Phenotypic and genotypic analysis of benzimidazole resistance in the ovine parasite Nematodirus battus.

Benzimidazole resistance is common amongst many ovine trichostrongylid nematodes species globally. Although anthelmintics have been used for over half a century in some areas of the world for the control of Nematodirus battus, resistance has never been detected. Veterinary investigations conducted in 2010 demonstrated reduced efficacy in a flock that had been treated previously with fenbendazole (FBZ), suggesting probable resistance in N. battus. Infective larvae (L3; designated MNba2) were generated from the original material to conduct a controlled efficacy test (CET). Faecal egg counts show

Demonstration of IgA-containing cells in the small intestine of lambs infected with Nematodirus battus.

A peroxidase anti-peroxidase immunohistochemical technique was used to demonstrate IgA-containing cells in the small intestine of lambs infected with Nematodirus battus. These cells were more numerous in the infected lambs than in the uninfected animals. The difference was greatest for the first three sites, 1 to 3 m distal to the pylorus, where the preponderance of the N battus infection is found. It is suggested that future immunohistochemical studies on the relationships between IgA and resistance to N battus should be directed to this area.

Acquired immunity mechanisms in sheep infected with Nematodirus oiratianus: key genes and host responses.

BACKGROUND: Controlling gastrointestinal nematode infections poses a major challenge in intensive farming, worsened by rising anthelmintic resistance. Developing innovative control strategies is critical. Sheep acquired immunity to Nematodirus oiratianus offers a valuable model for dissecting host-nematode interactions, though underlying immune mechanisms remain unclear. METHODS: This study employed multi-omics analyses (transcriptome and proteome) of lamb duodenum across infection stages, validated by RT-qPCR, iELISA, histopathological staining (HE), and electron microscopy. RESULTS AND CON

Refugia, climatic conditions and farm management factors as drivers of adaptation in Nematodirus battus populations.

Parasites are known for their ability to rapidly adapt to changing conditions. For parasitic helminths, changes in climate, along with farming and management practices associated with the intensification of livestock farming, provide novel challenges which can impact on their epidemiology and control. The sustainability of livestock production partially relies on effective control of helminth infection. Therefore, understanding changes in parasite behaviour, and what drives these, is of great importance. Nematodirus battus is an economically important helminth in the UK and temperate regions.

16S rDNA-based detection technology: use in lambs infected with Nematodirus oiratianus to analyze changes in intestinal flora.

The nematode Nematodirus oiratianus is associated with major economic losses in the livestock industry, as it is a common gastrointestinal parasites of cattle, sheep, and other ruminants. These parasites primarily obtain nutrients by consuming the blood of their host. This study aimed to investigate changes in the intestinal microbiota of lambs infected with N. oiratianus before and after infection, using 16S rDNA sequencing technology. We aimed to reveal the impact of N. oiratianus infection on lamb intestinal microecology and to provide scientific evidence for the prevention and control of r

Large scale screening for benzimidazole resistance mutations in Nematodirus battus, using both pyrosequence genotyping and deep amplicon sequencing, indicates the early emergence of resistance on UK sheep farms.

Benzimidazoles (BZ) have been the anthelmintic of choice for controlling Nematodirus battus infections since their release in the 1950s. Despite heavy reliance on this single anthelmintic drug class, resistance was not identified in this nematode until 2010 (Mitchell et al., 2011). The study aimed to explore the prevalence of BZ-resistance mutations in N. battus from UK sheep flocks using deep amplicon sequencing and pyrosequencing platforms. Based on evidence from other gastrointestinal nematodes, resistance in N. battus is likely to be conferred by single nucleotide polymorphisms (SNP) withi

In an effort to generate data for regulatory purposes on the therapeutic efficacy of eprinomectin 5% w/v extended-release injection (Eprinomectin ERI) (LongRange®, Boehringer Ingelheim) against infections of immature and adult stages of some nematode species (or stages) which are generally less common or predominantly seen in younger cattle, nine studies (minimum two per parasite and stage) were conducted in the USA and Germany. A total of 218 young cattle were included in seven experimentally induced infection studies (180 animals) and two studies with naturally acquired nematode infections (

Dynamics of endoparasites in farmed fallow deer (Dama dama) from birth to puberty.

