Tranexamic acid attenuates oleic-acid-induced pulmonary extravasation.

OBJECTIVE: Activation of fibrinolysis is implicated in the development of vascular injury in certain lung injuries. It has yet to be reported that activation of plasmin is involved in extravasation caused by oleic acid (OA). We examined whether or not plasmin is involved in pulmonary extravasation by OA. DESIGN: Prospective trial. SETTING: University laboratory. SUBJECTS: A total of 78 guinea pigs (498.9 +/- 10.6 g). INTERVENTIONS: Evans blue (EB) was administered to anesthetized guinea pigs. Subsequently four protocols were followed: (1) After 1 min, 60 micro l/kg of OA was injected. Perf

Hyaluronic acid conjugates with controlled oleic acid substitution as new nanomaterials for improving ocular co-delivery of cyclosporine A and oleic acid.

A structural conjugate (HOC) of polysaccharide, hyaluronic acid (HA) with different ratios of oleic acid (OA) via cystamine (CYS) linker as a new ocular biomaterial was developed. The HOCs with controlled degrees of substitution of OA (4.6 %, 8.3 % and 12.2 %) were synthesized to form self-assembled HA-CYS-OA nanoparticles (HONs, HON1, HON2, HON3). A poorly water-soluble cyclosporine A (CsA) to be used for the treatment of multifactorial dry eye disease (DED) was chosen as model drug. CsA-loaded HONs exhibited improved solution transparency via solubilizing capacity of HON, and increased in vi

Metabolomic profiling of HepG2/C3A liver-on-chip model exposed to oleic acid, palmitic acid and oleic/palmitic acids mixture.

Metabolic dysfunction associated steatotic liver disease (MASLD) is one of the most predominant liver disorders. MASLD is associated with dietary intake of fatty acids. Among them, oleic (OA) and palmitic (PA) acids are the most abundant. Furthermore, OA was reported to have an hepatoprotective effect against PA toxicity on primary hepatocytes and liver cell lines. In this work, to characterize the mechanisms related to fatty acids exposure, we analyzed the metabolomic signatures of HepG2/C3A using advanced organ-on-chip technology. The cells were exposed for 2 to 7 days to OA, PA and OA/PA mi

"Resolving the SCD1-oleic acid paradox: majority of oleic acid is converted to free cholesterol in colorectal cancer cells".

OBJECTIVE: This study aimed to elucidate the mystery of “disappearing” oleic acid (OA) in colorectal cancer (CRC) cells. The primary objective was to verify if OA is predominantly used as an energy source or as a substrate for the synthesis of other molecules, given the paradox of elevated stearoyl-CoA desaturase-1 (SCD1) expression and lower OA levels in CRC tissue. METHODS: Tissue samples from CRC patients were analyzed for fatty acid (FA) profiles (GC-MS) and SCD1 mRNA levels (real-time PCR). For in-vitro experiments, CRC cell lines (HT-29, WiDr) and a normal human colon cell line (CCD-881-

Protective role of oleic acid against palmitic acid-induced pancreatic fibrosis.

BACKGROUND: Obesity has been associated with several pancreatic disorders and is an important risk factor for pancreatic cancer. Nevertheless, the role of lipids in the early steps of carcinogenesis is unknown. Although we previously identified two types of pancreatic fatty infiltration with different lipid compositions that were associated with precancerous lesions and fibrosis, their mechanisms of action have not been clarified. METHODS: We hypothesized that saturated palmitic acid and mono-unsaturated oleic acid (OA and PA) could play diverse roles in the activation of pancreatic stellate

DDX17 protects hepatocytes against oleic acid/palmitic acid-induced lipid accumulation.

Hepatic lipid accumulation is an initiation factor in fatty liver disease, and promoting a reduction in hepatic lipid accumulation is an important treatment strategy. DEAD box RNA helicase 17 (DDX17) is a member of the DEAD-box family and a molecular chaperone. Previous studies have demonstrated that DDX17 is a transcriptional coregulator of tumorigenesis, inflammation, and macrophage cholesterol efflux. The liver is the main site for lipid metabolism, and metabolic (dysfunction)-associated fatty liver disease (MAFLD) is one of the most common chronic liver diseases. However, the impact of DDX

Association of meibum oleic acid with meibomian seborrhea.

