Models in neuroendocrinology.

The neuroendocrine systems of the hypothalamus are critical for survival and reproduction, and are highly conserved throughout vertebrate evolution. Their roles in controlling body metabolism, growth and body composition, stress, electrolyte balance and reproduction have been intensively studied, and have yielded a rich crop of original and challenging insights into neuronal function, insights that circumscribe a vision of the brain that is quite different from conventional views. Despite the diverse physiological roles of pituitary hormones, most are secreted in a pulsatile pattern, but arisi

Neuroendocrinology of ageing.

Many common problems encountered in the ageing patient can be related to neuroendocrine phenomena. These include Alzheimer's disease, dementia and cognitive dysfunction, depression, Parkinson's disease, hyponatraemia and the postmenopausal increase in both vascular risk and osteoporosis. This review concentrates on the hypothalamic neuroendocrine system, including the dopaminergic, noradrenergic, serotoninergic, cholinergic and neurohypophyseal systems and the roles of the anterior pituitary and monoamine oxidases, luteinizing hormone-releasing hormone, corticotrophin-releasing factor, the pro

Neuroendocrinology of the skin.

The skin is considered the mirror of the soul and is affected by neurohormonal triggers, especially stress. Hair follicles, keratinocytes, mast cells, melanocytes, and sebocytes all express sex and stress hormones implicating them in a local "hypothalamic-pituitary-adrenal axis." In particular, the peptides corticotropin-releasing hormone (CRH) and neurotensin (NT) have synergistic action stimulating mast cells and are uniquely elevated in the serum of patients with skin diseases exacerbated by stress. Addressing the neurohormonal regulation of skin function could lead to new targets for effec

Neuroendocrinology of bone.

The past decade has witnessed significant advances in our understanding of skeletal homeostasis and the mechanisms that mediate the loss of bone in primary and secondary osteoporosis. Recent breakthroughs have primarily emerged from identifying disease-causing mutations and phenocopying human bone disease in rodents. Notably, using genetically-modified rodent models, disrupting the reciprocal relationship with tropic pituitary hormone and effector hormones, we have learned that pituitary hormones have independent roles in skeletal physiology, beyond their effects exerted through target endocri

Neuroendocrinology of pyridoxine deficiency.

Dihydroxyphenylalanine decarboxylase and 5-hydroxytryptophan decarboxylase respectively have high and low affinities for pyridoxal phosphate. In the pyridoxine-deficient animal, hypothalamic serotonin content is significantly reduced without any change in catecholamine levels. Hypothalamic neurotransmitters affect the hypothalamo-pituitary-end organ axes. Specifically, the decrease in hypothalamic serotonin in the pyridoxine-deficient rat results in tertiary hypothyroidism. In addition, pineal function is affected in deficient animals due to decreased synthesis of melatonin.

Gene Targeting in Neuroendocrinology.

Research in neuroendocrinology faces particular challenges due to the complex interactions between cells in the hypothalamus, in the pituitary gland and in peripheral tissues. Within the hypothalamus alone, attempting to target a specific neuronal cell type can be problematic due to the heterogeneous nature and level of cellular diversity of hypothalamic nuclei. Because of the inherent complexity of the reproductive axis, the use of animal models and in vivo experiments are often a prerequisite in reproductive neuroendocrinology. The advent of targeted genetic modifications, particularly in mi

Neuroendocrinology of amphibian metamorphosis.

The timing of metamorphosis is a central amphibian life history trait and is controlled by the interplay of developmental progression, body size and condition, and environmental signals. These different processes and signals are integrated by the neuroendocrine system to regulate production of hormones by the thyroid gland. Thyroid hormone (TH) is the primary morphogen controlling metamorphosis, while corticosteroids (CSs) produced by the interrenal glands synergize with TH to promote metamorphic changes. The actions of TH are modulated by monodeiodinase enzymes expressed in TH target tissues.

Clinical aspects of neuroendocrinology.

Recent studies have identified structures of most of the hypothalamic hormones and clarified their actions. In addition, the importance of a variety of classical and peptide neurotransmitters or neuromodulators in the regulation of pituitary hormone secretion and other hypothalamic functions has been partly elucidated. These advances in the physiological aspects of the hypothalamus have greatly helped our understanding of the pathogenesis of a variety of hypothalamic syndromes.

