Inactivation of Nosema spp. with zinc phthalocyanine.

Most honey bee pathogens, such as Vairimorpha (Nosema), cannot be rapidly and definitively diagnosed in a natural setting, consequently there is typically the spread of these diseases through shared and re-use of beekeeping equipment. Furthermore, there are no viable treatment options available for Nosema spores to aid in managing the spread of this bee disease. We therefore aimed to develop a new method using novel Zinc Phthalocyanine (ZnPc) as a photosensitizer for the photodynamic inactivation of Nosema spores that could be used for the decontamination of beekeeping equipment. Nosema spores

Nosema ceranae, a new parasite in Thai honeybees.

Adult workers of Apis cerana, Apis florea and Apis mellifera from colonies heavily infected with Nosema ceranae were selected for molecular analyses of the parasite. PCR-specific 16S rRNA primers were designed, cloned, sequenced and compared to GenBank entries. The sequenced products corresponded to N. ceranae. We then infected A. cerana with N. ceranae spores isolated from A. florea workers. Newly emerged bees from healthy colonies were fed 10,000, 20,000 and 40,000 spores/bee. There were significant dosage dependent differences in bee infection and survival rates. The ratio of infected cells

Proteomics of Anatomical Sections of the Gut of Nosema-Infected Western Honeybee (Apis mellifera) Reveals Different Early Responses to Nosema spp. Isolates.

Honeybees play an important role in pollinating native plants and agricultural crops and produce valuable hive products. Within the last decade, honeybee colonies have been reported to be in decline, due to both biotic and abiotic stress factors including pathogens and pesticides. This study evaluated the impact of different isolates of Nosema spp. [Nosema apis spores (NA), Nosema ceranae from Apis mellifera from France (NF), N. ceranae from Apis cerana from Thailand (NC1), and N. ceranae from A. mellifera from Thailand (NC2)] on the different gut sections of newly emerged adult A. mellifera b

Nosema ceranae in Apis mellifera: a 12 years postdetection perspective.

Nosema ceranae is a hot topic in honey bee health as reflected by numerous papers published every year. This review presents an update of the knowledge generated in the last 12 years in the field of N. ceranae research, addressing the routes of transmission, population structure and genetic diversity. This includes description of how the infection modifies the honey bee's metabolism, the immune response and other vital functions. The effects on individual honey bees will have a direct impact on the colony by leading to losses in the adult's population. The absence of clear clinical signs could

First detection and dominance of Nosema ceranae in Hungarian honeybee colonies.

Microsporidiosis (nosema disease) of the European honeybee ( Apis mellifera L.) is present in bee colonies worldwide. Until recently, Nosema apis had been regarded as the causative agent of the disease, which may have many negative effects on the colony and cause heavy economic losses in apicultures. Another microsporidium species, Nosema ceranae , was reported to infest the Asian honeybee ( Apis ceranae ), but both honeybee species are susceptible to both microsporidia. In the European honeybee N. ceranae was first detected in Spain in the year 2006. As it is difficult to distinguish N. ceran

New insights into the genome and transmission of the microsporidian pathogen Nosema muscidifuracis.

INTRODUCTION: Nosema is a diverse genus of unicellular microsporidian parasites of insects and other arthropods. Nosema muscidifuracis infects parasitoid wasp species of Muscidifurax zaraptor and M. raptor (Hymenoptera: Pteromalidae), causing ~50% reduction in longevity and ~90% reduction in fecundity. METHODS AND RESULTS: Here, we report the first assembly of the N. muscidifuracis genome (14,397,169 bp in 28 contigs) of high continuity (contig N50 544.3 Kb) and completeness (BUSCO score 97.0%). A total of 2,782 protein-coding genes were annotated, with 66.2% of the genes having two copies an

Screening of Dietary Ingredients against the Honey Bee Parasite Nosema ceranae.

