Organ economy: organ trafficking in Moldova and Israel.

Organ trafficking is an illegal means of meeting the shortage of transplants. The activity flourishes for several interacting reasons, such as medical needs, poverty and criminality. Other factors are fundamental conceptual structures such as the dream of the regenerative body as well as the view of the body as an object of utility and an object of value. The article aims to go behind the normative discussions that usually surround organ trafficking. Why this is happening, and what the societal consequences are, is examined through ethnographic fieldwork. The focus is on the shadow economies t

Gut immune cell trafficking: inter-organ communication and immune-mediated inflammation.

Immune cell trafficking is a complex and tightly regulated process that is indispensable for the body's fight against pathogens. However, it is also increasingly acknowledged that dysregulation of cell trafficking contributes to the pathogenesis of immune-mediated inflammatory diseases (IMIDs) in gastroenterology and hepatology, such as inflammatory bowel disease and primary sclerosing cholangitis. Moreover, altered cell trafficking has also been implicated as a crucial step in the immunopathogenesis of other IMIDs, such as rheumatoid arthritis and multiple sclerosis. Over the past few years,

Physiological implications of phase separation in vesicle organization and trafficking.

Phase separation has emerged as a key mechanism of cellular compartmentalization, complementing classical membrane-bound organelles. In addition to forming membraneless compartments that provide confined environments for biochemical reactions, phase separation actively organizes and regulates membrane vesicles. Protein condensates interact with vesicular organelles through defined modes of communication, influencing their assembly, trafficking, and turnover. This review highlights recent advances in understanding how phase separation contributes to vesicle biology, including synaptic vesicle c

Trafficking and Membrane Organization of GPI-Anchored Proteins in Health and Diseases.

Glycosylphosphatidylinositol (GPI)-anchored proteins (GPI-APs) are a class of lipid-anchored proteins attached to the membranes by a glycolipid anchor that is added, as posttranslation modification, in the endoplasmic reticulum. GPI-APs are expressed at the cell surface of eukaryotes where they play diverse vital functions. Like all plasma membrane proteins, GPI-APs must be correctly sorted along the different steps of the secretory pathway to their final destination. The presence of both a glycolipid anchor and a protein portion confers special trafficking features to GPI-APs. Here, we discus

Role of membrane organization and membrane domains in endocytic lipid trafficking.

Lipid compositions vary greatly among organelles, and specific sorting mechanisms are required to establish and maintain these distinct compositions. In this review, we discuss how the biophysical properties of the membrane bilayer and the chemistry of individual lipid molecules play a role in the intracellular trafficking of the lipids themselves, as well as influencing the trafficking of transmembrane proteins. The large diversity of lipid head groups and acyl chains lead to a variety of weak interactions, such as ionic and hydrogen bonding at the lipid/water interfacial region, hydrophobic

Phosphatidic acid increases Notch signalling by affecting Sanpodo trafficking during Drosophila sensory organ development.

Organ cell diversity depends on binary cell-fate decisions mediated by the Notch signalling pathway during development and tissue homeostasis. A clear example is the series of binary cell-fate decisions that take place during asymmetric cell divisions that give rise to the sensory organs of Drosophila melanogaster. The regulated trafficking of Sanpodo, a transmembrane protein that potentiates receptor activity, plays a pivotal role in this process. Membrane lipids can regulate many signalling pathways by affecting receptor and ligand trafficking. It remains unknown, however, whether phosphatid

Trafficking and organization of cyst wall components into a robust biphasic structure in Entamoeba.

Entamoeba histolytica, a protozoan parasite, causes amebiasis. Amebiasis is mainly transmitted by oral ingestion of cysts. Cysts are produced in the large intestine of humans from proliferative trophozoites by a cell differentiation process called encystation. The Entamoeba cyst wall consists of chitins and proteins that include chitinase and Jacob and Jessie lectins. During encystation, these components are synthesized and layered around encysting Entamoeba cells. The structures of these components are well studied; however, the detailed timings of their synthesis (the transcription of the en

The GPR183-oxysterol axis in cancer: immune trafficking, spatial organization, and therapeutic potential.

