Structure and Dynamics of Confined Water Inside Diphenylalanine Peptide Nanotubes.

Diphenylalanine (FF) peptides exhibit a unique ability to self-assemble into nanotubes with confined water molecules playing pivotal roles in their structure and function. This study investigates the structure and dynamics of diphenylalanine peptide nanotubes (FFPNTs) using all-atom molecular dynamics (MD) and grand canonical Monte Carlo combined with MD (GCMC/MD) simulations with both the CHARMM additive and Drude polarizable force fields. The occupancy and dynamics of confined water molecules were also examined. It was found that less than 2 confined water molecules per FF help stabilize the

Controlled patterning of peptide nanotubes and nanospheres using inkjet printing technology.

Peptide nanostructures are expected to serve as a major tool in future nanotechnological applications owing to their excellent self-assembly properties, biological and chemical flexibility and structural simplicity. Yet one of the limiting factors for the integration of peptide assemblies into functional electro-organic hybrid devices is the controlled patterning of their assemblies. Here we report the use of inkjet technology for the application of peptide nanostructures on nonbiological surfaces. The aromatic dipeptides nanotubes (ADNT) which readily self-assemble in solution were used as an

Thermal and chemical stability of diphenylalanine peptide nanotubes: implications for nanotechnological applications.

The diphenylalanine peptide, the core recognition motif of the beta-amyloid polypeptide, efficiently self-assembles into discrete, well-ordered nanotubes. Here, we describe the notable thermal and chemical stability of these tubular structures both in aqueous solution and under dry conditions. Scanning and transmission electron microscopy (SEM and TEM) as well as atomic force microscopy (AFM) revealed the stability of the nanotubes in aqueous solution at temperatures above the boiling point of water upon autoclave treatment. The nanotubes preserved their secondary structure at temperatures up

Anti-Invasive Peptide-Functionalized Nanotubes for Selective c‑Met Targeting and Metal Chelation.

This work describes the development of a sustainable and biocompatible drug delivery system using halloysite nanotubes (HNTs). The goal was to create a material that could both deliver the chemotherapy drug doxorubicin (DOX) and selectively target the tumor cells. The synthesis process was made environmentally friendly by using tetrahydropyran (Thp) as a solvent. The HNTs were first functionalized with an amino group using (3-aminopropyl)-triethoxysilane (APTES), followed by the conjugation of two different peptides, P1 and P2, via EDC coupling. Successful functionalization was confirmed by FT

Hierarchical Peptide Functionalized Titania Nanotubes: Improving Stability, Promoting Osteogenesis, and Reducing Infection Risk.

In this study, a hierarchical surface functionalization strategy was developed that combines engineering of titania nanotubes (TiNTs) on the titanium surface, followed by dual-mode peptide functionalization. Fmoc-FF-OH dipeptide was first covalently anchored onto the TiNT surface, then a secondary layer of the self-assembling FF dipeptide was physically deposited. This technique harnesses the strength of covalent bonds and the extensive surface coverage provided by self-assembling peptides to create a long-lasting, bioactive, and functional surface that promotes superior biocompatibility. Self-assembled peptide nanotubes (SPNTs)/SnO2nanocomposites for high-performance NO2sensing at room temperature.

Nitrogen dioxide (NO2) is a major pollutant that poses significant risks to sustainable human life. As a result, a growing focus has been placed on the development of highly selective and sensitive gas sensors for NO2. Traditional cutting-edge non-organic NO2gas detectors often necessitate stringent production conditions and potentially harmful materials, which are not environmentally friendly, and these shortcomings have limited their widespread practical use. To overcome these challenges, we synthesized self-assembled peptide nanotubes (SPNTs) through a molecular self-assembly process. The S

Short peptide based nanotubes capable of effective curcumin delivery for treating drug resistant malaria.

BACKGROUND: Curcumin (Ccm) has shown immense potential as an antimalarial agent; however its low solubility and less bioavailability attenuate the in vivo efficacy of this potent compound. In order to increase Ccm's bioavailability, a number of organic/inorganic polymer based nanoparticles have been investigated. However, most of the present day nano based delivery systems pose a conundrum with respect to their complex synthesis procedures, poor in vivo stability and toxicity issues. Peptides due to their high biocompatibility could act as excellent materials for the synthesis of nanoparticula

Self-Assembling Cyclic Peptide Nanotubes for the Delivery of Doxorubicin into Drug-Resistant Cancer Cells.

Synthetic antimicrobial cyclic peptides conjugated to an antitumoral drug are used against drug-resistant cancer cells for a combined drug delivery strategy. The antimicrobial peptides are based on nanotube-forming cyclic peptides of alternating chirality, whose amphipathic and cationic characteristics determine their propensity to mainly interact with cell membranes rich in anionic phospholipids. This affinity triggers the formation of a supramolecular structure capable of destabilizing cell membranes such as those present in endosomes, thereby facilitating the delivery of the therapeutic age

Multifunctional cyclic biomimetic peptides: Self-assembling nanotubes for effective treatment of sepsis.

