Orthoreovirus avis in asymptomatic broiler chickens.

Orthoreovirus avis (previously named Avian othoreoviruses; ARVs) cause significant economic losses in the global poultry industry. Disease control is compromised by the emergence of variants that escape vaccine-induced immunity. The development of more effective vaccines requires knowledge of circulating strains. One hundred and twenty-four feces from asymptomatic and unvaccinated broiler chickens from two commercial poultry farms in the states of Rio de Janeiro and Espírito Santo, southeastern Brazil, were examined with the aim of detecting, genotyping and characterizing the σC protein of the

Outbreak of Orthoreovirus-induced meningoencephalomyelitis in baboons.

BACKGROUND AND PURPOSE: Spontaneous viral encephalitis is rare in the baboon; yet, during a 13-month period (1993-1994), eight juvenile baboons (Papio cynocephalus spp.) developed acute, progressive nonsuppurative meningoencephalomyelitis caused by an unknown agent. Clinical signs of disease included disorientation and truncal ataxia that rapidly progressed to hemiparesis or paraparesis. Clinicopathologic findings were not remarkable and appreciable gross lesions were not seen at necropsy. Microscopic examination revealed CNS lesions that were characterized by lymphoplasmacytic perivascular cu

Evidence of vertical transmission of novel duck orthoreovirus in ducks.

Since 2017, duck spleen necrosis caused by new variant duck orthoreovirus (N-DRV) infection had been observed in several provinces in China. This disease retards the growth and development of ducks, thereby reducing feed return rate. N-DRV infection causes damage to duck spleen and other immune organs, leading to immunosuppression and susceptibility to other pathogens. In this study, we successfully constructed a breeding duck artificial infection model and found that N-DRV infection can cause pathologic changes, such as ovarian hemorrhage, follicle atrophy, and fallopian tube bleeding in bree

Serological evidence of human infection by bat orthoreovirus in Singapore.

To determine whether Pteropine orthoreovirus (PRV) exposure has occurred in Singapore, we tested 856 individuals from an existing serum panel collected from 2005-2013. After an initial screen with luciferase immunoprecipitation system and secondary confirmation with virus neutralization test, we identified at least seven individuals with specific antibodies against PRV in both assays. Our findings confirm that PRV spillover into human populations is relatively common in this region of the world.

Toward Effective Vaccines Against Piscine Orthoreovirus: Challenges and Current Strategies.

Piscine orthoreovirus (PRV) is a globally distributed viral pathogen that causes heart and skeletal muscle inflammation (HSMI) in Atlantic salmon (Salmo salar) and affects other salmonids, yet no commercial vaccines are currently available. Major barriers to vaccine development include the inability to propagate PRV in cell lines and the low, variable immunogenicity of its proteins, particularly the outer capsid protein σ1, which mediates viral attachment. This protein is hypothesized to be immunologically relevant due to its homology with Mammalian orthoreoviruses. Recombinant σ1 expressed in

Characterization of a Goose-Origin Avian Orthoreovirus with Interferon Suppression Activity.

The emergence of variant strains of Avian orthoreovirus (ARV) has caused substantial economic losses in the poultry industry worldwide, but the molecular features of goose-origin strains and viral transmission among different avian species remain poorly understood. Here, we describe a goose-origin avian orthoreovirus strain, SD0407, associated with growth retardation and joint swelling. Complete genome analysis identified ten double-stranded RNA segments. Sequence comparison indicated that SD0407 is closely related to previously reported duck-origin reovirus strains. Phylogenetic and recombina

Synergistic pathogenicity of novel duck Orthoreovirus and salmonella typhimurium in ducks.

Co-infection with Novel Duck Orthoreovirus (N-DRV) and Salmonella Typhimurium (S. Typhimurium) poses significant threats to duck health. The disease associated with N-DRV is characterized by epizootic outbreaks featuring hepatosplenomegaly, hemorrhage, and necrosis. Ducklings infected with STM primarily exhibit symptoms such as fibrinous pericarditis, perihepatitis, and intestinal hemorrhage. In recent years, China has experienced outbreaks involving mixed infections of these two pathogens in various regions, leading to more complex and atypical clinical manifestations. This study investigated

Synergistic pathogenicity of avian orthoreovirus and Staphylococcus aureus on SPF chickens.

