Painting the evolutionary history of a multicompartmentalized syngameon in North American Castilleja (Orobanchaceae).

UNLABELLED: Understanding the evolutionary history of recent rapidly radiating groups remains a major challenge in systematics, especially when incomplete reproductive isolation and rampant interspecific gene flow obscure phylogenetic signal. Here, we present a comprehensive phylogenomic study of the genus Castilleja ("the paintbrushes", Orobanchaceae), integrating nuclear and chloroplast data to investigate patterns of diversification, hybridization, and geographic structure across its range. Our results provide the first evidence that Castilleja functions as a multicompartmentalized syngameo

A revision of the genus Cistanche (Orobanchaceae) in Israel, and considerations for its taxonomic circumscription in the Middle East.

The genus Cistanche is the subject of considerable taxonomic confusion due to reduced morphology, poor preservation in herbaria, and misidentification. Here we re-examine the Cistanche of Israel, a region that falls within the broader Middle Eastern centre of diversity for the genus, but has been neglected from systematic attention. In our reconsideration we address taxonomic confusion in the region and confirm the presence of C.tubulosa and C.violacea, report for the first time the occurrence of C.laxiflora plus a separate entity we refer to as C.tinctoria, and describe a new species, C.mimii

The complete mitochondrial genome of an important medicinal plant, Rehmannia glutinosa (Gaertn.) DC., 1845 (Lamiales, Orobanchaceae).

Rehmannia glutinosa, an extensively utilized Chinese herbal medicine, is highly valued for its medicinal properties. In this study, the complete mitochondrial genome (mitogenome) of R. glutinosa was sequenced and assembled for the first time. The mitogenome is 547,032 bp in length, with an overall GC content of 44.97%. The mitogenome contains 67 unique genes, comprising 43 protein-coding, three rRNA, and 21 tRNA genes, with six protein-coding and nine tRNA genes being chloroplast-derived. The phylogenetic analysis, based on the maximum-likelihood criterion, demonstrated that R. glutinosa is cl

Comprehensive Analysis of the Complete Mitochondrial Genome of Rehmannia chingii: An Autotrophic Species in the Orobanchaceae Family.

Rehmannia chingii is an important medicinal plant with immense value in scientific research. However, its mitochondrial genome (mitogenome) has not yet been characterized. Herein, based on whole-genome Illumina short reads and PacBio HiFi reads, we obtained the complete mitogenome of R. chingii through a de novo assembly strategy. We carried out comparative genomic analyses and found that, in comparison with the plastid genome (plastome) showing a high degree of structural conservation, the R. chingii mitogenome structure is relatively complex, showing an intricate ring structure with 16 conne

Targeting the dermis for melasma maintenance treatment.

Melasma relapse is almost common after discontinuation of conventional treatment. Recent studies suggesting that photoaging dermis is the main pathomechanism of melasma, emphasize the dermal targeting therapy. We investigated maintenance effect of microneedling radiofrequency (RF) for melasma treatment. Subjects with melasma were administered oral tranexamic acid and triple combination cream for 2 months and a randomly assigned half face was treated with RF. After discontinuation of conventional therapy, the half face RF continued monthly over 6 months. Modified melasma area severity index (mM

Pancreatic lipase-inhibiting polysaccharide from Stropharia rugosoannulata: Structural characterization and interaction mechanisms.

Obesity as a global epidemic concern is often considered as a risk factor for multiple metabolic syndrome. Exploring safe and effective natural ingredients exhibiting excellent pancreatic lipase (PL) inhibition has become a promising way to overcome this issue. This study aimed to investigate the structural characterization of Stropharia rugosoannulata polysaccharide (STR-II*) as well as its inhibition effect and mechanism on PL. The results showed that STR-II* with an average Mw of 8.5 kDa was composed of Gal, Glu, Man and GlcA at the molar ratio of 3.3: 82.6: 7.9: 6.2. Its backbone was proba

Bewitching the host: Striga disrupts host stomatal ABA signaling by impairing the tryptophan lock.

Parasitic plants of the Orobanchaceae family, particularly Striga hermonthica, rank among the world's most devastating agricultural weeds (Jamil et al., 2021). They inflict massive yield losses on staple cereal crops across Africa, including sorghum, millet, and rice, as well as globally on other essential crops such as maize, rapeseed, tomato, and legumes (Parker et al., 2009; Bouwmeester, 2018). These root parasites have evolved a specialized organ called the 'haustorium' that connects to the vascular system of the host to siphon water, minerals, and nutrients (Yang et al., 2015). A critical

Parasitic Plant-Host Interactions: Molecular Mechanisms and Agricultural Resistance Strategies.