Neuroendocrinology and the Genetics of Obesity.

The increase in the incidence of obesity has coincided with changes in lifestyle, diet, and environment. Comorbidities associated with obesity include cardiovascular disease, diabetes, musculoskeletal disorders, stroke, and thromboembolism, affecting public health. The effect of increased weight has recently become even more obvious, since obesity has been significantly associated with increased severity and higher mortality among COVID-19 patients. The need to decrease rates of obesity prompted a surge in the use of glucagon-like peptide-1 agonist medications. Twin studies, however, determine

[Neuroendocrinologic aspects of epilepsy].

Although the literature recognises the involvement of neuropeptide structures in the genesis of epilepsy, only tentative preliminary data are available at present. Experiments indicate that neuropeptides primarily involved in the genesis of epilepsy are leuencephalin, metencephalin and beta-endorphin. Their convulsion properties are apparently related to the specific levels, locations and convulsive potential of the individual substances. A qualitative and/or quantitative change to the GABA-neuropeptide balance may also be involved in convulsive disorders; decreased GABA fluid levels have been

The neuroendocrinology of sexual attraction.

Sexual attraction has two components: Emission of sexually attractive stimuli and responsiveness to these stimuli. In rodents, olfactory stimuli are necessary but not sufficient for attraction. We argue that body odors are far superior to odors from excreta (urine, feces) as sexual attractants. Body odors are produced by sebaceous glands all over the body surface and in specialized glands. In primates, visual stimuli, for example the sexual skin, are more important than olfactory. The role of gonadal hormones for the production of and responsiveness to odorants is well established. Both the an

[Neuroendocrinological aspects of aging].

Aging is accompanied by important changes in the production and secretion of hormones. These partly arise from changes in the peripheral endocrine glands themselves but are also partly the consequence of changes in the neuro-endocrine regulating centers of the hormone secretion. Considering e.g. the male gonads, we know that the reduced androgen secretion (and the diminished spermatogenesis) along the ageing process is the consequence of changes in the testes, such as the reduced number of Leydig- and Sertoli-cells, as well as a reduced testicular blood-perfusion. But on the other hand, it sho

Neuroendocrinology and neurobiology of sebaceous glands.

The nervous system communicates with peripheral tissues through nerve fibres and the systemic release of hypothalamic and pituitary neurohormones. Communication between the nervous system and the largest human organ, skin, has traditionally received little attention. In particular, the neuro-regulation of sebaceous glands (SGs), a major skin appendage, is rarely considered. Yet, it is clear that the SG is under stringent pituitary control, and forms a fascinating, clinically relevant peripheral target organ in which to study the neuroendocrine and neural regulation of epithelia. Fifty Years of Advances in Neuroendocrinology.

Importance of the neuroendocrine brain for health and happiness has become clear since the 1960s. Foundations laid 100 years ago culminated in Geoffrey W Harris's model of control by the brain of secretion of anterior and posterior pituitary gland hormones through, respectively, releasing factors secreted into the hypothalamic-hypophysial portal system, and directly from axon terminals into the systemic circulation. Confirmation, expansion and deepening of knowledge and understanding have followed increasingly sophisticated technology. This allowed chemical characterisation of the posterior pi

The neuroendocrinology of primate maternal behavior.

In nonhuman primates and humans, similar to other mammals, hormones are not strictly necessary for the expression of maternal behavior, but nevertheless influence variation in maternal responsiveness and parental behavior both within and between individuals. A growing number of correlational and experimental studies have indicated that high circulating estrogen concentrations during pregnancy increase maternal motivation and responsiveness to infant stimuli, while effects of prepartum or postpartum estrogens and progestogens on maternal behavior are less clear. Prolactin is thought to play a r

The neuroendocrinology of human puberty revisited.

The fundamental aspects of the hypothalamic luteinizing hormone-releasing hormone (LHRH)(1) [1]pulse generator-pituitary gonadotrophin-gonadal apparatus in mammals have striking commonalities. There are, however, critical, substantive differences in the neuroendocrinology of puberty among species. The onset of puberty in the human is marked by an increase in the amplitude of LH pulses, an indirect indicator of the increase in amplitude of LHRH pulses. The hypothalamic LHRH-pituitary gonadotrophin complex is functional by at least 0.3 gestation in the human foetus; the sex difference in the fet