Nosema ceranae is a major pathogen in the beekeeping sector, responsible for nosemosis. This disease is hard to manage since its symptomatology is masked until a strong collapse of the colony population occurs. Conversely, no medicaments are available in the market to counteract nosemosis, and only a few feed additives, with claimed antifungal action, are available. New solutions are strongly required, especially based on natural methods alternative to veterinary drugs that might develop resistance or strongly pollute honey bees and the environment. This study aims at investigating the nosemos

Nosema ceranae Infection Promotes Proliferation of Yeasts in Honey Bee Intestines.

BACKGROUND: Nosema ceranae infection not only damages honey bee (Apis melifera) intestines, but we believe it may also affect intestinal yeast development and its seasonal pattern. In order to check our hypothesis, infection intensity versus intestinal yeast colony forming units (CFU) both in field and cage experiments were studied. METHODS/FINDINGS: Field tests were carried out from March to October in 2014 and 2015. N. ceranae infection intensity decreased more than 100 times from 7.6 x 108 in March to 5.8 x 106 in October 2014. A similar tendency was observed in 2015. Therefore, in the Eur

Genetic detection and quantification of Nosema apis and N. ceranae in the honey bee.

The incidence of nosemosis has increased in recent years due to an emerging infestation of Nosema ceranae in managed honey bee populations in much of the world. A real-time PCR assay was developed to facilitate detection and quantification of both Nosema apis and N. ceranae in both single bee and pooled samples. The assay is a multiplexed reaction in which both species are detected and quantified in a single reaction. The assay is highly sensitive and can detect single copies of the target sequence. Real-time PCR results were calibrated to spore counts generated by standard microscopy procedur

Altered juvenile hormone dynamics mediate earlier foraging in Nosema-infected honey bees.

Parasites can profoundly alter host behavior, yet the physiological mechanisms underlying these changes are often unclear. In honey bees (Apis mellifera), microsporidian infection has long been associated with precocious foraging, a life-history shift that can destabilize colony function. Here, we identify the endocrine mechanism linking infection to behavioral maturation. Using Nosema apis, we show that infected workers initiate foraging significantly earlier than controls across four colonies (hazard ratio=2.69, P<0.001). This behavioral acceleration was accompanied by a ∼1.7-fold increase i

Ethanol Hormesis in Honeybees (Apis mellifera L.) Infected with Vairimorpha (Nosema) spp.

This study investigates the phenomenon of ethanol hormesis in honeybees (Apis mellifera) infected with Vairimorpha (Nosema) spp., a widespread parasite that significantly impacts bee health and colony survival. Hormesis refers to a biphasic response where low doses of potentially harmful substances may elicit beneficial effects, contrasting with the detrimental impacts observed at higher concentrations. We hypothesized that low ethanol concentrations could reduce Vairimorpha spp. infection severity and improve bee lifespan. In a controlled experiment, foraging bees were divided into groups of

Nosema ceranae infection reduces flight ability in queen and worker honey bees.

Nosema ceranae, a fungal pathogen that infects honey bees' midgut epithelial cells, is known to impact honey bees negatively in many aspects. Given that this pathogen changes honey bee physiology and causes a higher hunger status, we hypothesized that it may negatively impact the flight ability in honey bees. We fed newly emerged workers and virgin queens with 100,000 N. ceranae spores, let the bees age for 10 days, then tested their flight ability using flight mills. We found that the flight duration and distance of worker bees were significantly reduced by N. ceranae infection with a decreas

Nosema parkeri sp. n., a microsporidan from the argasid tick, Ornithodoros parkeri Cooley.

Nosema parkeri sp. n. is described from nymphs and adults of the argasid tick, Ornithodoros parkeri Cooley, from a laboratory colony. Schizogonic and sporogonic stages are described from various tick tissues. Spores are binucleate, measuring 3.2 (3-4) x 1.9 (1.8-2.5) micronm. Transmission is transovarial and transstadial. The parasite does not appear to affect adversely the development or reproduction of the tick. Dermacentor andersoni Stiles was experimentally infected. Attempts to infect Swiss mice by tick feeding or by injection of infected tick suspensions were unsuccessful. The microspori

Use of Thymol in Nosema ceranae Control and Health Improvement of Infected Honey Bees.

