The divorce of Sporothrix and Ophiostoma: solution to a problematic relationship.

One of the causal agents of human sporotrichosis, Sporothrix schenckii, is the type species of the genus Sporothrix. During the course of the last century the asexual morphs of many Ophiostoma spp. have also been treated in Sporothrix. More recently several DNA-based studies have suggested that species of Sporothrix and Ophiostoma converge in what has become known as Ophiostoma s. lat. Were the one fungus one name principles adopted in the Melbourne Code to be applied to Ophiostoma s. lat., Sporothrix would have priority over Ophiostoma, resulting in more than 100 new combinations. The consequ

Ophiostoma breviusculum sp. nov. (Ophiostomatales, Ascomycota) is a new species in the Ophiostoma piceae complex associated with bark beetles infesting larch in Japan.

A new species of the Ophiostoma piceae-complex was isolated from bark beetles and the insect galleries of Larix kaempferi in Japan. This species was described as Ophiostoma breviusculum. The fungus was morphologically similar to O. piceae and O. quercus. However the average length of the perithecial necks and synnemata were shorter than for O. piceae and O. quercus. Synnemata morphological characteristics also differentiated O. breviusculum from the other species of the Ophiostoma piceae-complex isolated from conifers. Mating tests demonstrated that this fungus did not produce perithecia with

Multi-gene phylogeny for Ophiostoma spp. reveals two new species from Protea infructescences.

Ophiostoma represents a genus of fungi that are mostly arthropod-dispersed and have a wide global distribution. The best known of these fungi are carried by scolytine bark beetles that infest trees, but an interesting guild of Ophiostoma spp. occurs in the infructescences of Protea spp. native to South Africa. Phylogenetic relationships between Ophiostoma spp. from Protea infructescences were studied using DNA sequence data from the beta-tubulin, 5.8S ITS (including the flanking internal transcribed spacers 1 and 2) and the large subunit DNA regions. Two new species, O. phasma sp. nov. and O.

Four new Ophiostoma species associated with hardwood-infesting bark beetles in Norway and Poland.

Ophiostoma spp. (Ophiostomatales, Ascomycota) are well-known fungi associated with bark and ambrosia beetles (Curculionidae: Scolytinae, Platypodinae). Fungi in the Ophiostomatales include serious tree pathogens as well as agents of timber blue-stain. Although these fungi have been extensively studied in the northern hemisphere, very little is known regarding their occurrence on hardwoods in Europe. The aims of the present study were to identify and characterize new Ophiostoma spp. associated with bark and ambrosia beetles infesting hardwoods in Norway and Poland, and to resolve phylogenetic r

The Ophiostoma clavatum species complex: a newly defined group in the Ophiostomatales including three novel taxa.

Two species of blue-stain fungi with similar morphologies, Ophiostoma brunneo-ciliatum and Ophiostoma clavatum, are associates of bark beetles infesting Pinus spp. in Europe. This has raised questions whether they represent distinct taxa. Absence of herbarium specimens and contaminated or mistakenly identified cultures of O. brunneo-ciliatum and O. clavatum have accentuated the uncertainty regarding their correct identification. The aim of this study was to reconsider the identity of European isolates reported as O. brunneo-ciliatum and O. clavatum by applying DNA-based identification methods,

Taxonomy and phylogeny of new wood- and soil-inhabiting Sporothrix species in the Ophiostoma stenoceras-Sporothrix schenckii complex.

Sporothrix, one of the anamorph genera of Ophiostoma, includes the important human pathogen S. schenckii and various fungi associated with insects and sap stain of wood. A survey of fungi from wood utility poles in South Africa yielded two distinct groups of Sporothrix isolates from different geographical areas. DNA sequence and morphological data derived in this study showed that isolates in these groups represent two novel species in the S. schenckii-O. stenoceras species complex. A new species isolated from pine poles and rosebush wood and phylogenetically closely related to S. pallida is d

Multigene phylogenies and morphological characterization of five new Ophiostoma spp. associated with spruce-infesting bark beetles in China.

