Most plant species can form symbioses with arbuscular mycorrhizal fungi (AMFs), which may enhance the host plant's acquisition of soil nutrients. In contrast to phosphorus nutrition, the molecular mechanism of mycorrhizal nitrogen (N) uptake remains largely unknown, and its physiological relevance is unclear. Here, we identified a gene encoding an AMF-inducible ammonium transporter, ZmAMT3;1, in maize (Zea mays) roots. ZmAMT3;1 was specifically expressed in arbuscule-containing cortical cells and the encoded protein was localized at the peri-arbuscular membrane. Functional analysis in yeast an

Metalliferous and aluminium (Al)-rich soils represent restrictive terrestrial environments where high concentrations of potentially toxic elements act as edaphic filters on plant communities. Since arbuscular mycorrhizal (AM) symbiosis can alleviate several environmental constraints, it may play an important role in plant performance in metalliferous and Al-rich soils. Here, we review current knowledge on the mechanisms and ecological relevance of AM symbiosis in naturally metalliferous and Al-rich ecosystems worldwide. Available evidence indicates that AM fungi (AMF) mitigate both trace metal

Uncovering the identity of "dark fungi" is crucial for understanding fungal taxonomy, diversity, and phylogenetic relationships. Previous exploratory analysis of the mycorrhizal fungi of the partially mycoheterotrophic orchid Nervilia nipponica revealed an association with a particular group of Agaricomycetes, tentatively designated "unidentified Agaricomycetes X" (uAX). Two culture strains were successfully isolated from the mycorrhiza, which enabled the phylogenetic position of this fungus to be determined using ultrastructural and multigene phylogenetic analyses. Transmission electron micro

Grassland degradation decreases ecosystem productivity and diminishes soil biodiversity, leading to the loss of beneficial microorganisms. Arbuscular mycorrhizal fungi (AMF) play a critical role in ecosystem functioning, being a key link between plant and microbial communities, soil, and vegetation. Here, we evaluated the potential of increasing the productivity of degraded grassland by AMF inoculation. A gradient of soil biodiversity: complete sterilization, low, moderate, and high biodiversity was established using the dilution-to-extinction approach. Grassland microcosms under greenhouse co

The rhizosphere is a narrow and dynamic soil zone where bacteria and fungi establish symbiotic and synergistic relationships with each other and with plants, collectively influencing nutrient cycling, soil health, and overall ecosystem functioning. Mycorrhiza Helper Bacteria (MHB) are a diverse group of bacterial taxa that promote mycorrhiza establishment, together with those that positively influence already established symbiotic associations. MHB have been largely described from the perspectives of interaction mechanisms, ecological functions, or agricultural applications, treating them as g

Most land plants engage in arbuscular mycorrhiza (AM) symbiosis with Glomeromycotina fungi for better access to mineral nutrients. The plant hormone ethylene suppresses AM development, but a molecular explanation for this phenomenon is lacking. Here we show that ethylene inhibits the expression of many genes required for AM formation in Lotus japonicus. These genes include strigolactone biosynthesis genes, which are needed for fungal activation, and Common Symbiosis genes, which are required for fungal entry into the root. Application of strigolactone analogs and ectopic expression of the Comm

The majority of land plants acquire soil nutrients, such as phosphorus and nitrogen, not only through the root surface but also through arbuscular mycorrhizal (AM) fungi. Soybean is the most important leguminous crop in the world. We found 16 ammonium transporter genes in the soybean genome, five of which are AM inducible. Among them, promoter-reporter analysis indicated that the most abundantly transcribed gene, GmAMT4.1, showed specific expression in arbusculated cortical cells. Moreover, the GmAMT4.1-green fluorescent protein fusion was localized on the branch domain of periarbuscular membr

In arbuscular mycorrhizal symbiosis, the belowground mycelium that develops into the soil not only provides extensive pathways for nutrient and signal fluxes, the occupation of different niches and dispersal of propagules but also has strong influences upon biogeochemical cycling. By providing a valuable overview of protein expression, shotgun proteomics can help decipher key metabolic pathways involved in the functioning of fungal mycelia. In this protocol, we describe the combination of extra-radical mycelium growth systems with gel-based extraction of fungal peptides amenable to shotgun pro

Increasing demands to reduce fertilizer and pesticide input in agriculture has triggered interest in arbuscular mycorrhizal fungi (AMF) that can enhance plant growth and confer mycorrhiza-induced resistance (MIR). MIR can be based on a variety of mechanisms, including induction of defense compounds, and sensitization of the plant's immune system (priming) for enhanced defense against later arriving pests or pathogens signaled through jasmonic acid (JA). However, growth and resistance benefits of AMF highly depend on environmental conditions. Low soil P and non-limiting light conditions are exp

Rhizosphere effect of nanoscale zero-valent iron (nZVI) is crucial but little reported. Maize seeds were dressed with four nZVI concentrations (0, 1.0, 1.5, 2 g kg-1 ) and inoculated with arbuscular mycorrhizal fungus (AMF) (Funneliformis mosseae). The SEM images illuminated that excessive nZVI particles (2 g kg-1 ) were agglomerated on the surface of hyphae and spore, causing severe deformation and inactivation of AMF symbionts and thereafter inhibiting water uptake in maize seedlings. This restrained the scavenging effects of enzymatic (superoxide dismutase, peroxidase) and non-enzymatic com