PURPOSE: We sought to determine whether patient meibum fatty acid unsaturation is associated with meibomian gland secretion consistency. METHODS: Meibomian gland secretions were dissolved in chloroform and separated into lipid classes by thin-layer chromatography. Fatty acids in individual lipid classes were characterized by gas chromatography after transmethylation. RESULTS: Meibum from patients with meibomian seborrhea was higher, and meibum from patients with meibomian keratoconjunctivitis (meibomianitis) was lower in the monounsaturated fatty acid, oleic acid, than that of other disease

Pathogenesis of oleic acid-induced lung injury in the rat: distribution of oleic acid during injury and early endothelial cell changes.

Oleic acid-induced lung injury, a well-described laboratory model for acute pulmonary injury in the rat and other species, causes morphologic and cellular changes similar to human adult respiratory distress syndrome (ARDS). Experiments were performed to test the hypothesis that the initial event of oleic acid lung injury is damage of the pulmonary vascular endothelium by oleic acid, with subsequent pulmonary damage and inflammation. Oleic acid levels were followed in the lung and other tissues by measuring accumulation of 14C-oleic acid; the direct effects of oleic acid and other fatty acids o

Acid-base status affects gas exchange in canine oleic acid pulmonary edema.

The effects of acidosis and alkalosis on pulmonary gas exchange were studied in 32 pentobarbital sodium-anesthetized intact dogs after induction of oleic acid (0.06 ml/kg) pulmonary edema. Gas exchange was assessed at constant ventilation and constant cardiac output, by venous admixture calculations and by intrapulmonary shunt measurements using the sulfur hexafluoride (SF6) method. Metabolic acidosis (pH 7.20) and alkalosis (pH 7.60) were induced with HCl and Carbicarb (isosmolar Na2CO3 and NaHCO3), respectively. Hypercapnia was induced by adding inspiratory CO2, whereas pH was allowed to cha

Mixed Oleic Acid-Erucic Acid Liposomes as a Carrier for Anticancer Drugs.

BACKGROUND: Liposomes are mostly known to be prepared from phospholipids and lipids and have a remarkable capacity to encapsulate both lipophobic and lipophilic molecules. However, there is little research on developing fatty acid liposomes for chemotherapy. OBJECTIVE: We have successfully prepared mixed fatty acid liposomes from two monounsaturated fatty acids, namely oleic acid and erucic acid, which stabilised by DOPEPEG2000. The Critical Vesicular Concentration (CVC) of liposomes was found to be within 0.09 to 0.21 mmol dm-3, with an average particle size of 400 nm. METHODS: Encapsulatio

Oleic acid derived metabolites in mouse neuroblastoma N18TG2 cells.

Oleamide is an endogenous sleep-inducing lipid that has been isolated from the cerebrospinal fluid of sleep-deprived mammals. Oleamide is the best-understood member of the primary fatty acid amide family. One key unanswered question regarding oleamide and all other primary acid amides is the pathway by which these molecules are produced. One proposed pathway involves oleoyl-CoA and N-oleoylglycine as intermediates: oleic acid --> oleoyl-CoA --> N-oleoylglycine --> oleamide. The first and third reactions are known reactions, catalyzed by acyl-CoA synthetase and peptidylglycine alpha-amidating m

Lipoprotein profile in canine pancreatitis induced with oleic acid.

Lipid and lipoprotein concentrations and apolipoprotein profile were investigated in canine pancreatitis induced by infusion with oleic acid (OA) into the accessory pancreatic duct. Pancreatitis was diagnosed by physical, hematological, biochemical and pathological examinations. In OA-treated dogs, serum triglyceride (TG) concentration was increased; however, there were no changes in serum total cholesterol (TC) and phospholipid (PL) concentrations. Serum concentrations of TG, TC, PL and total lipids (TL) in beta lipoprotein and those of TC, PL and TL in pre-beta lipoprotein were increased and

Apoptosis of Acanthamoeba castellanii Trophozoites Induced by Oleic Acid.

Acanthamoeba spp. can be parasitic in certain situations and are responsible for serious human infections, including Acanthamoeba keratitis, granulomatous amoebic encephalitis, and cutaneous acanthamoebiasis. We analyzed the fatty acid composition of Acanthamoeba castellanii trophozoites and tested the inhibitory activity of the main fatty acids, oleic acid and arachidonic acid, in vitro. Oleic acid markedly inhibited the growth of A. castellanii, with trophozoite viability of 57.4% at a concentration of 200 μM. Caspase-3 staining and annexin V assays showed that apoptotic death occurred in A.

