Validation of catheter semiquantitative culture technique for nonstaphylococcal organisms.

The catheter semiquantitative culture roll tip technique has been validated as a discriminator between non-catheter-related bacteremias and catheter-related bacteremias (CRBs) caused by Staphylococcus species. However, this technique has not been specifically validated when used for the evaluation of catheters infected with organisms other than staphylococci. We reviewed catheters that had been submitted for semiquantitative roll tip culture as well as hospital records to determine clinical correlates of infection. Local infection and CRB were defined by standard criteria. Catheter-related sep

Development of an organ culture technique capable of monitoring most pineal gland indole metabolites.

An intact pineal gland organ culture technique was developed which utilised radiolabelled tryptophan as the indolic precursor and two-dimensional thin-layer chromatography to separate the various indole metabolites produced. The method was capable of reproducibly separating and quantitating all tryptophan metabolites except 5-methoxytryptophan which co-chromatographed with tryptophan in all the solvent systems evaluated. Noradrenergic stimulation of cultured pineals led to a predictable increase in N-acetyl-serotonin and melatonin production, suggesting that the method was useful for biochemic

New uses for an old technique: live imaging on the slice organ culture to study reproductive processes†.

Reproductive processes are dynamic and involve extensive morphological remodeling and cell-cell interactions. Live imaging of organs enhances our understanding of how biological processes occur in real time. Slice culture is a type of organ culture where thick slices are collected from an organ and cultured for several days. Slice culture is a useful and easy-to-implement technique for live imaging of reproductive events at cellular resolution. Here we describe a pipeline of live imaging on slice culture to visualize the process of urethra closure in mouse embryonic penis as a proof of princip

Posterior lamellar keratoplasty using descemetorhexis and organ-cultured donor corneal tissue (Melles technique).

PURPOSE: To report the clinical results of posterior lamellar keratoplasty (PLK) using predissected organ-cultured donor corneal tissue implanted after stripping of the Descemet membrane. METHODS: Twenty-two eyes of 22 patients in whom a PLK procedure was performed for pseudophakic bullous keratopathy and/or Fuchs endothelial dystrophy were evaluated. In all eyes, the recipient Descemet membrane was excised by performing a descemetorhexis. Just after harvesting the donor tissue, each cornea had a posterior lamellar dissection made at approximately 80% stromal depth, and each cornea was preser

Endothelial quality of eye bank-prestripped DMEK prepared form organ-cultured corneas with the Muraine technique.

Our aim was to measure the endothelial quality of prestripped Descemet membrane endothelial keratoplasty (DMEK) 48 h after preparation in an eye bank with the Muraine technique and shipping in a distant center. Ten pairs of human corneas with similar eye bank endothelial cell density (ebECD) were stored in organ-culture (OC) for 25 days (20, 28) [median (10-90 percentiles)]. One cornea was then randomized to DMEK preparation using the Moria Muraine trephine, the other served as control. The grafts were left attached to the center of the cornea, immersed in the OC medium (without Dextran) and s

Mini organ culture of thyroid tissue: a new technique for maintaining the structural and functional integrity of thyroid tissue in vitro.

We have developed a culture technique by which the normal configuration of follicles and most other structural characteristics of the thyroid gland are well preserved. Small tissue fragments (diameter 0.5 to 0.9 mm) from various mammalian species were prepared and kept in hydrophobic culture dishes to prevent their attachment. It was essential to raise the oxygen concentration in the incubator to 50% and the D-glucose concentration to 5.6 mM. Under these conditions, mini organs were formed by the outgrowth of epithelial cells from follicles located on the fragment surface and opened during tis

Clinical Utility of PNA-FISH for Burn Wound Diagnostics: A Noninvasive, Culture-Independent Technique for Rapid Identification of Pathogenic Organisms in Burn Wounds.

