Nigericin selectively targets cancer stem cells in nasopharyngeal carcinoma.

Nasopharyngeal carcinoma (NPC) is prevalent in southern China, northern Africa, and Alaska. The prognosis for NPC patients at early stage is good, while it is poor for patients at late stages. Cancer stem cells (CSCs) have been proposed to be associated with tumor initiation, relapse and metastasis, and the poor prognosis of NPC likely results from residual CSCs after therapy. Study on the therapy targeting CSCs in NPC remains poor, though it received intensive attentions in other cancers. Here, we used NPC cell lines with high and low proportion of CSCs as models to explore the effect of nige

Nigericin is a potent inhibitor of the early stage of vaccinia virus replication.

Poxviruses remain a significant public health concern due to their potential use as bioterrorist agents and the spread of animal borne poxviruses, such as monkeypox virus, to humans. Thus, the identification of small molecule inhibitors of poxvirus replication is warranted. Vaccinia virus is the prototypic member of the Orthopoxvirus genus, which also includes variola and monkeypox virus. In this study, we demonstrate that the carboxylic ionophore nigericin is a potent inhibitor of vaccinia virus replication in several human cell lines. In HeLa cells, we found that the 50% inhibitory concentra

Activation of interleukin-1beta-converting enzyme by nigericin is independent of apoptosis.

Interleukin-1beta-converting enzyme (ICE) is believed to be one of the key proteases involved in apoptosis. Since the precursor form of interleukin-1beta (pre-IL-1beta) is one of the well known substrates for ICE, and a potassium/proton ionophore, nigericin, enhances IL-1beta processing, the authors hypothesized that nigericin induces apoptosis through the activation of ICE. In a lipopolysaccharide (LPS)-stimulated and nigericin-treated human monocytic cell line, THP-1, apoptosis was induced, as assessed as to a decrease in cell size, chromatin condensation, exposure of phosphatidylserine and

Nigericin Induces Paraptosis-Like Cell Death Instead of Pyroptosis in Corneal Keratocytes.

The purpose of this study was to examine the nature of the underlying molecular mechanisms of cell death in human keratocytes treated with nigericin, a known pyroptosis inducer. Human keratocytes were exposed to nigericin, and cell death was assessed through morphological analysis and detection of related molecular markers. Proteomic profiling was performed to identify cell death-related proteins, with key findings validated by western blot. Additionally, organelle disruptions were examined using immunostaining techniques. Pyroptosis-like cell death was observed morphologically in cultured ker

Nigericin is effective against multidrug resistant gram-positive bacteria, persisters, and biofilms.

Multidrug-resistant (MDR) bacteria pose a significant clinical threat to human health, but the development of antibiotics cannot meet the urgent need for effective agents, especially those that can kill persisters and biofilms. Here, we reported that nigericin showed potent bactericidal activity against various clinical MDR Gram-positive bacteria, persisters and biofilms, with low frequencies of resistance development. Moreover, nigericin exhibited favorable in vivo efficacy in deep-seated mouse biofilm, murine skin and bloodstream infection models. With Staphylococcus aureus, nigericin disrup

Genomic analysis and process optimization for nigericin production in a newly-isolated antimicrobial Streptomyces.

Streptomyces, the dominant species of actinomycetes, contribute to a large proportion of bioactive natural products. In our previous study, over 2400 culture extracts were subjected to bioassays against Candida albicans and Staphylococcus aureus. Among these, Streptomyces javensis named Inha503, was found to produce nigericin, which exhibits antimicrobial activity against gram-positive bacteria, such as Micrococcus luteus and S. aureus. This study aimed to understand the genomic features of S. javensis Inha503 and optimize the downstream process to improve the titer of nigericin derivatives. I

The Ionophores CCCP and Gramicidin but Not Nigericin Inhibit Trypanosoma brucei Aquaglyceroporins at Neutral pH.

Human African trypanosomiasis (HAT) is caused by Trypanosoma brucei parasites. The T. brucei aquaglyceroporin isoform 2, TbAQP2, has been linked to the uptake of pentamidine. Negative membrane potentials and transmembrane pH gradients were suggested to promote transport of the dicationic antitrypanosomal drug. Application of ionophores to trypanosomes further hinted at direct inhibition of TbAQP2 by carbonyl cyanide m-chlorophenyl hydrazone (CCCP). Here, we tested for direct effects of three classical ionophores (CCCP, nigericin, gramicidin) on the functionality of TbAQP2 and the related TbAQP

Nigericin Abrogates Maternal and Embryonic Oxidative Stress in the Streptozotocin-Induced Diabetic Pregnant Rats.

Hyperglycemic exposure in diabetic pregnancy can lead to many developmental changes, such as delayed development, fetal malformations, and fetal/embryo death. These detrimental complications are collectively known as diabetic embryopathy or teratogenesis. The current study focuses to discover the therapeutic properties of the nigericin against the STZ-stimulated diabetic embryopathy via alleviation of maternal and embryonic oxidative stress. The male and female rats at a 1:1 ratio were permitted to mate overnight to establish the course of pregnancy. The pregnant rats were distributed into fou

Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity.

Psoraleae Fructus (PF) is a well-known traditional herbal medicine in China, and it is widely used for osteoporosis, vitiligo, and other diseases in clinical settings. However, liver injury caused by PF and its preparations has been frequently reported in recent years. Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury (IDILI), but the mechanism underlying its hepatotoxicity remains unclear. This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity. Bav

Icariside I specifically facilitates ATP or nigericin-induced NLRP3 inflammasome activation and causes idiosyncratic hepatotoxicity.

