Callus mediated shoot organogenesis and regeneration of cytologically stable plants of Ledebouria revoluta: An ethnomedicinal plant with promising antimicrobial potency.

Ledebouria revoluta are important ethnomedicinal plant found in India and South Africa. Micropropagation via indirect shoot organogenesis had been established from three types of explant (i.e. scale leaf, leaf lamina and root) of L. revoluta. Scale leaf was found superior as compared to leaf lamina and root explant with respect to their organogenic callus induction potentiality. Murashige and Skoog (1962) [MS] media supplemented with 3.0 mg L-1 2,4-dichlorophenoxyacetic acid, 0.75 mg L-1 β-naphthoxyacetic acid were best effective for inducing organogenic callus. In Vitro Propagation of Plants via Organogenesis in Bambusa vulgaris Schrad. ex Wendl Using Temporary Immersion Systems.

The low multiplication and ex vitro survival rates during acclimatization in the culture house limit the in vitro mass propagation of B. vulgaris. Several scientific studies have described the development of different protocols for bamboo; however, not all of them address the effects of these systems on plant morphology, physiology, and biochemistry in vitro. In this chapter, a complete and optimized protocol is described for plants propagated via organogenesis in temporary immersion systems. In addition, the morphophysiological and biochemical characterization of the plants as well as the sur

Tomato DOR encodes a signal peptide peptidase-like protein required for adventitious organogenesis and rooting.

The identification of the tomato DOR gene that is essential for adventitious organogenesis provides a genetic target for improving regeneration efficiency and root system performance in tissue culture and plant breeding. Plant rooting and adventitious organogenesis represent major bottlenecks in the application of plant tissue culture techniques. In this study, we characterize the dor mutant (defective in organogenesis and rooting) identified in our collection of tomato T-DNA lines. This mutant exhibits normal callus proliferation but fails to differentiate adventitious buds. Additionally, it

Beyond elongation: The multifaceted roles of gibberellins in symbiosis and root development.

Plants regulate root development in response to fluctuating environmental conditions, including establishing symbiotic relationships with arbuscular mycorrhizal fungi and nitrogen-fixing bacteria under nutrient limitation. These processes are orchestrated by plant hormones, particularly gibberellins, and the repressors of gibberellin signalling, DELLA proteins. Gibberellin and DELLAs serve as critical regulators in symbiotic signalling and root organogenesis, integrating hormonal and environmental cues with cellular patterning to direct plant development. This review explores the current under

Spatiotemporal regulation of lateral root organogenesis in Arabidopsis by cytokinin.

The architecture of a plant's root system, established postembryonically, results from both coordinated root growth and lateral root branching. The plant hormones auxin and cytokinin are central endogenous signaling molecules that regulate lateral root organogenesis positively and negatively, respectively. Tight control and mutual balance of their antagonistic activities are particularly important during the early phases of lateral root organogenesis to ensure continuous lateral root initiation (LRI) and proper development of lateral root primordia (LRP). Here, we show that the early phases of

Autonomous differentiation of transgenic cells requiring no external hormone application: the endogenous gene expression and phytohormone behaviors.

The ectopic overexpression of developmental regulator (DR) genes has been reported to improve the transformation in recalcitrant plant species because of the promotion of cellular differentiation during cell culture processes. In other words, the external plant growth regulator (PGR) application during the tissue and cell culture process is still required in cases utilizing DR genes for plant regeneration. Here, the effect of Arabidopsis BABY BOOM (BBM) and WUSCHEL (WUS) on the differentiation of tobacco transgenic cells was examined. We found that the SRDX fusion to WUS, when co-expressed wit

Cytokinins and plant immunity: old foes or new friends?

Cytokinins are plant growth promoting hormones involved in the specification of embryonic cells, maintenance of meristematic cells, shoot formation and development of vasculature. Cytokinins have also emerged as a major factor in plant-microbe interactions during nodule organogenesis and pathogenesis. Microbe-originated cytokinins confer abnormal hypersensitivity of cytokinins to plants, augmenting the sink activity of infected regions. However, recent findings have shed light on a distinct role of cytokinins in plant immune responses. Plant-borne cytokinins systemically induce resistance agai

Marker-assisted in vivo imaging reveals prepatterning events prior to lateral root organogenesis.

Roots proliferate mainly through lateral root (LR) organogenesis events that succeed each other in a regular sequence. In the model plant Arabidopsis (Arabidopsis thaliana), live imaging of reporter lines containing the synthetic auxin-response DIRECT REPEAT5 (DR5) promoter have been instrumental for insight into prepatterning mechanisms that shape root architecture. However, depending on the reporter line in use, the course of prepatterning events leading to organogenesis may be described differently, causing confusion and misunderstandings in literature. In this study, we have setup a unique

Histological and transcriptomic analysis provides new insights into the regulatory mechanism of floral organogenesis in sunflower.

