Root morphology is essential for plant survival. NO₃- is not only a nutrient, but also a signal substance affecting root growth in plants. However, the mechanism of NO₃--mediated root growth in rice remains unclear. In this study, we investigated the effect of OsNRT2.1 on root elongation and nitrate signaling-mediated auxin transport using OsNRT2.1 overexpression lines. We observed that the overexpression of OsNRT2.1 increased the total root length in rice, including the seminal root length, total adventitious root length, and total lateral root length in seminal roots and adventitious roots u

Potassium (K+) and nitrogen (N) are essential nutrients, and their absorption and distribution within the plant must be coordinated for optimal growth and development. Potassium is involved in charge balance of inorganic and organic anions and macromolecules, control of membrane electrical potential, pH homeostasis and the regulation of cell osmotic pressure, whereas nitrogen is an essential component of amino acids, proteins, and nucleic acids. Nitrate (NO3 -) is often the primary nitrogen source, but it also serves as a signaling molecule to the plant. Nitrate regulates root architecture, st

Nitrate uptake in Neurospora crassa has been investigated under various conditions of nitrogen nutrition by measuring the rate of disappearance of nitrate from the medium and by determining mycelial nitrate accumulation. The nitrate transport system is induced by either nitrate or nitrite, but is not present in mycelia grown on ammonia or Casamino Acids. The appearance of nitrate uptake activity is prevented by cycloheximide, puromycin, or 6-methyl purine. The induced nitrate transport system displays a K(m) for nitrate of 0.25 mM. Nitrate uptake is inhibited by metabolic poisons such as 2,4-d

Identification of mechanisms that decrease cadmium accumulation in plants is a prerequisite for minimizing dietary uptake of cadmium from contaminated crops. Here, we show that cadmium inhibits nitrate transporter 1.1 (NRT1.1)-mediated nitrate (NO3 (-)) uptake in Arabidopsis (Arabidopsis thaliana) and impairs NO3 (-) homeostasis in roots. In NO3 (-)-containing medium, loss of NRT1.1 function in nrt1.1 mutants leads to decreased levels of cadmium and several other metals in both roots and shoots and results in better biomass production in the presence of cadmium, whereas in NO3 (-)-free medium,

Five NRT2 genes and three Nar2 genes, encoding putative high-affinity nitrate transporters, and the respective cDNAs were identified and characterized in Physcomitrella patens. The deduced moss NRT2 and NAR2 proteins were more similar to the corresponding proteins of higher plants than to those of the green alga Chlamydomonas reinhardtii. Expression of all the genes was inhibited by ammonium added to the medium. The regulation by ammonium was abolished by an inhibitor of glutamine synthetase, but the effect of this inhibitor was counteracted by an inhibitor of glutamate synthase. Negative corr

In vivo recycling of nitrate (NO(3)(-)) and nitrite (NO(2)(-)) is an important alternative pathway for the generation of nitric oxide (NO) and maintenance of systemic nitrate-nitrite-NO balance. More than 25% of the circulating NO(3)(-) is actively removed and secreted by salivary glands. Oral commensal bacteria convert salivary NO(3)(-) to NO(2)(-), which enters circulation and leads to NO generation. The transporters for NO(3)(-) in salivary glands have not yet been identified. Here we report that sialin (SLC17A5), mutations in which cause Salla disease and infantile sialic acid storage diso

There is continuing concern over potential impacts of widespread application of nutrients and pesticides on ground- and surface-water quality. Transport and fate of nitrate and pesticides were investigated in a shallow aquifer and adjacent stream, Cow Castle Creek, in Orangeburg County, South Carolina. Pesticide and pesticide degradate concentrations were detected in ground water with greatest frequency and largest concentrations directly beneath and downgradient from the corn (Zea mays L.) field where they were applied. In almost all samples in which they were detected, concentrations of pest

Describing the mechanisms of zinc (Zn) accumulation in plants is essential to counteract the effects of excessive Zn uptake in crops grown in contaminated soils. Increasing evidence suggests that there is a positive correlation between nitrate supply and Zn accumulation in plants. However, the role of the primary nitrate transporter NRT1.1 in Zn accumulation in plants remains unknown. In this study, a Zn stress-induced increase in nitrate uptake and an increase in NRT1.1 protein levels in wild-type (Col-0) Arabidopsis plants were measured using microelectrode ion flux and green fluorescent pro

The duality of nitrate is nowhere best exemplified than in human physiology - a detrimental pollutant but also a protective nutrient and signaling ion - particularly as connected to reactive nitrogen oxides. Aside from limited insights into nitrate uptake and storage, foundational nitrate biology has lagged. Genetically encoded fluorescent biosensors can address this gap with real-time imaging. However, imaging technologies for mammalian cell applications remain rare. Here, we set out to design and engineer a two-domain chimera fusing the split green fluorescent protein EGFP and the nitrate re

Microbial souring in oil reservoirs produces toxic, corrosive hydrogen sulfide through microbial sulfate reduction, often accompanying (sea)water flooding during secondary oil recovery. With data from column experiments as constraints, we developed the first reactive-transport model of a new candidate inhibitor, perchlorate, and compared it with the commonly used inhibitor, nitrate. Our model provided a good fit to the data, which suggest that perchlorate is more effective than nitrate on a per mole of inhibitor basis. Critically, we used our model to gain insight into the underlying competing

