Pathophysiological and parasitological studies on Ostertagia ostertagi infections in calves.

Friesian calves given a low level infection of the abomasal parasite Ostertagia ostertagi over a six week period displayed a mild diarrhoea with high faecal egg counts and elevated plasma pepsinogen values. At necropsy on day 23 abomasal lesions characteristic of ostertagiasis were widespread. At 42 and 84 days oedema and congestion were also prominent. Total worm burdens on days 23 and 42 were similar but a marked decrease had occurred by day 84. Feed digestibility and nitrogen economy were not markedly affected but radioisotopic measurements demonstrated an increase in albumin disappearance

Transfer of immunity to Ostertagia circumcincta and IgA memory between identical sheep by lymphocytes collected from gastric lymph.

Sheep rendered immune to Ostertagia circumcincta were challenged with 50,000 larvae and lymphocytes were collected from the gastric lymph up to eight days after challenge. The cells were transferred intravenously to genetically identical worm-free sheep which, together with controls, were challenged with 50,000 larvae and killed nine days later. Cells obtained during the donors lymphoblast response to challenge transferred partial immunity, measured either as stunting or loss of worms. Significantly less immunity was transferred by cells collected either before or after this response. Thus the

Anthelmintic activity of albendazole in white-tailed deer.

Albendazole at dose rates of 11 to 54 mg/kg (mean of 25 +/- 11 SD) of body weight was highly effective (greater than 99%) against Haemonchus contortus, Ostertagia odocoilei, O mossi, Trichostrongylus askivali and Nematodirus odocoilei in 22 white tailed deer (Odocoileus virginianus) in southern Texas. It was 38% effective against mature and immature deer liver flukes (Fascioloides magna). Toxicoses associated with treatment were not observed. Large numbers of abomasal nematodes (mean of 2,949 +/- 3.147 SD) were recovered from 16 untreated control deer, indicating these may have had a potential

Anthelmintics for cattle.

A number of anthelmintics are available for the control of gastrointestinal nematodes in cattle. In North America, O. ostertagi, Cooperia spp., lung worm, and F. hepatica probably cause the greatest losses in production. The older anthelmintics are often deficient in their action against some of these parasites. Recently, the Paratect morantel tartrate slow-release bolus has provided a mechanism for the prevention of infections with gastrointestinal nematodes and lung worm, to some extent, and this has been shown to produce considerable economic benefits. Fenbendazole removes arrested O. oster

Method for determining serum pepsinogen concentration, using pepsin standards and ultraviolet absorbance.

A simplification of the traditional hemoglobin methods for determining serum pepsinogen concentration was developed. In this method, 10% trichloroacetic acid solution was added to control samples, and hemoglobin substrate was added to controls and active enzyme samples; standards and samples were incubated for 18 hours, the proteins in the active tubes were precipitated with trichloroacetic acid and removed by filtration, and the absorbances of the supernatant of each standard and sample at 280 nm were measured. The major differences between this method and other methods for determining pepsin

Heterologous antibody-based immunity in cattle naturally infected with Ostertagia ostertagi and Haemonchus sp.

Heterologous antibody‑mediated immunity was evaluated in cattle naturally infected with Ostertagia ostertagi (Oo) and Haemonchus sp. (Hp). Antibody responses were assessed using ovalbumin (OVA) as unrelated model antigen in three groups of eight heifers, matched for eggs per gram of feces (EPG) and allocated to independent, nematode‑free pastures. Anti‑OVA titers were determined in serum by ELISA. EPG counts and nematode genera were determined in feces using the modified McMaster technique and pooled cultures, respectively. All variables were analyzed using mixed models. One group was previous

Control of some internal parasites of chinchillas.

An unidentified coccidia was found in chinchillas. Treatment with sulfamethazine apparently eradicated the infection from several herds. Ostertagia females and 1 Trichostrongylus colubriformis male were also found in chinchillas, causing emaciation. A dose of thiabendazole of 100 mg./kg. of body weight was effective against the roundworms, but it was toxic. A dose of trichlorphon, 100 mg./kg. of body weight, was effective and safe. Hymenolepis infection caused emaciation and death. Treatment with doses of niclosamide, 200 mg./kg. of body weight, was successful in removing tapeworms, ending dea

Seasonal dynamics of Ostertagia ostertagi infection in lactating Holstein-Friesian cows on a dairy farm in Hokkaido, Japan.

