Polyamine oxidase (PAO) catalyzes the oxidative catabolism of spermidine and spermine, generating hydrogen peroxide. In wild-type tobacco (Nicotiana tabacum 'Xanthi') plants, infection by the compatible pathogen Pseudomonas syringae pv tabaci resulted in increased PAO gene and corresponding PAO enzyme activities; polyamine homeostasis was maintained by induction of the arginine decarboxylase pathway and spermine was excreted into the apoplast, where it was oxidized by the enhanced apoplastic PAO, resulting in higher hydrogen peroxide accumulation. Moreover, plants overexpressing PAO showed pre

Since the protective activity of the Bordeaux mixture against plant disease caused by oomycetes was discovered, copper compounds have been used for more than a century as an effective plant protection strategy. However, the application of excessive copper can cause adverse effects through long-term heavy metal accumulation in soils. Therefore, it is necessary to develop new strategies to reduce or replace copper in pesticides based on organic and low-input farming systems. Organic acids are eco-friendly. In this study, we tested the antifungal and anti-oomycete activity of maleic acid (MA) and

Pathogenic oomycetes can infect hundreds of crops, leading to significant economic losses globally. This study synthesized polyethylenimine-coated MXene quantum dots (PEI-MQDs) and investigated their antioomycete potential using the vegetable oomycete pathogen Phytophthora capsici as a model system. PEI-MQDs demonstrated potent antioomycete activity against P. capsici by damaging cellular structures and subverting asexual reproduction (zoosporangium/zoospore formation). Transcriptomic profiling demonstrated that treatment with PEI-MQDs induced extensive transcriptional changes in P. capsici ge

In search of novel natural product-based fungicides, 24 cuminic acid derivatives containing the thiourea group were synthesized and characterized by 1H, 13C NMR, and HRMS spectra. The antifungal/oomycete activity bioassay showed that some of the title compounds displayed moderate to good antifungal activity against Botryosphaeria dothidea, Sclerotinia sclerotiorum, and Valsa mali. Compound 4k exhibited the best in vitro antifungal activity toward V. mali. The in vivo experiment identified that 4k possessed considerable anti-V. mali protective and curative effect at 200 mg/L. The invasive oomycete pathogen Aphanomyces astaci significantly threatens native European crayfish populations, prompting investigations towards the effects of protective immunostimulation on the immune response of the vulnerable noble crayfish (Astacus astacus). Here, we evaluate the effect of three oomycete-derived immunostimulant treatments: laminarin (β-1,3-glucan found within the Ap. astaci cell wall), inactivated Ap. astaci spores and Ap. astaci hyphal homogenate. Our findings reveal immediate changes in the noble crayfish total haemocyte count (THC), differential haemocyte count (DHC),

Crop diseases caused by fungal and oomycete pathogens threaten sustainable agricultural production, and the efficient identification of pathogen effectors is essential for understanding virulence mechanisms and developing disease-resistant crops. However, the rapid evolutionary diversification of effectors often obscures conserved motifs, limiting the effectiveness of conventional computational approaches and making experimental prioritization challenging. To address this, we developed DeepEffector, a high-precision predictor for fungal and oomycete effector identification. DeepEffector levera

Aphanomyces astaci is an invasive pathogenic oomycete responsible for the crayfish plague, a disease that has devastated European freshwater crayfish. So far, five genotype groups of this pathogen have been identified by applying random amplified polymorphic DNA analysis on axenic cultures. To allow genotyping of A. astaci in host tissue samples, we have developed co-dominant microsatellite markers for this pathogen, tested them on pure cultures of all genotype groups, and subsequently evaluated their use on tissues of (1) natural A. astaci carriers, i.e., North American crayfish species, and

INTRODUCTION: Late blight caused by Phytophthora infestans severely threatens potato production, necessitating novel natural fungicide.
METHODS: Ethanol extracts from 11 wild plants were screened for anti-oomycete activity. Active extracts from Artemisia lavandulaefolia and Clinopodium repens were fractionated. The petroleum ether fraction and aqueous fraction were evaluated for effects on mycelial growth, morphology, spore germination, pathogenicity, stress response, and synergy with fungicide Infinito. Components were analyzed by LC-MS.
RESULTS: A. lavandulaefolia and C. repens extracts sh

Botanicals are various plant-based products like plant extracts or essential oils. Anti-fungal activities of selected essential oils were tested on the pathogen causing potato and tomato late blight (Phytophthora infestans). Tests to evaluate anti-oomycete activities of commercial essential oils and their major compounds were carried out in vitro in microplate in liquid media. Anti-oomycete activities on Phytophthora infestans strain were obtained from essential oils/major compounds: Eucalyptus citriodora/citronellal; Syzygium aromaticum (clove)/eugenol; Mentha spicata/D-Carvone, L-Carvone; Or

