

Transmission of nocardiosis and molecular typing of Nocardia species: a short review.

Nocardia species are ubiquitous in the environment and may be found in the soil. They are generally responsible for sporadic pulmonary diseases acquired by inhalation of spores, with secondary localizations in the central nervous system and subcutaneous tissues. There is no absolute evidence for person to person transmission. Presumptive outbreaks of nocardiosis were observed in immunocompromised patients, more frequently in kidney transplant patients than in cardiac transplant patients. Nocardia spp., being present in dust particles, closure and disinfection of the transplantation unit with f

Molecular identification of pathogenic fungi.

Systemic fungal infections represent a major cause of morbidity and mortality in immunocompromised patients. The ever-increasing number of yeast species associated with human infections that are not covered by conventional identification kits, and the fact that moulds isolated from deep infections are frequently impossible to identify using classical methods due to lack of sporulation, has driven the need for rapid, robust molecular identification techniques. We recently developed a rapid method of preparing fungal genomic DNAs using Whatman FTA filters, which has greatly facilitated molecular

Importance of mycological confirmation of clinically suspected cases of tinea corporis, tinea pedis and tinea cruris.

Tinea corporis, tinea cruris, and tinea pedis are of the most prevalent dermatophytoses. Several conditions that mimic dermatophytoses and atypical and steroid modified forms of the disease usually present difficulties in diagnosis. Hence, the present investigation aimed at studying these conditions on mycological basis. The study included 163 cases clinically diagnosed as having tinea corporis, tinea pedis or tinea cruris. Specimens were taken by skin scraping. Samples were cultured on Sabouraud's dextrose agar and examined microscopically. The results revealed that, only 90.8% of cases were

Fungal Keratitis Caused by Scedosporium apiospermum in an Immunocompetent Patient: How Challenging Can Treatment get?

This case report underscores the complexities in managing fungal keratitis caused by Scedosporium apiospermum, even in an immunocompetent patient. The pathogen's resistance to many antifungal agents, its ability to invade deep ocular structures, and the presence of posterior chamber involvement added to the treatment challenges. Effective management requires not only timely diagnosis and targeted antifungal therapy, such as voriconazole, but also careful consideration of adjunctive treatments like corticosteroids and surgical interventions to address inflammation and posterior chamber involvem

Cryptococcus neoformans carried by Odontomachus bauri ants.

Cryptococcus neoformans is the most common causative agent of cryptococcosis worldwide. Although this fungus has been isolated from a variety of organic substrates, several studies suggest that hollow trees constitute an important natural niche for C. neoformans. A previously surveyed hollow of a living pink shower tree (Cassia grandis) positive for C. neoformans in the city of Rio de Janeiro, Brazil, was chosen for further investigation. Odontomachus bauri ants (trap-jaw ants) found inside the hollow were collected for evaluation as possible carriers of Cryptococcus spp. Two out of 10 ants we

Paracoccidioides brasiliensis. A mycologic and immunochemical study of two strains.

The authors conducted a mycologic, immunochemical and molecular biology study on two strains of Paracoccidioides brasiliensis, one of them, called IBIA, isolated from soil in the municipality of IBIA (Minas Gerais) by Silva-Vergara et al. (1996, 1998), and the other, BAT, cultivated from a human case of paracoccidioidomycosis in Ribeirão Preto (São Paulo/Brazil) by Freitas Da Silva (1996). Both strains showed cotton-like (M) and yeast-like (Y) forms and were pathogenic for testicularly inoculated guinea pigs, producing granulomatous and/or suppurative orchitis. Immunochemically was demonstrate

The Predominance of a Specific Genotype of Cryptococcus neoformans var. Grubii in China and Japan.

Источник пометил публикацию как отозванную

BACKGROUND: The extensive deployment of molecular genotyping methods is the top reliable keyword to characterize the population genetic structure of C. neoformans in the past decade. However, studies involving genotypic analysis of C. neoformans var. grubii from China and Japan are limited. OBJECTIVES: We address this challenge to determine the genotype distribution of C. neoformans var. grubii strains from China and Japan. METHODS: Genotypic analysis of 52 C. neoformans var. grubii isolates was performed using multilocus microsatellite typing (MLMT) based on the sequence analysis of 3 funct

Diagnosis of coccidioidomycosis by culture: safety considerations, traditional methods, and susceptibility testing.

The recovery of Coccidioides spp. by culture and confirmation utilizing the AccuProbe nucleic acid hybridization method by GenProbe remain the definitive diagnostic method. Biosafety considerations from specimen collection through culture confirmation in the mycology laboratory are critical, as acquisition of coccidioidomycosis by laboratory workers is well documented. The designation of Coccidioides spp. as select agents of potential bioterrorism has mandated strict regulation of their transport and inventory. The genus appears generally susceptible, in vitro, although no defined breakpoints

Prevalence, distribution and antifungal susceptibility profiles of Candida parapsilosis, Candida orthopsilosis and Candida metapsilosis bloodstream isolates.

The Candida parapsilosis group encompasses three species: C. parapsilosis, Candida orthopsilosis and Candida metapsilosis. These species are phenotypically indistinguishable, and molecular methods are needed for their detection. We analysed 152 unique blood culture isolates of the C. parapsilosis group obtained during 1997-2011. The isolates were screened by PCR amplification of the gene encoding secondary alcohol dehydrogenase, followed by digestion with the restriction enzyme BanI. Isolates with RFLP patterns distinct from those of the C. parapsilosis group were characterized as C. parapsilo

Rapid identification of black grain eumycetoma causative agents using rolling circle amplification.

Accurate identification of mycetoma causative agent is a priority for treatment. However, current identification tools are far from being satisfactory for both reliable diagnosis and epidemiological investigations. A rapid, simple, and highly efficient molecular based method for identification of agents of black grain eumycetoma is introduced, aiming to improve diagnostic in endemic areas. Rolling Circle Amplification (RCA) uses species-specific padlock probes and isothermal DNA amplification. The tests were based on ITS sequences and developed for Falciformispora senegalensis, F. tompkinsii,