G protein-coupled receptor 183 (GPR183; also known as EBI2, Epstein-Barr virus-induced G-protein-coupled receptor 2) has emerged as a central regulator linking 7α,25-dihydroxycholesterol (7α,25-OHC) gradients to immune cell trafficking and tissue organization. In this review, we synthesize the structural, physiological, and oncogenic dimensions of the GPR183 axis. We first discuss the enzymatic biogenesis of its oxysterol ligand and the structural mechanisms underlying receptor activation. We then examine the context-dependent roles of GPR183 within the tumor microenvironment (TME), including

Pathogenic podocin variants exhibit distinct defects in trafficking, membrane organization, and degradation pathways.

Nephrotic syndrome is frequently associated with pathogenic variants in NPHS2 (podocin), including the common and severe R138Q substitution. Using conditionally immortalized human podocytes expressing Myc-tagged podocin variants (G92C, V180M, R138Q, R238S, and R291W), we systematically compared variant-specific defects in plasma-membrane trafficking, detergent-resistant microdomain (DRM) localization, and protein stability. All variants displayed reduced plasma membrane abundance and altered DRM distribution. Among them, R138Q-podocin showed uniquely reduced protein stability. Consistent with

Expression of the self-marker CD47 on dendritic cells governs their trafficking to secondary lymphoid organs.

Dendritic cells (DCs) capture and process Ag in the periphery. Thus, traffic through lymphatic vessels is mandatory before DCs relocate to lymph nodes where they are dedicated to T-cell priming. Here, we show that the ubiquitous self-marker CD47 selectively regulates DC, but not T and B cell trafficking across lymphatic vessels and endothelial barriers in vivo. We find an altered skin DC migration and impaired T-cell priming in CD47-deficient mice at steady state and under inflammatory conditions. Competitive DC migration assays and active immunization with myeloid DCs demonstrate that CD47 ex

LRBA organizes distinct vesicular trafficking systems in distal nephron segments for water and sodium conservation.

Lipopolysaccharide-responsive and beige-like anchor protein (LRBA) deficiency is a rare genetic disorder characterized by immune dysregulation. The immune checkpoint molecule cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) fails to perform proper membrane trafficking in the absence of LRBA. In addition to immune cells, LRBA localizes to intracellular vesicles in various epithelial cells; however, its physiological roles have not been accurately deciphered. It was observed in this study that LRBA facilitates water and sodium transport by promoting vesicular trafficking of aquaporin-2 (AQP2

Multi-omic profiling reveals the ataxia protein sacsin is required for integrin trafficking and synaptic organization.

Autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS) is a childhood-onset cerebellar ataxia caused by mutations in SACS, which encodes the protein sacsin. Cellular ARSACS phenotypes include mitochondrial dysfunction, intermediate filament disorganization, and progressive death of cerebellar Purkinje neurons. It is unclear why the loss of sacsin causes these deficits or why they manifest as cerebellar ataxia. Here, we perform multi-omic profiling in sacsin knockout (KO) cells and identify alterations in microtubule dynamics and mislocalization of focal adhesion (FA) proteins, incl

α-Galactosidase A expressed in the salivary glands partially corrects organ biochemical deficits in the fabry mouse through endocrine trafficking.

Fabry disease is caused by an X-linked deficiency of the lysosomal enzyme α-galactosidase A (GLA) and has been treated successfully with enzyme replacement therapy (ERT). Gene therapy has been proposed as an alternative to ERT due to the presumed advantages of continuous, endogenous production of the therapeutic enzyme. GLA production in the liver and its therapeutic efficacy in the Fabry mouse have been demonstrated previously with various viral vector systems. In consideration of the potential advantages of using the salivary glands as endogenous GLA biosynthesis sites, we explored the feasi

Cadmium exposure enhances organic cation transporter 2 trafficking to the kidney membrane and exacerbates cisplatin nephrotoxicity.

