Characterization of the soluble allergenic proteins of cashew nut (Anacardium occidentale L.).

The allergens associated with cashew food allergy have not been well-characterized. We sought to identify the major allergens in cashew nut by performing IgE immunoblots to dissociated and reduced or nonreduced cashew protein extracts, followed by sequencing of the peptides of interest. Sera from 15 subjects with life-threatening reactions to cashews and 8 subjects who tolerate cashews but have life-threatening reactions to other tree nuts were compared. An aqueous cashew protein extract containing albumin/globulin was separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS

Antioxidant pine nut protein hydrolysate as a therapeutic oral immunotherapy agent in a murine model of pine nut allergy.

Oral immunotherapy (OIT) is one of the most promising treatments for food allergy, although no protocols have yet been described for pine nut allergy. We aim to produce a pine nut protein hydrolysate to be used as a treatment agent in OIT. Hydrolysates were produced using three proteases (pepsin, trypsin, and Alcalase) alone or in combination and their antioxidant activity, antigenicity, and immunomodulatory properties were determined in vitro. Pine nut-sensitized C57BL/6 mice were subjected to OIT with intact and hydrolysed pine nut protein. The trypsin hydrolysate showed higher antioxidant a

Immunodetection of NUT Protein: Implementation, Indications, and Results in a Tertiary Reference Center.

The immunodetection of NUT protein is a reliable tool to identify NUT carcinoma, a rare and still underdiagnosed tumor entity. The technique was implemented in 2017 in our department, a tertiary reference center with a large recruitment in all tumor types, including head and neck and thoracic tumors. We evaluated its use over a 6-year period (2017-2022) to (a) describe the indications for the technique, (b) determine the number of NUT carcinomas detected and confirmed by Fluorescence in situ hybridization, and (c) describe briefly the characteristics of these tumors. Over the study period, 382

Anaphylaxis to pine nuts and immunological cross-reactivity with pine pollen proteins.

Despite the wide use of pine nuts, the fruit of Pinus pinea, only a few reports of allergic reactions to them have been published. We present herein a case of food allergy to pine nuts in a patient who showed no clinical symptoms to pine pollen despite the presence in her serum of specific IgE antibodies. In order to verify whether the reaction against pine nuts was IgE mediated, specific IgE against pine nuts and pollen were evaluated by skin-prick test, prick by prick and RAST. Immunoblotting and immunoblotting-inhibition were used to evaluate the allergenic components of both extracts and t

Nuclear Protein in Testis (NUT) Carcinoma With Nasopharyngeal and Intracranial Involvement: A Case Report.

Nuclear protein in testis (NUT) carcinoma is a rare and aggressive malignancy characterized by NUT gene rearrangement. Previously classified as NUT midline carcinoma, this malignancy was reclassified to NUT carcinoma in order to reflect that it can occur outside midline structures of the body. This case report follows a 43-year-old male presenting with a nasopharyngeal mass invading the frontal brain region, ultimately diagnosed as NUT carcinoma. Here, we detail the clinical and molecular characteristics of this tumor to contribute to the growing body of literature on this rare malignancy, whi

Metabolic and Epigenetic Reprogramming in a Case of Nuclear Protein in Testis (NUT) Carcinoma of the Retroperitoneum.

Nuclear protein in testis (NUT) carcinoma is a rare but highly aggressive carcinoma, driven by genetic rearrangement of the NUT midline carcinoma family member 1 (NUTM1) gene on chromosome 15q14. Recently, a tight link has been suggested between genetic abnormalities and subsequent metabolic and epigenetic dysregulation to drive the progression of malignant tumors. However, it remains elusive whether such reprogramming could contribute to the pathogenesis of NUT carcinoma. We herein report an autopsy case of NUT carcinoma arising in the retroperitoneum of a 31-year-old male. Notably, reprogram

Tolerance of peanuts and tree nuts in Spanish children with exclusive sensitization to lipid transfer proteins.

BACKGROUND: Allergy to peanuts and tree nuts is a common cause of food allergy in Spain, with lipid transfer proteins (LTP) being the most frequently recognized panallergen. LTP sensitization often leads to multiple food group sensitivities, resulting in overly restrictive diets that hinder patient's quality of life. This study aimed to assess the tolerance of peanuts and tree nuts (hazelnuts and walnuts) in children sensitized to LTP, potentially mitigating the need for such diets. METHODS: This prospective study enrolled individuals diagnosed with allergy to peanuts, hazelnuts, or walnuts.

Mixed Nut Challenge Test (MixNut) as an Efficient Procedure in the Management of Lipid Transfer Protein Allergy.

Background/Objectives: Lipid transfer protein (LTP) syndrome is a leading cause of primary food allergy in Mediterranean countries, often associated with severe reactions. Due to in vitro cross-reactivity among plant foods, clinical manifestations are unpredictable, frequently requiring multiple oral food challenges (OFC) to assess nut tolerance. These procedures increase healthcare burden and patient anxiety. This study evaluated the safety and utility of a mixed-nut oral food challenge (MixNut) in LTP-sensitized patients. Methods: In this prospective observational multi-center study, patient

Antidiabetic effects of water-soluble Korean pine nut protein on type 2 diabetic mice.