Obligate parasitic plants, particularly members of the Orobanchaceae family, including Striga and Orobanche, greatly devastate crop production. Here, we synthesize recent advances in understanding the molecular and ecological dynamics underlying parasitic plant-host interactions, focusing on critical stages of parasitism: germination, host detection, haustorium formation, and resource extraction. Orobanchaceous parasites exploit host-derived strigolactones (SLs) to break seed dormancy, whereas Cuscuta species do not rely on SLs for germination. Instead, chemotropic responses to host-exuded com

Phytochemical analysis and antihyperglycemic activity of Castilleja arvensis.

Castilleja genus comprises approximately 211 species, some of them exhibiting potential in treating various diseases. Remarkably, despite its abundance, there is a significant lack of scientific studies that explore the chemical composition and/or therapeutic activity of this genus. In this work, the chemical composition of Castilleja arvensis was determined, and its antihyperglycemic activity was evaluated in vivo, in vitro, and ex vivo. Hydroalcoholic extract of C. arvensis (HECa) was obtained from the maceration of aerial parts. HECa was fractionated by liquid-liquid extractions to obtain t

Design and synthesis of strigolactone analogues and mimics containing indolin-2-one scaffold for the Phelipanche control.

BACKGROUND: The broomrapes are root-parasitic weeds widely distributed in the temperate zone area. The effective management on the Phelipanche and Orobanche parasitic weeds still remains challenging to date. RESULTS: Novel strigolactone (SL) analogues (X series) and mimics (O series) derived from indolin-2-one were designed and synthesized. Of them, compound O-3 showed nearly ten-fold higher seed germination activity (median effective concentration (EC50) = 0.0066 μm) towards Phelipanche aegyptiaca seeds compared to the control GR24. Moreover, it also showed prominent seed germination activit

The structure-activity relationship of aryloxyacetylthioureas for the inhibition of Orobanche minor radicle elongation.

Orobanchaceae root parasitic weeds cause significant damage to agriculture and become threats to global food security. Integrated pest management is a key concept in modern agriculture and requires chemicals with various modes of action. Planteose accumulates as a storage carbohydrate in the dry seeds of root parasitic weeds. In Orobanche minor seeds, planteose is hydrolyzed by an α-galactosidase, OmAGAL2, during germination. It was found that the OmAGAL2 inhibitor, PI-28, suppressed the radicle elongation of germinating O. minor seeds. This inhibitory activity against O. minor radicle elongat

The differentially regulated genes TvQR1 and TvPirin of the parasitic plant Triphysaria exhibit distinctive natural allelic diversity.

BACKGROUND: Plant parasitism represents an extraordinary interaction among flowering plants: parasitic plants use a specialized organ, the haustorium, to invade the host vascular system to deprive host plants of water and nutrients. Various compounds present in exudates of host plants trigger haustorium development. The two most effective haustorium inducing factors (HIFs) known for the parasitic plant Triphysaria versicolor (T. versicolor) are peonidin, an antioxidant flavonoid, and 2,6-dimethoxybenzoquinone (DMBQ), an oxidative stress agent. To date, two genes involved in haustorium initiati

Response of an obligate CAM plant to competition and increased watering intervals.

Climate change has exacerbated precipitation variability, profoundly impacting vegetation dynamics and community structures in arid ecosystems. There remains a notable knowledge gap regarding the ecological effects of altered precipitation on crassulacean acid metabolism (CAM) plants and their interactions with other photosynthetic types. This study investigated the response of the typical obligate CAM plant Orostachys fimbriata to extended watering intervals (WI4-WI8) and various competitive patterns (M1-M4) with the C3 grass Melilotus officinalis and the C4 grass Setaria viridis through gree

A new furofuran lignan diglycoside and other secondary metabolites from the antidepressant extract of Castilleja tenuiflora Benth.

Castilleja tenuiflora has been used for the treatment of several Central Nervous System (CNS) diseases. Herein we report the antidepressant activity of the methanol extract from the leaves of this medicinal plant. The oral administration of MeOH extract (500 mg/kg) induced a significant (p < 0.05) decrement of the immobility parameter on Forced Swimming Test (FST) and an increment in the latency and duration of the hypnosis, induced by administration of sodium pentobarbital (Pbi, 40 mg/kg, i.p.). Chemical analysis of this antidepressant extract allowed the isolation of (+)-piperitol-4-O-xylopy

Reduced strigolactone exudation as a key resistance mechanism in wild carrots against Phelipanche aegyptiaca.

Phelipanche aegyptiaca is a root parasitic plant that causes significant yield losses in many crops, including carrots (Daucus carota). This study investigates the resistance mechanisms of two wild carrot accessions, PI 21793 (Daucus glaber) and PI 341902 (Daucus littoralis), against P. aegyptiaca compared to a cultivated carrot (P0114; D. carota). Wild carrots induced lower germination rates of P. aegyptiaca seeds and fewer successful tubercles, indicating both pre-attachment and partial post-attachment resistance mechanisms. Strigolactone analysis revealed significant quantitative difference
