The nucleus raphe magnus modulates hypoxia-induced hyperventilation but not anapyrexia in rats.

The nucleus raphe magnus (NRM) is one of the brainstem cell groups involved in physiological responses to hypoxia. Thus, we tested the hypothesis that the NRM modulates hypoxia-induced hyperventilation and anapyrexia. To this end, we assessed the participation of NRM in the respiratory and thermoregulatory responses to hypoxia using ibotenic acid lesions produced in the NRM of rats. Our results demonstrated that, under resting breathing, NRM plays no role in ventilation or body temperature. Hypoxia caused hyperventilation and anapyrexia in all groups. NMR lesions elicited an increased ventilat

Nucleus raphe magnus serotonin neurones bidirectionally control spinal nociceptive transmission in mice.

BACKGROUND: Noxious stimuli are conveyed to and integrated in the dorsal horn of the spinal cord before being transmitted to supraspinal centres, where pain perception is generated. Descending pathways from the brainstem dynamically modulate this integration, either facilitating or inhibiting nociceptive information based on physiological, emotional, genetic and environmental factors. Serotonergic neurones in the nucleus raphe magnus (NRM), activating different spinal 5-hydroxytryptamine (5-HT) receptors, exert bidirectional control, both facilitatory and inhibitory, but the underlying mechani

Opioid sensitivity of nucleus raphe magnus ater analgesia by nonsteroidal anti-inflammatory drugs.

Our recent investigations have shown that microinjection of three non-steroidal anti-inflammatory drugs (NSAIDs) analgin, ketorolac and xefocam into the central nucleus of amygdala produce tolerance to these drugs and cross-tolerance to morphine. We have observed the same phenomenon in midbrain periaqueductal grey matter and nucleus raphe magnus. The medullar nucleus raphe magnus (NRM) is one of important parts of CNS circuit that controls nociceptive transmission at the level of spinal cord. It is functionally involved in descending pain modulation, and mainly consists of serotoninergic neuro

Medullary responses to chemoreceptor activation are inhibited by locus coeruleus and nucleus raphe magnus.

The effect of electrical and L-glutamate stimulation of the nucleus locus coeruleus (LC) and nucleus raphe magnus (NRM) on multiunit activity evoked in the nucleus tractus solitarius (NTS) by activation of arterial chemoreceptors (15-25 micrograms kg-1 of sodium cyanoborohydride, i.v.) was studied in rats anaesthetized with urethane (1.1 g kg-1 i.p.). Multiunit discharge of NTS neurones in response to cyanide injection was composed by spikes higher than 200 microV and about 10 Hz frequency. Electrical and L-glutamate stimulation of the LC and the NRM significantly reduced the frequency of the

An explanation for reflex blink hyperexcitability in Parkinson's disease. II. Nucleus raphe magnus.

Hyperexcitable reflex blinks are a cardinal sign of Parkinson's disease. The first step in the circuit linking the basal ganglia and brainstem reflex blink circuits is the inhibitory nigrostriatal pathway (Basso et al., 1996). The current study reports the circuits linking the superior colliculus (SC) to trigeminal reflex blink circuits. Microstimulation of the deep layers of the SC suppresses subsequent reflex blinks at a latency of 5.4 msec. This microstimulation does not activate periaqueductal gray antinociceptive circuits. The brainstem structure linking SC to reflex blink circuits must s

Nicotine withdrawal induces hyperalgesia via downregulation of descending serotonergic pathway in the nucleus raphe magnus.

Patients deprived of cigarettes exhibit increased pain sensitivity during perioperative periods, yet the underlying neuroanatomical and molecular bases of this hypersensitivity are unclear. The present study showed that both the mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were significantly decreased in a rat model of nicotine withdrawal. These rats showed less tryptophan hydroxylase 2 (TPH2) positive neurons and reduced TPH2 expression in the nucleus raphe magnus (NRM), and thus resulted in decreased 5-hydroxytryptamine (5-HT) levels in cerebrospinal fluid. Modulatory effects of repeated psychophysical stress on masseter muscle nociception in the nucleus raphe magnus of rats.