Korean pine nut protein (PNP) has a variety of biological activities, which are good for human health, but its ability to preventing diabetes has not been reported. This study evaluated the effects of water-soluble proteins of Korean pine nut obtained from a dilute alkali extract on carbohydrate metabolism of type 2 diabetic mice on a model of diabetes induced using a high fat diet combined with streptozotocin. The results showed that the hypoglycemic effect of PNP at a middle dose was the most significant, which was 38.7% lower than that of control. The extract significantly improved the oral

Adolescent dietary fiber, vegetable fat, vegetable protein, and nut intakes and breast cancer risk.

The importance of early-life exposures in breast cancer development is increasingly recognized. However, limited research has evaluated the relationship between adolescent diet and subsequent risk of breast cancer and reported inconsistent results. This population-based case-control study investigated the associations of dietary fiber, vegetable protein, vegetable fat, and nuts consumed during adolescence with adult breast cancer risk. Women, ages 25-74 years, who were diagnosed with first primary breast cancer between 2002 and 2003, were identified using the Ontario Cancer Registry. Controls

Retrospective analysis of nuclear protein in testis (NUT) midline carcinoma in the upper aerodigestive tract and mediastinum.

OBJECTIVE: Nuclear protein in testis (NUT) midline carcinoma (NMC) is a very aggressive tumor with limited survival, recently recognized as a subset of poorly differentiated squamous cell carcinoma. A simple chromosomal translocation results in NUT overexpression and malignant transformation. This study used immunohistochemistry to retrospectively diagnose and characterize NMC cases. STUDY DESIGN: Immunoperoxidase staining was performed according to a standard protocol and interpreted independently by two pathologists. Scores were based on nuclear staining with monoclonal NUT antibody (C52B1)

Acute tree nut-triggered food protein-induced enterocolitis syndrome in two infants: a case report.

INTRODUCTION: Food protein-induced enterocolitis syndrome (FPIES) is a non-IgE-mediated food allergy characterised by delayed repetitive vomiting and can be difficult to distinguish from infectious illness or even an IgE-mediated allergy. Tree nuts are uncommon but increasingly recognised triggers, and evidence regarding atypical FPIES with concomitant IgE sensitisation remains limited. CASE PRESENTATION: We describe two infants with severe acute reactions to tree nuts who were managed at a paediatric allergy centre. The first, a boy with eczema and cow's milk allergy, developed reproducible

Evaluation of the digestibility and antioxidant activity of protein and lipid after mixing nuts based on in vitro and in vivo models.

This study aimed to evaluate the change of digestibility and antioxidant activity of protein and lipid after mixing walnuts, cashews, and pistachios using in vitro and in vivo models. The results showed that mixed nuts significantly reduced the digested particle size and the degree of hydrolysis of protein and triacylglycerol compared to single nuts in vitro. As a consequence of co-digestion, bioaccessibility and antioxidant activity for amino acids and fatty acids were increased by 1.12-1.87 fold and 1.62-3.81 fold, respectively. In vivo studies, the mixed nuts diet increased the concentratio

Hard nut to crack: Solving the disulfide linkage pattern of the Neosartorya (Aspergillus) fischeri antifungal protein 2.

As a consequence of the fast resistance spreading, a limited number of drugs are available to treat fungal infections. Therefore, there is an urgent need to develop new antifungal treatment strategies. The features of a disulfide bond-stabilized antifungal protein, NFAP2 secreted by the mold Neosartorya (Aspergillus) fischeri render it to be a promising template for future protein-based antifungal drug design, which requires knowledge about the native disulfide linkage pattern as it is one of the prerequisites for biological activity. However, in the lack of tryptic and chymotryptic proteolyti

A sodium alginate-dual-protein double-network gel based on whey protein isolate and Tiger nut protein: Preparation, characterization and its sustained release properties of curcumin.

This study developed a sodium alginate-tiger nut protein/whey protein isolate double-network gel, and its sustained-release properties for curcumin were investigated. A series of sodium alginate (SA) concentrations (0-0.3%) were screened, and 0.25% SA was determined as the optimal level, which yielded superior gel hardness (401.67 ± 16.04 g) and water-holding capacity (81.49 ± 1.96%). Structural characterizations verified a double-network structure formation: one relied on protein aggregation, while the secondary network was formed via calcium ions-mediated cross-linking of SA. Owing to the du

Efficacy of cashew nut protein immunotherapy: protocol for a single-centre randomised controlled trial in a Polish paediatric population.

INTRODUCTION: The prevalence of food allergies, particularly IgE-mediated allergies, is rising in developed countries, with cashew nut allergy emerging as a significant public health concern due to its potential for severe anaphylaxis and frequent association with atopic disorders. Cashew nuts are among the most common allergens in Europe and Australia, often involving cosensitisation with pistachios, hazelnuts and other allergens. Diagnosis relies on clinical history, measurement of specific IgE (sIgE) levels, skin prick tests (SPT) and oral food challenges (OFCs). Current management strategi

Nuclear Protein in Testis (NUT) Carcinoma: Diagnostic Challenges in a Patient Requiring Rescue Venovenous Extracorporeal Membrane Oxygenation.

Nuclear protein in testis (NUT) carcinoma is a rare, highly aggressive malignancy defined by NUTM1 rearrangement, most commonly involving BRD4. Nonspecific presentation and poorly differentiated histology lead to frequent misdiagnosis. Patients often present with large mediastinal masses, making swift diagnosis both important and challenging. A 29-year-old woman presented with two months of progressive dyspnea, cough, and fatigue accompanied by new-onset hemoptysis. CT demonstrated a large right hilar and mediastinal mass compressing the trachea, pulmonary artery, and superior vena cava (SVC).