Ophiostoma spp. (Ophiostomatales, Ascomycota) are well-known fungi associated with bark beetles (Coleoptera: Scolytinae). Some of these are serious tree pathogens, while the majority is blue-stain agents of timber. In recent years, various bark beetle species have been attacking spruce forests in Qinghai province, China, causing significant damage. A preliminary survey was done to explore the diversity of the ophiostomatoid fungal associates of these beetles. The aims of the present study were to identify and characterize new Ophiostoma spp. associated with spruce-infesting bark beetles in Qin

Unraveling the regioselectivity of Ophiostoma piceae sterol esterase as a case study for lipases with wide acyl-binding tunnel entrances.

Lipase regioselectivity (sn-1(3) vs sn-2) is crucial for synthesizing structured lipids, but the structural basis for contrasting regioselectivities in lipases with acyl-binding tunnel remains incomplete. While the correlation between a narrow tunnel entrance and sn-2 regioselectivity has been previously established, the regioselectivity of Ophiostoma piceae sterol esterase (OPE), a lipolytic enzyme with a uniquely wide tunnel entrance, remained unclear. In this study, the chiral-phase resolution of oleic species confirmed that OPE exhibits a notable sn-1(3) regioselectivity (85.8%) against tr

Quorum sensing activity in Ophiostoma ulmi: effects of fusel oils and branched chain amino acids on yeast-mycelial dimorphism.

AIMS: For Ophiostoma (Ceratocystis) ulmi, the ability to undergo morphological change is a crucial factor for its virulence. To gain an understanding of quorum-sensing activity in O. ulmi as it relates to yeast-mycelium dimorphism control, this study examines the effects of branched-chain amino acids as well as their fusel alcohols and fusel acids as quorum sensing molecules. METHODS AND RESULTS: In a defined medium containing glucose, proline and salts, O. ulmi grew as yeasts when the culture was inoculated with a high density of spores (2 × 10(7)  CFU ml(-1) ) and as mycelia when inoculated

Four new Ophiostoma species associated with conifer- and hardwood-infesting bark and ambrosia beetles from the Czech Republic and Poland.

Fungi under the order Ophiostomatales (Ascomycota) are known to associate with various species of bark beetles (Coleoptera: Curculionidae: Scolytinae). In addition this group of fungi contains many taxa that can impart blue-stain on sapwood and some are important tree pathogens. A recent survey that focussed on the diversity of the Ophiostomatales in the forest ecosystems of the Czech Republic and Poland uncovered four putative new species. Phylogenetic analyses of four gene regions (ITS1-5.8S-ITS2 region, ß-tubulin, calmodulin, and translation elongation factor 1-α) indicated that these four

Chemical changes in Ulmus minor xylem tissue after salicylic acid or carvacrol treatments are associated with enhanced resistance to Ophiostoma novo-ulmi.

Application of endogenous plant hormone salicylic acid (SA) or essential oil component carvacrol (CA) in elms enhances tree resistance to the Dutch elm disease pathogen, although the effect of these compounds on tree metabolism is unknown. The chemical changes induced by SA or CA treatments in Ulmus minor were studied through gas chromatography-mass spectrometry (GC-MS) analysis of xylem tissues. Treatments consisted of fortnightly irrigating seedlings with water, SA or CA at 600 mg L⁻¹. The chemical composition of the xylem tissues sampled from treated trees was significantly altered dependin

Evolutionary relationships among putative RNA-dependent RNA polymerases encoded by a mitochondrial virus-like RNA in the Dutch elm disease fungus, Ophiostoma novo-ulmi, by other viruses and virus-like RNAs and by the Arabidopsis mitochondrial genome.

The nucleotide sequence (2617 nucleotides) of virus-like double-stranded (ds) RNA 3a in a diseased isolate, Log1/3-8d2 (Ld), of the ascomycete fungus Ophiostoma novo-ulmi has been determined. One strand of the dsRNA contains an open reading frame (ORF) with the potential to encode a protein of 718 amino acids, and the complementary strand contains two smaller ORFs with the potential to encode proteins of 178 and 182 amino acids, respectively. The large ORF contains 12 UGA codons which code for tryptophan in ascomycete mitochondria and has a codon bias typical of mitochondrial genes, consistent

Ex vivo determination of potentially virulent Sporothrix schenckii,

Hyphae from 30 isolants of Sporothrix and Ophiostoma species were washed, dried and pyrolyzed at 350 degrees C. Pyrolysis products were separated on a Carbowax column heated 7.5 degrees C/min to and maintained for 50 min at 160 degrees C. Hydrogen flame detector responses were recorded graphically. Fifteen clinical isolants of S. schenckii from geographically separated sources produced qualitatively identical pyrograms. S. foliorum, 8 avirulent S. schenckii and other Sporothrix species isolants from soils, and Sporothrix states of 6 Ophiostoma species yielded pyrograms readily distinguished fr

Sporothrix schenckii (sensu strict S. globosa) mating type 1-2 (MAT1-2) gene.