Rectal faecal samples from farmed fallow deer were examined for eggs, larvae and oocysts of gastro-intestinal and pulmonary parasites from July 1992, then the fawns were 6 weeks old, to September 1993 in four weekly intervals. The investigations showed a succession of species: Trichuris spp., Nematodirus roscidus and Capillaria bovis had their peaks in egg output in August 1992, September 1992 and February 1993, respectively. After reaching peaks the faecal output of Trichuris spp. and Nematodirus roscidus eggs showed a marked decline. The shedding of Dictyocaulus larvae had its highest peak i

Association between variation in faecal egg count for a natural mixed field-challenge of nematode parasites and TLR4 variation.

Research has shown that Toll-like receptor 4 (TLR4) is important in immune responses to some helminth parasites. In sheep, variation in the PAMP region of TLR4 may result in structurally and thus functionally different TLR4 molecules, and this may consequently lead to variation in the TLR4 response to parasite infections. This study involved three separate, but related sheep breeds (Merino, Polwarth and Corriedale sheep) and a total of 885 lambs from five New Zealand farms that underwent a mixed field-challenge from gastro-intestinal parasites. Faecal samples were collected at approximately 4

Effects of age, sex, lactation and social dominance on faecal egg count patterns of gastrointestinal nematodes in farmed eland (Taurotragus oryx).

The eland is a large African antelope that can be bred in a temperate climate, under similar conditions and production systems as cattle. However, knowledge of parasites in farmed elands outside the area of their native habitat is still limited, and information concerning factors that influence these parasites is lacking. Therefore, faecal samples from an entire herd of elands, including calves and adult females and males, were examined monthly over a one year period. Almost 84% of the animals were found to be positive for gastrointestinal nematodes. Strongyle-type eggs were most frequently de

Helminth infections in alpacas (Vicugna pacos), husbandry and worm control practices in South American Camelids in Italy.

The recent global expansion of alpaca breeding, including in Italy, has driven growing interest in this species, making further research necessary to support their health and management. Gastrointestinal helminths are among the main health concerns, causing severe disease that affects welfare and productivity. In Europe, data on helminth infections in South American Camelids (SACs) are still limited. This study aimed to assess the prevalence, distribution, and risk factors for helminth infections in alpacas in Italy, and evaluate the husbandry and worm control practices through a structured qu

Refugia or reservoir? Feral goats and their role in the maintenance and circulation of benzimidazole-resistant gastrointestinal nematodes on shared pastures.

Gastrointestinal nematodes threaten the productivity of grazing livestock and anthelmintic resistance has emerged globally. It is broadly understood that wild ruminants living in sympatry with livestock act as a positive source of refugia for anthelmintic-susceptible nematodes. However, they might also act as reservoirs of anthelmintic-resistant nematodes, contributing to the spread of anthelmintic resistance at a regional scale. Here, we sampled managed sheep and cattle together with feral goats within the same property in New South Wales, Australia. Internal transcribed spacer 2 (ITS-2) nema

Occurrence of gastrointestinal nematodes in lambs in Norway, as assessed by copromicroscopy and droplet digital polymerase chain reaction.

BACKGROUND: Gastrointestinal nematodes (GINs) have a major impact on sheep production, health, and welfare worldwide. Norway is no exception, but there are only a few studies on the prevalence of GINs in Norwegian sheep. The aim of this study was to investigate the current occurrence of the most important nematodes in sheep flocks in Norway. Faecal samples were collected from flocks in 2021/2022, mainly from three geographical regions in Norway, i.e., northern, eastern, and western. In each of 134 flocks included, individual samples from 10 lambs (autumn) were pooled. Third stage larvae (L3) w
