Neural GPI-anchored cell adhesion molecules.

When first identified, neural cell adhesion molecules (neural CAMs) were thought to act simply by providing cell surfaces with differential adhesion properties. In the decades following the identification of the first neural CAMs, it has been realized that these proteins are actually involved in very complex processes such as axon guidance, neuronal migration, neurite outgrowth and fasciculation, target selection, synapse formation, plasticity and more recently, the maintenance of the integrity of myelinated fibers. In this review we will summarize work relating to glycosylphosphatidylinositol

Developmental biology of a neural cell adhesion molecule.

A membrane glycoprotein isolated from embryonic brain has been shown to be a ligand in the formation of cell-cell bonds. The molecule, called neural cell adhesion molecule (N-CAM), participates in several aspects of neural development, including the formation of plexiform layers, neurite fasciculation and nerve-muscle interactions. Experiments in vivo indicate that N-CAM-mediated adhesion is essential to the orderly assembly of the visual system.

Multiple roles of neural cell adhesion molecule, neural cell adhesion molecule-polysialic acid, and L1 adhesion molecules during sensory innervation of the otic epithelium in vitro.

To explore the role of cell adhesion molecules in the innervation of the inner ear, antibody perturbation was used on histotypic co-cultures of the ganglionic and epithelial anlagen derived from the otocyst. When unperturbed, these tissues survived and differentiated in this culture system with outgrowth of fasciculated neuronal fibers which expressed neural cell adhesion molecule and L1. The fibers exhibited target choice and penetration, then branching and spreading within the otic epithelium as individual axons. Treatment of the co-cultures, or of the ganglionic anlagen alone, with anti-neu

Antibodies against cell adhesion molecules and neural structures in paraneoplastic neuropathies.

OBJECTIVE: Paraneoplastic neurological syndromes (PNS) are rare neurological disorders in which ectopic expression of neural antigens by a tumor results in an autoimmune attack against the nervous system. Onconeural antibodies not only guide PNS diagnosis but may also help detecting underlying malignancies. Our project aims to uncover new potential antibodies in paraneoplastic neuropathies (PN). METHODS: Thirty-four patients fulfilling diagnostic criteria of possible (n = 9; 26.5%) and definite (n = 25; 73.5%) PN without onconeural antibodies and 28 healthy controls were included in our study

Neural cell adhesion molecule expression on renal interstitial cells.

BACKGROUND: At early stages of kidney development, the neural cell adhesion molecule (NCAM) is highly expressed on cells of the metanephrogenic mesenchyme. During maturation of the fetal kidney, NCAM gradually disappears. So far, it has been widely accepted that NCAM in the adult kidney is only expressed by nerves, and not by other cell types. METHODS: NCAM expression was analysed in human adult healthy and diseased kidneys by immunohistochemistry and western blot analysis. NCAM+ renal interstitial cells were further characterized by double immunofluorescent staining using antibodies against

The neural cell adhesion molecule in synaptic plasticity and ageing.

By mediating cell adhesion and signal transduction, the neural cell adhesion molecule (NCAM) regulates neurite outgrowth, fasciculation and target recognition in the developing nervous system. In addition, a number of studies suggest an important role for the NCAM in regeneration and learning in the adult nervous system. NCAM-deficient mice are impaired in spatial learning. Moreover, by interfering with normal NCAM function by intracranial injections of NCAM-antibodies, long-term potentiation (LTP) in rat hippocampal slices and learning in rats and chicks have been inhibited. In the vertebrate

Overexpression of neural cell adhesion molecule in Chagas' myocarditis.

The expression of the neural cell adhesion molecule (NCAM) was studied in normal human myocardium and in Chagas' disease myocarditis. We found that NCAM is expressed in the conduction system as well as the myocardium in the fetal heart, but its expression is restricted to the conduction system and absent in the adult myocardium. Chagas' disease is an American endemic disease caused by the Trypanosoma cruzi parasite, which produces myocarditis and a blockade of the conduction system, resulting in cardiac dysfunction. We studied the expression of NCAM in paraffin-embedded human heart tissues fro

Cell adhesion molecules in neural plasticity and pathology: similar mechanisms, distinct organizations?

