Deep learning-driven MRI trigeminal nerve segmentation with SEVB-net.

PURPOSE: Trigeminal neuralgia (TN) poses significant challenges in its diagnosis and treatment due to its extreme pain. Magnetic resonance imaging (MRI) plays a crucial role in diagnosing TN and understanding its pathogenesis. Manual delineation of the trigeminal nerve in volumetric images is time-consuming and subjective. This study introduces a Squeeze and Excitation with BottleNeck V-Net (SEVB-Net), a novel approach for the automatic segmentation of the trigeminal nerve in three-dimensional T2 MRI volumes. METHODS: We enrolled 88 patients with trigeminal neuralgia and 99 healthy volunteers

Structure, development, and maintenance of the nerve net of the body column in Hydra.

The anatomy and developmental dynamics of the nerve net in the body column of Hydra viridissima were examined immunocytochemically with a monoclonal antibody (CC04) that recognizes an antigen in nerve cells and with an antiserum against vasopressin. CC04+ neuron cell bodies, their neurites, and vasopressin-like-immunoreactive (VLI+) neurites could be clearly visualized on whole-mount preparations. All neurites of the CC04+ neurons in the body column were VLI+. However, only half of the VLI+ neurites in the body column were CC04+. Immunocytochemical analysis of macerated preparations showed tha

Net optic nerve sheath diameter as a factor for predicting intracranial hypertension.

BACKGROUND: The assessment of intracranial pressure (ICP) is crucial in the management of neurological emergencies. Although invasive ICP monitoring has been widely used for the early diagnosis of intracranial hypertension, its routine application remains controversial for various reasons. This study aimed to evaluate the correlations between optic nerve parameters measured using cranial magnetic resonance imaging (MRI) and ICP, and to assess their diagnostic value in predicting intracranial hypertension. METHODS: The data of 112 patients with clinically suspected intracranial infections who

Automated Segmentation of the Median Nerve in the Carpal Tunnel using U-Net.

Nerve area and motion in carpal tunnel syndrome (CTS) are currently under investigation in terms of prognostic potential. Therefore, there is increasing interest in non-invasive measurement of the nerve using ultrasound. Manual segmentation is time consuming and subject to inter-rater variation, providing an opportunity for automation. Dynamic ultrasound images (n = 5560) of carpal tunnels from 99 clinically diagnosed CTS patients were used to train a U-Net-shaped neural network. The best results from the U-Net were achieved with a location primer as initial region of interest for the segmenta

Automatic segmentation and quantification of the optic nerve on MRI using a 3D U-Net.

PURPOSE: Pathological conditions associated with the optic nerve (ON) can cause structural changes in the nerve. Quantifying these changes could provide further understanding of disease mechanisms. We aim to develop a framework that automatically segments the ON separately from its surrounding cerebrospinal fluid (CSF) on magnetic resonance imaging (MRI) and quantifies the diameter and cross-sectional area along the entire length of the nerve. APPROACH: Multicenter data were obtained from retinoblastoma referral centers, providing a heterogeneous dataset of 40 high-resolution 3D T2-weighted M

Molecular Imaging-Derived Biomarker of Cardiac Nerve Integrity - Introducing High NET Affinity PET Probe 18F-AF78.

Background: Radiolabeled agents that are substrates for the norepinephrine transporter (NET) can be used to quantify cardiac sympathetic nervous conditions and have been demonstrated to identify high-risk congestive heart failure (HF) patients prone to arrhythmic events. We aimed to fully characterize the kinetic profile of the novel 18F-labeled NET probe AF78 for PET imaging of the cardiac sympathetic nervous system (SNS) among various species. Methods:18F-AF78 was compared to norepinephrine (NE) and established SNS radiotracers by employing in vitro cell assays, followed by an in vivo PET im

A giant nerve net with multi-effector synapses underlying epithelial adhesive strips in the mouth of Beroë (Ctenophora).