Psychophysical stress can cause neural changes that increase nociception in the orofacial region, particularly the masseter muscle (MM). The nucleus raphe magnus (NRM), which is located in the brain stem, serves the crucial role of regulating nociception through descending modulatory pain control. However, it remains unclear if neural activities in the NRM are affected under psychophysical stress conditions. This study conducted experiments to assess (1) whether neural activity, indicated by Fos expression in an NRM that has experienced MM injury, is affected by the stress of repeated forced s

Nitric oxide pathway in the nucleus raphe magnus modulates hypoxic ventilatory response but not anapyrexia in rats.

Nucleus raphe magnus (NRM) is one of the cellular groups of the brainstem that is involved in the physiologic responses to hypoxia and contains nitric oxide (NO) synthase. In the present study, we assessed the role of NO pathway in the NRM on the hypoxic ventilatory response (HVR) and anapyrexia (a regulated decrease in body temperature). To this end, pulmonary ventilation (VE) and body temperature (Tb) of male Wistar rats were measured before and after microinjection of N-monomethyl-L-arginine (L-NMMA, a nonselective nitric oxide synthase inhibitor, 12.5 microg/0.1 microl) into the NRM, follo

Role of opioidergic and GABAergic neurotransmission of the nucleus raphe magnus in the modulation of tonic immobility in guinea pigs.

Tonic immobility (TI) is an inborn defensive behavior characterized by a temporary state of profound and reversible motor inhibition elicited by some forms of physical restraint. Previous results from our laboratory have demonstrated that nucleus raphe magnus (NRM) is also a structure involved in the modulation of TI behavior, as chemical stimulation through carbachol decreases the duration of TI in guinea pigs. In view of the fact that GABAergic and opioidergic circuits participate in the regulation of neuronal activity in the NRM and since these neurotransmitters are also involved in the mod

Stimulation sites in periaqueductal gray, nucleus raphe magnus and adjacent regions effective in suppressing oral-facial reflexes.

Electrode penetrations were made in the mesencephalon and caudal brainstem at the levels of the periaqueductal gray matter (PAG) and nucleus raphe magnus (NRM) in chloralose-anaesthetized or decerebrate cats. In a systematic fashion, mesencephalic and brainstem loci at every 1 mm of vertical depth were electrically stimulated in a series of mediolateral or anteroposterior electrode penetrations to determine the lowest stimulation threshold at each locus for suppressing the digastric jaw-opening reflex evoked by tooth pulp or infraorbital nerve stimulation; at some loci, the threshold current r

Origin of monoaminergic innervation of the nucleus raphe magnus--a combined monoamine histochemistry and fluorescent retrograde tracing study in the rat.

By the use of the fluorescent retrograde tracer, Fast Blue (FB), in combination with monoamine fluorescence histochemistry, the origin of monoaminergic input to the region of the nucleus raphe magnus (NRM) was investigated in the rat. After microinjection of FB into the NRM, a great number of FB-labeled NA-containing cells were found in the region of the nucleus reticularis lateralis (corresponding to A1 NA areas), the reticular formation just dorsolateral to the nucleus olivaris inferior (corresponding to the A3 NA area), the ventral part of the locus coeruleus (A6 NA area), and the lateral p

Kappa-opioid receptor-mediated enhancement of the hyperpolarization-activated current (I(h)) through mobilization of intracellular calcium in rat nucleus raphe magnus.

The hyperpolarization-activated current (Ih) is important in the control of resting membrane potential, in the regulation of network firing pattern and in the modulation of presynaptic transmitter release in central neurons. Recent studies on native and cloned Ih channels have demonstrated that the Ih channel is commonly modulated by cAMP through a positive shift in its voltage dependence without a change in its maximum current. The present study demonstrates that activation of kappa-opioid receptors enhances Ih by increasing its maximum current in brainstem neurons in the nucleus raphe magnus

Ratio of excitatory and inhibitory synaptic processes in periaqueductal gray matter of the brain activated by the raphe magnus nucleus in a model of Parkinson's disease with hydrocortisone protection.