The initiation of floral organ primordia at the peripheral zone of the floral meristem (FM) is a fundamental process in flower development, yet its regulatory mechanisms in sunflower remain unclear. In this study, we characterized a sunflower space-mutagenized mutant, crazy broccoli 1 (cb1), in which FM-like structures fail to generate floral organs and maintain meristematic activity throughout development. This defect leads to floral development malformation, resulting in an indeterminate FM-like structure consisting of successive whorls of bracts and meristems. Through comparative transcript

In Vitro Propagation of Cannabis sativa L. and Evaluation of Regenerated Plants for Genetic Fidelity and Cannabinoids Content for Quality Assurance.

Cannabis sativa L. (Marijuana; Cannabaceae), one of the oldest medicinal plants in the world, has been used throughout history for fiber, food, as well as for its psychoactive properties. The dioecious and allogamous nature of C. sativa is the major constraint to maintain the consistency in chemical profile and overall efficacy if grown from seed. Therefore, the present optimized in vitro propagation protocol of the selected elite germplasm via direct organogenesis and quality assurance protocols using genetic and chemical profiling provide an ideal pathway for ensuring the efficacy of micropr

Quantitative Measurement of Basipetal Auxin Transport in Arabidopsis Roots Via 3H-IAA Labeling.

The polar transport of the phytohormone auxin regulates plant organogenesis, morphogenesis, and gravitropism, which are coordinately regulated by auxin influx and efflux carriers. To measure the basipetal (shootward) transport of auxin in roots, we applied radiolabeled IAA to the root tips of 7-day-old seedlings in the model plant Arabidopsis thaliana. A scintillation counter was then used to measure the radioactivity of the samples. Here, we describe a method for measuring auxin transport in A. thaliana roots via 3H-indole-3-acetic acid (3H-IAA).

Direct organogenesis of Mandevilla illustris (Vell) Woodson and effects of its aqueous extract on the enzymatic and toxic activities of Crotalus durissus terrificus snake venom.

In order to produce explants of Mandevilla illustris (Vell) Woodson for the "Cerrado in vitro", the Germplasm Bank of UNAERP, we carried out a micropropagation protocol using MS or MS/3 medium supplemented with different concentrations of 6-benzyladeninepurine (BA), Zeatin or 2-isopentenyladenine for nodal segment growth, and alpha-naphthaleneacetic acid, indole-3-butyric acid (IBA) or 1,4 dithiothreitol for rooting. For nodal segments, all the cytokinins tested yielded similar results. However, 2.22 micro M BA is more economical to use. MS/3 medium supplemented with 0.49 micro M IBA was the m

Hydrogen sulfide: A versatile gaseous molecule in plants.

Hydrogen sulfide (H2S) is a gasotransmitter and signaling molecule associated with seed germination, plant growth, organogenesis, photosynthesis, stomatal conductance, senescence, and post-harvesting. H2S is produced in plants via both enzymatic and non-enzymatic pathways in different subcellular compartments. Exogenous application of H2S facilitates versatile metabolic processes and antioxidant machinery in plants under normal and environmental stresses. This compound interacts with phytohormones like auxins, abscisic acid, gibberellins, ethylene, jasmonic acid, and salicylic acid. Furthermor

In vitro shoot organogenesis and hormone response are affected by the altered levels of Brassica napus meristem genes.

Arabidopsis shoot meristem activity is regulated by a molecular network involving the participation of several components, including SHOOTMERISTEMLESS (STM), CLAVATA1 (CLV1), and ZWILLE (ZLL). In an effort to identify the role of these genes during in vitro shoot formation Brassica and Arabidopsis plants were transformed with the Brassica napus (Bn) STM, CLV1, ZLL1 and ZLL2 identified in previous work [1]. In both systems shoot organogenesis was promoted by the over-expression of BnSTM, BnZLL1, and BnZLL2, and repressed by the over-expression of BnCLV1. This distinct regulation, analogous to t

Molecular cartography of root nodule organogenesis: insights into the role of transcription factors.

Nodule organogenesis is a complex developmental process which is induced post-embryonically following rhizobia infection and relies on coordinated action of a suite of transcription factors at multiple stages starting from its initiation, de novo meristem establishment and differentiation. The rewiring of nitrate-responsive NIN-LIKE PROTEIN (NLP) homolog, NODULE INCEPTION (NIN) transcription factor through genetic adaptations in the common ancestor of the nitrogen-fixing clade is crucial to the origin of nodulation trait. Moreover, nodule formation emerged through the co-option of evolutionary

Co-option of a conserved lateral-root development program by symbiotic signals.

Nod factors (NFs) are microbial signals originally identified for their key role in the nitrogen-fixing root nodule symbiosis in legumes. Beyond symbiosis, NFs also possess a conserved capacity to induce lateral-root formation across diverse plant species, including non-legumes. It is now well established that the nodule organogenesis program has co-opted several molecular mechanisms involved in root development, which raises the question of the developmental pathway controlled by NFs to trigger lateral-root formation and how it overlaps with nodule organogenesis in legumes. In Medicago trunca
