Modification of the anticonvulsant activity of 2-amino-5-phosphonovalerate by agents affecting different neurotransmitter systems.

The effects of atropine (5 mg/kg), baclofen (10 mg/kg), gamma-hydroxybutyrate (300 mg/kg), gamma-butyrolactone (100 mg/kg) and muscimol (1 mg/kg) upon the action of 2-amino-5-phosphonovalerate (APV; an antagonist at receptors for N-methyl-D-aspartate) on the threshold current for seizures induced by electroshock, were studied in mice. Neither APV, up to 100 mg/kg, nor the other agents produced any significant increase in the convulsive threshold when tested alone. Muscimol had no effect on the action of APV (50 and 100 mg/kg) and the combination of APV with the subthreshold doses of atropine,

Neurotransmitter amines and antioxidant agents in neuronal protection against methylmercury-induced cytotoxicity in primary cultures of mice cortical neurons.

Methylmercury (MeHg) is an environmental toxicant with detrimental effects on the developing brain and adult nervous system. The main mechanisms identified include oxidative stress, changes in intracellular calcium, mitochondrial changes, inhibition of glutamate uptake, of protein synthesis and disruption of microtubules. However, little is known about mechanisms of protection against MeHg neurotoxicity. We found that resveratrol (10 μM) and ascorbic acid (200 μM) protected MeHg-induced cell death in primary cultures of cortical neurons. In this work, we aimed at finding additional targets tha

Non-steroidal anti-inflammatory agents, tolmetin and sulindac, inhibit liver tryptophan 2,3-dioxygenase activity and alter brain neurotransmitter levels.

Hepatic tryptophan 2,3-dioxygenase (TDO) is one of the rate-limiting enzymes in tryptophan catabolism and plays an important role in regulating the physiological flux of tryptophan into relevant metabolic pathways. In this study, we determined the effect of the non-steroidal anti-inflammatory agents, tolmetin and sulindac, on rat liver TDO activity and the subsequent changes in the hippocampal and striatal neurotransmitter levels. The amount of melatonin produced by the pineal gland was also measured using high performance liquid chromatography (HPLC). Treatment of rats with tolmetin or sulind

Neurotransmitter System-Targeting Drugs Antagonize Growth of the Q Fever Agent, Coxiella burnetii, in Human Cells.

Coxiella burnetii is a highly infectious, intracellular, Gram-negative bacterial pathogen that causes human Q fever, an acute flu-like illness that can progress to chronic endocarditis. C. burnetii is transmitted to humans via aerosols and has long been considered a potential biological warfare agent. Although antibiotics, such as doxycycline, effectively treat acute Q fever, a recently identified antibiotic-resistant strain demonstrates the ability of C. burnetii to resist traditional antimicrobials, and chronic disease is extremely difficult to treat with current options. These findings high

Effects of Z-338, a novel gastroprokinetic agent, on the actions of excitatory and inhibitory neurotransmitters on neurons in area postrema.

We investigated the effects of the novel gastroprokinetic agent Z-338 on the actions of excitatory and inhibitory neurotransmitters on neurons in area postrema (AP). Iontophoretic applications of acetylcholine (ACh), AMPA and NMDA increased, while GABA suppressed the firing rates of AP neurons recorded by extracellular electrodes. Z-338 (10 microM) suppressed the ACh-induced acceleratory and GABA-induced inhibitory actions without affecting the excitatory actions of AMPA and NMDA. Under voltage-clamp conditions, nicotine, NMDA, kainic acid (KA) and ATP evoked inward currents in dissociated sin

Plasma membrane ordering agent pluronic F-68 (PF-68) reduces neurotransmitter uptake and release and produces learning and memory deficits in rats.

A substantial body of evidence indicates that aged-related changes in the fluidity and lipid composition of the plasma membrane contribute to cellular dysfunction in humans and other mammalian species. In the CNS, reductions in neuronal plasma membrane order (PMO) (i.e., increased plasma membrane fluidity) have been attributed to age as well as the presence of the beta-amyloid peptide-25-35, known to play an important role in the neuropathology of Alzheimer's disease (AD). These PMO increases may influence neurotransmitter synthesis, receptor binding, and second messenger systems as well as si

Modulation of the release of noradrenaline and of noncholinergic, nonadrenergic inhibitory neurotransmitter through presynaptic N-cholinergic receptors by means of depolarizing agents in cat internal anal sphincter.

The contractile activity of smooth-muscle strips (15 X 2 mm), isolated from the internal anal sphincter [IAS] of chloralose-anaesthesized cats, was recorded under isometric conditions. The relaxation effects of exogenously applied acetylcholine (Ach) and nicotine (Ns on cat IAS turn into contractile after treatment of the preparations with depolarizing agent (increased [K+]0 scorpion venom, ouabaine). These contractile effects are blocked completely by phentolamine, guanetidine, hexamethonium and they are antagonized by TTX. Relatively high N concentrations induce a three-phase effect whose se

[The effect of anthelmintics on the serotonin content of cestodes].

The effects of some anthelminthic agents, such as praziquantel, fenasal, albendazole, on the levels of the neurotransmitter serotonin in the tissues of Hymenolepis diminuta cestodes and Mesocestoides corti tetrathyridia were examined in vivo and in vitro. The finding showed lower serotonin levels in H.diminuta after praziquantel and albendazole and in M. corti after fenasal. It is suggested that the serotonin neurotransmitter system of helminths is involved in the mechanism of action of these agents.

Screening of neurotransmitter receptor modulators reveals novel inhibitors of influenza virus replication.