Brain plasticity and the mechanisms controlling plasticity are central to learning and memory as well as the recovery of function after brain injury. While it is clear that neurotrophic factors are one of the molecular classes that continue to regulate brain plasticity in the adult central nervous system (CNS), less appreciated but equally profound is the role of cell adhesion molecules (CAMs) in plasticity mechanisms such as long term potentiation, preservation of neurons and regeneration. Ironically, however, CAMs can also reorganize the extra-cellular space and cause disturbances that drive

Neural cell adhesion molecules belonging to the family of leucine-rich repeat proteins.

Leucine-rich repeats (LRRs) are motifs that form protein-ligand interaction domains. There are approximately 140 human genes encoding proteins with extracellular LRRs. These encode cell adhesion molecules (CAMs), proteoglycans, G-protein-coupled receptors, and other types of receptors. Here we give a brief description of 36 proteins with extracellular LRRs that all can be characterized as CAMs or putative CAMs expressed in the nervous system. The proteins are involved in multiple biological processes in the nervous system including the proliferation and survival of cells, neuritogenesis, axon

Neural cell adhesion molecules are present in the fetal human primary olfactory pathway.

Olfactory tissues from human fetuses (17.5-28 weeks of gestation) were stained by immunofluorescence for neural cell adhesion molecules (N-CAMs). Staining for N-CAMs was most prominent in the olfactory nerve bundles in the lamina propria, while in the olfactory epithelium, it was present on the olfactory receptor neurons and globose basal cells. The basal cells proper and supporting cells lacked N-CAMs. In the olfactory bulb, only the olfactory nerve and glomerular layers showed moderate labeling for N-CAMs. Western blot analysis showed that the N-CAMs of the fetal human primary olfactory path

Sera from 5 patients with IgM gammopathy and peripheral neuropathy reacted strongly by a sensitive immunospot enzyme-linked immunosorbent assay test with the myelin-associated glycoprotein (MAG) purified from human and bovine brains and the novel neural cell adhesion molecule J1 and associated glycoproteins from human brain. Two other members of the L2/HNK-1 family of cell adhesion molecules, the L1 glycoprotein and the neural cell adhesion molecule N-CAM, are not recognized significantly more strongly by these sera than by sera from patients with multiple sclerosis. Misregulation of cell adhesion molecules in the Ciona neural tube closure mutant bugeye.

Neural tube closure (NTC) is a complex multi-step morphogenetic process that transforms the flat neural plate found on the surface of the post-gastrulation embryo into the hollow and subsurface central nervous system (CNS). Errors in this process underlie some of the most prevalent human birth defects, and occur in about 1 out of every 1000 births. Previously, we discovered a mutant in the basal chordate Ciona savignyi (named bugeye) that revealed a novel role for a T-Type Calcium Channel (Cav3) in this process. Moreover, the requirement for CAV3s in Xenopus NTC suggests a conserved function a

Neural cell adhesion molecule (CD56)-positive B-cell lymphoma.

Expression of neural cell-adhesion molecule CD56 is a rare event in B-cell lymphoma. We described four cases of CD56-positive B-cell lymphoma, including follicular lymphoma and diffuse large B-cell lymphoma. These lymphoma cells expressed CD10 and bcl-6, which suggests germinal center-stage phenotype. Immunohistochemistry showed that CD56-positive cells aggregated and displayed a cohesive growth pattern, indicating that homotypic adhesion through the molecules might affect the manner of tumor growth and expansion. Although CD56 expression level varies among the cases, this molecule might play

Neural cell adhesion molecule expression in the human carotid body.

We studied by immunocytochemistry the expression of NCAM in human carotid bodies, sampled at autopsy from 16 adult subjects (mean age +/- SD: 44.3 +/- 3.4 years). No NCAM immunoreactivity was visible in type II cells. An high percentage (78.3 +/- 7.2%) of type I cells showed positive anti-NCAM immunoreaction. Statistically significant differences were not found in anti-NCAM immunostaining of light and dark cells (80.2 +/- 6.2% vs 74.7 +/- 13.4%, P>0.05). The high expression level of NCAM in the carotid body indicates a role in regulating adhesion between type I cells. The ascertained role of N

Neural cell adhesion molecule in breast, colon and lung carcinomas.

