Osmium tetroxide is a strong oxidizing agent used in electron microscopy. Eye exposure may cause severe burns, and after inhalation or ingestion damage to the respiratory or gastrointestinal tract occurs. Exposure to osmium and its compounds is extremely rare. Lobomycosis is a skin infection produced by the fungus Lacazia loboi, which mainly affects some indigenous and afro-descendant populations in Tropical America. The zinc iodide-osmium tetroxide (ZIO) fixation/staining method was applied for neurocytological studies and also to examine several other tissue samples including epidermal Langerhans cells, blood and bone marrow cells and lymphoid tissue. Although precise specificity cannot be attributed to the staining reaction, interesting staining patterns for different cell types were observed by using one of the ZIO staining solutions. The significance of ZIO positivity is briefly discussed.

Osmium tetroxide complexes with nitrogen ligands (L) are probes of DNA structure and electroactive labels of DNA. Aqueous solutions of osmium tetroxide (OsO4) have been injected into arthritic knees for the past 45 years to chemically destroy diseased tissue, in a procedure termed "chemical synovectomy." We describe a procedure for detection of fat emboli in formalin fixed tissue using osmium tetroxide postfixation. Intravascular fat in tissue postfixed in osmium tetroxide, embedded in epoxy or paraffin, and stained with toluidine blue, hematoxylin, or Oil Red O is more easily visualized than in frozen tissue that is stained with Oil Red O. With these methods, fat emboli may be detected years after the initial autopsy.

Reconstructions from serial ultrathin sections of microvascular endothelium suggest that gaps, which are induced by a range of stimuli, may pass through endothelial cells as well as between them.