Influenza presents a significant public health threat, as severe cases can lead to excessive inflammation and complications such as pneumonia or acute respiratory distress syndrome. Current antiviral agents targeting viral proteins may lead to the development of resistance, highlighting the need for new agents targeting host factors. Neurotransmitter receptors are vital for cellular signaling and cell cycle modulation, making them promising antiviral therapeutic targets. Recent research has demonstrated that screening libraries of compounds aimed at these receptors can help identify inhibitors

An experimental model combining microdialysis with electrophysiology, histology, and neurochemistry for exploring mechanisms of secondary damage in spinal cord injury: effects of potassium.

An experimental model for characterizing effects of agents suspected to cause damage secondary to spinal cord injury is presented. Microdialysis was used to administer candidate damaging agents and to sample the release of other substances in response to administered agents. Damage was assessed by monitoring the amplitudes of evoked potentials during candidate administration and by postmortem histologic examination. This approach enabled the correlation of electrophysiologic, histologic, and neurochemical parameters in the same experiment. Potassium was chosen as a candidate damaging agent, be

[Clinical implications of present knowledge respecting neuropharmacologic control of antipituitary hormones].

The secretion of the pituitary hormones is controlled by hypothalamic hormones which are synthesized by neurosecreting cells whose activity is modulated by different neurotransmitters such as dopamine, norepinephrine and serotonin. Centrally acting drugs (antiparkinson, antipsychotic, antidepressive and hypotensive agents) interfere with the activity of the neurotransmitters and may thus influence the secretion of hypothalamic and pituitary hormones. The action of different psychotropic drugs on the secretion of hormones of the anterior pituitary gland in man is reviewed, with particular empha

Elucidating the role of neurotensin in the pathophysiology and management of major mental disorders.

Neurotensin (NT) is a neuropeptide that is closely associated with, and is thought to modulate, dopaminergic and other neurotransmitter systems involved in the pathophysiology of various mental disorders. This review outlines data implicating NT in the pathophysiology and management of major mental disorders such as schizophrenia, drug addiction, and autism. The data suggest that NT receptor analogs have the potential to be used as novel therapeutic agents acting through modulation of neurotransmitter systems dys-regulated in these disorders.

Cerebral circulatory and metabolic effects of perivascular neurotransmitters.

In this article we review the most recent literature that concerns the various neurotransmitters that are known to innervate the cerebral circulation. The best characterized of these systems (the adrenergic and serotonergic pathways) are discussed extensively, but other putative neurovascular pathways (cholinergic and peptidergic nerves) was covered. The review will be divided into five major sections: the origin and nature of the perivascular nerve fibers in the cerebrovascular bed (this section encompasses both morphological and biochemical investigations); the response of isolated cerebral

Pharmacotherapy to enhance arousal: what is known and what is not.

Severe brain injury results in a disturbance among a wide range of critical neurotransmitter systems. Each neurotransmitter system places its own functional role while being interconnected to a multitude of other systems and functions. This chapter seeks to review the major neurotransmitter systems involved after severe acquired brain injury. While limited in their construct, animal models of brain injury have demonstrated agents that may assist in the recovery process and those that may further slow recovery. We review further the issue of laboratory evidence and what is transferable to the c

GABAergic, dopaminergic and cholinergic interactions in perphenazine-induced catatonia in rats.

Anticatatonic effects of systemic and intracerebroventricularly administered GABAergic agents (GABA, muscimol piracetam, sod. valproate) were studied in rats against perphenazine-induced catatonia. All GABAergic agents, except piracetam, were found to posses anticatatonic actions as they significantly blocked perphenazine-induced catatonia. When these GABAergic agents were administered simultaneously with anticholinergic (scopolamine) or dopaminergic (bromocriptine) substances there was potentiation of the anticatatonic effect. The protective effect of GABAergic agonists and GABA antagonist an

Effects of neurotoxicants on synaptic transmission: lessons learned from electrophysiological studies.

A number of environmentally-important neurotoxicants affect chemical synaptic transmission in the peripheral and central nervous system. These include heavy metals such as lead, mercury, cadmium and tin; organophosphates; pyrethroid insecticides, and 2,5-hexanedione. Electrophysiological techniques including intracellular microelectrode recording of nerve-evoked and spontaneously occurring synaptic potentials, iontophoresis of neurotransmitter, and voltage clamp of presynaptic and postsynaptic membrane ionic current have proven to be especially useful in analyzing the cellular mechanisms by wh

Treatment of schizophrenia in the 21st Century: beyond the neurotransmitter hypothesis.

Over the last six decades, the treatment of schizophrenia has focused primarily on interactions at monoamine neurotransmitter receptor sites, including those for dopamine and serotonin. While first-generation antipsychotics demonstrate antagonism at the dopamine 2 receptor, newer atypical agents involve multiple receptors at various neurotransmitter sites. Despite the advent of these newer agents, the treatment of schizophrenia continues to elude clinicians, perhaps owing to a lack of information about the factors contributing to the development of the disease. While the etiology is complex an

Etomidate and propofol inhibit the neurotransmitter release machinery at different sites.

The mechanism of general anaesthetic action is only partially understood. Facilitation of inhibitory GABAA receptors plays an important role in the action of most anaesthetics, but is thought to be especially relevant in the case of intravenous anaesthetics, like etomidate and propofol. Recent evidence suggests that anaesthetics also inhibit excitatory synaptic transmission via a presynaptic mechanism(s), but it has been difficult to determine whether these agents act on the neurotransmitter release machinery itself. In the present study we sought to determine whether the intravenous anaesthet
