The order Onygenales includes keratinophilic fungi, such as dermatophytes, that can cause onychomycosis. Although dermatophytes are the primary cause of these infections, some non-dermatophytic, keratinophilic onygenalean fungi have been reported as causing nail infections. Other such fungi are frequently isolated as surface contaminants of nails sent for culture, but are not etiologic agents of onychomycosis. Here we introduce a novel onygenalean fungus isolated in the Netherlands from a nail that was suspected to have a fungal infection. As only a single sample was available, etiologic invol

Paracoccidioides brasiliensis is the aetiological agent of paracoccidioidomycosis, the most important systemic mycosis in Latin America. In order to study the diversity of P. brasiliensis mitochondrial genes, to evaluate previous taxonomic proposals, and to explore the hypothesis that the previously described "divergent isolate" B30 (also called Pb01) could represent a new P. brasiliensis species, we undertook a molecular phylogenetic analysis based on five mitochondrial markers. Mitochondrial sequences of 59 P. brasiliensis isolates obtained from clinical and environmental samples, and the or

Paracoccidioides is a fungal pathogen and the cause of paracoccidioidomycosis, a health-threatening human systemic mycosis endemic to Latin America. Infection by Paracoccidioides, a dimorphic fungus in the order Onygenales, is coupled with a thermally regulated transition from a soil-dwelling filamentous form to a yeast-like pathogenic form. To better understand the genetic basis of growth and pathogenicity in Paracoccidioides, we sequenced the genomes of two strains of Paracoccidioides brasiliensis (Pb03 and Pb18) and one strain of Paracoccidioides lutzii (Pb01). These genomes range in size f

Dimorphic fungal pathogens are a significant cause of human disease worldwide. Notably, the dimorphic fungal pathogens within the order Onygenales are considered primary pathogens, causing disease in healthy hosts. Current changes in taxonomy are underway due to advances in molecular phylogenetics, population genetics, and new emerging dimorphic fungal pathogens causing human disease. In this review, we highlight evolutionary relationships of dimorphic fungal pathogens that cause human disease within the order Onygenales and provide rationale to support increased investment in studies understa

Coccidioides immitis and C. posadasii, the causative agents of the mammalian disease coccidioidomycosis, are dimorphic fungal pathogens distributed throughout desert-like environments in North and South America. Coccidioides spp. are members of the Onygenales, a diverse group of pathogenic and nonpathogenic fungi. Recently, full genomes have been published for Coccidioides and a number of other Onygenales species. Phylogenomic comparisons and additional studies in Coccidioides population genomics and gene expression have shed light on the ecology and pathogenesis of Coccidioides and the other

Ophidiomycosis, caused by Ophidiomyces ophidiicola, is a disease associated with variably high morbidity and mortality in both snakes under human care and free-living snakes. Within Illinois, USA, ophidiomycosis surveillance has primarily focused on snake populations in southern Illinois, adjacent to agricultural land. The purpose of this study was to determine the occurrence of O. ophidiicola DNA across multiple snake species and locations in an urban county of northeastern Illinois, containing historically diverse and abundant snake populations. During the summer of 2023, 51 snakes, represen

Knowledge of infectious diseases in wildlife provides important information for preventing potential outbreaks of zoonotic diseases. Adiaspiromycosis is a neglected human disease caused by dimorphic Onygenales fungi. The disease is produced by the inflammatory response against growing adiaspores, leading to granulomatous pneumonia. In humans, adiaspiromycosis is relevant in immunosuppressed patients. In animals, it is associated with pneumonia in fossorial species. Given the potential role of armadillos in the epidemiology of adiaspiromycosis, in this study, we sought to investigate the occurr

Ophidiomycosis (snake fungal disease) is caused by the fungus Ophidiomyces ophiodiicola. As ophidiomycosis is difficult to study in free-ranging snakes, a reliable experimental model is needed to investigate transmission, pathogenesis, morbidity, and mortality, and the effects of brumation and temperature on disease development. Our objective was to develop such a model via subcutaneous injection of O. ophiodiicola conidia in red cornsnakes (Pantherophis guttatus). The model was used to evaluate transmission and the effects of brumation and temperature in co-housed inoculated and noninoculated

Fungal pathogens pose a growing threat to vertebrate biodiversity. In snakes, Ophidiomyces ophidiicola (Oo) has garnered particular concern, although its impact in Europe remains poorly understood. We conducted a season-long, standardized survey of dice snakes (Natrix tessellata) along the northern shore of Lake Como (Italy) to quantify Oo and ophidiomycosis prevalence, identify the circulating strain, and explore the association with environmental, morphological and behavioral traits. Between March and October 2024, we collected 96 N. tessellata samples (23 sheds and swabs from 73 live indivi

Toenail onychomycoses are among the most common human superficial mycoses. The species spectrum of the causative agents, mainly nondermatophytes, is broad and varied. During 14 months (March 2007 to April 2008), Onychocola canadensis Sigler 1990, an uncommon nondermatophyte, was identified microscopically and by culture as the causative agent of nail infection in four patients. It is the first report of this species in the Slovak Republic. The experience gained with the identification of this uncommon, slow-growing arthroconidial fungus is presented.