Burn injury results in an immediate compromised skin state, which puts the affected patient at an immediate risk for infection, including sepsis. For burn patients that develop infections, it is critical to rapidly identify the etiology so that an appropriate treatment can be administered. Current clinical standards rely heavily on culture-based methods for local and systemic infection testing, which can often take days to complete. While more advanced methods (ie, MALDI or NAAT) have improved turnaround times, they may still suffer from either the need for pure culture or sensitivity and spec

Corneal grafting of donor tissue preserved for longer than 4 weeks in organ-culture medium.

In vitro viability of the endothelium of corneas stored in organ-culture medium is longer than in other storage techniques. This article reports on the clinical results of 20 penetrating keratoplasties performed using corneas stored for > 4 weeks in organ culture. Seventeen of the procedures were elective and three were emergencies. Mean preservation time was 35.6 +/- 7 days (mean +/- SD) and mean follow-up was 16.2 months. During preservation, an endothelial cell loss of 10% was noted. Seventeen grafts remained transparent, and three failed between 3 and 24 months postoperatively. Mean endoth

The role of hypertrophic cartilage in endochondral ossification.

Normal stages of histogenesis of long bones show that the hypertrophy of cartilage cells is the pre-requisite for the perichondrium to take up osteoblastic activity, (Fell 1925, Lutfi 1971). Cooper (1965) found the cartilage cells from epihysis of the long bones of chick failed to induce chondrogenesis in somites in mice and chick whereas flat cells and early Peripheral cells could do same. Fell and Landauer (1935) noticed that in avian phocomelia the hypertrophied cartilage cells fail to hypertrophy leading subsequently to deformities of long bones. Presently an attempt is made to analyse thi

Requisite instruments for the establishment of three-dimensional epidermal human skin equivalents-A methods review.

Human skin equivalents (HSEs) are three-dimensional skin organ culture models raised in vitro. This review gives an overview of common techniques for setting up HSEs. The HSE consists of an artificial dermis and epidermis. 3T3-J2 murine fibroblasts, purchased human fibroblasts or freshly isolated and cultured fibroblasts, together with other components, for example, collagen type I, are used to build the scaffold. Freshly isolated and cultured keratinocytes are seeded on top. It is possible to add other cell types, for example, melanocytes, to the HSE-depending on the research question. After

Preparation of Retinal Explant Cultures.

Organotypic retinal explants are routinely used as alternatives to in vitro cell culture and to replace the use of animals in modelling retinal neurodegenerative diseases. Retinal explants fill the gap between in vivo which are expensive, time consuming, and complex due to inaccessibility of target tissues. However, organotypic retinal explants are less expensive and rapid and retinal cell types in the explant maintain their morphologic interactions with other cells in the retina. Therefore, retinal explants have high potential to be used as tools to assess the pharmacological and other therap

Modulation of type III collagen synthesis in bovine corneal endothelial cells.

Bovine corneal endothelial cells in culture synthesize predominantly type III collagen, unlike rabbit corneal endothelial cultures which synthesize type IV collagen. In an attempt to document whether this type III collagen synthesis by bovine cells is a tissue culture-specific phenomenon, collagens synthesized by organ culture of bovine Descemet's membrane/corneal endothelium complex were compared with those of subsequent tissue culture cells, up to the eighth passage. The biosynthetically labeled collagens were analyzed on SDS electrophoresis. The soluble fractions of tissues extracted with n

Corneal endothelial cell therapy: feasibility of cell culture from corneas stored in organ culture.

In 2013, a clinical trial was initiated to investigate cell therapy for the treatment of corneal endothelial decompensation. Cultivating human corneal endothelial cells (CECs) while maintaining their functional phenotype is challenging; therefore, establishment of a confirmed protocol is pivotal for obtaining approval from regulatory authorities for use of cellular therapy products. In this study, we evaluated organ culture (OC) as a storage method for donor corneas used as a raw material for establishing CEC cultures. OC allows storage of corneal tissue for conventional corneal transplantatio

An untargeted metabolomics approach to study changes of the medium during human cornea culture.