Renal accumulation and exposure of cadmium originating from pollution in agricultural land and the prevalence of cigarette smoking remains an unneglectable human health concern. Whereas cadmium exposure has been correlated with increased incidence of a variety of kidney diseases, little is known pertaining to its effect on renal drug disposition and response in patients. Here, we report that cadmium exposure significantly increased the activity of organic cation transporter 2 (OCT2), a critical renal drug transporter recommended in United States Federal Drug Administration guidance for assessm

The Arp2/3 complex and WASp are required for apical trafficking of Delta into microvilli during cell fate specification of sensory organ precursors.

Cell fate decisions mediated by the Notch signalling pathway require direct cell-cell contact between adjacent cells. In Drosophila melanogaster, an external sensory organ (ESO) develops from a single sensory organ precursor (SOP) and its fate specification is governed by differential Notch activation. Here we show that mutations in actin-related protein-3 (Arp3) compromise Notch signalling, leading to a fate transformation of the ESO. Our data reveal that during ESO fate specification, most endocytosed vesicles containing the ligand Delta traffic to a prominent apical actin-rich structure (AR

Self-Etching of Metal-Organic Framework Templates during Polydopamine Coating: Nonspherical Polydopamine Capsules and Potential Intracellular Trafficking of Metal Ions.

Traditionally, containers made from steel or other metals are not good for making tea, probably due to the fact that polyphenol components in tea can chelate with metal ions. A similar reason might stand behind the observations as reported herein. During the coating of well-defined metal-organic framework (MOF) crystalline particles with polydopamine (PDA) via pH-induced self-polymerization of dopamine, we found that MOF templates automatically etch off during the coating, giving rise to nonspherical PDA capsules that inherit the morphologies of the templates. Such self-etching of MOF template

Inhibition of donor-derived T cells trafficking into target organs by FTY720 during acute graft-versus-host disease in small bowel transplantation.

In small bowel transplantation (SBTx), graft-versus-host disease (GVHD) is mediated by donor-derived T cells recognizing host major histocompatibility complex (MHC) alloantigens, and represents an important immunological event influencing life in experimental and clinical situations. We evaluated the possibility that a new sphingosine 1-phosphate receptor agonist, FTY720, could diminish GVHD in a rat SBTx model through traffic alteration of donor-derived T cells in target organs. Heterotopic SBTx was performed using a parent (WF)-into-F(1) (WF x ACI) rat combination. Recipient survival, body w

How People With Lived Experiences of Homelessness and Sex Trades Become Social Service Leaders in Sex Trafficking, Sex Trading, and Youth Organizations: A Community-Engaged Study.

Organizations that work with people who trade sex for compensation (including sex work and trafficking) and who are homeless employ lived experience experts to enhance the relevance and utility of services. We sought to understand how lived experience experts become social service leaders and the conditions that influence their pathways and well-being. Influenced by anti-capitalism and anti-white-supremacy, we used an intersectional, community-engaged, constructivist grounded theory approach to conduct semi-structured, in-depth interviews with 26 adults with lived experiences who were currentl

From intestinal immune niches to systemic inflammation: programming cellular plasticity and immune redistribution across organs.

Immune-mediated inflammatory diseases have traditionally been investigated within organ-specific frameworks. Although increasing evidence has revealed extensive communication along individual gut-organ axes, these interactions are often described through separate mechanisms, including microbial dysbiosis, barrier disruption, soluble mediators, and immune-cell redistribution. A central challenge remains to determine how intestinal immune states are established and how they are translated into organ-specific inflammatory responses. In this review, we propose that the intestine functions as a dyn

Combating human trafficking: pediatric surgeons as agents of change.

Human trafficking is a pervasive public health crisis that encompasses sex, labor, and organ trafficking. Although it affects individuals of all ages and identities, pediatric patients are particularly vulnerable. Up to 88% of trafficking victim-survivors access healthcare services during their exploitation, yet most healthcare professionals lack the training to identify and support these patients. Pediatric surgeons encounter trafficked patients presenting with trauma, abuse-related injuries, and complex medical conditions emphasizing the urgent need for anti-trafficking education and policy