The changing epidemiology and diagnostic challenges of coccidioidomycosis, histoplasmosis, and paracoccidioidomycosis in Latin America are reviewed. The agents responsible for these three systemic endemic mycoses are dimorphic fungi from the Onygenales order, all of which are classified as World Health Organization fungal priority pathogens. However, the mycoses they cause span a notable diversity of contrasting etiologies, target populations, geographic distributions, and factors (including climate changes) that determine their incidence. New, updated maps are presented that reflect the curre

Ophidiomycosis, or snake fungal disease, caused by the pathogen Ophidiomyces ophidiicola, poses a growing threat to wild snake populations in North America. We surveyed snake populations in central Virginia, USA in 2021-22; no clinical signs of ophidiomycosis had been observed in a survey in 2013-14. Across 82 individuals representing seven species, 16% tested O. ophidiicola-positive, including 17% of eastern wormsnakes (Carphophis amoenus), the most frequently encountered species. All O. ophidiicola-positive snakes exhibited skin lesions or abnormalities at the time of capture; no subclinical

Nondermatophyte moulds (NDMs) onychomycosis is often difficult to diagnose as NDMs have been considered contaminants of nails. There are several diagnostic methods used to identify NDMs, however, repeated laboratory isolation is recommended to validate pathogenicity. With NDM and mixed infection (dermatophytes plus NDM) onychomycosis on the rise, accurate clinical diagnosis along with mycological tests is recommended. Systemic antifungal agents such as itraconazole and terbinafine (e.g. pulse regimen: 1 pulse = every day for one week, followed by no treatment for three weeks) have shown effica

A newly described onygenalean fungus, Emydomyces testavorans, has been isolated from ulcerative shell and skin lesions of freshwater aquatic chelonians. To investigate the shell lesions associated with infection and determine if any lesional features were unique to E. testavorans, tissues from turtles housed in zoological institutions (n = 45) in the United States and free-living turtles (n = 5) submitted for diagnostic biopsy or necropsy were examined. Free-living turtles were from geographically distinct habitats in Florida (n = 1) and Washington (n = 4) at the time of sampling. Histologic s

Nannizziopsis spp., fungi responsible for emerging diseases, are rarely involved in human bone and joint infections. We present a rare case of septic arthritis with necrotizing cellulitis caused by N. obscura in a patient in France who had undergone kidney transplant. Rapid, aggressive medical and surgical management led to a favorable outcome.

A retrospective review of systemic or localized mycotic infections in captive snakes confirmed via biopsy or necropsy from 1983 to 2017 was performed at the Smithsonian's National Zoological Park. Quantitative polymerase chain reaction (qPCR) confirmed infection with Ophidiomyces ophiodiicola (Oo) in 36.8% (n = 14) of the 38 mycotic infections. Infections with Oo were evenly distributed over the 35-y period and lacked a sex predilection. There was a period prevalence of 4.5% of completed snake necropsy or biopsy cases that were Oo positive. Species affected included green anaconda (Eunectes mu

Ascosphaera (Eurotiomycetes: Onygenales) is a diverse genus of fungi that is exclusively found in association with bee nests and comprises both saprophytic and entomopathogenic species. To date, most genomic analyses have been focused on the honeybee pathogen A. apis, and we lack a genomic understanding of how pathogenesis evolved more broadly in the genus. To address this gap we sequenced the genomes of the leaf-cutting bee pathogen A. aggregata as well as three commensal species: A. pollenicola, A. atra and A. acerosa. De novo annotation and comparison of the assembled genomes was carried ou

We report the diagnosis and molecular characterization of lobomycosis-like lesions in a captive bottlenose dolphin. The clinical picture and the absence of growth in conventional media resembled the features associated with Lacazia loboi. However sequencing of ribosomal DNA and further phylogenetic analyses showed a novel sequence more related to Paracoccidioides brasilensis than to L. loboi. Moreover, the morphology of the yeast cells differed from those L. loboi causing infections humans. These facts suggest that the dolphin lobomycosis-like lesions might have been be caused by different a d