INTRODUCTION: Two main approaches (organ culture and hypothermia) for the preservation and storage of human donor corneas are globally adopted for corneal preservation before the transplant. Hypothermia is a hypothermic storage which slows down cellular metabolism while organ culture, a corneal culture performed at 28-37 °C, maintains an active corneal metabolism. Researchers, till now, have just studied the impact of organ culture on human cornea after manipulating and disrupting tissues. OBJECTIVES: The aim of the current work was to optimize an analytical procedure which can be useful for

Long-Term, Serum-Free Cultivation of Organotypic Mouse Retina Explants with Intact Retinal Pigment Epithelium.

In ophthalmic research, there is a strong need for in vitro models of the neuroretina. Here, we present a detailed protocol for organotypic culturing of the mouse neuroretina with intact retinal pigment epithelium (RPE). Depending on the research question, retinas can be isolated from wild-type animals or from disease models, to study, for instance, diabetic retinopathy or hereditary retinal degeneration. Eyes from early postnatal day 2-9 animals are enucleated under aseptic conditions. They are partially digested in proteinase K to allow for a detachment of the choroid from the RPE. Under the

Crustacean primary cell culture: A technical approach.

Crustacean cell culture has gained attention as a potent model to assist in the development of diagnostic reagents and probes for use in the shrimp, crayfish and lobster industries. The availability of such cellular tools is especially important to industries which use intensive aquaculture methods and thus have increased risk of disease problems. Indeed, crustacean cell cultures offer potential for studying the effects of pathogens in vitro and for increasing our knowledge on developmental and sexual maturation processes, or endocrine regulation in crustaceans. Although numerous attempts have

Developmental potency of cultured pineal cells: an approach to pineal developmental biology.

The pineal organ is still an enigma in regard to its developmental and phylogenetic origin. Little is known of the mechanism involved in determination and differentiation of pineal cells and virtually no studies have been done on the induction and tissue interactions during pinealogenesis. Interest is also centered on the evolutional transformation in structure and function, which may be related to the developmental alterations in pineal morphogenesis between the lower and higher vertebrate species. For developmental studies, avian embryos have great advantages for various experimental manipul

Organoid Culture of Different Intestinal Segments from Human and Mouse.

The intestine comprises distinct segments, each characterized by unique cell populations and functions. Intestinal organoids faithfully replicate the cellular composition and functions of the intestine. Over the past decade, the organoid model has garnered considerable attention for its application in investigation of organ development, renewal and functional performance. While the organoid culture systems for mouse small intestine and human large intestine have widely adopted, a comparison summary for different segments of the human or mouse intestine is lacking. In this study, we present a s

In vitro spermatogenesis: A century-long research journey, still half way around.

BACKGROUND: Spermatogenesis is one of the most complicated cellular differentiation processes in a body. Researchers struggled to find and develop a micro-environmental condition that can support the process in vitro. Such endeavors can be traced back to a century ago and are yet continuing. METHODS: Reports on in vitro spermatogenesis and related works were selected and classified into four categories based on the method used; organ culture, tubule culture, cell culture, and 3-dimensional cell culture methods. Each report was critically reviewed from the present point of view by authors who

Adenovirus types 8 and 19 infection of rabbit corneal organ cultures.

Adenovirus types 8 and 19 are responsible for a large number of cases of acute follicular conjunctivitis, mainly in the form of epidemic keratoconjunctivitis (EKC). This study successfully demonstrates the establishment of adenovirus infection in rabbit corneal organ cultures. Normal rabbit corneas were maintained in culture without significant morphologic alterations for up to 7 weeks. Adenovirus types 8 and 19 replication in rabbit corneas was demonstrated by viral growth curves and subpassage studies. Intracellular localization of adenoviral antigens was confirmed with immunofluorescent tec