Avian arthritis is a relatively common disease in the poultry industry, the cause of which is complex. Bacterial arthritis is often caused by infection of Staphylococcus aureus (S. aureus), whereas viral arthritis is caused by avian orthoreovirus (ARV). To investigate the infection of S. aureus and ARV in cases of avian arthritis, a total of 77 samples characterized by arthritis were collected and detection. The results showed that 68.83% of the samples were positive for ARV, and 66.23% were positive for S. aureus. Among them, the ARV mono-infection rate was 22.08%, the S. aureus mono-infectio

High Incidence of Mammalian Orthoreovirus Identified by Environmental Surveillance in Taiwan.

Wild poliovirus (WPV) persists in diverse locales worldwide, spreading outward from endemic areas. In response to the international threat of WPV transmission and changes in the national vaccination policy, we established an environmental surveillance system to monitor the circulation of wild and vaccine-related poliovirus in Taiwan. From July 2012 to December 2013, we collected sewage specimens every month from 10 sewage treatment plants located throughout Taiwan. The specimens were concentrated by the two-phase separation method and then inoculated into L20B, RD, and A549 cells for virus iso

Identification and characterization of a novel orthoreovirus from American eels, Anguilla rostrata.

Hemorrhagic gill necrosis disease (HGND) has resulted in substantial economic losses in the farming of American eels (Anguilla rostrata). Although prior research implicates viruses like American eel adomavirus (AEAdoV) and Anguillid herpesvirus (AngHV), the primary etiological agent remains elusive. In this study, a novel virus was isolated from the HGND samples of American eel using eel ovary (EO) cell lines. Following isolation, the virus was subjected to comprehensive characterization encompassing morphological features, genomic organization, phylogenetic relationships, pathogenic potential

eIF3i inhibits nelson bay orthoreovirus replication via interaction with the σNS protein.

L929 cells exhibit relatively strong resistance to infection by Nelson Bay orthoreovirus (NBV), primarily due to the unique virus-host interaction that suppresses NBV infection at multiple stages, including viral entry, genome replication, and virion assembly. This study investigates the role of host factor eIF3i in NBV replication. Co-immunoprecipitation and immunofluorescence co-localization assays were performed to verify whether eIF3i interacts with the σNS protein in vitro. The results demonstrated that eIF3i significantly inhibits NBV replication through specific binding to σNS. This fin

Pan-Viral Sequencing Surveillance Identifies Mammalian Orthoreovirus 2 in United States Wastewater.

Mammalian orthoreoviruses (MRVs) are segmented, double-stranded RNA viruses that infect a broad range of mammalian hosts, including humans. Although MRVs have been detected in wastewater in parts of Southeast Asia, they have not previously been reported in U.S. wastewater. Using the VirCapSeq-VERT pan-viral sequencing assay, we identified MRV type 2 (MRV-2) in a wastewater sample collected in March 2024 from the United States Air Force Academy. Complete genome sequences were recovered for all 10 segments. Phylogenetic analyses showed that the virus clustered most closely with an MRV isolate re

Characterization of inner capsid σA protein as a virulence factor of the pteropine orthoreovirus.

Pteropine orthoreovirus (PRV) is an emerging zoonotic virus that causes pneumonia in humans. We previously isolated the PRV strain Nachunsulwe-57 (N57) from a Zambian fruit bat and demonstrated its low virulence in laboratory mice. Here, we have attempted to identify factors responsible for differences in the virulence between strain N57 and the human-derived clinical strain Miyazaki-Bali/2007 (MB). Characterization of the virulence of recombinant monoreassortant PRVs derived from highly virulent MB and low virulent N57 strains in mice revealed that compared with wild-type (WT) MB, MB-based mo

Isolation and genomic characterization of a novel avian orthoreovirus strain in Korea, 2014.

In this study, we isolated a novel avian reovirus (ARV) strain, K738/14, from a broiler chicken with viral arthritis in South Korea. Genome sequence comparisons showed relatively low nucleotide identity with previously identified ARV strains. Phylogenetic analyses suggested multiple reassortment events between reovirus strain S1133 and reoviruses of Hungarian, Chinese, and US origin had occurred. In addition, recombination analyses showed evidence of intra-segmental recombination in the M2 and S2 genes. Based on our genetic analyses, multiple reassortment events, intra-segmental recombination,

Development of an oncolytic mammalian orthoreovirus expressing the near-infrared fluorescent protein iRFP720.