BACKGROUND: The involvement of antinociceptive centers in neurodegeneration within the brain, particularly in Parkinson's disease (PD), which is accompanied by chronic pain, is not yet well understood. METHODS: Electrophysiological recordings were conducted on 15 king albino rats across three experimental conditions: intact, a rotenone-induced PD model, and a PD model treated with hydrocortisone. Extracellular spike activity was recorded from 241 single neurons in the periaqueductal gray (PAG) in response to stimulation of the raphe magnus nucleus (RMG). RESULTS: The PD model exhibited signi

Involvement of 5-HT(2) serotonergic receptors of the nucleus raphe magnus and nucleus reticularis gigantocellularis/paragigantocellularis complex neural networks in the antinociceptive phenomenon that follows the post-ictal immobility syndrome.

The post-ictal immobility syndrome is followed by a significant increase in the nociceptive thresholds in animals and men. In this interesting post-ictal behavioral response, endogenous opioid peptides-mediated mechanisms, as well as cholinergic-mediated antinociceptive processes, have been suggested. However, considering that many serotonergic descending pathways have been implicated in antinociceptive reactions, the aim of the present work is to investigate the involvement of 5-HT(2)-serotonergic receptor subfamily in the post-ictal antinociception. The analgesia was measured by the tail-fli

Actions of GABA, glycine, methionine-enkephalin and beta-endorphin compared with electrical stimulation of nucleus raphe magnus on responses evoked by tooth pulp stimulation in the medial reticular formation in the cat.

In decerebrate, cerebellectomized cats, a comparison was made between the effects of electrical stimulation in nucleus raphe magnus (NRM) and iontophoretic application of GABA, glycine, met-enkephalin and beta-endorphin on the responses of neurones in the medial brain stem reticular formation to tooth pulp stimulation. NRM stimulation, GABA, glycine and enkephalin produced a short lasting inhibition of tooth pulp evoked responses whilst the time course of the inhibition produced by beta-endorphin was much slower, often lasting up to 1 h following a 3-7 min ejection period. The effects of GABA

Afferent projections to the rat nuclei raphe magnus, raphe pallidus and reticularis gigantocellularis pars alpha demonstrated by iontophoretic application of choleratoxin (subunit b).

The aim of the present study was to identify the specific afferent projections to the rostral and caudal nucleus raphe magnus, the gigantocellular reticular nucleus pars alpha and the rostral nucleus raphe pallidus. For this purpose, small iontophoretic injections of the sensitive retrograde tracer choleratoxin (subunit b) were made in each of these structures. In agreement with previous retrograde studies, after all injection sites, a substantial to large number of labeled neurons were observed in the dorsal hypothalamic area and dorsolateral and ventrolateral parts of the periaqueductal gray

Impact of adolescent ethanol binge on serotonin signaling and pain sensitivity post-withdrawal.

Adolescence is a critical neurodevelopmental period characterized by heightened neuroplasticity. While the acute effects of binge ethanol (EtOH) consumption are documented, its long-term impact on both pain sensitivity and microglial activation during adolescence remains unclear. Given serotonin's (5-HT) known involvement in pain processing and sensitivity to EtOH, this study examined the effects of adolescent EtOH binge on microglia-induced neuroinflammation in serotonergic nuclei, downstream 5-HT signaling, and pain sensitivity at different time points after EtOH withdrawal. Adolescent male

Involvement of the hypothalamus-raphe magnus-spinal defecation center axis in stress-induced defecation in rats.

Acute psychological stress stimulates colonic motility; however, the specific descending neural pathways linking the brain to the distal colon remain incompletely characterized. Here, we provide evidence that the hypothalamus‒raphe magnus‒spinal cord‒pelvic nerve pathway mediates stress-induced defecation in male Sprague-Dawley rats. Using urethane-anesthetized rats, we used viral tracing, pharmacological, and chemogenetic approaches to show that hypothalamic neurons projecting to the raphe magnus nucleus activate descending serotonergic neurons probably via oxytocinergic signaling, thereby st