Nosema ceranae is the most widespread microsporidian species which infects the honey bees of Apis mellifera by causing the weakening of their colonies and a decline in their productive and reproductive capacities. The only registered product for its control is the antibiotic fumagillin; however, in the European Union, there is no formulation registered for use in beekeeping. Thymol (3-hydroxy-p-cymene) is a natural essential-oil ingredient derived from Thymus vulgaris, which has been used in Varroa control for decades. The aim of this study was to investigate the effect of thymol supplementati

Engineered Enterococcus faecalis disrupts redox homeostasis to suppress Nosema bombycis in Bombyx mori.

Nosema bombycis, the causative agent of pébrine disease, is a major pathogen of the economically important silkworm Bombyx mori, resulting in substantial losses in sericulture. While conventional control strategies offer limited efficacy, symbiont-mediated RNA interference (RNAi) has emerged as a sustainable and targeted strategy for disease control. This study investigates the role of gut microbiota in host defense and evaluates the potential of engineered Enterococcus faecalis to inhibit N. bombycis proliferation by disrupting its antioxidant systems. Occurrence of Nosema ceranae, Ascosphaera apis and trypanosomatids in Vespa orientalis linneus 1771.

Vespa orientalis is spreading across the Italian and European territories leading to new interactions among species, which could lead to the transmission of pathogens between species. Detection of honey bee viruses in V. orientalis has already been revealed in both adults and larvae, while no information is available regarding parasitic occurrence. Sixty adult hornets collected across apiaries in the South of Italy were subjected to cytological, histopathological and biomolecular examination to evaluate the occurrence of Nosema ceranae, Ascosphaera apis, Lotmaria passim, Crithidia mellificae,

Interactions between ionizing radiation and Vairimorpha (Nosema) ceranae on the honeybee, Apis mellifera L.

The global decline of honeybee colonies represents a major ecological concern, primarily attributed to simultaneous exposure to multiple stressors. These include biotic pressures, such as parasitic infections, and abiotic pressures, such as exposure to ionizing radiation, which remains poorly understood. Assessing their combined effects provides novel insights into how biological and radiological stressors interact within the organism. Here, we investigated the individual and combined effects of Vairimorpha ceranae (formerly Nosema ceranae) infection and chronic gamma irradiation (14 µGy/h or

Detection of Nosema ceranae in Euglossa imperialis (Apidae) and a synthesis of host associations across Hymenoptera.

Pathogen spillover from managed to wild pollinators represents an emerging threat to global pollinator diversity. However, surveillance of microbial pathogens in tropical wild bee communities remains severely limited. We investigated Nosema ceranae detection prevalence in the orchid bee Euglossa imperialis across seasons at 2 ecologically contrasting sites in the Panama Canal corridor: a protected tropical forest and a semi-urban landscape. Using species-specific polymerase chain reaction (PCR) assays, we detected N. ceranae DNA in gut tissues of this euglossine bee. Detection prevalence was s

Single-cell analysis reveals the effects of glutaraldehyde and formaldehyde on individual Nosema bombycis spores.

Nosema bombycis (Nb) is the pathogen that causes pebrine in silkworms. Aldehydes are effective disinfectants commonly used in sericulture. However, the precise mechanism of their action on Nb spores remains unclear. Here, we used laser tweezers Raman spectroscopy to investigate the effects of glutaraldehyde and formaldehyde on individual Nb spores, as well as phase contrast microscopy imaging to monitor the germination dynamics of individual treated spores, to acquire a deeper understanding of the mechanism of action of aldehydes and to provide a theoretical reference for establishing an effec