Sporotix schenckii is a pathogenic fungus that causes human and animal sporotrichosis, and based on morphology of the sessile conidia and molecular analysis, it was recently recognized as a species complex comprising at least the following six sibling species: S. albicans, S. brasiliensis, S. globosa, S. luriei, S. mexicana and S. schenckii. However, apart from S. schenckii sensu strict, only S. brasiliensis, S. globosa and S. luriei are associated with human and animal infection. S. globosa has been most commonly isolated in Asia, Europe and the USA; therefore, molecular epidemiological study

Pine monoterpenes drive growth, competition, and volatile metabolism of symbiotic fungi associated with the mountain pine beetle.

Monoterpenes are key conifer defences that shape interactions between the mountain pine beetle and its symbiotic phytopathogenic fungi. Conifer monoterpenes vary both within and between tree species, yet how this variation affects symbiotic fungi remains poorly understood. We tested how monoterpene blends from three geographically distinct subspecies of Pinus contorta, including subsp. contorta, latifolia, and murrayana can affect the growth and volatile emissions of two fungal symbionts, Grosmannia clavigera and Ophiostoma montium, of the mountain pine beetle. We conducted a factorial experim

Nitrogen concentration in mountain pine beetle larvae reflects nitrogen status of the tree host and two fungal associates.

Individual lodgepole pines (Pinus contorta) were fertilized with urea at nitrogen (N) inputs equivalent to 0, 315, or 630 kg/ha. Four months after application of the fertilizer, inner bark tissue N concentrations were significantly higher in the trees that had received the low dose (315 kg/ha) fertilization treatment than in the control trees; trees that had received the high-dose treatment (630 kg/ha) were intermediate and not significantly different from either of the other treatments. There was a significant positive correlation between N concentration in inner bark tissue and larval mounta

Incidental Fungi in Host Trees Disrupt the Development of Sirex noctilio (Hymenoptera: Siricidae) Symbiotic Fungus and Larvae.

The woodwasp, Sirex noctilio Fabricius, is an exotic pest of Pinus L. in the southern hemisphere and North America, and it is an emerging threat in northeastern China. Adult woodwasps have numerous insect competitors for oviposition substrates, and developing larvae must compete for resources with other subcortical invaders. Its mutualistic fungus Amylostereum areolatum (Fr.) Boidon, is less competitive than many other fungal colonists present in pine ecosystems. This study investigated the effects of incidental, host-colonizing fungi on the growth and development of woodwasp larvae and A. are

Microbiome analysis and epigenetic patterns revealed distinct differences between two elm species with contrasting Dutch elm disease resistance.

BACKGROUND: Dutch elm disease (DED), caused by the fungus Ophiostoma novo-ulmi, has devastated elm species in Europe and North America for over a century. While the Chinese elm (Ulmus parvifolia) frequently displays notable resistance to DED, the European wych elm (Ulmus glabra) remains highly susceptible. Understanding microbiome interaction and epigenetic factors might help to explain this contrasting resilience and subsequently improve disease management. RESULTS: Seedlings of both elm species were germinated and maintained under identical environmental conditions. Comparative metabarcodin

Filamentous fungal associates of the alder bark beetle, Alniphagus aspericollis, including an undescribed species of Neonectria.

Bark beetles (Coleoptera: Curculionidae; Scolytinae) are tree-infesting insects that consume subcortical tissues and fungi. Species capable of killing their host trees are most commonly associated with conifers, as very few bark beetle species infest and kill hardwood hosts directly. The alder bark beetle, Alniphagus aspericollis, is a hardwood-killing bark beetle that colonizes and kills red alder, Alnus rubra. Conifer-killing bark beetles have well-known associations with symbiotic ophiostomatoid fungi that facilitate their life histories, but it is unknown whether A. aspericollis has any fu
