Difference in Serum Scuticocidal Activity Against Miamiensis avidus (Ciliophora, Scuticociliatida) Between Olive Flounder (Paralichthys olivaceus) and Starry Flounder (Platichthys stellatus).

Miamiensis avidus is an opportunistic parasite responsible for significant mortalities in marine aquaculture. Given the rising economic losses associated with scuticociliatosis in olive flounder (Paralichthys olivaceus) cultured in Korea, alternative species such as starry flounder (Platichthys stellatus), which demonstrates resistance to certain pathogens affecting olive flounder, are being considered for aquaculture. However, the underlying biological mechanisms influencing host susceptibility to M. avidus remain unclear. In this study, we investigated differences in susceptibility to M. avi

Removal of two pathogenic scuticociliates Miamiensis avidus and Miamiensis sp. using cells or culture filtrates of the dinoflagellate Alexandrium andersonii.

Scuticociliatosis, which is caused by parasitic protistan pathogens known as scuticociliates, is one of the most serious diseases in marine aquaculture worldwide. Thus, elimination of these ciliates is a primary concern for scientists and managers in the aquaculture industry. To date, formalin and other toxic chemicals have been used as anti-scuticociliate agents, but issues regarding their secondary effects often arise. Consequently, development of safer methods is necessary. To find out a safe method of controlling scuticociliate populations in aqua-tanks or small-scale natural environments,

Novel contribution to the taxonomy and molecular phylogeny of scuticociliates (Protozoa): Resolving the non-monophyly of the genus Paranophrys Thompson and Berger, 1965 and the establishment of the genus Anteparanophrys n. gen.

During a field survey of parasitic ciliates diversity in South Korea, a scuticociliate was found in a water sample collected during scuba diving. At first glance, the species looks similar to members of the genus Paranophrys especially P. magna but they differ mainly in the body size and the number of somatic kineties. Thus, integrative taxonomic approaches (i.e., silver impregnations, scanning electron microscopy, and molecular phylogeny) were applied and confirmed that the isolate is a new species. Detailed comparison between the new species and the three members of the genus Paranophrys rev

Phylogenomic analysis of Balantidium ctenopharyngodoni (Ciliophora, Litostomatea) based on single-cell transcriptome sequencing.

In this paper, we present transcriptome data for Balantidium ctenopharyngodoni Chen, 1955 collected from the hindgut of grass carp (Ctenopharyngodon idella). We evaluated sequence quality and de novo assembled a preliminary transcriptome, including 43.3 megabits and 119,141 transcripts. Then we obtained a final transcriptome, including 17.7 megabits and 35,560 transcripts, by removing contaminative and redundant sequences. Phylogenomic analysis based on a supermatrix with 132 genes comprising 53,873 amino acid residues and phylogenetic analysis based on SSU rDNA of 27 species were carried out

Phylogeny, morphology, and ultrastructure of sessilid peritrich ciliates in termite guts, Termitophrya africana and Doliophrys denislynni gen. nov., sp. nov.

Sessilids (Oligohymenophorea, Peritrichia, Sessilida) reportedly colonize the gut of certain "higher termites" (family Termitidae), but only a single species, Termitophrya africana from Jugositermes tuberculatus (subfamily Apicotermitinae), has been described based on a drawing. Similar ciliates were observed in other Apicotermitinae but remained unspecified. Our SSU rRNA gene-based survey of peritrich ciliates in a broad range of Termitidae recovered distinct phylotypes from several Apicotermitinae (Astalotermes, Jugositermes, and Phoxotermes), albeit only from samples collected in Cameroon a

Scuticociliate (Philasterides dicentrarchi) infection cluster in a multispecies marine aquarium system.

Scuticociliatosis, caused by ciliated protozoa of the subclass Scuticociliatia, has been associated with high mortalities in marine fish. Environmental factors such as an increase in water temperature can enhance this disease. The aim of the present report is to describe the occurrence of a cluster of cases of scuticociliatosis in a multispecies marine cold-water system in a public aquarium. Philasterides dicentrarchi was identified by PCR in formalin-fixed tissues of some of the fish showing meningitis or meningoencephalitis, dermatitis and myositis with intralesional protozoa. An increase in

Miamiensis avidus (Ciliophora: Scuticociliatida) causes systemic infection of olive flounder Paralichthys olivaceus and is a senior synonym of Philasterides dicentrarchi.

The scuticociliate Miamiensis avidus was isolated from olive flounder Paralichthys olivaceus showing typical symptoms of ulceration and hemorrhages in skeletal muscle and fins. In an infection experiment, olive flounder (mean length: 14.9 cm; mean weight: 26.8 g) were immersion challenged with 2.0 x 10(3), 2.0 x 10(4) and 2.0 x 10(5) ciliates ml-1 of the cloned YS1 strain of M. avidus. Cumulative mortalities were 85% in the 2.0 x 10(3) cells ml-1 treatment group and 100% in the other 2 infection groups. Many ciliates, containing red blood cells in the cytoplasm, were observed in the gills, ske

First Investigation of Trichodinid Species (Ciliophora: Trichodinidae) on Farmed Hybrid Yellow Catfish (Tachysurus fulvidraco × Tachysurus vachelli) in China.

Hybrid yellow catfish (Tachysurus fulvidraco♀ × Tachysurus vachelli♂) is a significant aquaculture variety in China. It is easy to be infected by trichodinids, causing serious economic losses. However, the species investigation of trichodinids on hybrid yellow catfish is relatively deficient, which seriously hinders the prevention and control of trichodiniasis. Hence, trichodinids were collected in a hybrid yellow catfish farm in Hubei Province from June 2022 to October 2023 and identified by morphological and molecular biology methods. A new species Tripartiella luhuensis sp. nov. and nine Tr

Characteristics of different scuticociliates from an infected olive flounder (Pararlichthys olivaceus) farm in South Korea.

