Human Carbonic Anhydrase (hCA) IX is a membrane-associated member of the CA enzyme family, involved in solid tumor acidification. This enzyme is a marker of tumor hypoxia and a prognostic factor for several human cancers. In a recent paper, we showed that CA IX interacts with cullin-associated NEDD8-dissociated protein 1 (CAND1), a nuclear protein involved in gene transcription and assembly of SCF ubiquitin ligase complexes. A functional role for this interaction was also identified, since lower CA IX levels were observed in cells with decreased CAND1 expression via shRNA-mediated interference

The molecular mechanism involved in the exocytosis of arginine vasopressin (AVP) is not fully known. Rabphilin-3A has been suggested as a novel autoantigen in infundibulo-neurohypophysitis (LINH), which leads to central diabetes insipidus through insufficient secretion of AVP. However, the role of rabphilin-3A in the pathogenesis of LINH remains unclear. Thus, the aim of the present study was to identify proteins binding rabphilin-3A in the posterior pituitary. Using glutathione S-transferase (GST)-pulldown assays and proteomic analyses, cullin-associated NEDD8-dissociated protein 1 (CAND1) wa

Nedd8 is a ubiquitin-like protein that covalently conjugates to target proteins through neddylation. In addition to cullin-RING ligases, neddylation also modifies non-cullin proteins to regulate protein activity, stability and localization. However, the roles of NEDD8 remain largely unknown in vivo Here, we found that loss of nedd8 in female zebrafish led to defects in oogenesis, disrupted oocyte maturation and stimulated growth of the breeding tubercles (BTs) on the pectoral fins. The BTs are normally present in males, not females. However, the loss of one copy of ar can partially rescue the

PURPOSE: Ischemic stroke (IS) causes severe brain tissue damage characterized by focal neuronal injury. Emerging evidence implicates ferritinophagy-mediated ferroptosis as a critical contributor to ischemic neuronal damage. This study aimed to investigate the role of NEDD8 in regulating ferritinophagy and ferroptosis in neurons under IS, with particular emphasis on elucidating the NEDD8/NRF2/FTH1 signaling axis in IS.
METHODS: Murine hippocampal neuronal cells (HT22) were subjected to OGD to simulate ischemic injury in vitro. NEDD8 expression was suppressed using siRNA-mediated knockdown. Cel

The objective of our study was to develop a genetically encoded biosensor for quantification of Nedd8, a post-translational modifier that regulates cellular signals through conjugation to other proteins. Perturbations in the balance of free (i.e., unconjugated) and conjugated Nedd8 caused by defects in Nedd8 enzymes or cellular stress are implicated in various diseases. Despite the biological and biomedical importance of Nedd8 dynamics, no method exists for direct quantification of free Nedd8, hindering the study of Nedd8 and activities of its associated enzymes. Genetically encoded biosensors

The pathogenetic bases of HAART-associated lipodystrophy are still poorly known, even if it is clear that adipose tissue and its metabolism are sensitive to antiretroviral therapy alone and/or in combination with HIV infection. The NEDD8 system is essential for the regulation of protein degradation pathways involved in cell cycle progression, morphogenesis and tumorigenesis. We investigated the possible involvement of NEED8 in adipogenesis and, consequently, in HIV-related lipodystrophy. One hundred HIV-1-infected patients were included in the study. Using an in vitro model of adipogenesis we

BACKGROUND: Radiotherapy is the first-line regimen for treating oral squamous cell carcinoma (OSCC) in current clinics. However, the development of therapeutic resistance impacts the anticancer efficacy of irradiation in a subpopulation of OSCC patients. As a result, discovering a valuable biomarker to predict radiotherapeutic effectiveness and uncovering the molecular mechanism for radioresistance are clinical issues in OSCC.
METHODS: Three OSCC cohorts from The Cancer Genome Atlas (TCGA), GSE42743 dataset and Taipei Medical University Biobank were enrolled to examine the transcriptional lev

Aloin (ALO), an anthraquinone derived from Aloe vera, exhibits antitumor activity; however, its precise mechanisms of action remain unclear. In this study, in silico molecular docking analysis first revealed that Aloin (ALO) bound effectively to ferroptosis-related proteins (SLC7A11, GPX4, ACSL4, and TFR1). Subsequently, in vitro ALO treatment triggered ferroptosis hallmarks in human cervical cancer cell line Hela and mouse colon cancer cell line MC38 at both transcriptional and protein levels-downregulating SLC7A11/GPX4, upregulating ACSL4/TFR1, with Fe2+/ROS accumulation, GSH depletion, and

Inflammation is a key factor leading to secondary brain injury after intracerebral hemorrhage (ICH). Neddylation, a post-translational modification that attaches NEDD8 (neuronal precursor cell-expressed developmentally downregulated protein 8) to targets, regulates diverse cellular processes. Neddylation is overactivated in various cancers and linked to atherosclerosis and inflammatory disorders, but its role in ICH remains unclear. In this study, mice were subjected to ICH by injection of collagenase. The NEDD8-activating enzyme inhibitor MLN4924 was administered subcutaneously post-ICH. We f