We present ultrastructural evidence for the first known example of a giant nerve net in the phylum Ctenophora. The giant fibre system in Beroë underlies paired strips of adherent epithelial cells that run inside the lips. Interlocking actin-lined cell junctions between opposing adhesive strips keep Beroë's large mouth closed while the ctenophore searches for prey. The giant neurons, up to 6-8 microns in diameter, form a continuous lattice-like plexus rich in vesicles, microtubules, and 'presynaptic triads'. A novel feature is that individual giant axons make synaptic contacts with more than on

Topical administration of a Rock/Net inhibitor promotes retinal ganglion cell survival and axon regeneration after optic nerve injury.

Intraocular pressure (IOP)-lowering ophthalmic solutions that inhibit Rho-associated protein kinases (Rock) and norepinephrine transporters (Net) are currently under clinical evaluation. Here we evaluate topical application of one such drug for its effects on retinal ganglion cell (RGC) survival and axon regeneration after optic nerve crush injury. We performed unilateral optic nerve crush on young rats (P18) and topically applied Rock/Net inhibitor AR-13324 or placebo 3 times a day for 14 days. IOP was measured starting 3 days before and up to 9 days after injury. On day 12, cholera toxin B (

Somatic nerve stimulation and cholera-induced net fluid secretion in the small intestine of the rat: evidence for an opioid effect.

The effects of somatic nerve stimulation on cholera toxin induced secretion was investigated in vivo in anaesthetised rats. Small intestinal secretion was induced with cholera toxin and measured by a gravimetric technique. Afferent stimulation (pulse frequency within train; 100 Hz; train duration: 50 ms; train frequency: 3 Hz) of the sciatic nerve over 30 min significantly reduced the net fluid secretion both during (P < 0.05) and after cessation of the stimulation (P < 0.01). The greatest effect was obtained immediately after the termination of the nerve stimulation when the secretion was rev

Catecholic Compounds in Ctenophore Colloblast and Nerve Net Proteins Suggest a Structural Role for DOPA-Like Molecules in an Early-Diverging Animal Lineage.

Ctenophores, or comb jellies, are among the earliest-diverging extant animal lineages. Several recent phylogenomic studies suggest that they may even be the sister group to all other animals. This unexpected finding remains difficult to contextualize, particularly given ctenophores' unique and sometimes poorly understood physiology. Colloblasts, a ctenophore-specific cell type found on the surface of these animals' tentacles, are emblematic of this difficulty. The exterior of the colloblast is dotted with granules that burst and release an adhesive on contact with prey, ensnaring it for consum

11-keto-β-boswellic acid and Z-guggulsterone suppress HMGB1/TLR4 pathway activity and modulate microglial polarization to remodel perineuronal nets after nerve injury.

INTRODUCTION: Remodeling of perineuronal nets is an emerging strategy for treating neuropathic pain (NP) and aligns with the traditional Chinese medicine concept of dispersing blood stasis dispersion and dredging collateral channels. Frankincense and myrrh are known for their ability to promote blood circulation, eliminate blood stasis, unblock collateral channels, and relieve pain. However, the precise pharmacological components and mechanisms underlying their analgesic effects remain unclear. OBJECTIVES: This study aimed to clarify the mechanisms of action of 11-keto-β-boswellic acid (KBA,

IMI2-PainCare-BioPain-RCT1: study protocol for a randomized, double-blind, placebo-controlled, crossover, multi-center trial in healthy subjects to investigate the effects of lacosamide, pregabalin, and tapentadol on biomarkers of pain processing observed by peripheral nerve excitability testing (NET).

BACKGROUND: Few new drugs have been developed for chronic pain. Drug development is challenged by uncertainty about whether the drug engages the human target sufficiently to have a meaningful pharmacodynamic effect. IMI2-PainCare-BioPain-RCT1 is one of four similarly designed studies that aim to link different functional biomarkers of drug effects on the nociceptive system that could serve to accelerate the future development of analgesics. This study focusses on biomarkers derived from nerve excitability testing (NET) using threshold tracking of the peripheral nervous system. METHODS: This i

Immunohistochemical localization of proteins involved in exocytosis of glutamate from P2X3 purinoceptor-expressing subserosal afferent nerve endings in the rat gastric antrum.