Annual wormwood (Artemisia annua L.) produces an array of complex terpenoids including artemisinin, a compound of current interest in the treatment of drug-resistant malaria. However, this promising antimalarial compound remains expensive and is hardly available on the global scale. Synthesis of artemisinin has not been proved to be feasible commercially. Therefore, increase in yield of naturally occurring artemisinin is an important area of investigation. The effects of inoculation by two arbuscular mycorrhizal (AM) fungi, Glomus macrocarpum and Glomus fasciculatum, either alone or supplement

Arbuscular mycorrhiza (AM) symbiosis occurs between obligate biotrophic fungi of the phylum Glomeromycota and most land plants. The exchange of nutrients between host plants and AM fungi (AMF) is presumed to be the main benefit for the two symbiotic partners. In this review article, we outline the current concepts of nutrient exchanges within this symbiosis (mechanisms and regulation). First, we focus on phosphorus and nitrogen transfer from the fungal partner to the host plant, and on the reciprocal transfer of carbon compounds, with a highlight on a possible interplay between nitrogen and ph

Variation in arbuscular mycorrhizal fungal (AMF) root colonization and associated root traits among plant species plays a crucial role in determining plant community responses to environmental change. However, species mean trait values often obscure substantial variation among individuals within a species. Because trait-based ecology has focused predominantly on interspecific differences, the intraspecific responses of AMF-associated traits to resource limitations remain poorly understood. To address this gap, we examined intraspecific variation in AMF-associated traits among genetically diver

Arbuscular mycorrhizal (AM) symbiosis in soil may be directly or indirectly involved in the reproductive process of sexually reproducing plants (seed plants), and affect their reproductive fitness. However, it is not clear how underground AM symbiosis affects plant reproductive function. Here, we reviewed the studies on the effects of AM symbiosis on plant reproductive fitness including both male function (pollen) and female function (seed). AM symbiosis regulates the development and function of plant sexual organs by affecting the nutrient using strategy and participating in the formation of

Lotus japonicus THIC is expressed in all organs, and the encoded protein catalyzes thiamine biosynthesis. Loss of function produces chlorosis, a typical thiamine-deficiency phenotype, and mortality. To investigate thiamine's role in symbiosis, we focused on THI1, a thiamine-biosynthesis gene expressed in roots, nodules, and seeds. The thi1 mutant had green leaves, but formed small nodules and immature seeds. These phenotypes were rescued by THI1 complementation and by exogenous thiamine. Thus, THI1 is required for nodule enlargement and seed maturation. On the other hand, colonization by arbus

Plants are often exposed to shade over different time scales and this may substantially affect not only their own growth, but also development and functioning of the energetically dependent organisms. Among those, the root symbionts such as arbuscular mycorrhizal (AM) fungi and rhizobia represent particularly important cases-on the one hand, they consume a significant share of plant carbon (C) budget and, on the other, they generate a number of important nutritional feedbacks on their plant hosts, often resulting in a net positive effect on their host growth and/or fitness. Here we discuss our

Arbuscular mycorrhizal (AM) fungi are known to have beneficial effects on host plants growing on contaminated soils. The present study aimed at investigating the influence of two different AM fungi (Rhizophagus intraradices and Funneliformis mosseae) on the growth of plants and Cu uptake by Mimosa pudica L. grown in polluted soils containing various levels of Cu (Control, 400, 500, or 600 mg kg-l soil) in pot experiments. Mycorrhizal colonisation rates by the two AM fungi decreased markedly with the increasing Cu levels in soils. This inhibition was more pronounced to F. mosseae than R. intrar

Heavy metal-contaminated land is increasingly becoming an important environmental, health, economic, and planning issue in Pakistan. The unplanned disposal of industrial effluent from tannery, for example, has resulted in a many fold increase in chromium (Cr) in the land near a tannery. This study was undertaken to compare the total and the DTPA-available Cr contents in the soil and the roots and leaves of tree species growing on it with those on the nearby noncontaminated reference site at Kala Shah Kakoo, Panjab, Pakistan. A very reduced plant cover on the tannery effluent-contaminated site

Arbuscular mycorrhizal fungi enhance plant resistance against necrotrophic fungi by priming jasmonic acid-dependent defences. Systemin is a solanaceous peptide hormone that mediates resistance against Botrytis cinerea, and JA signalling is critical for this systemin-mediated defence. Mycorrhizal symbiosis primes prosystemin expression in the tomato (Solanum lycopersicum)-B. cinerea pathosystem. Whether prosystemin or systemin mediates mycorrhiza-induced resistance (MIR) against B. cinerea remains unclear. We hypothesise that systemin mediates JA priming during MIR. We integrated transcriptiona

Molecular responses to cadmium (Cd) stress were studied in mycorrhizal and non-mycorrhizal Pisum sativum L. cv. Frisson inoculated with Glomus intraradices. Biomass decreases caused by the heavy metal were significantly less in mycorrhizal than in non-mycorrhizal plants. Real-time reverse transcriptase-polymerase chain reaction showed that genes implicated in pathways of Cd detoxification varied in response to mycorrhiza development or Cd application. Expression of a metallothionein-encoding gene increased strongly in roots of Cd-treated non-mycorrhizal plants. Genes encoding gamma-glutamylcys