Nitrate uptake in plants is mediated by coordinated transporter systems, which include NPF, NRT2 and NRT3 proteins. While these families have been extensively studied in angiosperms, their evolution and regulation in conifers are still not well understood. In this work, we examined the NRT3 family in maritime pine (Pinus pinaster) and in representative plant lineages. Phylogenetic analyses of nucleotide sequences and the NRT3 protein revealed a broad expansion in gymnosperms, particularly in conifers, while copy number increases among angiosperms appeared to be more lineage-specific. Nitric oxide (NO) is a vital immune molecule eliciting resistance to diverse microbial pathogens in humans and animals. However, its functional integration into plant immune networks remains incompletely characterized. In this study, we reveal that both endogenous induction and exogenous supplementation of NO significantly enhance resistance to rice stripe virus (RSV), a Bunyavirus that poses a huge threat to rice production. The nitrate transporter OsNPF6.1 potentiates virus resistance by upregulating the expression of nitrate reductase (OsNR2) and subsequent NO biosynthesis. Functional analy

The aims of this study are to investigate whether and how the nitrogen form (nitrate (NO3 (-)) versus ammonium (NH4 (+))) influences cadmium (Cd) uptake and translocation and subsequent Cd phytoextraction by the hyperaccumulator species Sedum plumbizincicola. Plants were grown hydroponically with N supplied as either NO3 (-) or NH4 (+). Short-term (36 h) Cd uptake and translocation were determined innovatively and quantitatively using a positron-emitting (107)Cd tracer and positron-emitting tracer imaging system. The results show that the rates of Cd uptake by roots and transport to the shoots

Magnesium (Mg) is fundamental for diverse physiological, biochemical, and molecular processes in higher plants because its deficiency or toxicity severely restricts development. The supply of Mg remodels the plant ionome and transporter profiles while simultaneously activating the nitrate reductase enzyme, which catalyzes nitrate reduction to nitrite and enhances nitrogen (N) assimilation. However, the specific mechanisms involving ion interactions and transporters in Mg metabolism that improve N metabolism in mulberry remain largely unexplored. To address this research gap, we employed physio

It is known that in Escherichia coli two dehydrogenases of the flavoprotein kind can participate in the transfer of hydrogens from sn-glycerol 3-phosphate (G3P) to nitrate and that possession of either enzyme is sufficient to permit anaerobic growth on glycerol as carbon source and nitrate as hydrogen acceptor. Results from this study show that under such a growth condition a protein with light-absorption characteristics of cytochrome b1 is induced. If G3P, nitrate, and adenosine diphosphate are added anaerobically to a particulate fraction prepared from these cells, four reactions can be dete

Residual plastic film in soil destroys the soil structure and changes the normal transportation and distribution of water and nutrient. In this work, we compared experiments on soil containing residual plastic film with a control experiment to investigate how residual film affects the transportation and distribution of water and nitrate (NO3-) in wetted soil. The experiment used Mariotte bottle, water tube, and soil tank to test six levels of residual-film concentration varying from 0 to 720 kg/hm2 in the soil. The wetting front, soil water content, and nitrate concentrations were all measured

Peroxyacetyl nitrate (PAN) is produced in the atmosphere by photochemical oxidation of non-methane volatile organic compounds in the presence of nitrogen oxides (NOx), and it can be transported over long distances at cold temperatures before decomposing thermally to release NOx in the remote troposphere. It is both a tracer and a precursor for transpacific ozone pollution transported from East Asia to North America. Here, we directly demonstrate this transport with PAN satellite observations from the infrared atmospheric sounding interferometer (IASI). We reprocess the IASI PAN retrievals by r

Identifying regional-scale surface water-groundwater interactions (SGI) is vital for predicting anthropogenic effects on surface water bodies and underlying aquifers. However, large-scale water and nutrient flux studies rely on surface water or groundwater-focused models. This study aims to model the effect of urbanization, which is usually accompanied by high groundwater abstraction and surface water pollution, particularly in the developing world, on a regional-scale SGI and nitrate loading. In the study area, the urban expansion increased by over 3% in the last decade. The integrated SWAT-M

Strigolactones (SLs) or their derivatives have recently been defined as novel phytohormones that regulate root development. However, it remains unclear whether SLs mediate root growth in response to phosphorus (P) and nitrogen (N) deficiency. In this study, the responses of root development in rice (Oryza sativa L.) to different levels of phosphate and nitrate supply were investigated using wild type (WT) and mutants defective in SL synthesis (d10 and d27) or insensitive to SL (d3). Reduced concentration of either phosphate or nitrate led to increased seminal root length and decreased lateral

Here, influx and efflux of (13)NO(3)(-), and net fluxes of (14)NO(3)(-) and (14)NO(2)(-), were measured in Aspergillus nidulans mutants niaD171 and niiA5, devoid of nitrate reductase (NR) and nitrite reductase (NiR) activities, respectively. Transcript and protein abundances of NrtA, the A. nidulans principal high-affinity NO(3)(-) transporter, were determined using semiquantitative reverse transcription-polymerase chain reaction and western blots, respectively. (13)NO(3)(-) influx in niaD171 was negligible relative to wild-type values, whereas efflux to influx ratios increased nine-fold. Neve