Parasitic diseases are considered to be a major problem in olive flounder (Paralichthys olivaceus) farms in South Korea, with scuticociliate infections being especially problematic due to their year-round occurrence and difficulty in control. In this study, we collected scuticociliates from flounder showing typical symptoms of scuticociliasis, such as ulcers, hemorrhages, and necrosis, at farms in Busan and Jeju, South Korea. Four species of scuticociliates were identified: Mesanophrys carcini, Miamiensis avidus, Paralembus digitiformis, and Pseudocohnilembus persalinus, the M. carcini and P.

Formulation of chitosan microsphere-based oral vaccine against the scuticociliate parasite Miamiensis avidus: Evaluation of its protective immune potency in olive flounder (Paralichthys olivaceus).

Miamiensis avidus is a parasitic pathogen that causes scuticociliatosis, a severe and often lethal marine infection that affects marine fishes worldwide, including olive flounder (Paralichthys olivaceus) in Korea. This parasite infects all size groups of flounder year-round, causing recurring mortalities and huge economic losses to the Korean flounder industry each year. However, few efforts have been made to implement effective remedial measures to control this parasite. Therefore, our study sought to develop a chitosan microsphere (MS)-encapsulated inactivated vaccine (IMa + chitosan) for or

Exuviotrophic apostome ciliates from freshwater decapods in southern Alabama (USA) and a description of Hyalophysa clampi n. sp. (Ciliophora, Apostomatida).

Apostome ciliates from crayfish and freshwater shrimp in southern and central Alabama were surveyed in this study. Hyalophysa bradburyae was found on both crayfish and shrimp from 16 sites in eight drainages. A new species, Hyalophysa clampi n. sp., was found infesting crayfish at one site and simultaneously infesting crayfish with H. bradburyae at two sites. Characteristics of the trophont ciliature of H. clampi n. sp. separate it from other species in the genus. Particularly, the contractile vacuole pore is oriented posterior to the ventral kineties xyz, kineties 1 and 2 are undivided, an ap

Chemotaxis of Miamiensis avidus to hyaluronic acid, a component of fish surface mucus.

Miamiensis avidus is a marine parasitic ciliate belonging to the order Scuticociliatida, the members of which are the causal agents of scuticociliatosis. This ciliate has resulted in considerable economic losses to the aquaculture industry in the Republic of Korea and Japan. Nevertheless, the development of vaccines and therapeutics has proven to be challenging, and there are currently no reports of such products commercially available in Korea or Japan. Furthermore, the aetiology and pathogenesis of M. avidus infection in fish remain unknown, as do the underlying reasons for the initial infec

Protozoan Co-Infection Drives Fish Mortality Event in Crete's Karteros River.

In August 2024, a significant fish kill involving hundreds of flathead grey mullet (Mugil cephalus) was reported in the delta region of the Karteros River, Crete, Greece. The investigation identified the primary cause of mortality as severe parasitic infections, specifically from the protozoan parasites Amyloodinium ocellatum and Trichodina sp., both of which heavily affected the gills of the fish. Concurrently, poor water quality in the area, likely due to reduced water volume and limited water renewal, created favourable conditions for parasite proliferation while weakening the fish's natura

MENINGOENCEPHALITIS IN STRANDED SMOOTH DOGFISH (MUSTELUS CANIS) INFECTED BY PHILASTERIDES DICENTRARCHI (PHILASTERIDA: PHILASTERIDAE) IN THE NORTHWESTERN ATLANTIC OCEAN.

During August through September 2018, strandings and mortalities of smooth dogfish (Mustelus canis [Mitchill, 1815 [Carcharhiniformes: Triakidae]) occurred on Brighton Beach and Coney Island Beach (Brooklyn, New York). Each of the 8 smooth dogfish examined grossly exhibited hemorrhagic meninges, turbid and pink to red cerebrospinal fluid, and soft and friable olfactory lobes. Wet mounts of the cerebrospinal fluid and brain showed intense infection by scuticociliates in each smooth dogfish. Histopathologic examination of the infected brain revealed intensity-dependent necrotizing meningoencepha

Isolation and Characterisation of Serotype III in Miamiensis avidus (Ciliophora: Scuticociliatida) From Olive Flounder (Paralichthys olivaceus).

Miamiensis avidus (Ciliophora: Scuticociliatida, syn. Philasterides dicentrarchi) is a significant pathogen causing scuticociliatosis in olive flounder (Paralichthys olivaceus) in Korea and Japan. In this study, the BB19 strain, isolated from olive flounder, was identified as M. avidus through phylogenetic analysis of cytochrome c oxidase subunit 1 (cox1) sequences and classified as cox1 genotype IV. Serotype-specific PCR confirmed BB19 as serotype III. Agglutination assay and indirect ELISA also showed that BB19 exclusively reacted with anti-serotype III (Mie0301 and BB19) sera, with no cross

Systematic reassessment of Ambiphrya and Apiosoma (Ciliophora, Peritrichia), with morpho-molecular characterization of two epibiotic ciliates from rainbow trout in Korea.

Many peritrich ciliates are parasites of, or epibionts on, aquatic animals. Among them, the genera Ambiphrya and Apiosoma are common fish ectocommensals in aquacultures. Here, we report infections by two poorly known forms, Ambiphrya cf. ameiuri and Apiosoma pseudopiscicola, on rainbow trout, a commonly farmed and economically important food fish in South Korea, and provide detailed descriptions and diagnostic illustrations of both species. Their SSU rDNA sequences were obtained, filling the molecular data gap for Apiosoma, a genus for which no sequences had previously been available. Our phyl