Gastrointestinal nematodes (GINs), such as Ostertagia ostertagi and Cooperia spp., are common in cattle worldwide. Although infected cattle are often asymptomatic, production is negatively affected. The present study investigated whether eggs per gram (EPG) of GINs in 32 lactating cows decreased during winter and increased as it became warmer. Larvae recovered by coprocultures mostly consisted of O. ostertagi in autumn, while three GINs, Ostertagia, Trichostrongylus and Meistocirrus, were collected in spring. Daily milk and bulk tank milk samples from cows previously infected with O. ostertagi

Internal parasites of Washington cattle.

The occurrence of internal parasites in Washinton cattle was estimated. During the survey period in 1972 and 1973, more than 3,700 fecal samples from 29 herds and 4 feedlots, and viscera from 55 cattle were examined. The percentage of fecal samples with parasite eggs or oocysts from herds was as follows: oocysts, 77%; "strongyline" eggs, 44%; Nematodirus eggs, 3%; Moniezia eggs, 3%; Fasciola eggs, 2%; Trichuris eggs, 2%; and Dictyocaulus larvae, 1%. The fecal samples from feedlots contained principally strongyline eggs which varied markedly in numbers among groups of cattle in the lots. The pr

Profiling Genome-Wide Methylation Patterns in Cattle Infected with Ostertagia ostertagi.

DNA methylation (DNAm) regulates gene expression and genomic imprinting. This study aimed to investigate the effect of gastrointestinal (GI) nematode infection on host DNAm. Helminth-free Holstein steers were either infected with Ostertagia ostertagi (the brown stomach worm) or given tap water only as a control. Animals were euthanized 30 days post-infection, and tissues were collected at necropsy. We conducted epigenome-wide profiling using a mammalian methylation array to explore the impact of infection on methylation patterns in the mucosa from abomasal fundus (FUN), pylorus (PYL), draining

Phytochemicals as inhibitors of pathogenesis to combat ostertagiosis, toxocariasis, trichostrongylosis and trichuriasis in cattle: A systematic review.

Highly pathogenic gastrointestinal nematodes (GIN) pose a severe global threat to cattle health and productivity, causing significant economic losses. With their widespread prevalence (96%) and growing anthelmintic resistance, phytochemicals (PC) present a promising alternative. This review evaluates studies from 2001 onward on herbal remedies as an effective alternative to combat the pathogenesis of ostertagiosis, toxocariasis, trichostrongylosis, and trichuriasis in cattle. A systematic literature review of electronic databases has identified 87 unique plant species from 45 publications agai

Rumen microbiome profiles of dairy cattle are affected by the presence of, and vaccination against, the abomasal parasitic nematode Ostertagia ostertagi.

Ostertagia ostertagi is a highly prevalent nematode that affects grazing cattle and impacts performance and welfare by reducing appetite and hindering weight gain. Despite its economic significance, the influence of the abomasal parasite O. ostertagi on the rumen microbiome remains unexplored. We examined the effects of subclinical O. ostertagi infection and vaccination on the rumen microbiome at taxonomic and functional levels. In an experimental trial, calves treated with vaccine or adjuvant-only were orally challenged with O. ostertagi larvae daily for 25 days; 4 groups of animals (UNF: unv

Characterisation of protective vaccine antigens from the thiol-containing components of excretory/secretory material of Ostertagia ostertagi.

Previous vaccination trials have demonstrated that thiol proteins affinity purified from Ostertagia ostertagi excretory-secretory products (O. ostertagi ES-thiol) are protective against homologous challenge. Here we have shown that protection induced by this vaccine was consistent across four independent vaccine-challenge experiments. Protection is associated with reduced cumulative faecal egg counts across the duration of the trials, relative to control animals. To better understand the diversity of antigens in O. ostertagi ES-thiol we used high-resolution shotgun proteomics to identify 490 u

Pathogenesis of simulated natural infections with Ostertagia ostertagi in calves.

The possibility of a mucosal hypersensitivity reaction and its relationship to the pathogenesis of simulated natural infections with Ostertagia ostertagi were studied in calves. Four groups of 4 calves each were used. One group was used as noninfected control; a 2nd group was given increasing doses of infective larvae; a 3rd group was given increasing doses of larvae and these were removed by succeeding treatment with an anthelmintic; and a 4th group was given an initial dose of larvae which was then eliminated with an anthelmintic. All calves given larvae became sensitized, as shown by an int
