We show that the optical force field in optical tweezers with elliptically polarized beams has the opposite handedness for a wide range of particle sizes and for the most common configurations. Our method is based on the direct observation of the particle equilibrium position under the effect of a transverse Stokes drag force, and its rotation around the optical axis by the mechanical effect of the optical torque. We find overall agreement with theory, with no fitting, provided that astigmatism, which is characterized separately, is included in the theoretical description. Our work opens the w

We present a remote focusing optical tweezer utilizing a 4f symmetrical optical system to compensate the high-order aberration during annular light refocusing. The position of the optical trap can be adjusted beyond the range of one hundred micrometers in the axial direction by means of tuning the position of the mirror placed in the focal region of the illumination objective lens. This optical tweezer can be combined with a sectioning microscope to realize three-dimensional (3D) imaging, e.g., a confocal microscope using a single water immersion objective lens. All optical elements are placed

This study introduces an indirect optical geometry measurement approach for transparent microchannels using optical tweezers to trap and manipulate a dielectric particle as a measuring probe. Traditional direct optical methods, such as white light interferometry and microscopy, face challenges in assembled microfluidic systems due to refractive index matching between the channel material and fluid, which reduces surface visibility. The proposed method overcomes these limitations by scanning along the axial direction with an optically trapped silica microsphere of 2 μm diameter as a probe and o

Laser cooling and trapping of atomic matter waves in optical potentials has enabled rapid progress in quantum science, particularly when combined with Rydberg excitation of the atoms to induce long-range interactions. Here, we propose the local manipulation and spatiotemporal sculpting of the electronic matter wave of a Rydberg atom by a laser field focused so that its beam width is smaller than the Rydberg electron orbit. We compute the electronic eigenstates in the presence of a sharply focused Gaussian laser beam, and find strong Rydberg state mixing leading to large kilo-Debye dipole momen

Optical tweezers (OT), a cornerstone of micromanipulation, are fundamentally constrained by substrate-induced adhesion and friction, limiting their application to mesoscale objects and fragile biological specimens where overcoming these resistive forces requires physiologically damaging laser powers. Here, we overcome this long-standing challenge by introducing dielectric levitation optical tweezers (DL-OT), a multiphysics platform that seamlessly integrates alternating-current dielectric levitation with optical traps. By using negative dielectrophoresis (n-DEP) to actively neutralize the norm

Optical tweezers, which leverage the forces exerted by radiation pressure, have emerged as a pivotal technique for precisely manipulating and analyzing microscopic particles. Since Arthur Ashkin's ground-breaking work in the 1970s and the subsequent development of the single-beam optical trap in 1986, the capabilities of optical tweezers have expanded significantly, enabling the intricate manipulation of biological specimens at the micro- and nanoscale. This review elucidates the foundational principles of optical trapping and their extensive applications in the biomedical sciences. The applic

Optical tweezers were used to scan individual Chronic Myelogenous Leukemia cells to determine if the cell death depends on the scanning conditions. Although increasing the scanning frequency or amplitude means greater force applied to the cells, their effects on cell death are not a simple increasing trend, as observed in the optical microscopy. Indeed, cell death sharply increased at particular screening frequencies and amplitudes, whereas other frequencies or amplitudes were less detrimental. These results suggest that cell damage was more sensitive to certain scanning conditions, rather tha

The ability to assess molecular binding kinetics in real time is critical for advancing our understanding of molecular interactions in biochemical and biotechnological systems. This work presents a novel optical tweezer (OT)-based method to monitor molecular affinity in real time, focusing on the high-affinity streptavidin-biotin system as a model. Transparent poly(methyl methacrylate) (PMMA) microparticles functionalized with streptavidin were trapped before, during, and after binding with biotinylated bovine serum albumin (biotin-BSA), enabling the analysis of forward-scattered signals to de

Currently, research on optical tweezers technology predominantly focuses on single-trap optical tweezers, which have a limited controllable range. Multi-trap optical tweezers effectively address these limitations. This paper proposes a method for developing a dual-trap optical tweezers system utilizing basic optical elements. Two optical traps are created by reflecting a laser beam off the front and rear surfaces of a beam splitter. The transition between single-trap and dual-trap configurations is facilitated by a lens group, which allows for the adjustment of the distance between the two tra

Bacterial flagellar swarming enables dense microbial populations to migrate collectively across surfaces, often resulting in emergent, coordinated behaviors. However, probing the underlying energetics of swarming at the single-cluster level remains a challenge. Here, we combine optical tweezers and multiparticle tracking within a stochastic thermodynamic framework to characterize the active motility of confined Proteus mirabilis clusters. Using the photon momentum method to directly measure trapping forces, we show that swarming clusters generate persistent, dissipative flows indicative of non