Sodium nitroprusside mitigates oleic acid-induced acute lung injury.

BACKGROUND: Acute lung injury (ALI) is associated with pulmonary hypertension, intrapulmonary shunting, and increased microvascular permeability, leading to altered oxygenation capacity. Oleic acid (OA) creates a significant ALI that physiologically mimics human adult respiratory distress syndrome (ARDS). It has been hypothesized that pulmonary vasodilatation may improve ALI. Studies in our laboratory using this model and nitric oxide (NO) have shown that NO inhalation is detrimental and worsens the effects of OA. We studied the effect of pretreatment with a potent vasodilator, sodium nitropru

Sequential cardiopulmonary changes after oleic-acid injury in dogs.

Oleic acid (OA) administered to experimental animals increases pulmonary vascular permeability and produces a condition that pathophysiologically resembles adult respiratory distress syndrome (ARDS) in humans. The present study examined the sequence of cardiorespiratory changes after OA infusion and their similarity to ARDS. After a baseline period, mechanically ventilated and anesthetized dogs were administered 0.18 ml/kg body weight OA into the pulmonary artery while hemodynamic and respiratory changes were monitored. After OA infusion, cardiac output fell by 39%, paralleling a 26% decrease

From dysbiosis to homeostasis: Oleic acid matters in the vagina.

The role of fatty acids in shaping vaginal microbiota remains unclear. In an issue of Cell, Zhu et al. use genomic and transcriptomic analyses to reveal that oleic acid (OA) selectively inhibits L. iners while promoting L. crispatus, suggesting new strategies for the treatment of bacterial vaginosis (BV).

Palmitic and oleic acids in type 2 diabetes mellitus.

The most common dietary and plasma fatty acids (FAs), palmitic and oleic acids, have opposing effects on the risk of developing insulin resistance and type 2 diabetes mellitus (T2DM). Palmitic acid has been strongly associated with the presence of T2DM, while oleic acid has not and may even counteract the detrimental effects of palmitic acid. Despite this, recent cohort studies have shown no association or even conflicting results, questioning these roles. This review summarizes the recently discovered molecular mechanisms by which palmitic acid inhibits insulin sensitivity and influences the

Postsynthetic Degradation of Toxic Quantum Dots via Oleic Acid Complexation.

Quantum-confined semiconductor nanocrystals (NCs) have found extensive applications in optoelectronics. However, their inherent toxicity, often due to the incorporation of heavy and toxic metals, poses significant environmental and health concerns. In this study, we introduce a postsynthesis degradation strategy for CdSe quantum dots (QDs) based on the use of oleic acid (OA), a long-chain fatty acid capable of complexing with heavy metals. The degradation of nanocrystals is monitored in situ using UV-vis spectroscopy, small-angle X-ray scattering (SAXS), and transmission electron microscopy (T

Ferroptosis inhibition by oleic acid mitigates iron-overload-induced injury.

Iron overload, characterized by accumulation of iron in tissues, induces a multiorgan toxicity whose mechanisms are not fully understood. Using cultured cell lines, Caenorhabditis elegans, and mice, we found that ferroptosis occurs in the context of iron-overload-mediated damage. Exogenous oleic acid protected against iron-overload-toxicity in cell culture and Caenorhabditis elegans by suppressing ferroptosis. In mice, oleic acid protected against FAC-induced liver lipid peroxidation and damage. Oleic acid changed the cellular lipid composition, characterized by decreased levels of polyunsatur

Mouse Model of Oleic Acid-Induced Acute Respiratory Distress Syndrome.

Acute respiratory distress syndrome (ARDS) is a significant threat to critically ill patients with a high fatality rate. Pollutant exposure, cigarette smoke, infectious agents, and fatty acids can induce ARDS. Animal models can mimic the complex pathomechanism of the ARDS. However, each of them has limitations. Notably, oleic acid (OA) is increased in critically ill patients with harmful effects on the lung. OA can induce lung injury by emboli, disrupting tissue, altering pH, and impairing edema clearance. OA-induced lung injury model resembles various features of ARDS with endothelial injury,
