Telencephalic afferents to the caudolateral neostriatum of the pigeon.

The pigeon caudolateral neostriatum (NCL) shares a dopaminergic innervation with mammalian frontal cortical areas and is implicated in the regulation of avian cognitive behavior. Retrograde tracing methods were used to identify forebrain projections to NCL and to suggest a possible role of this area in mediating spatial behavior. NCL receives telencephalic projections from the hyperstriatum accessorium, cells along the border of hyperstriatum dorsale and hyperstriatum ventrale, anterolateral hyperstriatum adjacent to the vallecula, confined cell groups within the anterior neostriatum, and subd

Molecular alterations in the neostriatum of human cocaine addicts.

Molecular changes in the neostriatum of human subjects who died with a history of cocaine abuse were revealed in discrete cell populations by means of the techniques of in situ hybridization histochemistry and in vitro receptor binding and autoradiography. Cocaine subjects had a history of repeated cocaine use and had cocaine and/or cocaine metabolites on board at the time of death. These subjects were compared to control subjects that had both a negative history and toxicology of cocaine use. Selective alterations in mRNA levels of striatal neuropeptides were detected in cocaine subjects comp

[Case of Creutzfeldt-Jakob syndrome with involvement of the neostriatum].

The authors describe a case of CJD in a man aged 45 years. The disease began with sluggishness of movements and speech difficulties, followed by development of pyramidal system damage and dementia. EEG findings were normal throughout the whole duration of the disease that is 3.5 years. Neuropathological examination disclosed major neuronal loss, spongy degeneration and astrogliosis in the striatum bilaterally. The cerebral and cerebellar cortex and brain stem showed similar but much less intense changes. This particular topography of the lesions was probably the cause of absent EEG changes.

Behavior-reactive neuron populations in the monkey neostriatum.

Comparative analysis of neuron activity in the monkey putamen during multistep behavior showed that putamen neurons are active during all the animal's behavioral actions. The difference between the number of active neurons at a given step of the behavior as compared with the preceding step was found to be significantly smaller than the number of neurons reorganizing their activity at this step. Reorganization of neuron activity in the putamen is regarded as a reflection of the efferent code controlling the behavior, while the extent of reorganization is regarded as a measure of the change in t

Early changes in the synapses of the neostriatum induced by perinatal asphyxia.

Perinatal asphyxia (PA) is a medical condition associated with a high short-term morbimortality and different long-term neurological diseases. In previous work we have observed at 6 months post-synaptic densities (PSDs) alterations compatible with neurodegeneration highly correlated with the increment in the ubiquitination. Although alterations in the synaptic organization and function have been related with neuronal death after hypoxia, little is known about the synaptic changes in young animals exposed to PA. The main aim of this work is to study the PSDs changes in striatum of 30-day-old ra

Presynaptic modulation by somatostatin in the rat neostriatum is altered in a model of parkinsonism.

Somatostatin (SST) is a peptide synthesized and released by a class of neostriatal local GABAergic interneurons, which, to some extent, are in charge of the feedforward inhibitory circuit. Spiny projection neurons (SPNs) make synapses with each other via their local axon collaterals, shaping the feedback inhibitory circuit. Both inhibitory circuits, feedforward and feedback, are related through SST, which, being released by interneurons, presynaptically inhibits connections among SPNs. Here, we studied SST presynaptic modulation of synapses among SPNs in the 6-hydroxydopamine (6-OHDA) rodent m

Morphofunctional studies of the interactions of the glutamatergic, cholinergic, and dopaminergic systems in the neostriatum.

Morphological, electrophysiological, and behavioral experiments were used to study the interactions of the glutamatergic, cholinergic, and dopaminergic systems in the neostriatum of white rats with unilateral lesions of the mesostriatal dopaminergic system induced by 6-hydroxydopamine. The neostriatum was shown to contain both synaptic and interneuronal nonsynaptic interactions between these neurochemical systems. It is suggested that glutamate, which is present in excess in conditions of prolonged dopamine deficiency, has toxic effects on corticoneostriatal synaptic connections.

Subcortical correlates of developmental language disorder: more than the neostriatum.

Developmental language disorder (DLD) is a common neurodevelopmental disorder that affects receptive and expressive language skills. In contrast to the wealth of evidence on acquired language disorders, we understand relatively little about the neural underpinnings of DLD. A recent meta-analysis across different types of structural brain analyses in DLD highlighted consistent anatomical differences in the anterior striatum, with other subcortical structures relatively spared. These findings are consistent with predictions from the procedural circuit deficit hypothesis (PCDH), namely that the a

[Myoclonic hyperkinesis induced by repeated administration of picrotoxin into rat neostriatum].

The effects of three-week daily intrastriatal 45 mcg GABA, 5 and 15 mcg picrotoxin and 5 bicuculline microinjections in rats were investigated. The inhibition of avoidance conditioning in shuttle box was registered in first days of GABA microinjection; stereotyped behaviour and paradoxal freezing in further GABA effects were developed. After the short orofacial dyskinesia as the acute picrotoxin effects, the choreo-myoclonic limb jerks, with the distinct generalization stage were observed. Picrotoxin dose increasing in toxic convulsive status results. No any dystonia, but strong avoidance cond

Synaptic potentials in the rat neostriatum in dissociated embryonic cell culture.