We previously reported the presence of P2X3 purinoceptors (P2X3)-expressing subserosal afferent nerve endings consisting of net- and basket-like nerve endings in the rat gastric antrum. These nerve endings may morphologically be vagal mechanoreceptors activated by antral peristalsis. The present study investigated immunoreactivities for vesicular glutamate transporter (VGLUT) 1 and VGLUT2 as well as exocytosis-related proteins, i.e., core components of the SNARE complex (SNAP25, Stx1, and VAMP2) and synaptotagmin-1 (Syt1), in whole-mount preparations of the rat gastric antrum using double immu

Transformer-Based Automated Segmentation of the Median Nerve in Ultrasound Videos of Wrist-to-Elbow Region.

Segmenting the median nerve is essential for identifying nerve entrapment syndromes, guiding surgical planning and interventions, and furthering understanding of nerve anatomy. This study aims to develop an automated tool that can assist clinicians in localizing and segmenting the median nerve from the wrist, mid-forearm, and elbow in ultrasound videos. This is the first fully automated single deep learning model for accurate segmentation of the median nerve from the wrist to the elbow in ultrasound videos, along with the computation of the cross-sectional area (CSA) of the nerve. Impact of Age and Nerve Impairment Interaction on Bell's Palsy Prognosis.

OBJECTIVES: Age as a prognostic factor for Bell's palsy remains debated. This study aimed to assess prognosis in older adults and clarify whether the interaction between aging and nerve-impairment severity, evaluated by the Nerve Excitability Test (NET), affects recovery. METHODS: This retrospective cohort study included 181 patients diagnosed with idiopathic peripheral facial nerve palsy (Bell's palsy) at Ehime University between 2011 and 2020 (older group, ≥ 65 years: 57; non-older group, < 65 years: 124). In multivariate logistic regression, factors associated with incomplete recovery were

An electrophysiological prognostic diagnosis for facial palsy.

Two electrophysiological tests for facial palsy-electroneurography (ENoG) and nerve excitability test (NET)-were reviewed. ENoG has advantages over NET in that it reflects the percentage of degenerated facial nerve fibers and can provide an accurate prognosis. However, as disadvantages, ENoG requires large, expensive equipment, and such supramaximal electrical stimulation can be quite painful for patients. NET is less painful due to weak stimulation with just enough current to meet the threshold, and the required equipment is compact and inexpensive to procure. However, it is impossible to cal

Distribution and morphology of P2X3-immunoreactive subserosal afferent nerve endings in the rat gastric antrum.

The present study investigated the morphological characteristics of subserosal afferent nerve endings with immunoreactivity for the P2X3 purinoceptor (P2X3) in the rat stomach by immunohistochemistry of whole-mount preparations using confocal scanning laser microscopy. P2X3 immunoreactivity was observed in subserosal nerve endings proximal and lateral to the gastric sling muscles in the distal antrum of the lesser curvature. Parent axons ramified into several lamellar processes to form net-like complex structures that extended two-dimensionally in every direction on the surface of the longitud

[Comparison of the prognostic value between the nerve excitability test and maximum stimulation test in evaluation of Bell's palsy].

In the present comparative study we applied the nerve excitability test (NET) and the maximum stimulation test (MST) to 131 patients with idiopathic facial nerve palsy. Our aim was to determine their value in the evaluation of facial nerve function. Results showed that the percentage of error of the NET in the prognosis of facial nerve palsy was 16%. In contrast, the MST was simple in its application and had a high percentage of reliability (97%). However, it remains a qualitative method, since it is dependent on the examiner's observations.

Automated 3D segmentation of human vagus nerve fascicles and epineurium from micro-computed tomography images using anatomy-aware neural networks.

Objective.Precise segmentation and quantification of nerve morphology from imaging data are critical for designing effective and selective peripheral nerve stimulation (PNS) therapies. However, prior studies on nerve morphology segmentation suffer from important limitations in both accuracy and efficiency. This study introduces a deep learning approach for robust and automated three-dimensional (3D) segmentation of human vagus nerve fascicles and epineurium from high-resolution micro-computed tomography (microCT) images.Methods.We developed a multi-class 3D U-Net to segment fascicles and epine