BACKGROUND: Epimedii Folium (EF) is commonly used for treating bone fractures and joint diseases, but the potential hepatotoxicity of EF limits its clinical application. Our previous study confirms that EF could lead to idiosyncratic drug-induced liver injury (IDILI) and hepatocyte apoptosis, but the mechanism remains unknown. Studies have shown that NLRP3 inflammasome plays an important role in the development of various inflammatory diseases such as IDILI. Specific stimulus-induced NLRP3 inflammasome activation may has been a key strategy for lead to liver injury. Therefore, main compounds d

The Sesn2/Nrf2 axis suppresses oxidative stress and pyroptosis-associated signaling in LPS/nigericin-challenged alveolar macrophages.

BACKGROUND: Pyroptosis is a key mechanism driving immune imbalance in sepsis, while the stress-induced protein Sestrin 2 (Sesn2) and Nuclear factor E2-related factor 2 (Nrf2) are core regulators of antioxidant defense. No existing studies have clarified the interactive regulatory effect of the Sesn2/Nrf2 axis on pyroptosis in alveolar macrophages. Therefore, this study aims to investigate the regulatory mechanism of the Sesn2/Nrf2 axis on pyroptosis and oxidative stress in LPS/nigericin-challenged alveolar macrophages, providing a theoretical basis for further understanding of sepsis. Nigericin decreases the viability of multidrug-resistant cancer cells and lung tumorspheres and potentiates the effects of cardiac glycosides.

Multiple factors including tumor heterogeneity and intrinsic or acquired resistance have been associated with drug resistance in lung cancer. Increased stemness and the plasticity of cancer cells have been identified as important mechanisms of resistance; therefore, treatments targeting cancer cells independent of stemness phenotype would be much more effective in treating lung cancer. In this article, we have characterized the anticancer effects of the antibiotic Nigericin in cells displaying varying degrees of stemness and resistance to anticancer drugs, arising from (1) routine culture cond

23Na- and 1H-NMR studies of the action of chlorpromazine and imipramine on nigericin-mediated Na+ transport across phosphatidylcholine vesicular membranes.

In order to elucidate the action of chlorpromazine (CPZ) and imipramine (IMP) on nigericin-mediated Na+ transport across phosphatidylcholine vesicular membranes, 23Na nuclear magnetic resonance was applied to the exchange system of Na+ ions present at the same concentration inside and outside unilamellar vesicles. CPZ and IMP added to the vesicles in micromolar concentrations produced an equal increase in the carrier-transport rate. The kinetic analysis, together with 1H-NMR observations of the reduction in membrane fluidity produced by the drugs and on the direct interaction between drugs and

Characterization and inhibition of inflammasome responses in severe and non-severe asthma.

BACKGROUND: Increased airway NLRP3 inflammasome-mediated IL-1β responses may underpin severe neutrophilic asthma. However, whether increased inflammasome activation is unique to severe asthma, is a common feature of immune cells in all inflammatory types of severe asthma, and whether inflammasome activation can be therapeutically targeted in patients, remains unknown. OBJECTIVE: To investigate the activation and inhibition of inflammasome-mediated IL-1β responses in immune cells from patients with asthma. METHODS: Peripheral blood mononuclear cells (PBMCs) were isolated from patients with no

Assessment of evidence for K+-H+ exchange in isolated type-1 cells of neonatal rat carotid body.

Intracellular pH (pHi) was measured in enzymically isolated, neonatal rat carotid body type-1 cells, using the fluorophore carboxy-SNARF-1 (AM-loaded), and using the nigericin technique for in situ fluorescence calibration (nigericin is a membrane-soluble K+-H+ exchanger). In CO2/HCO3--free media, inhibiting Na+-H+ exchange produced a prompt fall of pHi (background acid-loading), the rate of which was reduced by raising the extracellular K+ concentration, [K+]o. pHi recovery from an intracellular acid or alkali load was also sensitive to changes of [K+]o. These results are similar to those of

[Coupled transport of K+, Cl-, H+ and OH- ions in a liposome suspension].

By means of the system of ion-selective electrodes the coupling transport of K+, H+ and Cl- ions in liposome suspension was studied. Nigericin and tributyl tin (TBT) were used as inductors of cation and anion exchange. Effect of pH gradient created on the membrane on ionic transport was studied. The latter was shown to control the antiport exchange of both, cations and anions. At the joint effect of nigericin and TBT the yield of K+ and Cl- ions from liposomes independent of the pH gradient was observed.

Dysregulation of the KLF9/TXNRD2 Axis Leads to Mitochondrial DNA Oxidation and NLRP3 Inflammasome Activation in Ulcerative Colitis.

This study investigated the roles and mechanism of the transcription factor KLF9 and its downstream target gene TXNRD2 in ulcerative colitis (UC). An in vitro inflammatory model was established by stimulating human normal colonic mucosal epithelial cells (NCM460) with LPS and ATP. KLF9 or TXNRD2 expression was knocked down using RNA interference, and rescue experiments were performed using the NLRP3-specific agonist nigericin or by co-transfection with si-TXNRD2. A 3% DSS-induced UC model was established in C57BL/6 mice. Interventions were performed by tail vein injection of sh-KLF9 adeno-asso

Impaired NLRP3 inflammasome signaling diverts pyroptotic to apoptotic caspase activation in macrophages.

NLRP3 (NLR family pyrin domain-containing 3) inflammasome is a first line of defense of innate immunity, mediating caspase-1-dependent pyroptosis and cytokine release upon danger signaling. Intervention of NLRP3 innate surveillance may cause defects in this signaling pathway, while the host has evolved alternative ways to combat such intervention. Yet it remains incompletely understood whether NLRP3 sensing of danger signaling can divert pyroptosis to other forms of cell death in circumstances of impaired NLRP3 signaling. In this study, we adopted two macrophage models (delayed delivery of tri