Neostriatal cells of embryonic days 19-21 were grown in dissociated cell culture. To test whether the cultures contained predominantly neostriatal cells, a glyoxylic acid staining procedure was used which, after dopamine loading, stained neostriatal cells but not neurons of embryonic neocortical tissue. Whole cell current clamp recording was performed in the neurons after 1-2 weeks in cell culture. Although cells could be driven to discharge by direct depolarization, spontaneous activity was low. All cells responded to gamma-aminobutyric acid (GABA) (0.1-0.5 mM), and the majority of them respo

Gray matter atrophy patterns within the cerebellum-neostriatum-cortical network in SCA3.

OBJECTIVE: To investigate the spatial patterns and the probable sequences of gray matter atrophy in spinocerebellar ataxia type 3 (SCA3). METHODS: A total of 47 patients with SCA3 and 49 age- and sex-matched healthy controls participated in the study. High-resolution T1-weighted MRI were examined in all participants. We used the causal network of structural covariance (CasCN) to identify the sequence of gray matter atrophy patterns. This was achieved by applying Granger causality analysis to a gray matter atrophy staging scheme performed by voxel-based morphometry from the network level. RES

[The effect on rat behavior of phenamine contained in a cannula chronically implanted in the neostriatum].

Avoidance conditioning in a shuttle-box was studied in rats under conditions of daily three-week amphetamine microinjections (45 mcg) into the rostral striatum and in rats with chronically implanted intrastriatal amphetamine-containing cannulae (45 mgg/0.75 mcl). Amphetamine stimulatory behavioural effects were recorded in both groups of animals (improvement of avoidance conditioning, stereotyped hyperactivity etc.). The dopaminomimetic effects in rats with implanted cannulae were less expressed than in those with daily injections. High chemical stability of cannulated amphetamine was demonstr

Microstimulation of the primate neostriatum. I. Physiological properties of striatal microexcitable zones.

Microstimulation was carried out at over 1,250 sites in the putamen in four unanesthetized rhesus monkeys. At numerous sites, microstimulation resulted in movements of individual body parts including leg, arm, and face. Microstimulation-evoked limb movements were invariably contralateral to the stimulating electrode. In nearly all instances, the response at threshold was restricted to or maximal about a single joint. A small percentage of stimulation-evoked axial and orofacial movements were bilateral. The same motor response was frequently evoked over distances of up to 1,200 micron along a s

Protein ubiquitination in postsynaptic densities after hypoxia in rat neostriatum is blocked by hypothermia.

Synaptic dysfunction has been associated with neuronal cell death following hypoxia. The lack of knowledge on the mechanisms underlying this dysfunction prompted us to investigate the morphological changes in the postsynaptic densities (PSDs) induced by hypoxia. The results presented here demonstrate that PSDs of the rat neostriatum are highly modified and ubiquitinated 6 months after induction of hypoxia in a model of perinatal asphyxia. Using both two dimensional (2D) and three dimensional (3D) electron microscopic analyses of synapses stained with ethanolic phosphotungstic acid (E-PTA), we

Magnesium ions prevent the development of hyperkinesia evoked by administration of picrotoxin into the rat neostriatum.

Chronic experiments on rats were performed to study the effects of daily administration of the GABA(A) receptor antagonist picrotoxin (2 microg) into the rostral neostriatum in rats. Picrotoxin was injected in 1 microl of sterile apyrogenic physiological saline or in 1 microl of 1.0 M or 1.5 M MgCl(2); control animals received physiological saline or 1.0 M MgCl(2). Picrotoxin given in physiological saline induced impairments in conditioned reflex (avoidance) and free motor behavior and produced clear stereotypical imperative movements in the form of choreomyoclonic hyperkinesia of the paws, he

Skein-like inclusions in the neostriatum from a case of amyotrophic lateral sclerosis with dementia.

Skeins or skein-like inclusions (SLIs) in motor neurons detected by ubiquitin immunohistochemistry are a characteristic finding of amyotrophic lateral sclerosis (ALS). Here we report ubiquitinated SLIs in the putamen and caudate nucleus from a case of ALS with dementia. A 48-year-old Japanese man developed apathy and amimia. Mental and neurological examinations revealed severe character change, muscle atrophy and fasciculation of the distal upper extremities and the tongue, and an exaggeration of the deep tendon reflex. He subsequently showed dysphagia and dysarthria. He died at the age of 51

Mosaic pattern of gliosis in the neostriatum of a North American man with craniocervical dystonia and parkinsonism.

We present the case of a 51-year-old patient with a 31-year history of psychiatric symptoms, craniocervical dystonia, bulbar dysfunction, and parkinsonism. His dystonic movements included blepharospasm, jaw opening and lingual dystonia, and spasmodic retrocollis. Psychiatric symptoms included psychosis and depression, with onset years before the movement disorder. After his death by aspiration, examination of his brain revealed abnormalities limited to the neostriatum. Staining of brain sections, including Holzer, glial fibrillary acidic protein, and immunohistochemical stain for calbindin D28