The pathogenesis of Coxsackievirus B3 (CVB3), the causative pathogen for severe diseases such as myocarditis, pancreatitis, and meningitis, remains largely unknown. Neddylation, the covalent modification of proteins through the conjugation of NEDD8, a ubiquitin-like molecule, plays critical roles in regulating cellular activities. Our previous study demonstrated that 3Dpol, the RNA-dependent RNA polymerase of CVB3, was modified by ubiquitin. Since NEDD8 is highly homologous to ubiquitin in both the sequence and structure, we hypothesized that 3Dpol may also undergo neddylation. Here, we demons

PURPOSE: Keratoconus (KC) is an adolescent-onset vision-impairing corneal disorder with incompletely elucidated pathogenesis. This study aims to elucidate the regulatory role of NEDD8 in the Hippo-YAP signaling pathway and its influence on the corneal pathology, thereby providing a theoretical basis for mechanistic studies and targeted therapy of KC.
METHODS: Single-cell RNA sequencing (scRNA-seq) analysis of central corneas was performed to identify differentially expressed molecules and signaling pathways in KC. Then the expression changes and correlations of key regulatory molecules were v

A high-glucose microenvironment is a key feature of tumor metabolic reprogramming that promotes lung cancer cell progression. Previous studies have revealed that protein arginine methyltransferase 5 (PRMT5) is overactivated in various cancers and regulates neddylation of the tumor suppressor phosphatase and tensin homolog (PTEN). This study established a high-glucose (25 mM) in vitro model using human non-small cell lung cancer A549 cells. Cell proliferation, invasion and cell cycle progression were assessed using the cell counting kit-8, Transwell, 5-ethynyl-2'-deoxyuridine and flow cytometry

Neddylation is a ubiquitin-like post-translational protein modification. It occurs via the activation of the neural precursor cell expressed, developmentally downregulated protein 8 (NEDD8) by three enzymes: activating enzyme, conjugating enzyme, and ligase. NEDD8 was first isolated from the mouse brain in 1992 and was initially considered important for the development and differentiation of the central nervous system. Previously, the downregulation of neddylation was associated with some human diseases, such as neurodegenerative disorders and cancers. In recent years, neddylation has also bee

A variety of post-translational modifications of Plasmodium falciparum proteins, including phosphorylation and ubiquitination, are shown to have key regulatory roles during parasite development. NEDD8 is a ubiquitin-like modifier of cullin-RING E3 ubiquitin ligases, which regulates diverse cellular processes. Although neddylation is conserved in eukaryotes, it is yet to be characterized in Plasmodium and related apicomplexan parasites. We characterized P. falciparum NEDD8 (PfNEDD8) and identified cullins as its physiological substrates. PfNEDD8 is a 76 amino acid residue protein without the C-

Protein post translational modification (PTM) is the main regulatory mechanism for eukaryotic cell function, among which ubiquitination is based on the reversible degradation of proteins by the ubiquitin proteasome system to regulate cell homeostasis. The neural precursor cell expressed developmental downregulated gene 8 (NEDD8) is a kind of ubiquitin like protein that shares 80 % homology and 60 % identity with ubiquitin. The PTM process by covalently binding NEDD8 to lysine residues in proteins is called neddylation. The neddylation reaction could be regulated by NEDD8, its precursors, subst

Polycystic ovary syndrome (PCOS) is a common and complex endocrine and metabolic disorder whose etiology has not yet been fully elucidated. Emerging research has revealed an association between PCOS pathogenesis and post-translational modifications of proteins. However, the effect of neddylation on PCOS pathogenesis has not been investigated. In this study, key proteins of the neddylation pathway (Nedd8, neddylation-activating enzyme (NAE), and Cullin-1/2/3) were significantly upregulated in the granulosa cells of patients with PCOS; this was accompanied by enhanced apoptosis (increased Bax an

Diabetic nephropathy (DN) is a common and serious complication of diabetes, characterized by chronic fibro-inflammatory processes with an unclear pathogenesis. Renal fibrosis plays a significant role in the development and progression of DN. While recent research suggests that the neddylation pathway may influence fibrotic processes, its specific dysregulation in DN and the underlying mechanisms remain largely unexplored. This study identified the neddylation of RhoA as a novel post-translational modification that regulates its expression and promotes renal fibrosis in DN. We here demonstrated

NEDD8 is an ubiquitin-like molecule that covalently binds to target proteins through an enzymatic cascade analogous to ubiquitylation. This modifier is known to bind to p53 and p73, as well as all Cullin family proteins, which are essential components of Skp1/Cul-1/F-box protein (SCF)-like Ub ligase complexes. Here, we focused on a genomic analysis of the genes involved in the NEDD8 conjugation pathway in Schistosoma mansoni. The results revealed seven genes related to NEDD8 conjugation that are conserved in Schistosoma japonicum, Caenorhabditis elegans, Drosophila melanogaster and Homo sapien
