Anti-Candidal Activity of the Parasitic Plant Orobanche crenata Forssk.

Candida albicans (C. albicans) and Candida glabrata (C. glabrata) are part of the human microbiome. However, they possess numerous virulence factors, which confer them the ability to cause both local and systemic infections. Candidiasis can involve multiple organs, including the eye. In the present study, we investigated the anti-candidal activity and the re-epithelizing effect of Orobanche crenata leaf extract (OCLE). By the microdilution method, we demonstrated an inhibitory effect of OCLE on both C. albicans and C. glabrata growth. By crystal violet and 3-(4,5-dimethylthiazol-2-yl)-2,5-diph

The structure-activity relationship of aryloxyacetylthioureas for the inhibition of Orobanche minor radicle elongation.

Orobanchaceae root parasitic weeds cause significant damage to agriculture and become threats to global food security. Integrated pest management is a key concept in modern agriculture and requires chemicals with various modes of action. Planteose accumulates as a storage carbohydrate in the dry seeds of root parasitic weeds. In Orobanche minor seeds, planteose is hydrolyzed by an α-galactosidase, OmAGAL2, during germination. It was found that the OmAGAL2 inhibitor, PI-28, suppressed the radicle elongation of germinating O. minor seeds. This inhibitory activity against O. minor radicle elongat

Seed endophytic bacteria are involved in metal adaptation of Orobanche lutea: community dynamics and plant growth promotion traits.

BACKGROUND: Parasitic plants are affected by various abiotic stressors including water and drought stress, fluctuations or extremes of temperature, salinity, mineral deficiencies or toxic concentrations of heavy metals in soils. While the molecular mechanisms and the ecological roles of these parasitic angiosperms have been well-studied, their responses to abiotic stress remain poorly understood. This study explores the relationship between environmental metal stress and the seed endophytic bacterial community of the holoparasitic plant Orobanche lutea Baumg. (Orobanchaceae). RESULTS: Our find

The peroxidase OcPOD10 promotes the parasitism of sunflower broomrape (Orobanche cumana) via mediating reactive oxygen species (ROS) accumulation.

Orobanche cumana Wallr. is an obligate root parasite that mainly infects sunflower (Helianthus annuus L.), causing significant yield losses. Peroxidases are known to be involved in plant responses to abiotic and biotic stresses, while its role during parasitism has not been elucidated. This study investigated the role of OcPOD10, a peroxidase encoding gene from O. cumana, in the parasitic interaction between sunflower and O. cumana. OcPOD10 was specifically expressed in haustorium, an interface between sunflower root and O. cumana. Overexpression of OcPOD10 promoted parasite attachment, wherea

Sunflower HaWRKY29 dampens Orobanche cumana parasitism via transcriptional activation of HaLAC17 and lignin deposition.

Orobanche cumana Wallr. is a root holoparasitic plant that mainly parasitizes and, therefore, threatens sunflower (Helianthus annuus L.) production. Compared with other pathogens, the molecular mechanisms underlying host resistance to parasitic plants remain largely elusive. Here, we used 2 strategies to identify and functionally characterize sunflower genes in response to invading broomrape. First, we established a transient overexpression system via seed-soak agroinfiltration, providing a convenient expression system for functional genomics in sunflower. Second, transcriptome sequencing of 3

Changes in the activity of the alternative oxidase in Orobanche seeds during conditioning and their possible physiological function.

The appearance of the activity of the cyanide insensitive, alternative oxidase (AOX), pathway of oxygen uptake was followed in seeds of Orobanche aegyptiaca during conditioning. The pathway becomes operative during conditioning, up to day three as determined by inhibition of oxygen uptake of the seeds by propyl gallate. At the same time an increasing percentage of oxygen uptake is insensitive to cyanide and an increased oxygen uptake, responsive to propyl gallate, is induced by brief salicylic acid treatment of seeds. By day six of conditioning, these responses decrease and the AOX pathway cou

A Systematic Comparison of Two Host-Sourced Populations Suggests Tomato to be an Opportunistic Host for Orobanche cumana.

Orobanche cumana predominantly infects sunflower but often occurs in tomato fields in Xinjiang, China. However, it is suspected that the parasitic populations to those two host crops are different species or biotypes. The present study seeks to elucidate this issue through comparison of differences in morphology, genetics, and parasitic ability between populations from different host sources. A field survey showed that the infestation level in sunflower fields was remarkably higher than in tomato fields. Consistent with this, cross-infection experiments showed a preference for sunflowers by O.

Functional Analysis of the GH16 Domain-Containing XTH2 Homologs in Mediating Sunflower Response to Orobanche cumana Parasitism.

Sunflower (Helianthus annuus) is highly susceptible to infection by the root parasitic plant Orobanche cumana during its growth. In establishing connections with the sunflower root system, O. cumana induces the hydrolysis and remodeling of the host cell wall. Xyloglucan endotransglucosylase/hydrolase (XTH), a member of glycoside hydrolase family 16 (GH16), is a key enzyme involved in the hydrolysis and synthesis of xyloglucan, playing a critical role in cell wall modification and reconstruction. However, the involvement of XTH families in the interaction between sunflower and O. cumana remains

Resistance of red clover (Trifolium pratense) to the root parasitic plant Orobanche minor is activated by salicylate but not by jasmonate.

BACKGROUND AND AIMS: Obligate root holoparasites of the genus Orobanche attack dicotyledonous crops and cause severe losses in many parts of the world. Chemical induction of plant defence systems such as systemic acquired resistance was proposed to be an available strategy to control the root parasite, but the detailed mechanisms involved have not been clarified. The aim of this study was to elucidate the effects of salicylic acid (SA), jasmonic acid (JA) and their analogues on resistance of red clover to Orobanche parasitism. METHODS: Roots of red clover grown in plastic chambers were applie