The oomycete genus Phytophthora includes many important plant pathogens for which extensive genome data exist, but lacking is an inducible expression system to study contributions of their genes to growth and pathogenicity. Here the adaptation of the reverse tetracycline transactivator (rtTA) system to P. infestans is described. Vectors were developed containing rtTA expressed from an oomycete promoter, and beta-glucuronidase (GUS) controlled by TetR binding sites fused to a minimal oomycete promoter. Transformants were obtained in which GUS was expressed in a dose-dependent manner by the rtTA

The causative agent of crayfish plague, Aphanomyces astaci (Saprolegniales, Oomycota), is one of the 100 world's worst invasive alien species and represents a major threat to freshwater crayfish species worldwide. A better understanding of the biology and epidemiology of A. astaci relies on the application of efficient tools to detect the pathogen and assess its genetic diversity. In this study, we validated the specificity of two recently developed PCR-based approaches used to detect A. astaci groups. The first relies on the analysis of mitochondrial ribosomal rnnS (small) and rnnL (large) su

Zoospores from the crayfish plague fungus Aphanomyces astaci could germinate and grow in vitro on scales of Salmo salar. The fungicidal activity of different compounds was tested against the fungus growing on fish scales as well as in lake water. Malachite green was the most effective fungicide, but scale mucus protected the fungus from this compound to some extent. For complete inhibition of spore germination in scale mucus 2 mg1-1 was required compared to 1 1 mg1-1 in lake water.

BACKGROUND: 2E,4E-decadienoic acid (DDA) is an aliphatic compound with potent anti‑oomycete activity against Phytophthora nicotianae (Pn), a plant pathogen responsible for major agricultural losses. Mechanistic studies revealed that DDA targets mitochondria and requires intracellular delivery to exert its activity. To improve its stability and bioavailability, we developed a reactive oxygen species (ROS)-responsive, TAT‑peptide (P) ‑modified liposomal (Lipo) delivery system (DDA@P‑ROS‑Lipo) containing thioketal (TK) linkages that cleave under elevated ROS conditions during pathogen infection,

Soil oomycetes contain some of the most devastating plant pathogens, and are widespread within the soil microbiome, yet how individual plant species structure oomycete communities in natural soils, and whether any such structuring is consistent across space, remains poorly resolved. Because host-specific pathogen accumulation is a central prediction of plant-soil feedback theory but is rarely tested outside greenhouse conditions, we focused on whether the host-specific fraction of these communities is preferentially pathogenic. Using oomycete-specific ITS amplicons, we sequenced rhizosphere-as

Plants are able to detect insect eggs deposited on leaves. In Arabidopsis, eggs of the butterfly species Pieris brassicae (common name large white) induce plant defenses and activate the salicylic acid (SA) pathway. We previously discovered that oviposition triggers a systemic acquired resistance (SAR) against the bacterial hemibiotroph pathogen Pseudomonas syringae. Here, we show that insect eggs or treatment with egg extract (EE) induce SAR against the fungal necrotroph Botrytis cinerea BMM and the oomycete pathogen Hyaloperonospora arabidopsidis Noco2. This response is abolished in ics1, al

BACKGROUND: The fungus-like organism Pythium insidiosum is the causative agent of a life-threatening tropical infectious disease, pythiosis, which has high rates of morbidity and mortality. A lack of reliable diagnostic tools and effective treatments for pythiosis presents a major challenge to healthcare professionals. Unfortunately, surgical removal of infected organs remains the default treatment for pythiosis. P. insidiosum is an understudied organism. In-depth study of the pathogen at the molecular level could lead to better means of infection control High quality genomic DNA (gDNA) is nee

Oomycete pathogens in freshwaters, such as Saprolegnia parasitica and Aphanomyces astaci, are responsible for fish/crayfish population declines in the wild and disease outbreaks in aquaculture. Although the formation of infectious zoospores in the laboratory can be triggered by washing their mycelium with natural water samples, the physico-chemical properties of the water that might promote sporulation are still unexplored. We washed the mycelia of A. astaci and S. parasitica with a range of natural water samples and observed differences in sporulation efficiency. The results of Partial Least

Numerous oomycetes colonise the crayfish cuticle, the best known being the crayfish plague pathogen Aphanomyces astaci. Although other oomycetes associated with crayfish complicate the isolation and molecular detection of A. astaci, their diversity is little known. To improve this knowledge, we analysed 95 oomycete isolates obtained during attempts to isolate A. astaci from crayfish presumably infected by this pathogen. We characterized the isolates morphologically and by sequencing of the nuclear internal transcribed spacer (ITS) region. We identified 13 taxa by molecular analysis. Ten of the

A cascade formed by phosphorylation events of mitogen-activated protein kinases (MAPKs) takes part in plant stress responses. However, the roles of these MAPKs in resistance of potato (Solanum tuberosum) against Phytophthora pathogens is not well studied. Our previous work showed that a Phytophthora infestans RXLR effector targets and stabilizes the negative regulator of MAPK kinase 1 of potato (StMKK1). Because in Arabidopsis thaliana the AtMPK4 is the downstream phosphorylation target of AtMKK1, we performed a phylogenetic analysis and found that potato StMPK4/6/7 are closely related and are