Neural cell adhesion molecules (NCAM) play an important role in embryogenesis and in some tumors, especially of neuroectodermal origin. In this study, 18 cases of invasive breast carcinoma, 7 cases of sigmoid colon carcinomas and 17 cases of the non-small-cell lung carcinoma were immunostained for NCAM. NCAM expression, usually focal, was observed in some cases only. NCAM was expressed in the membranes, in a fine granular pattern. In 3 cases of breast cancer cytoplasmic localisation of NCAM was also observed, which may suggest its cytoplasmic formation. Furthermore, in 3 cases expression of NC

Binding properties of a cell adhesion molecule from neural tissue.

We have previously identified and purified a cell surface glycoprotein from retina and brain, called neural cell adhesion molecule or N-CAM, that appears to be involved in neural cell--cell adhesion, the fasciculation of neurites, and the formation of normal tissue patterns in the retina. The present studies reveal that artificial vesicles containing lipid and purified N-CAM bind to different cell types with a specificity similar to that of nerve cells. The same results were obtained with soluble N-CAM that had been briefly exposed to pH 3. In both cases the binding altered the rate of aggrega

An alcohol binding site on the neural cell adhesion molecule L1.

Prenatal ethanol exposure causes fetal alcohol spectrum disorders (FASD) in part by disrupting the neural cell adhesion molecule L1. L1 gene mutations cause neuropathological abnormalities similar to those of FASD. Ethanol and 1-butanol inhibit L1-mediated cell-cell adhesion (L1 adhesion), whereas 1-octanol antagonizes this action. To test the hypothesis that there are alcohol binding sites on L1, we used 3-azibutanol and 3-azioctanol, the photoactivatable analogs of 1-butanol and 1-octanol, to photolabel the purified Ig1-4 domain of human L1 (hL1 Ig1-4). 3-Azibutanol (11 mM), like ethanol, in

Expression of NCAM (neural cell adhesion molecule) in mitochondrial myopathy.

In muscle, the neural cell adhesion molecule (NCAM) is known to be expressed in denervated and regenerating fibers. Our present study, performed with immunohistochemical detection procedures, attempts to demonstrate the expression of NCAM in mitochondrial myopathy. Biopsy specimens from 29 patients were investigated. The following conditions were represented: nonweak control subjects (8 cases), chronic progressive external ophthalmoplegia (7 cases), Kearns-Sayre syndrome (5 cases), unclassified mitochondrial myopathy (2 cases), idiopathic polymyositis (7 cases). We demonstrate the expression o

Influences of the neural cell adhesion molecule on axon growth and guidance.

The neural cell adhesion molecule (NCAM) has been shown to be a ligand in the formation of cell-cell bonds. This molecule is present on essentially the entire surface of differentiated nerve cells, including the cell body, neurite shaft, and growth cone. In mediating membrane-membrane adhesion, NCAM appears to ligate with itself, and one of its most obvious functions is in the self-association of nerve fibers to form fascicles. In most cases fasciculation occurs by the successive elongation of axons along other axons and, therefore, is likely to represent a growth cone-neurite shaft interactio

Differential spectrum of expression of neural cell adhesion molecule isoforms and L1 adhesion molecules on human neuroectodermal tumors.

A series of four medulloblastomas, seven neuroblastomas (Nb), two ependymomas, and three gliomas, human neuroectodermal tumors, were screened for their expression of adhesion molecules L1, carcinoembryonic antigen, neural cell adhesion molecule isoforms (N-CAM) and HNK1 epitope by Western blotting and double immunofluorescence labeling. All seven neuroblastomas, whatever their differentiated state, expressed L1, a neural cell surface developmental antigen, whereas all other tumors tested were negative. All tumors expressed N-CAM; however, a large diversity was observed among the isoforms. Low