Fluorescence-guided surgery (FGS) is a useful method for removing invasive tumor tissues. For this, near-infrared (NIR) fluorescence probes are suitable for visualizing cancer cells due to their low autofluorescence, and an oncolytic mammalian orthoreovirus (MRV) expressing an NIR fluorescent protein is expected to be a novel tool for FGS. In this study, we identified the optimal insertion site of the NIR fluorescent protein gene iRFP720 (915 nt) in the MRV genome. We constructed genome plasmids for the L1, M1, and S2 segments, where a gene cassette comprising iRFP720 and T2A self-cleaving pep

Synergistic pathogenicity in sequential coinfection with fowl adenovirus type 4 and avian orthoreovirus.

Hydropericardium hepatitis syndrome (HHS) is a fatal disease caused by fowl adenovirus serotype 4 (FAdV-4). Avian viral arthritis is an infectious disease characterized by movement disorders caused by avian orthoreovirus (ARV). In the early 2019, our epidemiologic survey on poultry diseases in eight commercial broiler farms in China showed that FAdV-4 and ARV have a high coinfection rate, accounting for 63 % of all ARV-positive samples. We designed chicken embryo and animal models to investigate the synergistic pathogenicity of FAdV-4 and ARV. Weakness and inappetence were observed in all spec

Mammalian orthoreovirus capsid protein σ3 antagonizes RLR-mediated antiviral responses by degrading MAVS.

UNLABELLED: Mammalian orthoreovirus (MRV) outer capsid protein σ3 is a multifunctional protein containing a double-stranded RNA-binding domain, which facilitates viral entry and assembly. We reasoned that σ3 has an innate immune evasion function. Here, we show that σ3 protein localizes in the mitochondria and interacts with mitochondrial antiviral signaling protein (MAVS) to activate the intrinsic mitochondria-mediated apoptotic pathway. Consequently, σ3 protein promotes the degradation of MAVS through the intrinsic caspase-9/caspase-3 apoptotic pathway. Moreover, σ3 protein can also inhibit t

Isolation and characterization of a novel reassortant strain of Orthoreovirus mammalis from diarrheic pig.

Orthoreovirus mammalis (MRV) can infect a wide range of mammals, including humans and bats, and the cross-species transmission of the virus poses a potential threat to the health of humans and other animals. In this study, we identified a novel MRV strain from diarrheic pigs in Hunan Province, China. Genetic analysis revealed that the virus is a reassortant, containing segments derived from human, porcine, and mink MRVs. Designated as MRV3-HNU-XXS-2020, the strain clusters within Lineage IV of MRV3 and shows the highest nucleotide identity (98.79%) and amino acid identity (98.02%) to the MRV3

Evaluation of porcine intestinal organoids as an in vitro model for mammalian orthoreovirus 3 infection.

BACKGROUND: Mammalian orthoreovirus type 3 (MRV3), which is responsible for gastroenteritis in many mammalian species including pigs, has been isolated from piglets with severe diarrhea. However, the use of pig-derived cells as an infection model for swine-MRV3 has rarely been studied. OBJECTIVES: This study aims to establish porcine intestinal organoids (PIOs) and examine their susceptibility as an in vitro model for intestinal MRV3 infection. METHODS: PIOs were isolated and established from the jejunum of a miniature pig. Established PIOs were characterized using polymerase chain reaction

Isolation and Molecular Characteristics of a Novel Recombinant Avian Orthoreovirus From Chickens in China.

In recent years, the emergence of avian orthoreovirus (ARV) has caused significant losses to the poultry industry worldwide. In this study, a novel ARV isolate, designated as AHZJ19, was isolated and identified from domestic chicken with viral arthritis syndrome in China. AHZJ19 can cause typical syncytial cytopathic effect in the chicken hepatocellular carcinoma cell line, LMH. High-throughput sequencing using Illumina technology revealed that the genome size of AHZJ19 is about 23,230 bp, which codes 12 major proteins. Phylogenetic tree analysis found that AHZJ19 was possibly originated from