Given that red blood cells (RBCs) are the most abundant cells in blood, their morphology and mechanics strongly affect blood rheology. Furthermore, changes in the physiological functions and health status of an organism can also affect RBC mechanics. Therefore, understanding the mechanical properties of RBCs holds substantial research value in the biomedical field. The technology of optical tweezers (OT) has become a crucial method for measuring and analyzing the mechanical properties of RBCs, owing to their unique advantages such as non-contact manipulation and piconewton-level force sensitiv

Determining the optical potential is one of the crucial aspects of the analysis and control of optical tweezers. It requires finding the scale factor of displacements in an optical tweezer recording, which typically is hard to obtain directly through a photodetector, and instead is usually done by fitting the statistical features of the data to the Langevin dynamics predictions. We argue that this procedure is harder for measurements made using a Segmented Quadrant Detector, for which these scale factors can vary along different axes due to image distortions. Subsequently, it can lead to a fal

Precise quantification of microscopic forces in optical trapping is fundamental to a wide range of single-molecule biophysics. Conventional calibration techniques, however, require extensive datasets (typically exceeding 104 samples) to achieve statistical convergence in the presence of thermal noise and nonequilibrium artifacts. In this work, we employ the stochastic force inference (SFI) method, which combines information theory with statistical physics, to reconstruct deterministic force fields from noisy stochastic trajectories in optical traps. We demonstrate that the SFI-based calibratio

Optical tweezers are a powerful tool for the precise positioning of a variety of small objects, including single neutral atoms. Once trapped, atoms can be cooled to the motional ground state of the tweezers. For a more advanced control of their spatial wave function, we report here a simple method to squeeze their motion, and the protocol to measure the squeezing factor based on momentum spreading estimation. We explore the limitations set by the technical imperfections of the tweezers, as well as the more fundamental limit set by their anharmonicity, and finally demonstrate a squeezing of 5.4

Optical tweezers are widely used in materials assembly1, characterization2, biomechanical force sensing3,4 and the in vivo manipulation of cells5 and organs6. The trapping force has primarily been generated through the refractive index mismatch between a trapped object and its surrounding medium. This poses a fundamental challenge for the optical trapping of low-refractive-index nanoscale objects, including nanoparticles and intracellular organelles. Here, we report a technology that employs a resonance effect to enhance the permittivity and polarizability of nanocrystals, leading to enhanced

Optical trapping and patterning cells or microscopic particles is fascinating. We developed a light sheet-based optical tweezer to trap dielectric particles and live HeLa cells. The technique requires the generation of a tightly focussed diffraction-limited light-sheet realized by a combination of cylindrical lens and high NA objective lens. The resultant field is a focussed line (along x-axis) perpendicular to the beam propagation direction (z-axis). This is unlike traditional optical tweezers that are fundamentally point-traps and can trap one particle at a time. Several spherical beads unde

Lamins are nucleoskeletal proteins of mammalian cells that stabilize the structure and maintain the rigidity of the nucleus. These type V intermediate filament proteins which are predominantly of A and B types provide necessary tensile strength to the nucleus. Single amino acid missense mutations occurring all over the lamin A protein form a cluster of human diseases termed as laminopathies, most of which principally affect the muscle and cardiac tissues responsible for load bearing functionalities of the body. One such mutation is A350P which causes dilated cardiomyopathy in patients. It is p

Beads trapped in optical tweezers are aligned along the optical propagation direction, which makes it difficult to determine the number of beads with bright-field microscopy. This problem also dramatically influences the measurement of the optical trapping based single-molecule force spectroscopy. Here, we propose a video processing approach to count the number of trapped micro-objects in real time. The approach uses a normalized cross-correlation algorithm and image enhancement techniques to amplify a slight change of the image induced by the entry of an exotic object. As tested, this method

We report the experimental implementation of a new method for generating multiple dynamical optical tweezers, where each one of them is generated with an independent linear polarization state with arbitrary orientation. This also allows an independent simultaneous polarization-rotation control. The laser beam, both for generating multiple traps and polarization control, has been modulated using a single reflective nematic liquid crystal with parallel alignment. We present experimental results of controlled displacement, orientation and rotation of birefringent particles. In addition, a simple

Red blood cells (RBCs) rely on their mechanical and electrostatic properties to maintain microvascular circulation and oxygen delivery. Oxidative stress, a hallmark of many diseases, disrupts these properties by damaging membrane lipids and cytoskeletal proteins. In this study, RBCs from healthy donors were incubated with three oxidative agents, i.e., phenylhydrazine (PHZ), tert-butyl hydroperoxide (t-BHP), and diamide, to investigate how oxidative stress affects the combination of single-cell mechanical, morphological, biochemical, and electrical properties under different oxidizing condition