Antibiotic abuse has led to an increasingly serious risk of antimicrobial resistance, developing alternative antimicrobials to combat this alarming issue is urgently needed. Rhesus theta defensin-1 (RTD-1) is a theta-defensin contributing to broad-spectrum bactericidal activity via the mechanisms of membrane perturbation. Intriguingly, human defensin-6 (HD6), an enteric defensin secreted by Paneth cells without direct bactericidal effect, could self-assembled into fibrous networks to trap enteric pathogens for assistance of innate immunity. The direct bactericidal action of RTD-1 and the bacte

Glucose derivatives substitution and cyclic peptide diameter effects on the stability of the self-assembled cyclic peptide nanotubes; a joint QM/MD study.

Dynamical behavior and the stability of eighteen nanostructures composed of cyclic peptide (CP) with the general structure of the cyclo(CO(CH2)n=4, 6, 10COCyst), in the gas phase, water and chloroform were investigated during 50ns molecular dynamic (MD) simulations. CP dimers and cyclic peptide nanotubes (CPNTs) are more stable in chloroform than water and this stability is reversely correlated with the ring size of the CP units. Also the effect of glucose derivatives substitution, d-glucose (S1) and N-methyl-d-glucamine (S2), on the stability and other physicochemical properties of the CP dim

Synthetic non-metallic nanoparticles (NMNPs) such as carbon nanotubes (CNTs), carbon nanodots (CNDs), and cell-penetrating peptides (CPPs) have been explored to treat harmful algal blooms. However, their strain-specific algicidal activities have been rarely investigated. Here we determined their acute toxicity to nine freshwater cyanobacterial strains belonging to seven genera, including Microcystis aeruginosa UTEX 2386, M. aeruginosa UTEX 2385, M. aeruginosa LE3, Anabaena cylindrica PCC 7122, Aphanizomenon sp. Carbon nanotube composite peptide-based biosensors as putative diagnostic tools for rheumatoid arthritis.

This work reports on the fabrication and performance of a simple amperometric immunosensor device to be potentially used for the detection of serum anti-citrullinated peptide antibodies (ACPAs), which are specific for rheumatoid arthritis (RA) autoimmune disease. Sera of RA patients contain antibodies to different citrullinated peptides and proteins such as fibrin or filaggrin. Herein, a chimeric fibrin-filaggrin synthetic peptide (CFFCP1) was used as a recognition element anchored to the surface of a multiwalled carbon nanotube-polystyrene (MWCNT-PS) based electrochemical transducer. Fucosyltransferase 4-derived peptide bioconjugates on carbon nanotubes enhance antitumor immunity in an ovarian cancer mouse model.

Ovarian cancer is characterized by a profoundly immunosuppressive tumor microenvironment, which has limited the clinical success of immunotherapy approaches. Antigen-based nanovaccines represent a promising strategy to enhance antitumor immunity by improving antigen delivery and immune activation. In this study, we evaluated carbon nanotubes (CNTs) bioconjugated with a peptide derived from tumor-associated antigen fucosyltransferase 4 (FUT4) as an immunomodulating platform in an immunocompetent ovarian cancer mouse model. FUT4 expression was confirmed in the ID8-Defb29/Vegf-a (ID8DVLuc) ovaria

Multivalent Display of ApoAI Peptides on the Surface of Tobacco Mosaic Virus Nanotubes Improves Cholesterol Efflux.

Atherosclerosis is a progressive cardiovascular disease in which cholesterol-rich plaques build up within arteries, increasing the risk of thrombosis, myocardial infarction, and stroke. One promising therapeutic approach is the use of high-density lipoprotein (HDL) biomimetic formulations based on ApoAI peptides that promote cholesterol efflux from plaques, ultimately leading to cholesterol excretion. Here, we describe the multivalent display of ApoAI peptides on the surface of protein nanotubes derived from the plant virus tobacco mosaic virus (TMV) and protein nanoparticles using virus-like

Separation of chloroform from a dilute solution using a cyclic peptide nanotube: A molecular dynamics study.

This work firstly explored the potential application of a cyclic peptide nanotube (CPNT) in the separation of chloroform from a dilute solution. Four hydrophobic CPNTs of 8 × (WL)4,5 and 8 × (AL)4,5 all exhibit excellent adsorption characteristics to chloroform. The CPNT diameter, side chain structures and the concentration of chloroform in a solution all affect the adsorption characteristics of chloroform. CHCl3 molecules are overwhelmingly adsorbed on the surfaces of these CPNTs as a cluster, and sporadically reside inside the channels, consistent with the chloroform's potentials of mean for

Antifouling Cyclic Peptide Nanotubes Combined with Rh-Ti3C2TX Multifunctional Nanozyme for the Sensitive Detection of CA125 in Complex Biological Samples.

Although linear peptides exhibit advantageous physicochemical properties and biocompatibility for antifouling biosensing applications, the intrinsic structural flexibility of natural linear peptides makes them susceptible to enzymatic degradation, compromising their long-term practical applications. To address these limitations, cyclic peptide nanotubes (CPNTs) were prepared via a self-assembly strategy of cyclic peptides, which imparted exceptional enzymatic stability due to their intramolecular hydrogen bonds and rigid tubular structure. Moreover, the densely arranged hydrophilic residues on
